Objective To prove the mechanism of diethylstibestrol(DES) induced abnormal spermatogenesis, this study on the changes of proto-oncogenes bcl-2 and p53 proteins as well as cadherin adherens molecules expression in adult male hamster spermatogenic cells. Methods Adult male golden hamsters were treated daily olive oil vehicle either alone(control)or containing 1 mg/kg body weight of DES (experiment)by subcutaneous injection for 7 days. The changes of testes histological morphology were observed under light and electron microscope. The expressions of bcl-2 and p53 and cadherin in the testes seminiferous epithelium were detected by immunohistochemistry. Results DES induced adult hamster testes abnormal spermatogenesis. As compared to the control group animals,in DES-treated group animals the expression of both bcl-2 and p53 significantly increased in spermatogenic cells, especially in spermatocytes and round spermatids. However, the expression of cadherin adherens molecules obviously decreased. Conclusion It is one of reasons of DES inducing abnormal spermatogenesis that DES can result in increasing in the expression of proto-oncogenes bcl-2 and p53,and reducing cadherin adherens molecules expression.

Objective To investigate the dose-response of micronuclei (MN) frequency in the lymphocytes irradiated with or without combination of α-particles and γ-rays. Methods Human lymphoblast cells HMy2.CIR were irradiated with 0 - 1 Gy of α-particles,0 - 5 Gy of γ-rays,and 0.025 -0.5 Gy of α-particles followed by different doses of γ-rays,respectively.The micronuclei (MN) in the irradiated cells were measured with the cytokinesis block technique, and the dose-responses of MN were established under different irradiation conditions. Results For γ-ray irradiation, the dose-response of MN was well-fit by the linear-quadratic model with an equation Y =c + αD + βD2.For α-particle irradiation, the MN induction increased linearly with the dose less than 0.250 Gy. But when the dose of α-particles increased continually, the dose-response curve bended and could be well fit with the BaD model Y =c + αD + σ[ 1 - exp( - δD) ] exp( - βD) where radiation-induced bystander effect (RIBE) was indicated. For the combined exposure, the dose-response of MN was similar to that of γ-irradiation when the dose of α-particles was lower than 0.1 Gy,but it was similar to that of α-irradiation when the dose of α-particles was higher. When the dose of α-particles was 0.2 and 0.5 Gy,MN induced by the mixed radiation were significantly higher than the sum of corresponding irradiation alone ( t =5.22 - 11.86,P < 0.05 ).Conclusions The radiation damage of α-particles differs from that of γ-rays,where RIBE may be involved. The combination irradiation of α-particles and γ-rays has a synergistic effect on radiation damage of lymphoblast cells.

AIM: To clone human ?-globin gene carrying a thalassemic mutation IVS II654(C→T) and establish a eukaryotic expression system for high-level expression of human ? IVS II654 gene in mouse erythroleukaemia(MEL) cells. METHODS: The fragments of human ? 654 gene isolated from the ? thalassemia patients homozygous for the ? 654 mutation were amplified by PCR, and cloned to plasmid pBGT51. Then, the human ? LCR and ? 654 gene were subcloned from plasmid pBGT51 to the stable mammalian expression vector pcDNA3.1+ together, and the MEL cells were transfected with this vector using commercially available cationic lipid FuGENE6. The MEL cells were induced for further maturation by DMSO and the expression of human ? 654 gene in the MEL cells was identified by RT-PCR. RESULTS: A mammalian expression system of human ? thalassemic mutation ?IVS II654(C→T) was established. CONCLUSION: The level and the reliability of expression of human ? 654 gene in the MEL cells in vitro are similar to that in vivo in human body. This may be a valuable gene therapy model for human ? thalassemic mutation ?IVS II654(C→T).

Objective:To explore the effect of intermittent theta-burst stimulation (iTBS) of the cerebellum on the excitability of the M1 zone in the bilateral motor cortex and the duration of any effect. Methods:Twenty healthy youths were randomly assigned to a left or a right cerebellum iTBS group, each of 10. The resting motor threshold and motor evoked potential (MEP) of the motor cortex were determined before giving 600 pulses of iTBS. Then MEP was measured at the 5th, 10th, 15th, 20th, 25th, 30th, 35th and 40th minute after the intervention ended. Any changes in MEP amplitude were also analyzed.Results:①The average MEP amplitude in the right motor cortex had increased significantly in the left cerebellum iTBS group from the 5th to the 15th minute. The increase lasted at least 10 minutes. Then it had returned to the baseline value at the 35th minute. ②The MEP amplitude in the left motor cortex decreased slightly after iTBS of the left cerebellum began, but there were no significant changes. ③The MEP amplitude in the left motor cortex had a tendency to decrease after iTBS was administered to the right cerebellum, but that change too was not significant. ④The MEP amplitude of the right motor cortex decreased significantly only for the first 30 minutes. Conclusions:iTBS of the cerebellum can increase the excitability of the contralateral motor cortex. The effect can last 25 minutes. It can also decrease the excitability of the ipsilateral motor cortex for 30 minutes.

Objective To identify the effects of different release paths on public dose in the aftermath of a NPP accident. Methods RASCAL4.3 software was used to simulate the source terms of PWR NPPs and the characteristics of dose distribution in different release path scenarios of long term station blackout (LTSBO) accident. Results It had been shown that radioactivity was the highest in Scenario 2,which was the release path for releases of radionuclides into the environment due to steam generator tube rupture.Radioactivity was the lowest in Scenario 1,which was the release path for radionuclides to be released into the environment in the case of containment leakage or failure.TEDE dose was mainly distributed in the region within 40 km of the release point in different scearios.TEDE dose in the most sectors within 80 km was low than 10 mSv in Scenario 1,which was the release path via containment in containment leakage or failure and in Scenario 3,which was the release path by containment bypass. It was greater than 50 mSv in Scenario 2,which was the release path due to steam generator tube rupture. Conclusions For the environmental impact assessment,it should be paid more attention to the fact that the public health impact imposed by radionuclide release into the environment via the release path due to steam generator tube rupture in the aftermath of LTSBO accident at a NPP.

Background:In April 2013, a hospital in Suzhou City notified authorities of a patient with nitrite poisoning with two other family members who had similar toxic symptoms five days prior. We investigated the event to identify the cause, source and possible route of contamination. Methods:A case was defined as any person living in the Yang Shan Hua Yuan community who had been diagnosed with cyanoderma and food poisoning symptoms from 15 to 25 April 2013. Active case finding was conducted by interviewing community residents and reviewing medical records from local clinics; information was then retrospectively collected on the patient’s food history, cooking procedures and food sources. Results:We identified three nitrite poisoning cases, one male and two females, from the same family. The time between dinner and onset of illness was less than an a hour. A retrospective survey showed that a substance presumed to be sugar mixed with asparagus on 17 April and with stir-fried asparagus on 21 April was the suspected contaminant. The presumed sugar came from a clean-up of a neighbouring rental house. Nitrite was detected in a vomitus sample, the sugar substance and two leftover food samples. Conclusion:This family cluster of nitrite poisoning resulted from the mistaken use of nitrite as sugar to cook dishes. We recommend that sodium nitrite be dyed a bright colour to prevent such a mistake and that health departments strengthen food hygiene education to alert people about the danger of eating unidentified food from an unknown source.
