1.The effect of sepsis serum on intercellar adhesion molecule-1 of rat intestinal mucosa microvascular endothelial cells and the intervention of Adrenomedullin

Objective To observe the levels of intercellar adhesion molecule-1(ICAM-1)of rat intestinal mucosa microvascular endothelial cells in sepsis rat,and to investigate the possible protective effect of adrenomedullin on the expression of intercellar adhesion molecule-1(ICAM-1)of rat intestinal mucosa microvascular endothelial cells,and to confirm whether adrenomedullin can decrease released inflammatory factors to slow down or inhibit the occurrence and development of sepsis.Methods The rat intestinal mucosa microvascular endothelial cells were isolated and cultured in vitro,and it was divided into sham operated(sham operated serum),sepsis serum(with addition of sepsis serum)and ADM serum(with 0.lng/kg adrenomedullin 6h after sepsis treatment)groups.The soluble ICAM-1(sICAM-1)content in RIMMVECs culture supernatant was measured by ELISA at the 24th hour after serum stimulation.Results The ICAM-1 content in the supernatant of sepsis group [(0.33 ±0.04)ng/L] was obviously higher than that in sham operated [(0.15 ± 0.02)ng/L] and ADM [(0.17 ± 0.04)ng/L] group(P < 0.05),but ADM group was no significantly different compared with sham operated group(P > 0.05).Conclusions ADM could decrease the expression of ICAM-I in RIMMVECs,and the low level of ICAM-1 could suppress its downstream inflammation factors and inhibit the occurrence and development of sepsis.

s: Cyclocarya paliurus (Batal.) Iljinsk. polysaccharide (CPPC) has various biological activities including hypolipemic effect, hypoglycemic effect, anti-oxidant effect, anti-tumor effect and immunomodulatiry;especially that hypolipemic effect and hypoglycemic effect have become the research hotspots in recent years. This article reviewed extraction, purification, content determination and biological activity of CPPC in the latest decade, with a purpose to provide references for the deep research and industrial development CPPC.
3.Comparative Study on Trace Elements in Gentiana macrophlla and Gentiana officinalis from Gansu

Objective To determine the contents of elements Fe, Mn, Ni, Cu, Zn, Ca, Mg and Cr in Gentiana macrophlla and Gentiana officinalis. Methods The contents of elements were determined by flame atomic absorption spectrometry. Results The recovery rates obtained by standard addition method were between 88.1%~114.5%, and the RSDs were lower than 3.18%. Gentiana macrophlla is rich in the trace elements such as Fe, Ni, Cu, Ca, Mg, and Gentiana officinalis is rich in the trace elements such as Mn, Zn, Cr. Conclusion There are some difference between wild and planted Gentiana officinalis in element content, that may be related to their growing environment.

A sample investigation is conducted among 109 political affairs staff from different scales of hospitals in Yancheng city.By analyzing the investigation data,this paper points out there is a lack of humanities qualification among hospital political affairs staff.Some countermeasures are also come up with as follows.The enhancement of humanities knowledge should be the first step to improve qualification of political affairs staff.Political affairs staff should take a positive role in adapting to the challenges of information economics.The political affairs work should be initiated with humanities culture in order to keep pace with social progress.The mutual improvement in humanities education and political affairs work lies a solid foundation for the construction of a harmonious physician-patient relationship.
5.Natural CD4~+CD25~+ regulatory T cells express α7-nicotinic acetylcholine receptor subunits

