1.Genetic Toxicity of Europium Nitrate to Vicia faba Root Tip Cells

Objective To evaluate the genotoxicity of europium nitrate to the Vicia faba L root-tip cells. Methods The primary roots of Vicia faba L were cultured for 6 hours in the solution of Eu series(64,32,16,8,4,2,1 ?g/ml) at 25℃.And then the roots were repair-cultured in double distilled water for 24 h. The root tips of Vicia faba L were dissected and fixed,stained and the micronucleus rate,mitotic index and the rate of chromosomal aberrations were determined. Results From 1 ?g/ml to 32 ?g/ml,the rate of micronucleus increased with the increasing of the europium nitrate dosage(D),presented a dosage-effect relationship(r=0.949). From 2 ?g/ml to 64 ?g/ml,mitotic index decreased with the increasing of the europium nitrate dosage,presented a dosage-effect relationship(r=-0.852). From 1 ?g/ml to 64 ?g/ml,chromosome fragments(CF) and frequency of chromosomal aberrations(FCA) increased with the increasing of the europium nitrate dosage with a dosage-effect relationship significantly(rD-CF=0.915,rD-FCA=0.872). Conclusion Europium nitrate has the certain genetic toxicity to the Vicia faba L root-tip cells. The chromosome fragments may be one of the reasons to produce micronucleus.
2.Effect of manual massage on relapse rate and quality of life of grand mal epilepsy patients

Objective The aim of the study is to study the effect of manual massage on relapse rate and quality of life of grand mal epilepsy patients.Methods Using prospective research method,clinical data of patients with epileptic seizures and without attack up to 3 years and undergoing withdrawal of drugs for one year were collected.The grand mal epilepsy patients were randomly divided into the experimental group and the control group (76 cases in each group).The experimental group was given manual massage.The control group was not given manual massage.Both groups were given follow-up for 1 year.The relapse rate and quality of life were compared between two groups.Results The relapse rate of the ex perimental group was 17%,and that of the control group was 33%,the difference was significant.The score of life quality in the experimental group was significantly better than that of the control group.Conclusions Manual massage can reduce relapse rate and improve quality of life for patients with epileptic seizures after withdrawal of drugs,which is worthy of wide application.

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis complex and is mainly manifested as pulmonary tuberculosis. Mycobacterium tuberculosis is characterized by high pathogenicity and drug resistance due to its high viability and lethality, and multidrug-resistant tuberculosis caused by it has become a global public health problem. Early and effective treatment is essential to prevent the emergence of drug-resistant strains. Commonly employed methods for the screening and diagnosis of TB include: clinical signs, imaging examinations (chest X-ray, computed tomography), sputum culture (L-J medium, BACTEC liquid culture system), and immunological tests (lipoarabinomannan antigen test, tuberculin skin test, interferon-gamma release test). In recent years, emerging molecular diagnostic methods such as GeneXpert MTB/RIF assay, loop-mediated isothermal amplification (LAMP), linear probe assay, and whole-genome sequencing have been used to diagnose and characterize TB. These methods not only identify Mycobacterium tuberculosis but also detect mutation sites associated with resistance to first-line anti-tuberculosis drugs (isoniazid, rifampicin, etc.), providing new possibilities for the diagnosis of drug-resistant Mycobacterium tuberculosis. In this paper, the advantages and disadvantages of some commonly used detection methods for tuberculosis are first reviewed, aiming at assisting clinicians to diagnose and treat the disease in a timely manner. Secondly, this paper explores the prospects of the latest high-throughput detection methods for drug-resistant Mycobacterium tuberculosis.
4.Clinical characteristics of 1126 cases of malignant lymphoma

Objective To analyze the clinical characteristics of 1126 cases of malignant lymphoma in our hospital from 2005 to 2009. Methods Age, sex, pathological classification, onset locus and clinical staging of the malignant lymphoma were analyzed and summed up, based on the data of the malignant lymphoma patient information in hospital case database from 2005 to 2009 and excluded the cases as pathology was not sure and the repeated cases. Results A male:female ratio of malignant lymphoma in inpatient was 1.94:1. The majority of Hodgkin lymphoma (HL) manifested in 20-40 years old, mixed cellularity (64.16 %) and nodular sclerosis (29.48%) was most common. The majority of non-Hodgkin lymphoma (NHL) manifested in 50-70 years old, the most common pathological classifications were diffuse large B-cell lymphoma (53.31 %), extranode nasal-type NK/T cell lymphoma (7.35 %), mantle cell lymphoma (6.40 %), B chronic lymphocytic leukemia/small B-cell lymphoma (4.30 %), anaplastic large cell lymphoma (4.09 %), precursor T cell lymphoblastic leukemia/lymphoma (3.88 %), peripheral T cell lymphoma but not otherwise specified (3.46 %), angioimmunoblastic lymphoma(3.04 %), follicular lymphoma(2.94 %), Burkitt lymphoma (2.52 %). The onset locus of both HL and NHL most often involved lymph nodes of the cervical region. Conclusion The gender, age, pathological classification, onset locus and clinical staging of malignant lymphoma were different between HL and NHL.

