1.Study on HBV DNA “a” deteminant mutation in viral carrier children following universal HB vaccination

Objective To investigate hepatitis B virus mutation rate after the universal immunization in China. Methods DNA sequence analysis of the “a” determinant was performed on sera from 72 viral carrier children after immunoprophylaxis selected through a sampling survery. Results Four mutants were detected, the mutation rate was 5.56%. One strain showed Gly-to-Arg mutation at the 145 codon of surface antigen. Animo acid substitutions at the amino acid 126 and 140 were noted in the other three children. The epidemiological HBV serotype shifted from previous adw infection to more recent adr was also observed. Conclusions These observations indicate that vaccination is probably the cause of mutation, whereas the mutation is low and it will certainly not be a reason for doubting that the vaccination is the method of main choice for preventing hepatitis B infection that still devastates the developing countries.
2.THE THERAPEUTIC EFFECTS OF RECOMBINANT HUMANIZED ANTI-HER2 ANTIBODY (HERCEPTIN) ON HER-2/ NEU-OVEREXPRESSING METASTATIC IRJMAN BREAST CANCER

To investigate the therapeutic effects of recombinant humanized anti-HER2 antibody (Herceptin) on Her-2/neu-overexpressing metastatic human breast cancer, 7 patients with Her-2/neu-overexpressing metastatic human breast cancer were involved in this study. The results indicated that the therapeutic effective rate was 71. 4%. CR. PR, SD. PI) were seen in 2. 3, 1, 1 cases, respectively. No side reactions were observed. The results revealed that the therapeutic effect of Herceptin is intimately linked to Her-2/neu expression of breast cancer.
3.CLINICAL TRIALS OF PARTIAL RADIOIMMUNITY TARGETING THERAPY WITH ~(131)I MONOCLONAL ANTIBODY FOR HEPATOCELLULAR CARCINOMA (HCC)

To explore the therapeutic effects and toxic side-effect of 131I-McAb (S102, T9, CL-3) in the treatment of hepatocellular carcinoma (HCC). 131I-McAb, which was labelled by IODO-GEN method, with 50-70mCi of 131I, was percutaneously injected into the tumor under B-ultrasonic guidance in 116 patients. The results showed that the effective rate (CR+PR) was 29.3% (34/116), the overall effective rate (CR+PR+MR) was 53.4% (62/116), the one-year survival rate was 38.2% (39/102), the mean survival time was 34.7 weeks, and the mean tumor developing phase was 12 weeks. The results suggested that the use of 131I-McAb in the treatment of HCC is a low toxic, effective and minimal invasive operation.

Objective To investigate the possibility of using 5-aminolevulinic acid(5-ALA)induced protoporphyrin IX(PpIX)for photodynamic therapy(PDT)of nasopharyngeal carcinoma(NPC)cell line CNE2 cultured in vitro.Methods CNE2 cells cultured were incubated in a medium containing various concentrations of 5-ALA(0.01,0.05,0.1,0.25,0.5,1.0,2.0,4.0mmol/L)for 6 hours,then illuminated with different light doses(20,10,5,2J/cm2)using a semiconductor laser at 630nm.After 6,12,24 hours incubation with 5-ALA-PDT,the survival rates of CNE2 cells were analyzed by MTT assay.Results 5-ALA-PDT may effectively kill the CNE2 cells.The killing degree was positively correlated with the incubation time after irradiation,concentration of 5-ALA and the dosage of radiation(P
5.Anti-angiogenic effect of sorafenib combined with heparin

Objective To observe and compare the anti-angiogenic effect of sorafenib, unfractionated heparin(UFH)and low molecular weight heparin (LMWH), singly used or in combination, with chick chorioallantoic membrane assay. Methods Sixty 8d-developed chick embryos were randomly divided into 6 groups (10 each): control group, sorafenib group, UFH group, LMWH group, sorafenib+UFH group, and sorafenib+LMWH group. Drugs were given respectively and incubation continued. The dosage of drugs for each embryo was 20 nmol of sorafenib, 5U of UFH and 5U of LMWH, respectively, and H-DMEM was applied to the embryo in control group in the same volume (2?l). After continuing incubation for another 72h, the anti-angiogenic effect in each group was observed and the vascular number in the chick chorioallantoic membrane was counted. Objective To investigate the clinical significance of IL-27 in hepatocellular carcinoma ( HCC) in the early stage , and to compare it with alpha-fetoprotein ( AFP) in diagnostic performance .Methods Enzyme-linked immunosorbent assay was used to detect the serum level of IL-27 in the HCC group and control group .Receiver operator characteristic ( ROC) curve was used to ana-lyze two indicators and the logistic regression model predicted value ( PRE) was obtained .Results The study indicated the concentra-tion of IL-27 in HCC group [(364.19 ±177.55)pg/ml] was significantly higher than the control group [(255.49 ±94.33)pg/ml], the area under the curve (AUC) of IL-27 and AFP were (0.804) and (0.818), respectively.Logistic regression obtained the regres-sion model PRE that contains AFP and IL-27, with a predicted area under the ROC curve for HCC (0.901), while the diagnostic sen-sitivity (84.8%) and specificity (96.7%) were significantly higher than the capability of individual diagnosis of each indicator .Con-clusions In this study we obtained the model which can be used for clinical diagnosis of hepatocellular carcinoma in future .
8.Regulatory Effect of Buyang Hhuanwu Decoction on Rat Myocardial H9C2 Cell Activity Inhibition Induced by Trastuzumab

