OBJECTIVETo explore the feasibility of the transdiferentiation of human breast adipose-derived stem cells (hbASCs) into mammary epithelial-like cells after co-culturing in Transwell in vitro.
METHODSThe third passage hbASC and the HBL-100 cell line were co-cultured in a Transwell culture system for 15 days. The hbASCs were observed and identified by inverted phase contrast microscope and transmission electron microscopy, and immunocytochemistry staining in the induced and control groups.
RESULTSBoth the third passage hbASCs and the HBL-100 cell line cells could adhere and grow rapidly after co-culture in the Transwell system. After co-culture for 15 days, the morphology of some induced hbASCs changed into epithelial-like cells. Some induced hbASCs showed positive expression of CK18, CK19 by immunocytochemistry staining, and typical epithelium cells with microvilli, desmosomes and tonofilaments observed under TEM. The positive rate of CK18 and CK19 was (24.4 +/- 12.0)% and (21.6 +/- 16.4)% in experimental group, and (1.8 +/- 1.7)% and (1.1 +/- 0.6)% in control group.
CONCLUSIONThe data suggests that hbASCs may have the potential to transdifferentiate into human mammary epithelial-like cells after co-culturing in Transwell in vitro.

Objective To measure serum and tissue levels of soluble syndecan-1 (SDC1) in patients with cutaneous squamous cell carcinoma (CSCC),and to explore the relationship between the expression of SDC1 and clinicopathologic features of CSCC as well as between the serum and tissue levels of SDC 1.Methods An immunohistochemical study was performed to measure SDC 1 expression in the epidermis of lesional specimens from 93 patients with CSCC and normal skin specimens from 30 healthy human controls, and enzyme-linked immunosorbent assay (ELISA) to detect serum levels of soluble SDC1 in 81 patients with CSCC and 30 healthy human controls. Results The expression of SDC1 was significantly lower in CSCC tissues than in normal skin tissues (Z=3.85,P< 0.01).The expression intensity of SDC 1 decreased with the increase in tumor thickness but with the decrease in degree of differentiation of CSCC (x2 =11.66,12.51 respectively, both P < 0.01).Furthermore,the expression of SDC1 was significantly lower in lesional tissues of CSCC with lymph node metastasis than in those without (Z =2.20,P < 0.05).As ELISA showed, serum levels of soluble SDC1 were significantly higher in patients with CSCC than in healthy controls (Z=4.12,P< 0.01),and gradually increased with the increase in tumor thickness and with advancing clinical stages of CSCC.In addition, serum levels of SDC1 were significantly up-regulated in patients with invasive CSCC compared with those with CSCC in situ (Z =3.02,P < 0.01),but were not significantly different among patients with invasive CSCC at different degrees of differentiation (P > 0.05).CSCC patients with lymphatic metastasis showed significantly higher serum levels of SDC1 compared with those without (Z =5.30,P < 0.01).The serum levels of soluble SDC1 were significantly negatively correlated with its tissue levels in CSCC patients (rs =-0.625,P < 0.01).Receiver operating characteristic (ROC) curve analysis showed that the best cutoff point of serum SDC 1 levels was 65.5 μg/L for the diagnosis of lymphatic metastasis, with the sensitivity, specificity and area under the curve (AUC) being 73.7%,87.1% and 0.904 (0.840-0.968) respectively. Conclusion The downregulated tissue expression but up-regulated serum levels of SDC 1 may be associated with the invasiveness of CSCC,and elevated serum SDC 1 levels are somewhat valuable to the diagnosis of lymphatic metastasis.

Objective:To explore the effect of mild cognitive impairment on the subjective well-being of the elderly and the mediating role of self-efficacy in it. To provide an effective basis for health management and clinical intervention in the elderly with mild cognitive impairment. Methods:A total of 164 patients with mild cognitive impairment from December 2018 to June 2019 were surveyed by the General Self-efficacy Scale, the Montreal Cognitive Assessment Scale, and the Memorial University of Newfoundland Scale of Happiness Scale. Results:The overall score of self-efficacy was 21.41±3.98, and subjective well-being was 26.44±7.35. There was a positive correlation among cognitive function, self-efficacy and subjective well-being, except that there was no significant correlation between self-efficacy and cognitive function( r values were 0.263-0.687, P<0.01). Self-efficacy played a part in mediating between cognitive function and subjective well-being in patients with mild cognitive impairment, accounting for 37.8% of the total effect. Conclusions:Mild cognitive impairment can also have a negative impact on the subjective well-being of the elderly. Among them, self-efficacy plays a particularly important role in mediating. Interventions about self-efficacy on the elderly with mild cognitive impairment may have more significant effects.

Journal of Biomedical Engineering 2008;25(4):934-949
It is important for cardiac therapy devices such as the automated external defibrillator to discriminate different cardiac disorders based on Electrocardiogram analysis. A phase space analysis based algorithm is proposed to detect cardiac arrhythmias effectively. Firstly, the phase space of the signal is reconstructed. Then from the viewpoint of geometry and information theory, the distribution entropy of the point density in the two-dimensional reconstructed phase space is calculated as the features in the further classification. Finally the nearest-neighbour method based on Mahalanobis distance is used to classify the sinus rhythm (SR), supraventricular tachyarrhythmia (SVTA), atrial flutter (AFL) and atrial fibrillation (AF). To evaluate the sensitivity, specificity and accuracy of this proposed method in the cardiac arrhythmias classification, the MIT-BIH arrhythmias database and the canine endocardial database are studied respectively. Experiment results demonstrate that the proposed method can detect SR, SVTA, AFL and AF signals rapidly and accurately with the simple computation. It promises to find application in automated devices for cardiac arrhythmias therapy.
