1.Advanced study on microRNAs involved in prevention and treatment of respiratory system diseases

Recently, along with the harmful factors in air in-creasing, the incidence of respiratory system diseases has been gradually rising. Therefore, more studies are focused on impro-ving the prevention and treatment on respiratory system diseases. Current researches have shown that microRNAs ( miRNA ) con-tribute to the prevention and treatment on respiratory system dis-eases, including virus infectious respiratory disease, pulmonary fibrosis, asthma, COPD, pulmonary tuberculosis and lung canc-er. The up-regulation and down-regulation of microRNAs could effectively regulate the biological process of these diseases. The review analyzes the recent years′study results of respiratory sys-tem diseases and microRNAs in depth, and discuss the role of microRNA in the treatment and prevention of respiratory disea-ses. Moreover, in this review we can provide new knowledge a-bout respiratory diseases and development of more potent drug targets for the treatment and prevention of respiratory diseases.
2.Nephroblastoma with distant metastasis in children:Clinical analysis of 9 cases

[Abstract ] Objective Nephroblastoma is one of the solid malignancies with fair prognosis in children .The lungs, liver, and bones are the common organs for distant metastasis of the tumor and important factors influencing the therapeutic effects as well .This study was to investigate the clinical characteristics and prognosis of nephroblastoma with distant metastasis in children . Methods We retrospectively analyzed the clinical characteristics , pathological features , and prognosis of 9 cases of nephroblastoma with distant metastasis treated in Nanjing Children Hospital from September 2011 to May 2015 . Results The patients presented with abdominal masses, distention and pain at preliminary diagnosis and confirmed with nephroblastoma , with pulmonary metastasis in 7 cases, liver metastasis in 1, and lung and liver metastases in the other .All were treated by surgery, radiotherapy, and chemotherapy and followed up for a median time of 34 months.By the end of the follow-up, 1 patient died of recurrence , 1 was undergoing chemotherapy , and the other 7 were being recovered, including complete disappearance of lung metastasis in 4, obvious reduction or calcification of lung metastasis in 2, evident disappearance of liver metastasis in 1, but relapse in none. Conclusion Lung and liver are common sites of nephroblastoma metastasis in children .As for the treatment of the disease , relatively good results can be achieved by radiotherapy , chemotherapy , and reoperation .
3.HPLC fingerprint of amino acids compounds of Xiasangju Granules

AIM:To establish the fingerprint chromatogram of amino acids compounds of Xiasangju Granules(Spica Prunellae,Flos Chrysanthemi indici,Folium mori.).METHODS:To apply 6-aminoqtiinoly-N-hydroxysuccinimdyl carbamate(AQC) pre-column derivatization HPLC method.Separation was performed on SYMMETRY C_(18)(150 mm?(4.6) mm,5 ?m) analytical column with mobile phase consisting of acetate(pH=(5.05)) and 60% acetonitrile with gradient elution with the flow rate(1.0) mL/min and the column temperature at 37 ℃.The Fluorescence wavelength used for detection was set at 250 nm(Excitation wavelength) and 395 nm(Emission wavelength) and the analysis time was 50 min.RESULTS:12 co-peaks on the HPLC fingerprints of Xiasangju Granules were indicated.The similarities were determined by the coefficients of cosine and correlation.The results of similarity analysis were(0.95)-(1.00).CONCLUSION:Perfect fingerprints were obtained which can be used for the quality control of Xiasangju Granules.
4.Fingerprint of Xiasangju Granules and its application in quality control

