1.Study on Immunoregulation by Method of Promoting Blood Flow and Dissipating Phlegm among Rats with Chronic Pelvic Inflammatory Disease

This study was aimed to explore the effect on immunological function of T lymphocyte subpopulation and erythrocyte by the method of promoting blood flow and dissipating phlegm of rat with chronic pelvic inflammatory dis-ease (PID). A mixture of bacteria combined with mechanical injury was used in the establishment of a total of 75 Wistar chronic PID rat models. The rats were divided into the normal group, sham-operation group, model group, Gui-Zhi Fu-Ling Capsule (GZFLC) group, Shao-Fu Zhu-Y u Capsule (SFZYC) group, Jin-Gang-Teng Capsule (JGTC) group, and the control group of GZFLC, SFZYC, and JGTC. Intragastric administration of each medication was given according to different groups. The percentage of amount of CD3+, CD4+, CD8+and CD4+CD25+regulatory T cell (Treg) in the spleen of rats was observed in each group. The RBC-C3bRR and RBC-ICR in the blood serum of rats were also observed. The results showed that medications used in all treatment group significantly increased the percentage of amount of CD3+, CD4+ and CD4/CD8 ratio (P< 0.05) and reduced the percentage of amount of CD8+(P< 0.05). GZFLC has significantly decreased the abnormal increased percentage of the amount of CD4+CD25+ Treg. GZFLC and SFZYC significantly increased RBC-C3bRR (P< 0.05) and decreased RBC-ICR (P< 0.05). It was con-cluded that the prescription of promoting blood flow and dissipating phlegm can treatment chronic PID by adjusting immunity of T lymphocyte subpopulation and erythrocyte.

Objective To investigate the possible mechanism of Gongqing Granule(Granule for Clearing the Uterus) on shortening the duration of vaginal bleeding after medical abortion.Methods The 120 unwanted pregnancy women within 49d of intrauterine gestation were randomly divided into three groups.Gongqing Granule combined with mifepristone and misoprostol was administered orally in Group A(treatment group).Qianzhi Capsule combined with mifepristone and misoprostol was administered orally to Group B(control group),and only mifepristone plus misoprostol was administered orally to Group C(blank group).Other 40 women(Group D,induced abortion group),were treated with vacuum aspiration for abortion.Chorionic villi and decidual tissue were collected and expression of Bax and Bcl-2 were detected by immunohistochemical method and Bax mRNA and Bcl-2 mRNA by situ hybridization.Results The expression of Bax,Bcl-2,Bax mRNA and Bcl-2 mRNA was found in all groups.The expression of Bax and Bax mRNA in the treatment group was higher than those in the control group,then the blank group,and then induced abortion group.While the expression of Bcl-2 and Bcl-2 mRNA of the treatment group was lower than those in the control group,then the blank group,and then induced abortion group(P
3.Application effect of family intervention model in early Alzheimer's disease patients' family

Objective To investigate the impact of Friedman family intervention model on the personal life ability and psychological status of caregivers of early Alzheimer's disease patients.Methods 66 Alzheimer's disease patients,according to the random sequence,were divided into the study group and the control group with 33 patients in each group respectively.The control group received routine geriatric care,the study group was given Friedman family intervention model on the basis of routine geriatric care.Two groups were estimated with Life Scale Index and ADL before intervention and two months after intervention respectively,and the matching caregiver were evaluated with Symptom Checklist-90 (SCL-90).Results Compared with the control group,the quality of daily life after the intervention of the study group was significantly improved,and the differences of daily living skills between two groups were statistically significant.The differences of the SCL-90 total score and depression,anxiety,somatization,interpersonal factor scores of caregivers between the two groups were statistically significant after two months of intervention.Conclusions The application of Friedman family intervention model plays a significant role in the improvement of the quality of daily life and daily living skills in patients with Alzheimer's disease,and can improve the psychological status of the caregivers of AD patients.
4.Establishment and assessment of a rat model of persistent intestinal mucosal injury induced by methotrexate

