Objective To investigate the expression of matrix metalloproteinases 9 (MMP 9) and tissue inhibitor of metalloproteinases 1(TIMP 1) mRNA in the endometrium during mid luteal phase in women with unexplained infertility and its steroidal regulation Methods In situ hybridization method was utilized to detect the location of MMP 9 and TIMP 1 mRNA. Reverse transcription polymerase chain reaction (RT PCR) method was utilized to detect and quantitate MMP 9 and TIMP 1 mRNA expression levels in the endometrium during mid luteal phase. Serum concentration of estradiol (E 2) and progesterone (P) were measured by radioimmunoassay. Thirty eight patients with unexplained infertility were included as study group, and 20 women with male factor infertility or normal volunteers were selected as control group. Results MMP 9 and TIMP 1 mRNA were diffusely distributed in the cytoplasm and neuclei of glandular and stromal cells. Their staining in control group were more abundant than those in study group. Endometrial MMP 9 and TIMP 1 mRNA expression during mid luteal phase showed by RT PCR in the study group were significantly lower than those in the control group (0.42?0.19 Vs 0.57?0.19, P

4.Emission Rate of Formaldehyde in Artificial Woodiness Boards

Objective To understand the emission rate and the attenuation rule of formaldehyde in the artificial woodiness boards for sell in China and to study the efficiency of formaldehyde-eliminator. Methods With the small Climate-Chamber and 4160 formaldehyde-analyzer, quantity of formaldehyde released from some kinds of woodiness boards was analyzed under the different condition of temperature, relative humidity and ventilation cycle. Results The emission rate of formaldehyde in most veneers was over 1.2 mg/(m2?h), in most of moderate density fibreboard, it was 0.7~1.0 mg/(m2?h) and in the veneer for medical application, it was 0.3~0.9 mg/(m2?h). Formaldehyde emission rates of all the three kinds of woodiness boards investigated in the present study showed a fall of 75% in a period of time after made, and met the National Standard of China. The formaldehyde eliminator could reduce the emission rate of formaldehyde remarkably, however the effect perhaps weakened in a short time. Conclusion The formaldehyde emission rates in all kinds of woodiness boards investigated in the present paper are different, they exceed the National Standard of China while the boards made immediately and will reduce after 10-15 days in a given condition. The formaldehyde eliminator used in the study shows a remarkable effect but lasts only a short time.
5.Deguelin treatment delays embryonic development of zebrafish:A gene chip analysis

BACKGROUND:Anticancer drug deguelin delays the embryonic development of zebrafish, but its exact mechanism is not yet clear.
OBJECTIVE:To explore expression differences in genes and signaling pathways in deguelin-treated zebrafish embryos.
METHODS:Zebrafish embryos were harvested from zebrafish fed according to the Zebrafish Book. Zebrafish embryos at 2-cel stage were selected and divided into two groups:control group treated with dimethyl sulfoxide incubator liquid, and experimental group treated with 0.6μmol/L deguelin. Total RNA was extracted from the two groups at sphere stage. Then, gene chip technique was used to detect differential y expressed genes in the deguelin-treated zebrafish embryos. Real-time quantitative PCR was employed to validate microarray cluster analysis and pathway analysis to explore the mechanism of action of deguelin.
RESULTS AND CONCLUSION:Chip results showed that 407 genes were upregulated more than three times and 461 genes were downregulated more than three times after deguelin treatment. PCR validation results were consistent with those of the chip. Fourteen pathways were identified by KEGG pathway analysis. Deguelin may play an important role by intervening cel metabolism growth and differentiation.
6.Analysis for influence of coronary artery lesions position and degree in 224 patients with acute ST eleva-tion myocardial infarction

Objective:To explore influence of position of myocardial infarction and coronary artery lesions in patients with acute ST elevation myocardial infarction (STEMI) ,and the relationship among heart failure ,arrhythmia and severity of coronary artery lesion .Methods :Clinical data of 224 patients ,who hospitalized in our hospital because of STEMI and received coronary angiography from Jan 2009 to Jun 2011 ,were retrospectively analyzed .General data of patients were collected ,and SYNTAX score was used to reflect severity of coronary artery lesion ,and the rela‐tionship among heart failure ,arrhythmia and SYNTAX scores were analyzed .Results:Incidence rate of heart fail‐ure in patients with infarction relate artery left anterior descending artery (LAD) AND/or left main coronary artery (LM) was significantly higher than that of patients with right coronary artery (RCA) (57.0% vs .39.7% , P=0.017) ,incidence rate of arrhythmia in patients with RCA was significantly higher than that of patients with left circumflex artery (LCX) (37.0% vs .6.3% , P=0.016);incidence rates of arrhythmia (48.4% ) ,shock (54.8% ) were highest in patients with inferior wall/right ventricle than those of other position , P<0.05 or <0.01.SYNTAX scores in patients with heart failure and arrhythmia were significantly higher than those of patients without heart failure and arrhythmia respectively [ (18.7 ± 9.1) scores vs .(15.4 ± 8.6) scores ,(19.7 ± 9.0) scores vs .(16.1 ± 8.8) scores , P<0.01 both] .Conclusion:Incidence rates of heart failure ,cardiogenic shock and arrhythmia are related to coronary artery lesions position and degree and myocardial infarction position in STEMI patients .
7.Influence factor analysis of 656 cases with AMI undergoing emergency PCI