Objective To investigate whether CD4~+ CD25~+ regulatory T cells (Treg) from C57BL/6J mice express alpha7 nicotinic acetylcholine receptor (α7nAChR). Methods CD4~+ CD25~+ regulatory T cells were isolated from mouse splenocytes with a CD4~+ CD25~+ regulatory T Cell isolation kit (Mihenyi Bio-tee). The purity of isolated Tregs was analyzed by flow eytometry. Expressions of α7nAChR in mouse CD4~+ CD25~+ Tregs were examined by immunofluorescence staining, Western blotting, and reverse transeription-PCR, respectively. Results It was revealed by flow cytometry that Tregs could bind alpha-bungarotoxin (α-BGT)-F/TC, a specific α7 nAChR antagonist. Moreover, a positive binding to α-Bgt was also observed on the cell surface of Treg, as viewed by fluorescent confoeal microscopy. In addition, a clear band of a7nAChR with a molecular mass of approximately 55 kD was found from Tregs by Western blotting analysis, and α7nAChR mRNA was expressed with the expected size of 199 bp from Tregs by reverse transcription-PCR. Conclusion Natural CD4~+ CD25~+ Tregs from mice express α7nAChR.
6.Effect of centrifugal force on osteoblastic differentiation of bone marrow stroma cells in vitro

BACKGROUND: Centrifugal force is a contributing factor inducing osteoblastic differentiation from bone marrow stroma cells.OBJECTIVE: To explore whether centrifugal force promote osteoblastic differentiation from rabbit marrow stroma cell seeded on polylactic-co-glycolic acid (PLGA) scaffolds. METHODS: Rabbit bone marrow stroma cells were isolated and cultured by whole bone marrow method, purified by attachment method, and digested by trypsin-EDTA at 80% confluency. The cell concentration was adjusted to 1×10~9/L. PLGA was cut into pieces, 5 mm×5 mm, soaked in serum-conditioned culture medium for 24 hours. The third passage of bone stroma cell suspension at a density of 300 μL was respectively seeded into the PLGA material. The scaffold/cell compound was placed in centrifuge tube, with cell at the upper layer and cultured in osteoblastic induced medium containing antiscorbic acid, β-sodium glycerophosphate, and dexamethasone respectively under centrifugal force every 12 hours (1 000 r/min for 30 minutes, relative centrifugal force 132 g) and static condition. Alkaline phosphatase activity, osteocalcin content and calcium content as well as observation by light microscopy were used to evaluate osteoblastic differentiation. RESULTS AND CONCLUSION: After 16 days of in vitro culture, the scaffolds of centrifugal force group were coated by multiplayer cells and mineralized matrix, but only a thin layer of cells were observed on the scaffold of control group. The centrifugal force system resulted in a significant decrease in alkaline phosphatase activity at day 2 (P < 0.05) but significant increase at day 4 compared with the static culture condition (P < 0.05). During the whole culture time, osteocalcin secretion remained low in control group. At days 12 and 16, a significant enhancement in osteocalcin secretion was observed for centrifugal force culture compared with static culture conditions (P < 0.05). Moreover, after 16 days of culture a significant increase in calcium deposition was observed in the scaffolds subjected to centrifugal force compared with static culture condition (P < 0.05). Centrifugal force can enhance osteoblastic differentiation and mineralized matrix production of bone marrow stroma cell seeded in PLGA.
7.Nursing of patients with mechanical ventilation after percutaneous endoscopic gatrostomy

Objective To investigate the nursing points of the mechanical ventilated patients after percutaneous endoscopic gatrostomy (PEG). Methods The nursing approaches of 28 mechanical ventilated patients after percutaneous endoscopic gatrostomy in our department was analyzed retrospectively. Results The nutritional status of the 28 patients was improved to different degree after enteral nutrition (EN) support by PEG. Thereby, the nutritional requirement of the mechanical ventilated patients was ensured effectively. Among these patients, 4 patients could take food by mouth after taking off ventilator and extubation and stoma tube was pulled out, 15 patients were discharged carrying with stoma tube and convalescent care was continued, 3 patients died of primary diseases. Conclusions The PEG is a simple,safe and cost-effective approach of nutritional support for EN of mechanical ventilated patients. The key point of insuring EN of severe patients is intensive nursing and to prevent and reduce the complication of PEG.
8.Study on Antioxidant Activities of Ethanol Extract of Paeonia lactiflora and Different Polar Parts in vitro