Objective To establish a rat model of persistent intestinal mucosal injury.Methods The rat model of persistent intestinal mucosal injury was induced by intraperitoneal injection of methotrexate.The food intake and body weight of all the rats were recorded.Pathological changes were observed using HE staining, the level of D-lactate and diam-ine oxidase in plasma, and myeloperoxidase and malondiadehyde in the intestinal tissue were measured by biochemistry. Results After modeling, the rat body weight and food intake were decreased.On day 4, the scores of mucosal damage, the levels of plasma D-lactate, DAO, MPO and MDA were significantly increased (P<0.05).On day 5, the intestinal damages of rats began to restore, and there was no significant difference among groups on day 6.The symptoms after the secondary injection were similar to those after the first injection, and the rats recovered gradually at day 12.Conclusions Intestinal mucosal injury in rats induced by 20 mg/kg MTX is an acute injury process, the course only lasts for 4-5 days. Intermittent injections twice of 10 mg/kg MTX can cause persistent intestinal mucosal injury in rats.This persistent injury model is more suitable for nutritional therapy evaluation in medium-and long-term studies of nutritional therapy.
5.Effects of peptide-based enteral nutrition on inflammatory response and immune function in intestinal mucositis rat

Objective To evaluate the effects of peptide-based enteral nutrition (PBEN) on inflammatory response and immune function in rats with intestinal mucositis.Methods 48 Sprague-Dawley male rats were randomly divided into 6 groups (all n =8):basal feed group (BFG),PBEN group (PBENG),intact protein enteral nutrition group (IPENG),methotrexate (MTX) + BFG,MTX + PBENG,and MTX + IPENG.The rats in MTX + BFG,MTX + PENG,and MTX + IPENG were intraperitoneal injected with MTX 10 mg/kg on day 0 and day 6 to induce sustained intestinal injury.From day 1,BFG and MTX + BFG were fed with basal feed,PBENG and MTX + PBENG with PBEN,IPENG and MTX + IPENG with IPEN.The daily energy intake of each rat was 1.80 kJ/g body weight.All the rats were sacrificed on day 11.The pathological changes of intestinal tissue were observed with HE staining,the levels of tissue myeloperoxidase (MPO),nitric oxide (NO),inducible nitric oxide synthase (iNOS),the thymus index and spleen gland index of intestinal tissue were measured using colorimetry,and the serum levels of immunoglobulins IgG,IgA,and IgM were determined with enzyme-linked immunosorbent assay.Results There were no significant differences among BFG,PBENG,and IPENG in each index.Serious injury of intestinal mucosa was observed in MTX groups.Significant differences were noted in all indexes between MTX + BFG and BFG (all P <0.05).The mucosal damage score (Chiu score) and the level of MPO and iNOS in MTX + PBENG were significantly lower than those in MTX + BFG [2.3 ± 0.69vs.2.96 ± 0.75,P =0.003 ; (2.30 ± 0.42) U/g tissue vs.(2.98 ± 0.23) U/g tissue,P =0.040 ; (0.37 ±0.06) U/mg prot vs.(0.44 ±0.10) U/rag prot,P =0.030] ; the serum levels of IgG and IgA were significantly higher than those in MTX + BFG (P =0.015,P =0.021) ; however,the levels of NO and IgM were not significantly different between the 2 groups (P =0.597,P =0.160).There were no statistically significant differences between MTX + IPENG and MTX + BFG in terms of the indexes (all P > 0.05).Conclusion PBEN can reduce the inflammation response and improve the immune function in intestinal mucositis rat.
6.Determination of Tetrodotoxin in Human Urine and Plasma after Immunoaffinity Column Clean-up Using Ultra-Performanc Hydrophilic Interaction Chromatography Coupled with Triple Quadrupole Mass Spectrometry

Anultra-performancehydrophilicinteractionliquidchromatography-triplequadrupolemass spectrometric ( UPLC-MS/MS) method was developed for the determination of tetrodotoxin ( TTX) in human urine and plasma. After the sample was cleaned-up and concentrated by immunoaffinity column, the separation of the TTX was carried out on an Acquity UPLC BEH amide column (100 mm×2. 1 mm, 1. 7 μm) with gradient elution using mobile phases of 0. 1% ( V/V) formic acid in water and acetonitrile. The analyte was detected by positive electrospray ionization mass spectrometry in the multiple reaction monitoring ( MRM) mode, and quantified by external solvent standard calibration. The measuring ranges of TTX in urine and plasma were 0. 05-400 μg/L. The average recoveries were 92%-95% and 91%-96% for TTX respectively spiked in urine and plasma with relative standard deviations of 3 . 3%-7 . 2% and 3 . 9%-7 . 8% ( n=5 ) . The limits of detection (LOD, S/N=3) and limits of quantitation (LOQ, S/N=10) of TTX were 0. 02 μg/L and 0. 05μg/L for urine and plasma, respectively. This method is suitable for the detection of TTX in urine and plasma for both forensic and clinical purposes.

