1.Preparation of anti-Rh(D) reagent serum by using Rh(D) positive erythrocyte absorption

Objective To develop a new method of preparing anti-Rh(D) reagent serum in order to help overcome the shortage of Rh(D) negative red blood cells(RBCs). Methods Anti-Rh(D) containing type A and B sera were mixed. Through dilution and neutralization, the titer of complete antibodies decreased to 2. Anti-Rh(D) saturated Rh(D)positive RBCs were prepared by adding anti-Rh(D) containing serum to Rh(D) positive RBCs so that all the Rh factors on the cells were saturated. The anti-Rh(D) saturated type A and type B RBCs were added to the mixed plasma to absorb the remaining anti-A and anti-B. Results The acquired regent sera had a high anti-Rh(D) titer with a high specificity. Conclusion Anti-Rh(D) saturated Rh(D) positive RBCs absorption after neutralization can help with the production of human origin anti-Rh(D) reagent serum and save a large amount of Rh(D) negative evythvocytes.
2.~(137)Cs gamma irradiation for sterilization of red blood cells contaminated by escherichia Coli

Objective To study the bacteriological effect of different dosages of gamma irradiation on Escherichia Coli (E. Coli) in red blood cells (RBC) products,and assess the appropriate dose of gamma irradiation. Methods Prepare RBC samples inoculated with(102-106)/ml of E. Coli. The contaminated RBCs were irradiated by 0 (as control),15,20,25,30 and 35Gy of gamma irradiation. E. Coli in the RBCs were tested on days 0,7, and 14 after irradiation. Results The quantities of E. Coli changed after irradiation. The RBCs inoculated with less than 103cfu/ml of E. Coli were completely sterilized by 25 Gy irradiation. Conclusion 25 Gy gamma irradiation can eradicate less than 103cfu/ml of E. Coli in the RBC products.
3.The difference of intensity of A,B,H antigen on erythrocytes between neonates and adult

Objective To detect the difference of intensity of A ,B ,H antigen on erythrocytes between neonates and adult .Meth-ods Anti-A ,anti-B and anti-H serum were diluted in multiple proportions ,then add the sample erythrocytes into the test tube to react with the serum above .Observe the agglutination between the erythrocytes and the serum ,and score for every agglutination re-action .The total of the score in different dilution of every sample were brought into statistics analysis by SPSS software .Results The A ,B ,H antigen intensity on the surface of erythrocyte of neonates were less than that of adult (P<0 .05) .And no matter neo-nates or adult ,the A antigen intensity on the surface of type A erythrocytes were higher than that of B antigen on the surface of type B erythrocyte(P<0 .05) .The A ,B ,H antigen intensity have no significant difference between adult male and female (P>0 .05) .Conclusion There is a significant difference of A ,B ,H antigen intensity on erythrocytes between neonates and adult .It also can be concluded that the varity of the antigen has effect on intensity of A ,B ,H antigen in erythrocytes ,except sex .
4.Study of the effects of three different types of transportations on the quality of red cell suspension

Objective To study the effects of aircraft, railway and highway transportations on the quality of red cell suspension.Methods Red cell suspensions from aircraft, railway and highway transportations were randomly selected, 10 units each, and stored for about 20 days as test groups, Another 10 unites of red cell suspensions were collected and stored for 20 days to serve as the control group. Biochemical indicators, including lactic dehydrogenase(LDH), Na+, K+ and Cl-were tested by automatic biochemistry analyzer. Free hemoglobin (FHb) was measured by chromatometry of 3 wavelengths. PO2 was detected by blood gas analyzer. The rate of red blood anaerobic glycolysis and osmotic fragility were also tested. The microstructure of red blood cells (RBCs) was observed by scanning electron microscope.Results The rates of anaerobic glycolysis of aircraft and railway transported red cell suspensions were (42.9?5.6)% and (42.2?3.1)%, respectively, significantly lower than those of highway transportation and the control group, which were (60.4?5.9)% and (70.1?7.8)%, respectively. FHb concentrations in erythrocyte suspension from aircraft and railway transportations were (1.37?0.31) mg/ml and (1.28?0.41) mg/ml, respectively, significantly higher than those of the highway transportation and the control group, which were (0.37?0.05) mg/ml and (0.31?0.03) mg/ml, respectively. LDH level and Na+ concentrations in erythrocyte suspension from aircraft and railway transportations were significantly higher than those from highway transportation and the control group. PO2 was decreased significantly in red cell suspension from aircraft transportation, and the osmotic fragility was increased. K+ was increased significantly in red cell suspension from railway transportation. Cl-was not changed. The aircraft and railway transported RBCs showed agglutination, spherical changes and ragged edges. Many thorn RBCs could be found in these samples. The morphology of RBCs after highway transportation had normal shape , but a few were spherical and ragged at edges. The RBCs of the control group was in double-faced concave disc shape and equably suspended.Conclusion The quality of red cell suspension is closely related to the different ways of transportation. Under certain circumstances, the quality of red cell suspension from highway transportation is superior to that from aircraft and railway transportations.
5.Quality evaluation of artificial musk based on its inhibitory effect on cyclooxygenase-2.

