1.Application of next generation sequencing in the study of hematologic neoplasms: reports from the 56th American Society of Hematology annual meeting

In comparison with the American Society of Hematology (ASH) annual meeting last year,more studies of hematologic malignancies using next generation sequencing (NGS) are published in the 56th ASH meeting this year.NGS was mainly used in the diagnosis and classification,risk stratification,MRD detection,clonal evolution,prognosis and therapy guidance.This short report will address the application of NGS in the study of hematologic malignancies by summarizing several representative publications from the 56th ASH meeting.

4.Detection and isolation of cancer stem cells in primary and metastatic carcinoma tissues of colorectal cancer

Purpose To detect and isolate cancer stem cells ( CSC) in fresh specimens of primary and metastatic carcinoma tissues from 10 cases of colorectal cancer to study the biological functions and mechanisms of migrating cancer stem cells. Methods Suspen-sions of primary and metastatic colorectal cancer cells were labeled with antibodies CD133-PE and CD44-FITC, and then flow cytometry was used to detect and isolate the CD133-PE-positive and CD44-FITC-positive cells. Results There were certain percentages of CD133-positive and CD44-positive cells present in primary and metastatic colorectal cancer and the proportion of CSC in metastatic colorectal cancer was higher than that in primary cancer tissue (P<0.05). Conclusions There are CSC in both primary and meta-static colorectal cancer and the proportion of CSC in metastatic colorectal cancer is significantly higher than that in primary cancer tis-sue, suggesting that CSC play an important role in the metastasis of colorectal cancer.
5. Effect of antidepressant puyu capsule on sexual l function of male rats and its mechanism

Aim To evaluate the effect of PuYu Cap-sule on sexual function in male rats in comparison to that of mirtazapine and SSRI antidepressant fluoxetine, so as to explore the pharmacological features of PuYu Capsule. Methods Sixty male rats were divided into control group, fluoxetine group, mirtazapine group, PuYu Capsule of high, medium and low dose group. After repeated administration of PuYu Capsule, the effects of the drugs on sexual function of male rats were verified by the mating behavior model and non-contact behavior model of rats. After repeated administration of PuYu Capsule, WAY100635 and buspirone were added respectively to verify the mating behavior model and non-contact behavior model. Furthermore, the effects of PuYu Capsule on rat androgen were verified by rat androgen ELISA kit after chronic administration. Re-suits In the mating behavior test and non-contact penile erection experiment of male rats, PuYu Capsule yielded no effect on the sexual function of male rats. WAY100635 and buspirone reversed the sexual dysfunction induced by fluoxetine, but had no effects on other medicines. None of the medications had any effect on androgen. Conclusions PuYu Capsule has no significant effect on androgen and sexual function of male rats, which is better than that of fluoxetine. The antidepressant mechanism of PuYu Capsule may be that it does not affect 5-HT.
6.Research progress of primary open angle glaucoma, 24h intraocular pressure and ocular perfusion pressure

Primary open angle glaucoma ( POAG) is one of the eye diseases which can lead to blindness. The early symptom of POAG is unconspicuous, however, it will result in irreversible optic never damage and visual field defect with the disease progress. As an index for diagnosing and evaluating the therapeutic effect of POAG, intraocular pressure ( IOP ) is simple and important. Clinically, although some treated patients have a target IOP at clinic time, the optic never damage still get worse. Researches indicate that higher nocturnal IOP and 24h IOP variation and lower nocturnal ocular perfusion pressure ( OPP) may be the reasons for this phenomenon. The following essay will give an overview of POAG,IOP and OPP according to the relevant literatures so that we can have a better understanding.
7.Expression of HMGB1 and the antagonistic effects of ethyl pyruvate on synovium in collagen-induced arthritis rats