OBJECTIVE:To study antioxidant activities of ethanol extract of Paeonia lactiflora (CREt) and different polar parts in vitro. METHODS:CREt was prepared with 95% ethanol. CREt (extracted by 50% ethanal) was extracted with aether petrolei,acetic ether and n-butyl alcohol to obtain aether petrolei part (CRP part),acetic ether part (CRE part),n-butyl alcohol part(CRB part)and water part(CRW part). The ability of CREt and different polar parts eliminating DPPH·,O2-· and ·OH were investigated (mass concentrations of CREt and different polar parts were respectively 0.75-12,0.5-6,1.25-15 mg/ml in above 3 tests) and compared with ascorbic acid (VC,0.2 mg/ml) group. RESULTS:The maximum elimination rate of CREt to DPPH·, ·OH and O2-· were(97.55±0.25)%,(81.45±0.20)% and(75.28±0.41)%,IC50 were 1.629,1.789 and 5.268 mg/ml;those of CRE part to those radicals were (82.54 ± 0.36)%,(77.74 ± 0.42)% and (72.16 ± 0.73)%,IC50 were 2.481,1.918 and 6.005 mg/ml;that of CRB part to ·OH reached to(62.53±0.83)%,IC50 was 7.232 mg/ml,but to DPPH·、O2-· were less than 45%;those of CRP part and CRW part to those radicals were all lower than 40%. Each part could eliminate 3 radicals in dose-dependent manner,but were all poorer than VC group,with statistical significance (P<0.01). CONCLUSIONS:The CREt and CRE part show strongest antioxidant activities in vitro,and other parts have weak antioxidant effect.
9.The expression of splice variant of progesterone-induced blocking factor in hepatocellular carcinoma

Objective To study the expression of splice variant of progesterone-induced blocking factor (PIBF) of healthy adult and hepatocellular carcinoma (HCC) patients. Methods Western blot was used to find out the expression in 3 normal livers and tumor and para-tumor tissue of 8 HCC patients. Use T Test to compare the difference. Immunohistochemistry was used to detect the expression in tumor and para-tumor tissue of 48 HCC patients. The splice variant of PIBF serum concentrations of 103 healthy adult, pre-operation and part of 5 days post-operation of 109 HCC patients was tested with ELISA. Result The expression of HCC tissue is lower than para-tumor tissue. The serum concentrations of healthy adult and HCC patients is 73.04 ± 6.29 ng/mL and 49.10 ± 4.07 ng/mL, and the difference was statistically significant (P=0.010). The analysis of blood concentrations of pre-operation and part of 5 days post-operation of 15 HCC patients indicate that the expression of pre-operation is lower than post-operation, and the difference was statistically significant (P = 0.008). Conclusion The splice variant of PIBF is low expressed in HCC tissue and serum. PIBF is a potential protein plays important role in cancer.
10.Study on association of the Beta 3-adrenergic receptor gene wit h obesity

Objective To investigate the association betwee n the β3-adrenergic receptor gene (β3-AR) and obesity. Methods 147 patients with obesity and 106 controls were selected. Genotype of the β 3-AR was determined by polymerase chain reaction (PCR) and restriction fragme nt length polymorphism methods (RFLP). Body mass index (BMI) was detected. Results The frequency of Arg64 allele of β3-AR gene in the obesit y g roup (20.3%) was significantly higher than those in the control group (10.2%) ( P=0.001) . The distribution of Trp64Arg and Arg64Arg genotype in the patient group (38.7% and 0.9%) were both higher than that in the control group (19.0% and 0.7%). In the female subj ects, the Trp64Arg mutation and frequency of Arg64 allele of the β3-AR gene in the obesity group(40.6% and 20.3%) were also higher than those in control (16 . 7% and 9.5%)(P<0.01). Conclusions These results suggest th at the Trp64Arg mutation of the β3-adrenergic-receptor gene may be associat ed with increased body fat.
