Objective To observe the distributive characteristics of glial cell line-derived neurotrophic factor(GDNF)in brain tissue during cerebral ischemia-reperfusion in rats,and its role in ischemic brain damage.Methods The model of focal cerebral ischemia was made by occluding middle cerebral artery(MCA) for 2 h and reperfusion for 0.5~48 h, HE Staining was used to investigate the histological features of ischemic cerebral damage,the immunohistochemical method was used to observe the distributive characteristics of GDNF in brain tissue in rats. Results The focal ischemic areas (including preoptic area, striatum and cortex) presented at 0.5 h of reperfusion and peaked at 24 h. The neurons presented irreversible degeneration at 6 h of reperfusion. At 24 h, the ischemic area in the preoptic area developed into infarct form. At 0.5 h of reperfusion, neurons in ischemic cortex showed GDNF weak positive, neurons in peri-ischemic regions showed GDNF moderate positive. During reperfusion 3~48 h, neurons in ischemic regions showed GDNF negative. 2.The Applied Value of Selected Intraoperative Cholangiography in Cholecystectomy

Objective To evaluate the applied of selected intraoperative cholangiography(SIOCG) during cholecystectomy.Methods The SIOCG data of 39 patients who had indications for exploration of common bile duct were analysed retrospectively.Results Among 39 cases with SIOCG,abnormal findings were detected in 19 cases(48.72%),in which choledocholith in 16 cases;stenosis of common bile duct at the third part in 3 cases.Positive findings in exploration of common bile duct were 14 cases(73.68%),in which,choledocholith were 12 cases inflammatory stenosis 1 case and tumor of common bile duct at the third part 1 case.Negative patients in exploration of common bile duct were 5 cases(26.32%).Normal imaging in 20 patients who did not underwent exploration of common bile duct had no disorders in follow-up.Conclusion SIOCG is safe,convenient,and useful in preventing the remnant stones and biliary tract damage and improving the quality of cholecystectomy.
3.Immunohistochemical Study of Insulin-Like Growth Factor-I in Human Fetal Organs

Objective To study the distribution of insulin-like Growth Factors (IGFs) in liver、 spleen、kidney、thymus and supraadrenal glands of fetus from 16 to 24 weeks.Method Using ABC immunohistochemistry. Results IGF-I positive cells were found in these organs during this period of fetal life, but the differences of number and reaction intensity of IGF-I positive cells appeared in different organs and individuals.Conclusion These organs tissues can express IGF-I during fetal development and play paracrine and/or autocrine role in cellular proliferation and differentiation of these fetal tissue.
4.Development and design of a new sonography rigid bronchoscopy and corollary vacuum-assisted biopsy device system.

The present study was to develop and design a new sonography rigid bronchoscopy and corollary vacuum-assisted biopsy device system with less injury and complication. The system combined ultrasonic-probe with ultrasound catheter, a new medical ultrasound technique, and rigid bronchoscopy (RB) which is improved with an auxiliary vacuum-assisted biopsy device. The principle of the device is vacuum suction and rotary knife. The reduced outer diameter of the RB led to less pain and lower complications for the patient. With the help of ultrasonic-probe (30 MHz), lesions and blood vessels can be identified clearly and unintentional puncture and damage to blood vessels can be avoided. Plenty of lesions can be obtained quickly through the vacuum-assisted biopsy device without getting puncture needle in and out repeatedly. The novel endobronchial sonography rigid bronchoscopy and matched vacuum-assisted biopsy device has many remarkable advantages. It can enlarge the applied range of the RB from endobronchial to mediastinal lesions, avoiding unintentional puncture of vessels. Obtaining multiple samples with a higher accuracy rate than that by other sampling techniques, minimizing operation time, alleviating pain and decreasing the complication rate, the system makes up the technical deficiency for the diagnosis and treatment of the mediastinal lesions, to a certain degree.

5.Difference of morphology and proliferation ability between diabetes mellitus-derived ADSCs and normal ADSCs

Objective To investigate the morphological characteristics and proliferation ability of diabetes mellitus-derived adipose-derived stem cells (ADSCs) by comparing with normal-derived ADSCs.Methods The ADSCs could be achieved from diabetes mellitus' adipose tissue by the similar method of normal ADSCs isolation and culture,and then the differnce was compared between the two groups about their morphologies under microscope,and the proliferation ability of two groups was determined by CCK-8.Results ADSCs in patients with diabetes were obvious hypertrophy and irregular in morphology.A values of ADSCs in diabetes and non-diabetes were 0.210+0.002 and 0.390+0.006 in 1st day;0.250+0.015 and 0.443+0.023 in 2nd day;0.368+0.014 and 0.726+0.033 in 3rd day;0.368+0.014 and 0.726+0.033 in 4th day;1.767+0.072 and 3.153+0.067 in 5th day and 1.810+0.072and 3.170+0.021 in 6th day,respectively.The difference was statstically significant at the beginning from 4th day (P<0.05).Conclusions There is obvious difference of the microscopic morphology between the diabetes mellitus-derived ADSCs and normal-derived ADSCs.The proliferation ability of the diabetes mellitus-derived ADSCs is lower than normal-derived ADSCs.These difference could be closely related to ulcerous non-healing wounds in patients with diabetes mellitus.
6.Outcomes of open and minimally invasive esophagectomy for esophageal cancer