Objective To observe the influence of Buyang Huanwu Decoction ( BYHWD) on the inhibition of rat myocardial H9C2 cell activity and SH2-containing tyrosine phosphatase-1 ( SHP-1) activity induced by trastuzumab, and to explore the possible regulatory mechanism after observing the intervention of BYHWD on rat myocardial H9C2 cell transfected with SHP-1 or SHPC/S-1 gene. Methods The eukaryotic expression vectors pcDNA3.1 (+)- SHP-1 and pcDNA3.1 (+) -SHPC/S–1 were constructed and then were transfected to rat myocardial H9C2 cells using the method of liposome transfection. The cells with positive clones were screened out with G418, and then were cultured with trastuzumab for maintaining growth. Using quantitative RT-PCR, we detected the expression of SHP-1 gene and SHPC/S - 1 gene in rat myocardial H9C2 cells. The phosphatase activity analysis was used for observing the regulatory effect of BYHWD on SHP-1 in myocardial cells. Furthermore, we observed the apoptosis of rat myocardial H9C2 cells by methyl thiazolyl tetrazolium (MTT) assay after treatment with BYHWD. Results Sequencing results indicated the successful construction of eukaryotic expression vectors, which had stable expression in myocardial H9C2 cells even under the intervention of trastuzumab. The results of phosphatase analysis showed that H9C2-SHP-1 had the highest activities of phosphatase, but the activities were decreased after the intervention with BYHWD ( P<0.05) . The results of MTT assay also showed the apoptotic rate of H9C2-SHP-1 cells was decreased after treatment with BYHWD ( P <0.05) . Conclusion BYHWD can promote the proliferation of myocardial H9C2 cells inhibited by trastuzumab, and can regulate the expression of SHP-1 in myocardial cells, which will supply reference to the further study of treatment of trastuzumab-induced cardiac toxicity.
9.Forensic Analysis of 25 Cases of Unnatural Death in Custody.

OBJECTIVES:
To screen and collect the cases of unnatural death in custody and analyze the influences and forensic characteristics.
METHODS:
Total 25 cases of unnatural death in detainees in custody form 2000 to 2015 were collected. Some forensic characteristics such as gender, age, yearly incidence, causes of death, manner of death were analyzed. The public security custodies were also compared with the prisons.
RESULTS:
All dead involved were male, mostly were young and middle-aged adults. It showed that the number of cases tended to decrease year by year. The incidence of the injury cases were higher in public security custodies (64.7%) than that in the prisons (12.5%). However, there was a higher suicide rate in prisons (62.5%) than that in public security custodies (23.5%). The mainly cause of death were injury and asphyxia, there were also some cases died from intoxication and electricity.
CONCLUSIONS
The cases of unnatural death in custody expose some problems such as the imperfectness of law enforcement standardization, supervision loopholes and poor medical standards. A comprehensive and detailed autopsy has important implications for the identification of cause of death in custody.

10.Cloning and eukaryotic expressing of GPI-B7-1 in CHO

AIM: To construct human GPI-B7-1 fusion protein and investigate the therapeutic potentials in the treatment of tumors. METHODS: The chimeric GPI anchored-B7-1 gene was obtained by overlap PCR and inserted into expressing vector pcDNA3.1, named pc3.1/GPI-B7-1. pc3.1/GPI-B7-1was transfected into CHO cells by lipofectamine ~2 000 reagent. The CHO cells, expressing GPI-B7-1 on membranes, were obtained after selecting by G418. That was confirmed by flow cytometry, SDS-PAGE and Western blot. RESULTS: Recombinant vector pc3.1/GPI-B7-1 was successfully constructed and sequence result indicated that it was identical with reference sequence. The protein on transfected CHO cell membrane selected by G418 was confirmed to be GPI-anchored protein by flow cytometry, and GPI-B7-1 approximately 60 kD was conformed by SDS-PAGE and Western blot. CONCLUSION: A large amount of GPI-B7-1 fusion protein was obtained and will be further studied in the treatment of tumors.2? [
