1.Clinical significance of the ban nationality and kazak hepatitis B virus large surface protein detection

ObjectiveTo discuss the clinical significance of han nationality and kazak hepatitis B virus large surface protein detection.MethodsEnzyme linked immunosorbent assay(ELISA) method was used to examine the HBV-LP、HBV markers and quantitative real-time PCR methods were used to detect the HBV DNA in 270 patients with Hepatitis B.ResultsAmong the 270 cases,there was no significant difference between the levels of HBV DNA and HBV-LP( P > 0.05 )in HBe Ag-positive patients,which was not affected by nationality.Significant difference of positive rate was observed between HBV-LP and HBV DNA( P <0.05) in HBe Ag-negative ones,which was not affected by nationality.HBV-Lp expression was significantly correlated with the logarithm of HBV DNA level ( r =0.986,P < 0.05).ConclusionThere was higher coincidence rate between the levels of HBV-LP and HBV DNA in HBeAg--positive patients.The positive rate of HBV-LP was higher than that of HBV DNA in HBe Ag-negative patients.HBV-LP could serve as a reliable marker in the reflection of HBV the replication at protein level,and it was valuable to monitor HBV replication and prognosis of the disease,especially in HBe Ag-negative HBV infected patients.

3.Insulin enhances glucose transporter 1 gene expression of low-flow ischemic myocardium

AIM: To investigate the mechanism underlying insulin-stimulated increase in glucose uptake during low-flow myocardial ischemia. METHODS: The expression of myocardial GLUT1 polypeptide was determined by semiquantitative immunoblotting. The expression of GLUT1 mRNA was determined by semiquantitative Northern blotting. RESULTS: After infusing insulin during low- flow myocardial ischemia for 8 h,the expression of both GLUT1 mRNA and GLUT1 polypeptide was significantly higher in experimental myocardium than that in normal myocardium. The glucose uptake was upregulated at the same time in the exprimental myocardium. CONCLUSION: Insulin enhances the expression of GLUT1 mRNA and GLUT1 polypeptide in ischemic myocardial regions. GLUT1 expression may be an important mechanism by which myocardial cells enhance glucose uptake and metabolism during low-flow myocardial ischemia.
4.Additive e ffects of hyperinsulinemia and ischemia on canine myocardial GLUT4 gene expression in vivo

Objective: To investigate whether there is additi ve effects of hyperinsulinemia and ischemia on expression of canine myocardial G LUT4 gene in vivo. Methods: The expression of myocardial GLU T4 was determined by semiquantitative immunoblotting.The expression of GLUT4 mRN A was determined by semiquantitative Northern blotting. Results: Dramatic changes were seen in GLUT4 mRNA and GLUT4 expression in the ischemic hearts.After infusing insulin for 8 h,regional GLUT4 mRNA and GLUT4 levels in is chemic hearts were 2.5, 2.3-fold that of expression in normal hearts(P<0.01 ). Myocardial glucose uptake in ischemic hearts was increased by 4-fold when co mpared with normal hearts(P<0.01). Conclusion: There are not only additive effects of hyperinsulinemia and low-flow ischemia on canine myoc ardial GLUT4 mRNA and GLUT4 expression in vivo, but also increase of myocar dial glucose uptake. Enhanced GLUT4 expression may be an important protective m echanism by which myocardial cells enhance glucose uptake and metabolism during low-flow ischemia.

8.Treatment and prognosis of squamous cell carcinoma of thyroid.

OBJECTIVE:
To investigate the treatment and prognosis of the patients with squamous cell carcinoma of thyroid.
METHOD:
The clinical data of all patients with squamous cell carcinoma of the thyroid in our hospital from Dec. 1994 to Dec. 2008 were analyzed retrospectively.
RESULT:
The overall 1-year, 3-year survival rates were 36.4% and 9.1% respectively. The median survival time was 8 months. Survival analysis showed the patients receiving surgery and postoperative radiotherapy had a better prognosis.
CONCLUSION
The prognosis of squamous cell carcinoma of thyroid was poor; the patients with squamous cell carcinoma of thyroid maybe get a better prognosis by receiving surgery and postoperative radiotherapy.

9.Deep learning-based drug screening for the discovery of potential therapeutic agents for Alzheimer's disease

Alzheimer's disease(AD)is gradually increasing in prevalence and the complexity of its pathogenesis has led to a lengthy process of developing therapeutic drugs with limited success.Faced with this challenge,we proposed using a state-of-the-art drug screening algorithm to identify potential therapeutic com-pounds for AD from traditional Chinese medicine formulas with strong empirical support.We developed four deep neural network(DNN)models for AD drugs screening at the disease and target levels.The AD model was trained with compounds labeled for AD activity to predict active compounds at the disease level,while the acetylcholinesterase(AChE),monoamine oxidase-A(MAO-A),and 5-hydroxytryptamine 6(5-HT6)models were trained for specific AD targets.All four models performed excellently and were used to identify potential AD agents in the Kaixinsan(KXS)formula.High-scoring compounds underwent experimental validation at the enzyme,cellular,and animal levels.Compounds like 2,4-di-tert-butyl-phenol and elemicin showed significant binding and inhibitory effects on AChE and MAO-A.Additionally,13 compounds,including α-asarone,penetrated the blood-brain barrier(BBB),indicating potential brain target binding,and eight compounds enhanced microglial β-amyloid phagocytosis,aiding in clearing AD pathological substances.Our results demonstrate the effectiveness of deep learning models in devel-oping AD therapies and provide a strong platform for AD drug discovery.