Objective To compare the safety,feasibility,and long-term outcomes between combined thoracoscopic and laparoscopic esophagectomy (TLE) and open three-field esophagectomy (OE) for esophageal cancer.Methods The data of 183 patients with esophageal cancer who accepted TLE (n =94) or OE (n =89) from February 2009 to December 2011 were retrospectively collected and compared.Results Demographics,pathologic data in each cohort were almost identical.No significant differences were observed in operative time,the number of lymph node harvested,rates of anastomotic leak,thoracic abscess,chyle chest,re-operation,vocal cord paralysis,gastric emptying dysfunction and morbidity.A significant difference was observed in blood loss (86.6 ±38.3 ml in TLE group vs.217.4 ±87.2 ml in OE group,P=0.000),rate of transfusion (1.1% vs.6.7%,P =0.045),hospital stay (13.9 ±7.5 d vs.17.1 ±10.2 d,P =0.017),rate of overall surgical complications (23.4% vs.38.2%,P =0.030),rate of pulmonary complications (9.6% vs.27.0%,P =0.002) and rate of arythmia (4.1% vs.12.4%,P =0.046).Median follow-up time was 28.0 ±2.0 months (95% CI:24.2-31.8).No difference between the two groups concerning survival was observed (P =0.954).Conclusion The TLE for esophageal cancer is safe and feasible.Although it cannot improve the survival of long-term,it is worth spreading in intraoperative and postoperative advantages compared with OE group.
7.Inhibitory effect of melatonin on murine foregastric carcinoma cells via membrane receptors MT2

Objective To investigate the inhibitory effect of melatonin on the proliferation activity of murine foregastic carcinomac ( MFC) cells via melatonin membrane receptors MT 2 and its relationship with the signaling pathways of mitogen-activated protein kinases ( MAPKs), phosphatidylinositol 3-kinase ( PI3K)-Akt.Methods Using siRNA technology to silence MT2 expression, we examined the ability of melatonin to inhibit the proliferation activity of MFC cells and its influence on the phosphorylation of ERK 1/2 and Akt.Results We found two interesting effects of SiRNA-mediated silencing of MT2 expression.Firstly, it significantly antagonized the inhibitory effect of melatonin on the proliferation activity of MFC cells .Secondly , it partially blocked the inhibitory effect of melatonin on the phosphorylation of ERK 1/2 and Akt.Conclusion Our results suggest that melatonin can inhibit the phosphorylation of ERK 1/2 and Akt via MT2 receptors , thereby inhibiting the proliferation of gastric cancer cells .
9.Study of optimal discharge pressure of compressor in CO2 refrigerating trans-critical cycle

In this paper, a carbon dioxide trans-critical refrigerating system which is different from a conventional subcritical refrigerating cycle was studied. The trans-critical carbon dioxide refrigerating systems are based on the Gustav Lorntzen cycle. Emphasis was focused on how to determine the optimal discharge pressure of compressor in CO2 trans-critical cycle. The factors related with the optimal discharge pressure were analyzed. A formula was developed based on cycle simulation, which could be used to predict the optimal discharge pressure of a basic CO2 trans-critical cycle. After further studies on CO2 trans-critical cycles with a regenerator or expander, two more formulas were also developed. These formulas could provide an access to improve the COP of CO2 trans-critical cycle.

Objeetive:To study the expression of cell adhesion molecules on the lymphatic endothelial cells In the rectum cancer and peritumoral lymph nodes as well as their relationship with the diffusion of cancer cellS. Methods: In this Study the immunhistochemistry method was employed to observe the expression of CEA, ICAM-1 and CD31 on rectum cancer lymphatic endo-thelial cells. Results :The results demonstrated that CEA and lCAM-1 were highly expressed on the lynphatio endothelial cells in the peritumoral rectum tissues and lymph nodes. It was, howev-er,negative in normal lymphatic endothelial cells. CD31 was expressed on both lymphatic endothe-Ilal cells in the periturmral rectum tissues and lymph nodes and vessel endothelial cells of the normal human. Conelusion: The study suggests that there are relationships between the dis-semination of cancer cells and excession of CEA, lCAM-1 and CD31 in lymphatic endothelial cells.The CAE and ICAM-1 seem to induce stable adhesion between oancer cells and endothelial cells.
