1.Clinical Study on the Treatment of Non-alcoholic Fatty Liver Disease with Pingganjian Decoction:A Report of 54 Cases

Objective To evaluate the effect of Pingganjian Decoction (Decoction for liver diseases) on the non-alcoholic fatty liver diseases (NAFLD).Methods Totally 105 NAFLD patients were randomized into a treatment group (54 cases) and a control group (51 cases).The former was treated with Pingganjian Decoction while the latter treated with Fufang Yiganling Tablets and Zhibituo Tablets for 3 months.Changes in the symptoms scores,body mass index (BMI),waist-hip ratio (WHR),CT ratio of liver-spleen,homeostasis model assessment insulin resistance (HOMA-IR),liver functions,uric acid and blood-fat before and after treatment were observed.Results After treatment,in the treatment group,the symptom score was significantly decreased,CT ratio of liver-spleen was significantly increased,and the level of HOMA-IR was decreased.Compared with those of the control group,the difference was significant (P
2.Endoscopic sinus surgery combined with mitomycin in nasal inverted papillom

ObjectiveTo explore the effect of endoscopic sinus surgery combined with mitomycin in nasal inverted papilloma (NIP). MethodThe clinical data of 75 patients with NIP was retrospectively analyzed and divided into treatment group (endoscopic sinus surgery combined with mitomycin treatment, 38 cases) and control group (endoscopic sinus surgery treatment, 37 cases) by random digits table. Results There was no complication in treatment group, the symptoms disappeared after treatment, good recovery and surgery epithelization was found after follow-up, 1 case of grade Ⅲ recurred at 3 months after treatment and followed up for 5 years without recurrence. In control group, 37 cases of conventional dressing endoscopic treatment, recurrence within 1 year in 3 cases, 4 cases of recurrence in the second year, 5 cases of reoperation cured, 2 cases of malignant transformation. The efficacy was no statistically significant between two groups( X2 =9.86,P < 0.05). ConclusionEndoscopic sinus surgery combined with mitomycin in treatment of NIP, intraoperative and postoperative adjuvant therapy given to the local mitomycin, inhibit their growth and development, can inhibit the NIP recurrence and improve the cure rate.
3.Application of hospital teaching rounds in clinical teaching practice in hospitals for infectious diseases

Great difficulties were existed in clinical teaching in hospitals for infectious diseases. Therefore,a centralized hospital teaching rounds,a new clinical teaching practice was taken in a hospital for infectious diseases. During seven years' practice,a basic mode of centralized hospital teaching rounds was established,which contains many aspects such as organizational form,time arrangement,case se-lection and operation procedures. Its effectiveness was evaluated accoding to the following aspects:number of people attending hospital teaching rounds,composition of these people,satisfaction of these people, comparison of ward rounds data recorded nowadays and 7 years before and assembling all the rounds records into a book. Results indicated that this measure overcame the limitations of the specialized hospi-tals to some extent and played an important role in improving clinical teaching quality.
4.Association between dietary patterns and metabolic syndrome among elderly people in Zhejiang province

Objective To understand the relationship between the dietary patterns and metabolic syndrome among elderly people in Zhejiang province,in order to provide scientific basis and reference for nutrition interventions.Methods Data were obtained from the 2010-2012 Chinese National Nutrition and Health Survey,and factor analysis was used to obtain food patterns among elderly people in Zhejiang province.The Logistic regression was used to explore the association of the food patterns with metabolic syndrome and its components.Results The intakes of bean products,nuts,fruits,eggs,and milk in elderly people were generally low,and the intakes of livestock and poultry meat,cooking oil and salt were relatively high.5 dietary patterns were derived by factor analysis:high-quality protein diet,mediterranean diet,condiments diet,traditional dietary pattern and western dietary pattern.The medical test results in 780 elderly people showed that the standardized prevalence of metabolic syndrome in the elderly was 28.8%,standardized central obesity rate was 41.2%,standardized hypertension rate was 50.3% and standardized hyperglycemia rate was 39.5%.Logistic regression analysis showed that different dietary patterns had different effects on the metabolic syndrome and its components.Conclusions The prevalence rate of metabolic syndrome and the abnormal rates of its components are high in the elderly in Zhejiang.The intakes of fruits and milk are generally low,and intakes of livestock and poultry meat,cooking oil and salt are relatively high.The relationship between dietary patterns and MS or its components is complicated,so it is necessary to guide the elderly people to adopt reasonable diet by referring to the Diet Pagoda in order to reduce the risk of MS or its components.
5.ROS-EGFR-JNK-AP-1 signaling pathway involves in angiotensin Ⅱ-induced human mesangial cells proliferation

