Objective: To establish an experimental model of atherosclerosis in rats and to study the atherosclerotic lesions in the aorta, heart and liver. Methods: The rats in experimental groups were injected with a single dose of vitamin D (600 000 IU/kg) and loaded with high fat diet;control group was given saline and basic food. The pathological changes were observed in the aorta, heart and liver after 2,4,6 weeks. The scores were evaluated according to the pathological degrees. Results: No changes were observed after 2 weeks, but after 4 weeks atheroslerotic plaques were seen in the aorta and the score of the lesions were 0.50?0.39; a little lipid were found in coronary arteries;deposition of lipid was seen in the myocardium; many positive red pellets were found in the plasma of cells in the liver. After 6 weeks, more atheroslerotic plaques were observed in the aorta, and atheroslerotic plaques in coronary arteries were formed (pathological score 1.12?0.48); abundant positive red pellets were found in the plasma of the cardiac myocyte. The pathological changes occurred in rats were very similar to that of man. Conclusion: The experimental model of atherosclerosis in rat may be conveniently established by injection of vitamin D with loading of high fat diet, which can be used for the pathological and pharmacological study of atherosclerosis.
4. Effects of bisphenol A exposure during embryonic phase on development and neurobehavior of zebrafish
Journal of Shanghai Jiaotong University(Medical Science) 2019;39(5):458-462
Objective • To investigate the effects of bisphenol A (BPA) on the development and neurobehavior of zebrafish. Methods • The zebrafish fertilized eggs that developed to 4 hpf (hours post-fertilization) were treated with different concentrations of BPA (0, 2.5, 25, 250, and 2 500 μg/L). Survival rates, malformation rates, hatching rates, 24 hpf autonomic movement, and 48 hpf heart rates in the groups were observed and recorded. And the neurobehavioral test of larval zebrafish that developed to 144 hpf was performed. Results • The BPA exposure did not affect the survival rates, malformation rates and hatching rates of zebrafish. Compared with the control group, the autonomic movement in 24 hpf and heart rate in 48 hpf of zebrafish embryos that exposed to 2 500 μg/L BPA decreased (P=0.000), and the total distance and average speed of larval zebrafish that developed to 144 hpf also decreased (P=0.000). The behavior of larval zebrafish exposed to 250 μg/L and 2 500 μg/L BPA changed on light-evoked startle escape response activity, mainly including the decrease of total distance under strong light stimulation (P<0.05). Conclusion • BPA exposure during embryonic phase has developmental toxicity and neurotoxicity on zebrafish.
5. Significance of EphA2 receptor expression in human gliomas
Tumor 2007;27(8):646-650
Objective: To investigate the expression and significance of erythropoietin-producing hepatocellular A2 (EphA2), a receptor of tyrosine kinase in human gliomas and the correlation between EphA2 expression and tumor angiogenesis. Methods: Immunohistochemistry was used to examine the EphA2 expression in 66 surgically resected human glioma tissues, 2 human glioblastoma multiforme (GBM) cell lines(U251 and U87) and 10 normal brain tissues. Next, we performed Western blotting to further investigate the expression of EphA2 in high-grade GBM and normal brain tissues. Microvessels in the glioma tissues were counted after immunostaining for CD34, and the c orrelation between EphA2 expression and microvessel counts (MVCs) in glioma tissues were assessed. Results: Our immunohistochemical analyses demonstrated variable levels of EphA2 expression in 63 of 66 cases examined (63/66, 95.5%). But EphA2 expression was not detected in all 10 normal brain samples and 3 cases of low-grade gliomas (I-II). There was statistical difference in EphA2 expression between human gliomas and normal brain samples (P < 0.01). The positive rate of EphA2 expression was significantly higher in high-grade gliomas (WHO III-IV) than in low-grade gliomas (WHO I-II) (P < 0.01). The specific EphA2 staining was observed in both glioma tissues and U251 cells. The expression level of EphA2 protein was significantly lower in U87 cells than that in U251 cells. It was not detected in normal brain tissues. Microvessel density (MVD) significantly correlated with expression level of EphA2 protein (P <0.01). The MVD was higher in EphA2-strong positive tumor areas. Conclusion: EphA2 is specifically over-expressed in high-grade gliomas, but is not detected in normal brain tissues. EphA2 may be a novel marker of malignant glioma. It can be used as a molecular target for therapeutic intervention against high-grade glioma. EphA2 is involved in angiogenesis and plays an important role in the initiation and progression of glioma.

Many studies have shown that chronic inflammation occurs in the brain of patients with Alzheimer's disease (AD). It is well known that long-term administration of non-steroidal anti-inflammatory drugs (NSAIDs) can alleviate the cognitive decline of AD patient and elderly. Several inflammatory cytokines produced in the metabolism of arachidonic acid (AA) are closely related to inflammatory diseases. Lipoxygenases (LOXs) and cyclooxygenases (COXs) play a crucial role in the AA network, the products eicosanoids have an important impact on the progression of AD. Although there are many arguments and conflicting evidence, currently LOXs and COXs are still the hot topics in the research on AD pathogenesis and drug development. Here, we review the progress in research on COXs and LOXs, including their actions on CNS and their association with AD, and explore the feasibility of LOXs and COXs as targets for the drugs to prevent and/or treat AD.
