Objective: Accumulating evidence shows that hypertension is a chronic low-grade inflammatory disease, and inflammatory reaction is involved in the formation and progress of target-organ damage in hypertension. The authors aimed to explore the changes in the serum levels of interleukin-6(IL-6),soluble CD40 ligand(sCD40L),metalloproteinase-9(MMP-9),tissue inhibitor of metalloproteinase-1(TIMP-1),lipid profile and left ventricular structure in patients with essential hypertension. Methods: The serum levels of IL-6,sCD40L,MMP-9 and TIMP-1,along with blood lipids were determined and echocardiography was performed to evaluate the cardiac structure and function of 158 hypertension patients and 22 age-and sex-matched controls. Results: The hypertension patients exhibited higher levels of IL-6(P

Objective To investigate the efficacy of medial plantar flaps with cutaneous nerves on fixing palm skin defects for patients injured while working. Methods All cases with palm skin defects who received medial plantar flaps grafting from June 2003 to January 2013,were analyzed retrospectively. The data included 13 males and 10 females, with the mean age of 36 years old (range,18-61 years old). There were 16 cases with right hand injury and 7 cases with left, whose skin defects ranged from 3.6 cm×2.8 cm to 7.0 cru×5.5 cm.All cases had skin and soft tissue injury while 5 cases with vascular and nerve injury,3 cases with tendon involvement and being exposed,6 cases with open metacarpal fractures.15 patients were treated with emergency surgery within 8 hours after injury, while 8 patients were treated 10 days after debridement surgery. Results Altogether,23 patients who received medial plantar flaps grafting operations, with palm skin defects, were included in this study. All cases were followed up for 3 to 36 months with a average period of 18 month. The grafted flaps range from 4.6 cm×3.8 cm to 8.0 cm×6.5 cm.Among the patients,19 of them recovered excellently with healing by first intention, with 7-14 d healing time,fully-retained hand functions and palm appearance, and 4-8 mm for between two points distinguish test.Flaps survived fairly well in 3 patients because partial necrosis happened at plantar skin graft donor site. After being debrided and dressed, the wound healed at second phase with a healing time of 12-21 d and 5-9 mm for between two points distinguish test. The operated hands could flex and extend functionally in the follow-up period.Only one patient did not recovered from medial plantar flap grafting and the flap did not survive operationally. The patient recovered after another operation with groin skin grafting later.According to Gu Yudong et al' evaluation criteria, such as improved hand function,19 cases were scored as excellent recovery,3 cases as good, and 1 as poor. The excellent rate was 95.7% (22/23).Conclusion Medial plantar flaps with cutaneous nerves are ideal flaps for palm skin defects restoration, which characterized by the advantages of fixed anatomical position, high survival rate and being good to the functional recovery and palm appearance.

Objective To investigate the correlation between the measurement methods of carotidcerebral pulse wave velocity (ccPWV) and the traditional method of brachial-ankle pulse wave velocity (baPWV).Methods A total of 136 healthy volunteers were divided into a youth group (20-39 years),a middleaged group (40-59 years),and an elderly group (more than 60 years) according to their ages. While detecting baPWV,transcranial Doppler ultrasound was used to simultaneously monitor the ipsilateral common carotid artery and the terminal segment of internal carotid artery. The time differences of the beating points of their cardiac cycles and the distanceses of the skin surfaces between the 2 probes were measured and ccPWV was calculated. Results The systolic blood pressure, pulse pressure and mean arterial pressure in the elderly group were signifificantly higher than those in the middle-aged group and the young group. The ccPWVs in the youth,middle-aged and elderly groups were 418 ± 52 cm/s,489 ±54 cm/s,and 599 ± 58 cm/s,respectively.The elderly group was significantly faster than the middle-aged group (t =7.308,P <0.001),and the middle-aged group was significantly faster than the youth group (t=6.758,P<0.001).A Pearson correlation analysis showed that ccPWV was significantly positively associated with the age (r=0.847,P<0.001) and baPWV (r =0.548,P <0.001).The multiple linear regression analysis showed that ccPWV was significantly positively associated with the age and diastolic blood pressure (partial correlation coefficients were 0.742 and 0.293respectively,P <0.001 and <0.010 respectively).Conchlusions ccPWV is a new measurement method for cerebrovascular stiffneas, and it has a good correlation with the traditional measurement method.

Objective:In this study,a series of experiments were conducted to research the mechanism of anticancer and preliminary molecular effects of PAMs on the HEPG-2 cancer cells. Methods:Morphological observation and MTT assay were used to explore the inhibition and killing effect of PAMs acting on HEPG-2.AO/EB staining and Annexin V-FITC/PI staining were employed to observe the apoptosis of HEPG-2 treated with PAMs.The expression level of Foxm1,bcl-2 and others genes in HEPG-2 cells were detected by using qRT-PCR and western blot.Wound healing and transwell experiments determined if PAMs can inhibit the migration of HEPG-2.Results:PAMs can inhibit and kill HEPG-2 cells in time and dose-dependent manners, and the cytotoxic effects were closely related to the cell apoptosis. The mRNA expression of foxm1,bcl-2 and surviving gene were remarkably decreased in HEPG-2 cells after the treatment of PAMs.PAMs decreased the FoXM1 protein expression in HEPG-2 cells, while up-regulating thep53 protein expression.,and it could also inhibit the migration of cancer cells. Conclusions:The possible molecular mechanism for the killing of HEPG-2 cancer cells by PAMs was proposed.By down-regulating the expression of foxm1 and up-regulating the expression of p53,the transcriptional expression of their downstream target genes survivin and bcl-2 was inhibited or reduced, hence enhancing the cancer cell apoptosis. This study provides an important foundation for the development of anti-cancer Chinese folk medicine based on PAMs.

In this study, the leukemia K562 cell line was used as a model to elucidate the anticancer effects and preliminary mechanisms of PAMs. MTT assay showed that PAMs could cause cytotoxicities in K562 cells in dose- and time-dependent manners. AO-EB, Annexin-FITC/PI staining showed that the killing effects of PAMs in K562 cells were related to apoptosis, which was further confirmed by the following molecular and enzymatic assay. The mRNA levels of pro-apoptotic genes caspase-3, caspase-9 and bax were remarkably increased while the anti-apoptotic gene bcl-2 was significantly decreased determined by fluorescent quantitative PCR. Western blotting disclosed that PAMs could up-regulate caspase-3 and down-regulate anti-apoptotic survivin protein expression. The latter was also consistent with the results that PAMs could increase the enzymatic activities of both caspase-3 and caspase-9. All these results suggested that PAMs could effectively inhibit the proliferation of K562 cells and the mechanisms may be closely related to apoptosis induction. The work provides evidence basis for PAMs to be potentially developed as anti-cancer leukemia Chinese medicine.