Objective To investigate the efficacy of different adding times,treatment courses and doses of bifidobacterium and lactobacillus triple live bacteria in Helicobacter pylori (Hp) eradication.Methods A total of 280 patients Hp-infected were enrolled and randomly assigned to five groups.Group A received lansoprazole 30 mg,clarithromycin 500 mg and amoxicillin 1,000 mg bid for 14 days;group B received bifidobacterium and lactobacillus triple live bacteria 2,000 mg tid for 14 days followed by regimen of group A for another 14 days;group C1 received regimen of group A with addition of bifidobacterium and lactobacillus triple live bacteria 2,000 mg bid for 14 days;group C2:regimen of group A with addition of bifidobacterium and lactobacillus triple livc bacteria 2,000 mg tid for 14 days;and group D received regimen of group C2 followed by bifidobacterium and lactobacillus triple live bacteria 2,000 mg tid for another 14 days.4 weeks after end of treatment,Hp eradication was assessed by 13C-urea breath test.Adverse effects during the courses of treatment were recorded.Results A total of 252 (90.0%) patients completed the treatment.The completion rate in group A,B,C1,C2,and D were 78.6% (44/56),92.9% (52/56),87.5% (49/56),96.4% (54/56),and 94.6% (53/56) respectively;the completion rate was significantly higher in group B,C2 and D than in group A (P < 0.05),but there were no differences among groups B,C2 and D (P > 0.05).According to intention-to-trcat (ITT) analysis,the eradication rate was 62.5%,80.4%,69.6%,85.7%,and 87.5% in groups A,B,C1,C2,and D respectively.The eradication rate in groups B,C2 and D was significantly higher than that in group A (x2 =4.375,P =0.036;x2 =7.864,P =0.005;x2 =9.333,P =0.002),and the eradication rate was higher in group C2 than in group C1 (x2 =4.171,P =0.041),but there were no differences among groups B,C2 and D (P >0.05).As for per-protocol (PP) analysis,the eradication rate was 79.5%,86.5%,79.6%,88.9% and 92.5% in groups A,B,C1,C2,and D respectively,but no significant statistical differences were found among the five groups (P > 0.05).Adverse effects included nausea,bloating,taste distortion,anorexia and constipation.The rate of adverse effects in groups A,B,C1,C2 and D was 67.9% (38/56),26.8% (15/56),35.7% (20/56),21.4% (12/56),and 17.9% (10/56) respectively.The incidence rate was significantly lower in groups B,C2 and D than in group A (P < 0.05),but no significant statistical differences were found among groups B,C2,and D (P > 0.05).Conclusions The triple therapy combined with bifidobacterium and lactobacillus triple live bacteria can obviously decrease the adverse effects and improve patient compliance,thereby increasing the rate of Hp eradication.14-day therapy with probiotics is the best regimen.
6.Quality evaluation of artificial musk based on its inhibitory effect on cyclooxygenase-2.

The inhibitory ratio (1%) of artificial musk on cyclooxygenase-2 (COX-2) was determined by enzyme immunoassay (EIA). The dose-effect relationship between concentrations of artificial musk and 1% was established. It was found that artificial musk had obvious inhibitory action on COX-2. The concentration for 50% of maximum inhibitory effect (IC50) was about 2.26 mg x mL(-1). There was a good relationship between the logarithm concentrations of artificial musk and 1% when the concentrations of artificial musk ranged from 0.31-20.0 mg x mL(-1). The results indicated that this EIA method could be applied to evaluate the anti-inflammatory activity of artificial musk quickly, conveniently, sensitively and exactly. This paper provided a novel method and foundational research for the bioassay of artificial musk.
7.Influences of apolipoprotein CII concentrations on high-density lipoprotein subclass distribution