8.The application of ultrasonography in evaluating cervical malignant lymph nodes

Background and purpose: Metastatic carcinoma and lymphoma are usually the causes of cervical malignant lymph nodes. This paper was aimed to investigate the different appearances of cervical malignant lymph nodes by 2 dimension and color doppler flow imaging (CDFI) and discuss the role of ultrasenography in diagnosing cervical malignant lymph nodes. Methods: Ninety-nine patients and 40 healthy people were selected as our subjects, among which 54 cases were diagnased as metastatic tumor and 45 cases as lymphoma according to pathology examination. With high frequency transducers, ultrasonography was used to document the shape of cervical lymph nodes, the ratio of long/short axis (L/S), types of echo and hilus of lymph node. CDFI features were also recorded and measured in order to analyze the parameters. Results: Compared with image of normal lymph nodes, the image of cervical malignant lymph nodes showed that the shape tended to be round and the ratio of L/S decreased. In addition, the majority of echo types of malignant lymph nodes was a low level echo and easily became fused. Meanwhile, the image of hilus of lymph node tended to disappear or shift. The lymph node of cases with metastatic tumor was shown to be either calcified or liquefied. The rate of blood flow in the artery was observed to be of high velocity and high resistive index (RI). Between metastatic tumor and lymphoma tumor case, there was significant difference in terms of ultrasonographic and CDFI features. All the above results showed that the features of malignant lymph nodes were significantly different from the normal ones. Conclusion: Ultrasonography could be regarded as one method of evaluating cervical malignant lymph nodes.
9.Imaging diagnosis of chondromyxoid fibroma

Objective To analyze the imaging manifestations of chondromyxoid fibroma,so as to improve diagnostic accuracy.Methods The X-ray,CT and MRI manifestations of 8 cases with chondromyxoid fibroma confirmed by surgical pathology were analyzed retrospectively.Results All of 8 cases were in the long tubular bones.X-ray showed oval,well defined,eccentric,radiolucent lesion with surrounding sclerosis.The thick dense bony or coarse reticular septation were seen in 6 cases.CT scan showed osteolytic,surrounding sclerosis,and 1 case with foci of calcification.MRI images showed a low or medium signal on T1WI,high signal on T2WI,1 case with a equal T1WI and slight high T2WI signal intensity peripheral rim around.Two cases with diffused enhancement on T1WI after intravenous injection of gapentetate acid meglumine injection and a peripheral liner enhancement in 1 case.Conclusion Chondromyxoid fibroma has certain imaging characteristics,but should be combined with X-ray,CT and MRI to differentiate from other bone tumors with similar imaging manifestations.
10.Low dosage sufentanil for blind nasotracheal intubation

Objective:To evaluate the effects of low dosage sufentanil used for blind nasotracheal intubation.Methods:Sixty cases for maxillofacial surgery were divided into 3 groups randomly with 20 in each group. Patients in groupⅠwere administered with fentanyl and midazolam by vein, those in groupⅡ with fentanyl and droperidol by vein,those in group Ⅲ with sufentanil at 0.1~0.2 ?g/kg. Intramuscular premedication of atropine-midazolam and blind nasotracheal intubation were performed in all cases after surface anesthesia for routine. Blood pressure(BP), heart rate (HR), mean artery pressure (MAP), blood oxygen saturation of pulse (SpO_2), intubation complication(IC), intubation achievement ratio(IAR), sedation score (Ramsay score), patient satisfaction(PS), and the incident rate of amnesia(IRA) in the three groups at T1 (before administering drug), T2 (after administering drug) and T3 (when tracheal tube was inserted into tracheal) were measured and observed. Results:In groups I and Ⅱ Ramsay score increased to 3~5, MAP and SpO_2 decreased (P0.05).Conclution:The low dose sufentanil can be applied for the sedation and analgesic before blind nasotracheal intubation.