Objective In this study,we elucidated the role of high mobility group box chromosomal protein 1 (HMGB1) in collagen-induced arthritis (CIA) rat and the antagonist role of ethyl pyruvate by using a rat model of CIA as the research object by comparing the expression of HMGB1 in normal control group,CIA model group and ethyl pyruvate group.Methods Thirty-six rats were randomly divided into 3 groups (n=12):normal control group,CIA group and ethyl pyruvate group.Then the 6 rats were dissected at the 6th,9th week respectively.Thc expression of HMGB1 was analyzed by immunohistochemistry and Pathology-image analysis software in the cytoplasma.The expression of HMGB1 mRNA with real time-polymerse chain reaction (PCR) was evaluate,and the HMGB1 expression of each group were compared with t-test.Results The immunohistochemical results of HMGB1 showed that the expression intensity in the normal control group,CIA model group and ethyl pyruvate group was 2.1±0.6,7.3±1.2,6.0±1.2 respectively at the 6th week; and 2.2±0.7,12.4±4.5,5.5±1.0 at the 9th week respectively.The HMGB1 mRNA real time-PCR results had shown that the relative quantification of the normal control group and CIA model group were 1,2.865,2.602respectively at the 6th week and 1.005,4.694,1.729 at the 9th week.At those two points, the HMGB1 expressions of HMGB1 antagonist group were significantly higher than those of the normal controls (P<0.05).In addition,there was statistical significant difference(P<0.05) in the HMGB1 expression when compared with the placebo group.Furthermore, when the degree of HMGB1 expression among the three groups was compared,the HMGB1 antagonist group was decreased significantly (P<0.05).Conclusion The results has demonstrated that HMGB1 could induce inflammation in the synovial tissue of CIA rats,and has provided the rationale that HMBG 1 could be the target of treating rheumatoid arthritis (RA).The results of this study have shown that ethyl pyruvate could antagonize the effect of HMGB1.This finding may provide a new therapeutic target for the treatment of RA.
9.Roles of ROS and TGF-?1 in aldosterone-induced production of PAI-1

Objective To explore the roles of reactive oxygen species(ROS) and TGF-?1 in aldosterone-induced PAI-1 production.Methods Quiescent rat mesangial cells (MCs) were treated by aldosterone.The level of ROS in MCs induced by aldosterone was measured by confecal laser scanning microscopy and the TGF-?1 activity in the supematant of culture was measured by mink lung epithelial cell (Mvllu) proliferation inhibition MTT assay.Then,before the addition of aldosterone,MCs were pretreated with NAC or TGF-?1 neutralizing antibody to decrease cellular ROS or inhibit activity of TGF-?1 induced by aldosterone respectively.PAI-1 mRNA was examined by semi-quantification RT-PCR and PAI-1 protein by Western blotting.Results The intracellular ROS induced by aldosterone increased by 5-fold compared to that of control group,and the activity of TGF-?1 stimulated by aldosterone increased markedly.TGF-?1 neutralizing antibody and NAC effectively decreased aldosterone-induced PAI-1 mRNA expression by 30% and 32%,and PAI-1 protein expression by 21% and 11%,respectively.However,neither TGF-?1 neutralizing antibody nor NAC alone could regulate aldosterone-induced PAI-1 mRNA and protein expression to normal level in 24 hours.Conclusions ROS and TGF-?1 play important roles in up-regulation of aldosterone- induced PAI-1 in MCs.ROS and TGF-?1 are not the exclusive pathway of PAI-1 expression induced by aldosterone in MCs.
10.Paraspinal elicited somatosensory evoked potentials in the detection of spinal cord lesions

Objective To explore the role of somatosensory evoked potentials elicited by paraspinal stimulation in the detection of spinal cord lesions.Methods Patients with clinically suspected spinal cord lesions underwent somatosensory evoked potentials(SEPs)and spinal cord conduction velocity(SCCV)test using paraspinal stimulating method.96 patients aged 15 to 75 years old with suspected spinal cord lesions of various aetiologies were tested.36 patients had demyelinating disease of the spinal cord,26 had sub-acute combined degeneration of the spinal cord,19 had myelopathy,10 had acute myelitis,5 had spinal cord corhpression.Results Paraspinal stimulation elicited somatosensory evoked potentials were Performed on all 96 patients,68 of whom underwent spinal MRI as well.SEPs and SCCV were found abnormal in a high ratio in all kinds of spinal cord lesions ; in general,78 among the 96 patients had abnormal SEPs with a sensitivity of 81.25%.27 out of 36 with demyelinating disease of the spine had abnormal SEPs with a sensitivity of 75.00%,23 out of 26 with subacute combined degeneration of the spinal cord had abnormal SEPs with a sensitivity of 88.46%,8 out of 10 with acute myelitis had abnormal SEPs,15 patients with myelopathy having abnormal findings,all 5 patients with spinal cord compression had abnormal SEPs.42 of 68 patients undergoing MRI revealed to be abnormal,in which 35 patients also had abnormal SEPs.The other 26 patients had normal spinal MRI,in which 21 patients had abnormal SEPs.Conclusions Paraspinal stimulation somatosensory evoked potentials and spinal cord conduction velocity may objectively document the abnormalities of electrophysiology,which occurs earlier than those of anatomy and radiological finds, therefore it may detect the dysfunction of spinal cord at an early stage.It is even more useful in the detecting of the metabolic myelopathy,which can hardly be detected by MRI.This technique is simple,inexpensive, and maybe useful in the diagnosis for patients with suspected spinal cord lesions.