AIM: To investigate the influence of serum apolipoprotein (apo) CII concentrations on the distribution of serum high-density lipoprotein (HDL) subclasses. METHODS: Serum HDL subclasses in 247 subjects were determined by two dimensional gel electrophoresis-immunodetection. RESULTS: With the increase in serum apolipoprotein CII levels, age, BMI, the contents of TG, TC, apoB100, apoCII, apoCIII, apoE, preβ_1-HDL, preβ_2-HDL, HDL_3b and HDL_3a increased significantly, but the contents of HDL-C, HDL_(2a) and HDL_(2b) decreased remarkably. The contents of preβ_1-HDL increased with the rise in apoCII and apoA I levels, whereas the content of HDL_(2b) increased with the rise in serum apoA I level in the same apoC II group, but decreased with the increase in serum apolipoprotein CII level in the same apoA I group. With the increase in the ratio of apoCII/ apoCIII, the content of preβ_1-HDL elevated, but the content of HDL_(2b) decreased. The correlation analysis illustrated that the apoCII level was positively correlated with preβ_1-HDL (r=0.186, P<0.01), but inversely correlated with HDL_(2b) (r=-0.149, P<0.05). The apoA I level was positively associated with all HDL subclasses (r in the range of 0.349-0.587, P<0.01). In addition, the apoCIII level was positively correlated with preβ_1-HDL (r=0.184, P<0.01) and preβ_2-HDL (r=0.178, P<0.01), while the apoE level was positively correlated with HDL_(3a) (r=0.040, P<0.05). The apoB100 level was inversely correlated with HDL_(2a) (r=-0.102, P<0.05). CONCLUSION: The particles of HDL show a general shift towards smaller size with the increase in apoCII levels, indicating that the maturation of HDL is abnormal. Whereas the contents of apoA I level correct the effect of apoCII on the distribution profile of HDL subclasses. The ratio of apoCII/apoCIII might also been taken as one of the indexes reflecting the distribution profile of serum HDL subclasses.
8.Development of the outflow tract ridge in the embryonic mouse heart

Objective To investigate the origin of α-SMA positive cells in the outflow tract ridge of the embyonic mouse heart and to explore the ultrastructure change of the mesenchymal cells during the ridges fusion. Methods Sections of embryonic day 10(E10d) to E14d mouse embryonic hearts were stained by immunohistochemistry assay with monoclonal antibodies against α-smooth muscle actin (α-SMA), α-sarcomeric actin(α-SCA) and in situ hybridization method with PlexinA2 probe. The outflow tract ridges fusion was observed by transmission electron microscopy at E12.5d. Results From E10d to E11d, PlexinA2 positive cells were seen in the neural tube with mesenchymes around it, the aortic sac and aortic arch. These cells also migrated into the outflow tract ridge along the aortic sac wall and part of them expressed α-SMA. At E12d, PlexinA2 was expressed in the spinal ganglia, the pharyngeal mesenchyme, the aorto-pulmonary septum and the ascending aorta and pulmonary trunk. The septum showed α-SMA strongly positive. But only a few of α-SMA positive cells were observed in the ascending aorta and pulmonary trunk. At E12.5d, two clusters of condensed mesenchymal cells in the outflow tract ridges formed and began to express PlexinA2 weakly and α-SMA strongly. When the ridges began to fuse, the endothelial cells formed a cellular seam, which rapidly broke into pieces and disappeared and were replaced by the sparsed mesenchymal cells containing a few of microfilaments. Two clusters of condensed mesenchymal cells gradully moved to merge. It was noted that some mesenchymal cells contained plenty of microfilament bundles, mitochondria and focal contacts between them. Some mesenchymal cells only had a few of microfilaments and plasma membrane fusion was seen between them. Conclusionα-SMA positive cells in the outflow tract cushion may be derived from cardiac neural crest. The endothelial cells might participate in the fusion of the outflow tract ridges by endothelial-mesenchymal transformation. Mesenchymal cells of the condensed cell mass contain plenty of microfilament bundles and focal contacts or membrane fusion, which contribute to the ridges fusion.

10.A Study on the Recombinant 26 kDa Glutathione-S-Transferase as a Vaccine Candidate:Dynamics of Antibodies in Immunized Buffaloes and Protection against Schistosoma japonicum Infections

ObjectiveTo observe the dynamics of antibodies and protection against Schistosoma japonicum infections in buffaloes after immunized with recombinant 26 kDa glutathione S transferase (reSjc26GST). Methods Buffaloes in 2 villages endemic for schistosomiasis japonica were selected as test and control groups, respectively.In test group initially 96 buffaloes were vaccinated with reSjc26GST, and 90 buffaloes in the control group did not experience vaccination. The indicators included levels of antibodies to reSjc26GST in buffaloes before and after infection with S japonicum and changes in infection rate. Results Specific antibodies, which showed a trend of trapezoid increase, were induced in buffaloes after immunized with reSjc26GST. Twenty months after immunization, the infection rate of the test group was decreased by 62 2% when compared with that before vaccination,and by 67 7% when compared with that of the control in the corresponding period.Conclusion Specific antibodies and a certain extent of protection were induced in buffaloes after immunized with reSjc26GST, which played an significant role in ameliorating morbidity.
