Objective To select a simple, stable and reliable mouse model of alcoholic liver disease. Methods The mouse models of alcoholic liver disease were induced by oral gavage ethanol or Lierber?DeCarli ethanol liquid diet for 8 weeks. The food intake and body weight were recorded. Pathological changes were examined using HE staining. Liver injury was assessed by the activities of serum ALT, AST, AKP and γ?GT, and serum and hepatic TC and TG. Results After modeling, both models showed significantly increased activities of serum ALT, AST, AKP, and contents of serum and hepatic TG (P<0?05), indicating the successful development of alcoholic steatohepatitis. However, oral ethanol gavage led to body weight loss and weak mental state. Ethanol liquid diet less affected the body weight and mental state. Ethanol liquid diet enhanced liver to?body weight ratio and serum TC, but oral gavage of ethanol did not. The changes of serum ALT, AST, serum and hepatic TG, and hepatic steatosis in the ethanol liquid diet models were more severe than those in the oral gavage ethanol models, suggesting that Lierber?DeCarli ethanol liquid diet led to more serious liver injury than oral gavage ethanol. Conclusions Lierber?DeCarli ethanol liquid diet model is better than oral gavage ethanol model, and is more suitable for studies on mechanisms and evaluation of hepato?protective drugs for alcoholic liver disease.

OBJECTIVETo investigate the effect of Trx-LHRH, a new GnRH crasis protein, on antibody production and male reproductive function.
METHODSTrx-LHRH produced in vitro with a new crasis gene which crasised Trx gene and GnRH gene together, was used as vaccine, and hydroalaminum base as adjuvant, in adult SD rats. After 5 weeks of the first treatment, the same dosage was used again to enhance the effect of vaccine. Antibody level was measured by ELISA, and androgen level by RIA.
RESULTSTrx-LHRH induced successfully the polycolonal antibody at the level of 1 :1 280 approximately 2 560 after 4 weeks of the first treatment, and 1 : 2 000 after 6 weeks of the enhanced treatment. Testosterone level was reduced significantly (P < 0.01) by ELISA, but there was reasonable variation among individuals. Sperm count was also reduced by Trx-LHRH treatment.
CONCLUSIONTrx-LHRH can be used as effective vaccine to induce antibody production, and at the same time, restrain the function of hypothatamas-pituitary-testis axis in vivo.

OBJECTIVETo study clinicopathologic features, immunohistochemical profile, diagnosis and differential diagnosis of childhood central nervous system primitive neuroectodermal tumors (CNS PNETs) with the features of ependymoblastoma and neuroblastoma.
METHODSThe clinical data, morphologic and immunohistochemical features were analyzed in 4 cases of pediatric CNS PNETs with features of ependymoblastoma and neuroblastoma. EnVision method immunohistochemistry was applied.
RESULTSFour patients including three boys and one girl presented at the age from 12 month to 4 years and three tumors located in cerebrum, one in brain stem. All tumors showed typical combined histological patterns of ependymoblastoma and neuroblastoma, demonstrating zones of true rosettes, occasional pseudovascular rosettes, and undifferentiated neuroepithelial cells in a prominent background of mature neuropils. There was focal expression of glial fibrillary acidic protein (GFAP) consistent with glial differentiation and epithelial membrane antigen (EMA) consistent with ependymal differentiation. Necrosis was seen in three cases and calcification was present in one case. Immunohistochemically, the rosettes and undifferentiated neuroepithelial cells were positive for vimentin, partially positive for GFAP and EMA but negative for synaptophysin. The tumor cells were also positive for synaptophysin in neuropils. The Ki-67 label index ranged from 20% to 60%.
CONCLUSIONSCNS PNETs with the features of ependymoblastoma and neuroblastoma is a rare tumor with poor prognosis. The tumor primarily occurs in childhood, especially infant and belongs to the family of embryonal tumors of the CNS. The morphologic, immunohistochemical and genetic features are important in differential diagnosis from other tumors of the CNS.

Chinese Medical Journal 2014;127(13):2475-2478
BACKGROUNDThe prevalence reported of maternal depression from the first to the third trimester was 7.4%, 12.8%, 12.0% respectively, which implies that around one-tenth of pregnant women suffer from psychological disorder during the whole pregnancy. It is assumed that during pregnancy the maternal-fetal circulation unit is also affected by maternal psychological status. The aim of this study is to explore the association between maternal psychological status and fetal dynamic blood flow circulation during pregnancy.
METHODSWe recruited 102 singleton low risk pregnant women between 30 to 33 gestational weeks. The Symptom Checklist-90 (SCL-90) was used to assess maternal psychological status. Ultrasound Doppler was used to measure blood flow changes in maternal-fetal circulation. Pulsatility index (PI) value was measured in umbilical artery (UA), fetal middle cerebral artery (MCA) and maternal uterine arteries (UTA). Peak systolic velocity (PSV) was measured in MCA and velocity was measured in umbilical vein (UV). Statistical analysis was performed with standard nonparametric Mann-Whitney U tests. Two-tailed P values <0.05 were considered statistical significance.
RESULTSOf the 102 pregnant women, 12 patients show high levels of poor mental health. Women with poor mental health have higher umbilical artery PI values than good mental health patients (P = 0.020). A higher MCA PI value is found in poor mental health patients but this does not reach statistical significance (P = 0.053). Women with hostility show lower placental scores (P = 0.030). Women with somatization demonstrate higher values in UA PI, MCA PI and MCA PSV (P = 0.049, 0.030 and 0.035 respectively). A higher MCA PSV value is also found in phobic anxiety patients (P = 0.046).
CONCLUSIONSPoor mental health during pregnancy is found to have an adverse effect on maternal-fetal circulation. Umbilical artery and fetal cerebral circulation are more sensitive and affected by maternal psychological disorder.