Objective:To analyze influence factors in patients with acute myocardial infarction (AMI) undergoing e‐mergency percutaneous coronary intervention (PCI ) .Methods:Clinical data of 656 patients ,who received emer‐gency PCI because of AMI in our hospital from Jan 2005 to Jan 2015 ,were retrospectively analyzed .Short -term prognosis ,including incidence rates of heart failure (HF) ,cardiogenic shock (CS) ,arrhythmias and mortality , were compared and analyzed among these patients from onset related factors ,coronary disease and vascular reope‐ning time etc .Results:Mean onset age of men was significantly younger than that of women (P<0.05) .Incidence rate of AMI in winter and spring was significantly higher than those in summer and autumn (χ2 = 6.244 , P=0.012) .According to complicated with hypertension ,DM ,dyslipidemia and smoking or not ,patients were divided into corresponding disease group and normal group ;only incidence rates of CS (37.78% vs .29.62% ) and arrhyth‐mia (47.78% vs .38.24% ) in DM group were significantly higher than those of non -DM group ,mortality rate of dyslipidemia group was significantly lower than that of normal blood lipid group (0.73% vs .3.69% ) , P<0.05 or< 0.01 ,there were no significant difference in incidence rates of other events between normal group and disease group (P>0.05 all) .Incidence rate of arrhythmia in RCA group was significantly higher than those of LAD group , LCX group and multi -vessel group (56.36% vs .31.55% ,37.50% ,34.38% ) ,mortality of LM group was signifi‐cantly higher than those of LAD group ,RCA group and LCX group (25.00% vs .1.79% ,0.91% ,0% ) , P<0.05 or <0.01. Incidence rate of HF in 10~12h group was significantly higher than those of 0~3h ,4~6h and 7~9h group (79.46% vs .61.70% ,66.81% ,64.78% ) ,incidence rate of arrhythmia was significantly lower than those of 0~3h ,4~6h and 7~9h group (32.14% vs .55.32% ,43.81% ,44.65% ) ,and incidence rate of CS was significant‐ly higher than that of 0~3h group (35.27% vs .21.28% ) , P<0.05 or <0.01. Conclusion:Onset age ,season and DM ,coronary disease extent ,vascular reopening time are risk factors influencing short‐term prognosis of AMI .
8.Effects of infliximab induced clinical remission of active Crohn's disease on the level of interleukin-21 and interleukin-21 receptor

Objective To investigate the changes of interleukin-21 (IL 21) and interleukin-21 receptor (IL 21R) expression level in Crohn's disease (CD) patients before and after accepted infliximab (IFX) treatment.Methods From June 2009 to July 2011,twenty-two CD patients met the research criteria were recruited at Tenth People's Hospital of Tongji University.Patients were treated with infliximab at weeks 0,2,6,and 16 healthy individuals were set as healthy control group at same time.Peripheral blood of healthy control group was taken at regular physical examination and blood of CD patients was taken before treatment and 10 weeks after treatment,intestinal mucosa biopsy samples were taken under colon endoscopy examination.The changes of Crohn's disease activity index (CDAI),erythrocyte sedimentation rate (ESR),C-reactive protein (CRP) in CD patients were observed.The change of IL-21R in peripheral blood CD4+ T lymphocytes was detected by flow cytometry.The change of IL-21 expression at mRNA level in intestinal mucosa was determined by realtime quantitative polymerase chain reaction (PCR).The data were analyzed by t test.Results Before treatment,the level of IL21R in peripheral blood CD4+ T lymphocytes of CD patients (12.25%±3.25%) and the expression of IL-21 at mRNA level in inflamed intestinal mucosa (1.38±0.32) were both significantly higher than those of healthy controls (4.25 % ± 1.41%,0.44±0.18),the differences were statistically significant (F=15.88,6.75 ; both P<0.05).At 10th week,the level of IL-21R in peripheral blood CD4+ T lymphocytes of CD patients (8.12% ± 2.05%) and the expression of IL-21 at mRNA level in intestinal mucosa (0.77 ± 0.24) were both significantly lower than those before treatment,the differences were statistically significant ( t=4.880,8.019; both P<0.01).Before treatment,ESR,CRP and CDAI of CD patients was (46.8±11.4) mm/1 h,(52.4±11.5) mg/L and 319±74,which was (23.5±9.0) mm/1 h,(11.6±4.6) mg/L and 113±42 after treatment,the differences were statistically significant (t=9.485,16.458,11.100; all P<0.05).Conclusion The IL-21 expression of active CD patients decreases after IFX treatment,which indicates that IL-21 may involve in IFX induced clinical remission of active CD.
9. Influence of brown adipose tissue activity on 18F-FDG uptake in tumors