Objective To elucidate whether Ang Ⅱ indnces the proliferation of mesangial cells through ROS-EGFR-JNK-AP-1 signaling pathway. Methods The incorporation of 3H-thymidine (3H-TdR) and cell count were used to measure mesangial cell (MC) proliferation. ROS production was determined by DCFDA fluorescence. EGFR and JNK activation was assayed by Western blot. Results Ang Ⅱ significantly enhanced ROS production in mesangial cells, which was up-regulated by 2.26 folds of control group after incubation with Ang Ⅱ for 60 min. Ang Ⅱ induced EGFR phosphorylation in dose- and time-dependent manner, with the peak (3.96 folds increase) at 30 min. EGFR phosphorylation was significantly blocked by AT1R antagonist losartan, antioxidant NAC, and NADPH oxidase inhibitor apocynin and DPI. EGFR antagonist AG1478 significantly inhibited Ang Ⅱ-induced mcsangial cell proliferation. Losartan, NAC, apocynin, DPI, and AG1478 ahnost abolished Ang Ⅱ-induced JNK activation. Conclusions ROS-EGFR-JNK-AP-1 signaling pathway is involved in Ang Ⅱ-induced mesangial cell proliferation. Apocynin and AG 1478 may be used as new therapy.
6.Construction and functional characterization of alentiviralvector mediating RNA interference of DEK gene

Objective To construct the lentiviral vector of RNA interference(RNAi) for DEK,and to detect its effect on breast cancer cell growth.Methods The DEK siRNA was designed and constructed based on DEK sequence using a lentiviral vector.The lentivirul vector containing DEK siRNA was named PSIH-H1-DEK as confirmed by PCR and sequenceing.PSIH-H1-DEK was then packaged with accessory plasmids into lentivirus in 293T cells and selected for 2 weeks with puromycin ( puro ) before the mixed colonies stably expressing DEK siRNA were obtained and the DEK expression was detected by real time PCR( RT-PCR) and Western blotting.The effect of DEK siRNA on ZR75-1 cell growth was determined by cell counting kit.Results Western blot and RT-PCR showed that PSIH-H1-DEK siRNA could suppress DEK gene expression.Suppression of DEK could markedly inhibit the growth of ZR75-1 cells.Conclusion The lentivirus-mediated DEK siRNA is obtained,which will facilitate further research on DEK function in breast cancer development.
7.Influence of vitrified versus fresh embryo transplantation on the growth and development of mouse offspring

Objective Assisted reproductive technology ( ART) is associated with an increased incidence of offspring de-fects.The aim of this study was to investigate the impact of embryo vitrification in in vitro fertilization ( IVF) on the growth and devel-opment of the offspring by comparing the development status of mouse offspring from fresh embryo versus vitrified embryo transplanta-tion. Methods This study included three groups of mouse offspring , from natural fertilization ( n=12 ) , fresh embryo transplanta-tion (FET, n=9), and vitrified embryo transplantation (VET, n=9).We compared the body weight and body fat content in the sex-ually mature stage among the three groups of mice . Results No obvious differences were found in the morphology of the embryos be-tween the FET and VET groups .The body weight of the mouse offspring was significantly higher in the VET than in the natural control group at 0 postnatal (PN) week (P<0.01) and 12 PN weeks (P<0.01), and so was it in the FET group than in the control at 0 PN week (P<0.01) and 6 PN weeks (P<0.05).The body fat content at sexual maturity was remarkably higher in the VET than in the FET and control groups ([9.32 ±4.34] vs [5.24 ±2.56] and [2.80 ± 0.48] g, P<0.05), and so was the muscle content in the VET than in the control ([33.77 ±4.22] vs [27.50 ±1.79] g, P<0.01). Conclusion In IVF, fresh embryo transplantation has less interference with the growth and development of mouse offspring than vitri-fied embryo transplantation .
8.Expression of cyclooxygenase-2 in IL-1?-stimulated mesangial cells is medicated by NF-?B/I?B signal pathway