The inhibitory ratio (1%) of artificial musk on cyclooxygenase-2 (COX-2) was determined by enzyme immunoassay (EIA). The dose-effect relationship between concentrations of artificial musk and 1% was established. It was found that artificial musk had obvious inhibitory action on COX-2. The concentration for 50% of maximum inhibitory effect (IC50) was about 2.26 mg x mL(-1). There was a good relationship between the logarithm concentrations of artificial musk and 1% when the concentrations of artificial musk ranged from 0.31-20.0 mg x mL(-1). The results indicated that this EIA method could be applied to evaluate the anti-inflammatory activity of artificial musk quickly, conveniently, sensitively and exactly. This paper provided a novel method and foundational research for the bioassay of artificial musk.
6.Research on psychological resilience and its influencing factors among parents of children with cancer

Objective To probe into the psychological resilience and its influencing factor among parents of children with cancer.Methods A total of 125 parents were investigated by self-designed general information questionnaire,Connor-Davidson psychological resilience scale (CD-RISC),Chinese version of Parents' Perception Uncertainty in Illness Scale (PPUS),Social Support Rating Scale (SSRS),self-rating depression scale (SDS) and self-rating anxiety scale (SAS).Results The total score of psychological resilience was (54.86±12.88) and the illness uncertainty score was (91.52±14.66).The social support score was moderate with a total score of (39.86±7.99).The anxiety and depression among parents got a total score of (40.08±7.19) and (44.61±7.38) respectively.Psychological resilience was negatively correlated with illness uncertainty and depression,and was positively correlated with the social support.The depression,illness uncertainty,residence,children's age,clinical diagnosis entered into the multiple linear regression equation of psychological resilience,predicting 30.1% of total variation in psychological resilience.Conclusions Social support,illness uncertainty and depression are the important factors which affect psychological resilience among parents of children with cancer.Nurses should assess the social support that parents receive,the level of depression and the disease feeling of the parents,and provide targeted interventions to relieve it,help to establish effective coping style and improve their psychological state.
7.Rationality of the processing methods of aconiti lateralis radix (Fuzi) based on chemical analysis.

In this study, we explored the rationality of processing methods and mechanism of Aconiti Lateralis Radix (Fuzi) through comparing the chemical contents of diester alkaloids (DAs) and monoester alkaloids (MAs) in the raw material of Fuzi and its processed products. The results showed that the toxicity potency of MAs is at least lower than 1/64 to 1/180 of the toxicity potency of DAs. The contents of DAs in processed Fuzi decreased to 1/76.5 to 1/38.3 of the value of raw Fuzi. The contents of MAs in processed Fuzi significantly increased by 4.6 to 5.2 fold or basically the same as that of the raw Fuzi. The values of MAs/DAs of processed Fuzi were enhanced by 30 to 390 fold of the raw Fuzi. It was found that the contents of DAs were insignificantly different between "Wu dan fu pian" (steaming or stir-frying without Danba) and "Dan fu pian" (steaming or stir-frying with Danba). The result suggested that the abilities of "eliminating toxicity" of different processing methods were equivalent at all. In contrast, the contents of MAs contained in "Wu dan fu pian" were of 5.3 to 8.7 fold higher than the values in "Dan fu pian". This result suggested the processing method by steaming or stir-frying without Danba might have better effect for "conserving property" than the method processed with Danba stipulated by China Pharmacopoeia. We believe that the new processing method without Danba can be recommended in further application due to it offers a simple procedure and it will not introduce inorganic impurities in the products.

OBJECTIVE The abnormal amyloid-β(Aβ)and oxidative stress assiociated with the progression of Alzheimer disease(AD).Quercetin has been reported to possess antioxidant and anti-inflammatory properties in neurodegenerative disorders.In this present study,we designed to characterize the mechanisms by which quer-cetin exerts neuroprotective effects in murine neuroblas-toma N2a cells stably expressing human Swedish mutant amyloid precursor protein(N2a/APP).METHODS N2a/APP cells were treated with quercetin at concentrations of 10,20 and 50 μ mol·L-1 for 24 h.Cell viability was examined with CCK-8 assays.The protein levels of ERK1/2 and Akt were detected by Western blotting.Intra-cellular reactive oxygen species(ROS)was detected by a fluorescent probe 2,7-dichlorofluorescein diacetate(DCFH-DA).The mitochondrial membrane potential(Δψ m)in N2a/APP cells was detected by using JC-1 staining method.Immunofluorescence was used to detect the generation of 8-hydroxy-2′-deoxyguanosine(8-OHdG)and 4-hydroxynonenal(4-HNE).RESULTS Quercetin attenuated the enhancement of p-ERK1/2,reductions of p-Akt,and decreased levels of APP expression.More-over,quercetin alleviated loss of mitochondria membrane potential(MMP)since it attenuates these oxidative stress,as reflected in the levels of ROS,4-HNE and 8-OHdG,was elevated in N2a/APP cells and these effects were again ameliorated by quercetin.CONCLUSION Neuroprotection by quercetin in N2a/APP cells involves normalizing the impaired mitochondrial function and reducing oxidative stress via inactivation of the ERK1/2 and activation of the Akt pathways.
