1.Probable Sporadic Creutzfeldt-Jakob Disease:A Study of Glucose Metabolism byStatistical Parameter Mapping

PurposeTo analyze fluorodeoxyglucose (FDG) PET/CT images in patients with probable sporadic Creutzfeldt-Jakob disease (sCJD) and the affected encephalic regions by statistical parameter mapping (SPM) so that image information can be provided for early clinical diagnosis.Materials and MethodsSeven patients with probable sCJD diagnosed according to the WHO criteria and 2009 clinical diagnosis criteria and 7 controls at the matched age were enrolled in the study. Both groups underwent FDG PET/CT scan and the images were analyzed visually and by SPM.ResultsThe visual analysis showed that the patients with probable sCJD had a hypometabolism in the wide pallium and basal ganglia region and that a portion of patients mainly had a lateral decrease. The SPM analysis exhibited that patients in sCJD group had a pattern of hypometabolism that affected bilateral parietal, frontal, occipital cortices and head of caudate (P<0.05) and indicated that the patients with lateral hypometabolism existed crossed cerebella diaschisis (P<0.05).ConclusionFDG PET/CT image features of sCJD patients present a hypometabolism in the wide pallium and basal ganglia region, which may be helpful in the diagnosis of sCJD in certain clinical situations.
2.Subjective well-being of immigrant children in Beijing

Objective: To explore the subjective well-being (SWB) of immigrant children and the differences between urban children and immigrant children in Beijing. Methods: The SWB Self-evaluating Scale,Positive Affect and Negative Affect Seale-Revised Edition, Student Life Satisfaction Scale, and Multidimensional Student Life Satisfaction Scale were administrated to a sample of 1018 immigrant children and 447 urban children in 4 immigrant children schools, 3 mixed public schools and 4 public schools in Beijing. Results: (1) On the whole,immigrant children from school for farmer-worker's children (6.9±2.7) and from mixed public school (7.4±2.3) scored lowest on SWB, urban children from mixed public school (8.2±2.1) were placed in the middle,and urban children from public school (8.4±1.9) scored highest (F=16.27, P<0.001). (2) Immigrant chlldren's general well-being score was (7.0±2.6), and their positive affect score was higher than that of negative affect [(3.0±0.5) vs.(2.3±0.6), P<0.001] . (3) Boys scored higher in environment satisfaction and gen-eral life satisfaction than girls [(3.7±0.9) vs.(3.5±0.9), (2.9±0.7) vs.(2.8±0.7), Ps<0.05], but lower in school satisfaction [(4.5±0.9)vs.(4.7±0.9), P <0.001] . (4) Higher grade children had more negative affect and self-satisfaction, but lower in the other aspects. Conclusion: Generally speaking, immigrant children's subjective well-being are above the average level, and immigrant children in mixed public schools have higher score than those in pure immigrant children schools, but both kinds of immigrant children score lower than urban children in subjective well-being.
3.Treatment of nonunion of tibial fractures with local muscle flap transfer and injection of autogenous bone marrow

Objective To report the treatment outcome of nonunion of tibial fractures with local muscle flap transfer and injection of autogenous bone marrow. Methods 15 patients who had suffered from nonunion of tibial fracture 10 to 42 (mean 22) months after the initial injury underwent open reduction, internal fixation with locking intramedullary nail, wound coverage with locally transferred muscle flap and injection of autogenous bone marrow into the fracture site 12 to 15 days after operation. Results The follow-ups revealed bone union in all the cases of this series with a mean healing time of 22 (3 to 11) months. Except for limited necrosis of the skin edge in 2 cases which healed after dressing changes, the wounds healed primarily in all the cases without infection and implant failure. Conclusion Besides stable internal fixation and bone graft, coverage of fracture site with locally transferred muscle flap and injection of autogenous bone marrow can be used to treat nonunion of tibial fractures with satisfactory results.
4.Effects of the human 21 .5 kDa MBP gene silencing on the proliferation and apoptosis of glioma cells

Objective To investigate the effects of silencing of the human cerebral 21 .5 kDa myelin basic protein (MBP) gene on the proliferation and apoptosis of the glioma U251 cells .Methods The 21 .5 kDa MBP sequence‐specific short hair‐pin RNA (shR‐NA) recombinant plasmids pGenesil‐1‐MBP‐3 were transfected into the human glioma cell line(U251) ,the cells of U251 was used as MBP silencing group ,the cells transfected with negative control plasmids used as negative control group ,and the cells transfected with liposomes used as blank control group .Real‐time PCR and Westernblot were used to detect the expression levels of the 21 .5 kDa MBP mRNA and protein in each group ,and the cell proliferation curve was measured by CCK‐8 assay ,the apoptosis rate was a‐nalysised by Flow cytometry .Results Both the mRNA and the protein expression levels of the 21 .5 kDa MBP of MBP silencing group were significantly lower than those in the control groups (P<0 .05);the cellular proliferation activity of the MBP silencing group decreased significantly (P<0 .05)while the cellular apoptotic rate increased significantly (P<0 .05) .Conclusion Silencing of the human cerebral 21 .5 kDa MBP gene may playa dual role in the inhibition of proliferation and the promotion of apoptosis of the glioma U251 cells .