AIM: To investigate the role of NF-?B/I?B signal pathway in the regulation of (cyclooxygenase-2) (COX-2) expression in human mesangial cells (HMC). METHODS: The PGE_2 concentration in supernatants of HMC was measured by radioimmunoassay. COX-2 mRNA and protein expression were determined by RT-PCR and Western blot. Electrophoretic mobility shift assay (EMSA) and Western blot were used to detect the activity of NF-?B and degradation of I?B. RESULTS: IL-1? significantly upregulated COX-2 expression and PGE_2 production in HMC. Significant up-regulation of NF-?B activation, nuclear translocation of p65 subunit, and degradation of I?B ? and I?B ? were observed in IL-1?-induced HMC. CONCLUSION: Expression of COX-2 in IL-1?-induced HMC is mediated by NF-?B/I?B signal pathway. [

Objective To construct eukaryotic expressing vector of mouse soluble CD160 and stably transfect into CHO cells for eukaryotic expression.Methods Recombinant soluble CD160(rsCD160) was constructed by gene recombination.Total RNA was extracted from the spleen of C57BL/6 mice.cDNA was amplified for the soluble form of CD160.Then,the PCR product was cloned tO pcDNA3.1 and pEGFP-N1.The recombinant plasmid was identified by restriction map and sequence analy-sis.The soluble CDl 60 expression in CHO cells transfected with recombinant psCDl 60 was verified by RT-PCR and Westernblot.The binding ability of psCD160 tO its ligand was detected by FACS.Results 520 bp mouse soluble CD160 gene was obtained.Recombinant mouse psCD160 was successfully constructed.After transfection,soluble CD160 expression in the culture supernatant of CHO cells was successfully detected.FACS analysis indicated that soluble CD160 could bind tO its ligand.Conclusion Recombinant mouse psCD160 is successfully constructed,which will benefit our further study on soluble CD160 for immune therapy against tumor in the future experiments.
10.Suppression of EphB4 improves the inhibitory effect of mTOR shRNA on the biological behaviors of ovarian cancer cells by down-regulating Akt phosphorylation.

The aim of the present study was to examine the effects of suppression of EphB4 and/or mTOR on the biological behaviors of ovarian cancer cells, and the potential regulatory pathways. Antisense EphB4 vectors and shRNA vectors targeting mammalian target of rapamycin (mTOR) were constructed and transfected into A2780 and SKOV3 cells (two ovarian cancer cell lines). The effects of the antisense EphB4 vectors and the shRNA vectors on the proliferation, apoptosis and invasion of ovarian cancer cells were measured, and the expression of EphB4, mTOR and Akt detected. The results showed that transfection with mTOR shRNA could inhibit growth, induce apoptosis, and reduce invasive ability of ovarian cancer cells, which was accompanied by downregulation of EphB4, mTOR and Akt. The inhibitory effects on cell growth caused by mTOR shRNA alone were weaker than those by antisense pEGFP-C1-EphB4. In the antisense pEGFP-C1-EphB4-transfected cells, it was found that EphB4 knockdown could decrease the mTOR expression and slightly reduce the Akt phosphorylation. Significant suppressive effects on cell growth were observed in cells co-transfected with antisense pEGFP-C1-EphB4 and mTOR shRNA. In co-transfection group, the expression levels of EphB4, mTOR and Akt were distinctly lower than those in other groups. It was concluded that suppression of EphB4 may inhibit the growth of ovarian cancer cells by downregulation of the PI3K/Akt/mTOR pathway, and reverse Akt phosphorylation induced by mTOR shRNA. Inhibition of EphB4 and mTOR combined may cooperatively suppress the biological behaviors of ovarian cancer cells.
