1.MiR-31 inhibit the invasion of breast cancer by Dock180

Purpose To investigate the correlation of Dock180 and miR-31 expression in breast cancer cells,and to observe the effect of miR-31 on the invasion of breast cancer cells by Dock180.Methods MiR-31 was transfected into breast cancer cells by liposome transfection technique.The actual binding site of miR-31 to the 3'-untranslated region of Dock180 was confirmed through luciferase assay.Western blot was performed to detect the expression of Dockl80 and other related proteins.Real-time PCR was used to measure the expression of Dock180.Matrigel invasion were performed to detect the invasion of breast cancer cell lines with miR-31 increased.Resuits The protein levels of Dock180 in breast cancer cell lines negatively correlated with miR-31 expression,and Dock180 was directly targeted by miR-31.Dock180 downregulation and miR-31 overexpression reduced breast cancer cells invasion.Conclusion Dock180 modulated by miR-31 plays an important function in breast cancer cell lines invasion.
2.Raptor induces migration and invasion of breast cancerthrough epithelial-mesenchymal transition

Aim To study the molecular mechanism of Raptor in the migration and invasion of breast cancer cells and provide the clinical theory basis for prevention of breast cancer invasion and metastasis.Methods Western blot was used to detect the expression of Raptor protein in MCF-7 cells and MDA231 cells.The siRNA plasmids were used to transfect MDA231 cells.At the same time,the plasmid pcDNA3.1-Raptor was transfected into MCF-7 breast cancer cells.And Western blot was used to analyze the protein expression level of E-cadherin and Vimentin.Transwell was used to test the ability of invasion.Nucleus mass separation experiment was used to test the expression of Snail.Results The expression of Raptor protein in MDA231 cells was higher than in the MCF-7 cell.When the control group of Scr/MDA231 cells compared with siRaptor/MDA231,Raptor protein expression was decreased obviously after plasmid transfection interference,accompanied by reduction of Vimentin protein expression and increase of E-cadherin protein expression.Compared with MCF-7/Con,Raptor protein expression significantly increased in MCF-7/Raptor,accompanied by increase of Vimentin protein expression and reduction of E-cadherin protein expression.The number of cells through the artificial basilemma Transwell in siRaptor/MDA231 cells was significantly reduced(P<0.05),and the number of cells through the Transwell chambers artificial basilemma was significantly increased in MCF-7/Raptor(P<0.05).Nucleus mass separation experiment results showed that the expression of Snail decreased obviously in the siRaptor/MDA231 than in the Scr/MDA231,however,the expression of Snail was obviously higher in the MCF-7/Raptor.Conclusions Raptor can promote the occurrence of EMT in breast cancer,thus it promotes invasion and metastasis of breast cancer.
3.Effect of exogenous insulin like growth factor 1 on postoperative cognitive function in aged rats

Objective To evaluate the effects of exogenous insulin like growth factor 1(IGF-1) on postoperative cognitive function in aged rats. Methods One hundred and twenty pathogen-free healthy male Sprague-Dawley rats, aged 18-20 months, weighing 500-600 g, were divided into 4 groups(n=30)using a random number table: control group(group C), sham operation group(group S), opera-tion group(group O)and exogenous IGF-1 group(group I). Splenectomy was performed, IGF-1 50 μg∕kg was subcutaneously injected at the same time point every day for 7 consecutive day starting from the end of surgery on the day of surgery in group I, and splenectomy was performed, and the equal volume of normal saline was given instead in group O. Morris water maze test was performed on 1, 3 and 7 days after surgery, and the escape latency and swimming distance were recorded. The rats were sacrificed after the end of Morris water maze test, and hippocampi were isolated for determination of the expression of amyloid-β(Aβ), amyloid precusor protein(APP)and β-secretase 1(BACE-1)using immunohistochemistry and Western blot methods. Results Compared with group C, the escape latency and swimming distance were significantly prolonged at 1, 3 and 7 days after surgery, and the expression of Aβ, BACE-1 and APP was up-regulated in O and I groups(P<005), and no significant change was found in the parameters mentioned above in group S(P>005). Compared with group O, the escape latency and swimming dis-tance were significantly shortened at 1, 3 and 7 days after surgery, and the expression of Aβ, BACE-1 and APP was down-regulated in group I(P<005). Conclusion Exogenous IGF-1 can improve postoperative cognitive function in aged rats.
4.Relationship between schizotypal personality traits and creativity in college students: mediating role of cognitive flexibility

ObjectiveTo explore the relationship between schizotypal personality traits and creativity in college students and the mediating role of cognitive flexibility. MethodsSchizotypal Personality Questionnaire (SPQ), Cognitive Flexibility Inventory (CFI) and Williams Creative Aptitude Test (WCAT) were used to assess 471 college students. Thereafter, Spearman correlation analysis was used to explore the relationship among the variables and the Bootstrap methodology was used to estimate the mediating role of cognitive flexibility. ResultsThe total SPQ, positive and disorganized schizotypal traits scores, and CFI score were all positively correlated with WCAT score (r=0.241~0.313, P<0.01). The total SPQ, positive and disorganized schizotypal traits scores were also positively correlated with CFI score (r=0.111~0.128, P<0.05). Cognitive flexibility mediated the relationship between positive schizotypal traits and creativity [indirect effect=0.052 (95% CI: 0.016~0.112, P<0.01), accounting for 11.93% of the total effect]. Cognitive flexibility mediated the relationship between disorganized schizotypal traits and creativity [indirect effect=0.075 (95% CI: 0.020~0.161, P<0.01), accounting for 11.50% of the total effect]. ConclusionSchizotypal personality has a direct impact on the creativity of medical students and also cause an indirect impact on their creativity through the mediating role of cognitive flexibility.