6.An improved fixation method of the indwelling needle inserted in the rat tail vein

Objective To improve the fixation method of indwelling needle inserted in the tail vein in rats to keep the inserted needle for a longer indwelling time .Methods One hundred healthy Wistar rats ( age 5~6 months, male:female=1:1) were randomly divided into two groups: the experimental group and the control group .Rats in both groups received the same tail vein indwelling needle puncture and cannulation .The control group got the traditional fixation , namely fixed by sticking with 3M transparent dressing paste .The experimental group received in addition to the traditional fixation, a 0.1 mm-thick aluminum tube placed outside the needle fixation site .The indwelling time in the rats were recorded and analyzed .Results The indwelling time was (166.86 ±9.03) h and (20.24 ±5.04) h in the experimental and control groups, respectively (t =68.546, P<0.01).Conclusions Our improved method is safe and reliable.It can prolong the indwelling time of the punctured intravenous indwelling needle , and provides a useful rat model in studies on the complications of intravenous indwelling needle kept for a longer time .
7.Correlation Between Breast Imaging Reporting and Data System of Ultrasound and 18F-lfuorodeoxyglucose PET/CT in Diagnosis of Breast Diseases

Purpose To evaluate the breast imaging reporting and data system (BI-RADS) of ultrasound and 18F-lfuorodeoxyglucose (FDG) PET/CT in diagnosis of breast tumor, and to analyze the correlation between them. Materials and Methods 18F-FDG PET/CT images and ultrasound images of 103 patients with suspected breast cancer were retrospectively analyzed to get correlation between the maximum standardized uptake values (SUVmax) and BI-RADS. Sensitivity, speciifcity, positive predictive value, negative predictive value were compared with histology or follow-up results as golden standard. Results Of the 103 lesions, 46 were benign and 57 were malignant. Pearson correlation coefifcient was 0.464 (P<0.01). The sensitivity, speciifcity, positive predictive value, and negative predictive value of PET/CT were 89.47%, 73.91%, 80.95%and 84.99%, respectively;those of BI-RADS were 94.70%, 69.60%, 79.42%and 91.38%, respectively. The sensitivity, speciifcity, positive predictive value, and negative predictive value in patients with BI-RADS 3-4 were 88.90%, 71.40%, 66.65%, and 90.91%, respectively. The sensitivity, speciifcity, positive predictive value, and negative predictive value for BI-RADS grading diagnosis were 88.90%, 46.40%, 51.60%and 86.67%, respectively. Conclusion There is no signiifcant correlation between SUVmax and BI-RADS. BI-RADS has low speciifcity for patients with BI-RADS grade 3-4, while PET/CT can make up this shortcoming. Combined diagnosis in the breast disease can be potentially recommended in clinics.
8.The therapeutic effect of elderly dialysis patients with refractory hypertension

Objective To investigate the treatment of elderly dialysis patients with refractory hypertension.Methods Sixty-six patients were randomly divided into two groups of A group and B group,with 33 cases in each group.Patients in A group were treated by routine hemodialysis 3 times per week and hemoperfusion 1 time every 2 weeks.Patients in B group were treated by routine hemodialysis 2 times per week and hemodiafiltration 1 time per week plus hemoperfusion 1 time every 2 weeks.The levels of blood pressure,plasma endothelin (ET),angiotensin-Ⅱ (AT Ⅱ),renin (RA),parathyroid hormone (PTH) in two groups before and after treatment were compared.Results Before and after treatment,the levels of blood pressure,ET,AT Ⅱ,RA and PTH in A group were (183.42 ± 16.20)/(109.15 ± 7.69) and (151.45 ± 3.31)/(88.71 ±4.95) mmHg(1 mmHg =0.133 kPa),(173.45 ±41.25) and (160.38 ±25.31) ng/L,(69.21 ± 18.47)and(57.25 ± 23.34) ng/L,(1.21 ± 0.15) and (0.87 ± 0.14) μ g/L,(530.27 ± 43.28) and (272.43 ± 34.19) ng/Lrespectively.Before and after treatment,the levels of blood pressure,ET,AT Ⅱ,RA and PTH in B group were (185.34 ± 12.26)/ (112.38 ± 6.24) and (141.59 ± 4.15)/ (80.39 ± 2.73) mmHg,(178.21 ± 34.35) and (149.12 ±21.34) ng/L,(68.38 ±23.57) and (47.17 ± 15.61) ng/L,(1.19 ±0.29) and (0.69 ±0.41) μg/L,(528.62 ± 42.53) and (253.15 ± 31.62) ng/L respectively.The levels of blood pressure,ET,AT Ⅱ,RA and PTH in two groups after treatment were significantly decreased compared with those before treatment (P <0.05),and there were significant differences between two groups (P < 0.05).Conclusion Combined with hemoperfusion in the treatment of elderly hemodialysis patients with refractory hypertension,the effect of combined hemodiafiltration is significant,and it is worthy of promotion.

10.TLC identification and determination of baicalin in Qubanjieyan Capsules

Objective: To establish the quality standard for Qubanjieyan Capsules(Radix Scutellariae, Radix Bupleuri, Radix Angelicae Sinensis, Radix Paeoniae Rubra, Flos Carthami, etc.) Methods: Baicalin in Qubanjieyan Capsules was determined by HPLC with methol H 2O H 3PO 4(85∶15∶0.2) as the mobile phase, and the detection wavelength was set at 280nm, while Radix Bupleuri, Radix Angelicae Sinensis, Radix Paeoniae Rubra and Flos Carthami were identified by TLC. Results: The average recovery rate and RSD were 102.20% and 2.18%, respectively. The TLC spots developed were fairly clear, and the blank test showed no interference. Conclusion: The results are fast and simple with good reproducibility, and the method can be used for quality control of the Qubanjieyan capsules.