Objective: To evaluate the influence of BAT (brown adipose tissue) activity on 18F-FDG (18F-fluoro-2-deoxyglucose) uptake in tumors in cancer-bearing mice by using PET/CT (positron emission tomography/computed tomography) imaging. Methods: The model of mice bearing NCI-H1299 human non-small cell lung cancer was established. The activity of BAT in mice was adjusted through regulating the experimental conditions. Two groups were designed in this study, including BAT-activation group (the activity of BAT in mice was stimulated by cold exposure and food diet during the period of 18F-FDG uptake) and BAT-inhibition group (the activity of BAT in mice was inhibited by fasting, anesthesia and heating). The mice were scanned with micro-PET/CT and the uptake of 18F-FDG in tumors in the two groups was analyzed and compared. The SUV (standard uptake value) was calculated. Results: The tumor in BAT-activation group was nearly invisible on PET image and its 18F-FDG uptake value (SUV 0.15±0.06) was significantly lower than that in the BAT-inhibition group (SUV 0.58±0.20) (P < 0.05). Conclusion: The enhancement of BAT activity will significantly decrease the uptake of 18F-FDG in tumors and influence the imaging of the tumors. Copyright © 2013 by TUMOR.
10.Expression and its clinical significance of microRNA-10a in inflammatory bowel disease

Objective To investigate the expression of microRNA (miRNA)-10a in the intestinal mucosa,serum and peripheral blood mononuclear cell (PBMC) of patients with inflammatory bowel disease (IBD) and explore its role and relevance in the pathogenesis of the disease.Methods The intestinal or colonic mucosal biopsy specimens of nine active ulcerative colitis (UC) patients,11 active Crohn's disease (CD) patients and eight patients with negative colonoscopy result as control were collected.The sera of 12 active UC patients,13 active CD patients and nine healthy controls were collected.The PBMC of nine active UC patients,11 active CD patients and eight healthy controls were collected.The expression of miRNA-10a in the intestinal mucosa,sera and PBMC and the expression of IL-12/IL-23 p40 in the intestinal mucosa were detected by real-time polymerase chain reaction (PCR).Each 8 cases of active UC and CD patients were collected.The intestinal mucosa before infliximab (IFX) treatment and six weeks after three times of IFX treatment were collected.And at same time,the intestinal mucosa of 11 active UC patients and 10 active CD patients were collected and cultured for 18 hours stimulated with IFX in vitro and then the expression of miRNA-10a in the intestinal mucosa was tested.One-way analysis of variance was used for comparison in three samples.Paired t-test was used for two samples comparison.Spearman test was used for correlation analysis.Results Compared with healthy controls,the expression of miRNA-10a in the intestinal mucosa,serum and PBMC of UC and CD patients significantly decreased (F=38.45,30.46 and 14.74,all P<0.05).There was no statistic significance between UC and CD groups.The expression of IL-12/IL-23 p40 in the intestinal mucosa of UC and CD patients significantly increased (F=32.90,P<0.05).The expression of IL-12/IL-23 p40 was negatively correlated with the expression of miRNA-10a in the intestinal mucosa of CD patients.After three times of IFX treatment,the expression of miR-10a in the intestinal mucosa of IBD patients significantly increased (t=3.341,3.382,both P<0.05).After stimulated with IFX in vitro,the expression of miRNA-10a in the intestinal mucosa significantly increased (t=3.095,7.193,both P<0.05).Conclusions miRNA-10a was closely correlated with the inflammation of IBD patients and with the role of targeting IL-12/IL-23 p40.miRNA-10a might be a new target for the IBD treatment.