BACKGROUND:Previous brain studies have mostly focused on adults and fetuses, and the developmental characteristics of young children's brainstems have rarely been studied. OBJECTIVE:To observe the brainstem development characteristics of healthy young children and to explore the age-related differences and their correlation with sex. METHODS:From January 2019 to April 2022,a retrospective study of 3.0T MRI images of 174 children aged 2 to 6 years in the Affiliated Hospital of Inner Mongolia Medical University was conducted, and the median sagittal diameter, area and angle of the brainstem(including midbrain, pons and medulla oblongata)were measured. RESULTS AND CONCLUSION:There is an age-related increase in the anterior and posterior diameters of the midbrain, pons and medulla oblongata in the 2-5 years old group as well as in the longitudinal diameter and area of the midbrain, pons and medulla oblongata in the 2-6 years old group.Except for the longitudinal diameter of the medulla oblongata, all others show a positive correlation with age(r>0,P<0.05).In the 2-3 years old group and 4-5 years old group, the children are in the rapid growth and development stage, and these two age groups can be used as the key observation indicators for the development of young children. The anterior-posterior diameter, longitudinal diameter, area of the pons and total brainstem area are strongly correlated with age, which can be used as the key observation indicators for the brainstem development in young children.

【Objective】 To evaluate the ability of different disinfection methods to remove nucleic acid pollution in 2019-nCoV so as to obtain the best removal scheme. 【Methods】 2019-nCoV positive quality control nucleic acid of 50 μL was applied to plastic, metal and glass with medical cotton swabs, respectively. After drying, we dropped 50 μL of 750 mL/L alcohol (ethanol), chlorine-containing disinfectant (2 000 mg/L and 5 500 mg/L), and PCR Cleaner, respectively. After 1 min, the contaminated area was wiped with medical cotton swabs and soaked in 300 μL of pure water. After shaking and mixing, 5 μL was taken as a template. The Ct values of ORF1ab and N genes and IC genes of internal standard fragment in the amplified target area of 2019-nCoV after wiping with different disinfection methods were compared to evaluate the effect of eliminating nucleic acid pollution, and each experiment was repeated for three times. Similarly, the effects of ultraviolet irradiation for 0, 1, 2, 3, 4, and 5 hours on the removal of nucleic acid pollution were compared. 【Results】 After 2 000 mg/L and 5 500 mg/L chlorine-containing disinfectant wiped the contaminated area, the Ct values of ORF1ab and N genes and IC genes of internal standard fragment in the amplified target area in 2019-nCoV were all 0, and the Ct values of all genes in the contaminated area in groups 3, 4 and 5 h after UV irradiation were all 0, which completely cleared the pollution and had a strong effect. The effect of PCR Cleaner was second, and 750 mL/L ethanol was the worst. 【Conclusion】 2 000 mg/L and 5 500 mg/L chlorine-containing disinfectant and ultraviolet irradiation for 3 hours have the best effect of eliminating nucleic acid pollution, which is worth popularizing under appropriate conditions.

Objective:To investigate the association between metabolic syndrome and the risk of early renal function injury in chronic kidney disease(CKD) in the elderly. Methods:A retrospective cohort was established based on health check-up data of 4 495 elderly residents in Mengzi City, Yunnan Province from January 2016 to December 2018. The medial history, living habits, and related physical examination information were collected. Cox hazard regression model was used to explore the association between metabolic syndrome, along with its components, and the early renal function injury in CKD. Results:The median age of the elderly was 71.00(67.00, 75.00) years, with metabolic syndrome detection rate of 21.98%. Early renal function injury of CKD developed in 1 300(28.92%) subjects during the follow-up. Univariate Cox regression showed that the number of metabolic syndrome components was associated with the risk of early kidney development in CKD. The HRs were 1.23 (95% CI 1.03-1.47, P=0.022) with 1 component, 1.54 (95% CI 1.28-1.84, P<0.001) with 2, and 1.38 (95% CI 1.14-1.67, P<0.001) with 3 or more. Multivariate Cox regression showed that elevated fasting triglycerides( HR=1.20, 95% CI 1.07-1.36, P=0.003) and lower high density lipoprotein-cholesterol(HDL-C; HR=1.25, 95% CI 1.09-1.43, P=0.002) were risk factors for early kidney injury in CKD, while doing some physical activity( HR=0.57, 95% CI 0.33-0.98, P=0.042), or on daily basis( HR=0.57, 95% CI 0.49-0.66, P<0.001) was a protective factor for early kidney injury in CKD. Conclusion:The abnormality of one or more metabolic components can significantly increase the risk of early kidney injury in the elderly with CKD. Elevated triglyceride and decreased HDL-C may be the risk factors.