AIM: To establish an effective and convenient method for applying HPLC fingerprints to quality control in the production of Xiasangju Granules(Spica prunellae,Folium mori,Flos chrysanthemi). METHODS: Komasil Sunfrie C_(18)(150 mm?4.6 mm,5 ?m) analytical column was used and eluted with a gradient program consisted of phase A(1% acetic acid) and phase B(methanol) and detected at 290 nm.The fingerprints of aqueous extract,alcohol-precipitated extract,concentrate and finished product were compared with. RESULTS: The fingerprint method for Xiasangju Granules was established.The similarity among 10 batches of Xiasangju Granules was no less than 0.970.The difference between extracts and finished product of Xiasangju Granules was obvious. CONCLUSION: This validated method is available for quality evaluation and quality control in Xiasangju Granule's production.
5.Study on Quality Standards for Shenchang Injection

OBJECTIVE:To study the quality standards for Shenchang injection.METHODS:TLC was applied to identify the properties of Borneol and volatile oil of Rhizoma Acori Tatarinowii in the formula.HPLC was adopted to determine the content of ?-asarone in Shenchang injection.RESULTS:?-asarone showed good linear relationship in the range of 0.632 3~ 1.264 6? g.The average recovery of ammonium ?-asarone was 99.92%(RSD=0.91%).CONCLUSION:The standard can be used for the quality control of Shenchang injection.

8.Study on HPLC Fingerprint of Amino Acids Composition in Buzhong Yiqi Pill

Objective To establish the HPLC fingerprint of amino acids compounds from Buzhong Yiqi Pill.Methods A pre-column derivatization HPLC method was adopted with praline as a reference.Separation was performed on a AccQ? TagTM C18(3.9? 150 mm)analytical column with mobile phase consisting of 0.15 mol/L sodium acetate buffer solution and 60 % acetonitrile,gradient elution with the flow rate of 1.0 mL/min.The column temperature was at 37 ℃.The Fluorescence detecting wavelength was set at 250 nm(excitation wavelength)and 395 nm(emission wavelength),the analysis time was 35 min.Results Twelve common peaks on the HPLC fingerprints of Buzhong Yiqi Pill were presented.The similarity was determined by the correlation coefficient,included angle cosine coefficient and evaluation software of chromatogram fingerprint similarity.The results showed the similarity for 10 bathes of samples was in 0.97~ 1.00.Conclusion Good fingerprints were obtained,which can be used for the quality control of Buzhong Yiqi Pill.

Objective Primitive neuroectodermal tumor ( PNET) is a small round cell tumor occurring mostly in children or young adults and categorized into the Ewing sarcoma family of tumors, the purpose of the study was to investigate the clinical features, treatment and prognosis of Children PNET. Methods A retrospective study was performed on clinical data of 13 children with PNET hospitalized in our hospital from March 2010 to Octorber 2014.In order to analyze the clinical effects and prognostic results, statistical analysis was made on their clinical manifestations, CT and ultrasonic results, therapeutic schemes, postoperative pathology and immu-nohistochemical staining results, along with telephone and outpaitient follow-up of average 19 months. Results Of the 13 patients, 4 cases were central nervous system PNET(CNS-PNET) and 9 cases were peripheral PNET(pPNET).The clinical manifestations of the former were headache, vomiting, convolsion, movement disorder or decreased muscle strength, while the latter mainly showed mas-ses and abdominal distention.3 cases with metastasis to lung, lymph nodes and bones respectively showed masses on surface or in cavi-ty in imaging.Homer-Wright pseudorosette textures were found in 12 patients.Immunohistochemical results showed 11 cases with CD99(+), 9 cases with VIM(+) and 8 cases Syn( +).Among 11 patients underwent chemotherapy, 2 cases were lost to follow-up, 7 cases were in stable condition without occurrence and the other 2 cases recurred 4-6 months after chemotherapy.3 of 4 cases with CNS-PNET survived with the sequelae of decreased muscle strength, convolsion and movement disorder.1 case with CNS-PNET and 1 case with pPNET died 7 months and 16 months after diagnosis respectively. Conclusion PNET in children is a very highly malignant tumor with low long-term survival rate.Immunohistochemi-cal examination including CD99 and VIM is of important diagnostic value and CNS-PNET is prone to sequelae.
