1.Analysis of diagnose accordance rate of thyroid function tests

Objective To investigate diagnose accordance rate of thyroid function tests .Methods Clinical data of 15 579 patients suspected with thyroid diseases ,treated during 2011 and 2013 ,were collected ,who were divided into hyperthyroidism group ,hypot‐hyroidism group ,normal group and other diseases group .Proportions of each type of patients were calculated .In each group ,100 pa‐tients were randomly selected to analyze the diagnose accordance rate of thyroid function tests .Results Among all of the 15 579 pa‐tients ,the proportion of hyperthyroidism(24 .73% ) ,hypothyroidism(9 .83% ) and other thyroid diseases(7 .56% ) were statistically different with the proportion of normal thyroid function (57 .89% ,P< 0 .05) .The diagnose accordance rates of thyroid function tests were 81 .00%‐99 .00% .Conclusion Thyroid function tests could be helpful for the diagnosis and differential diagnosis of thy‐roid diseases ,but still with probability of missed diagnosis .
2.Expression of RCAS1 in tissues of normal cervix,cervical intraepithelial neofolasia and cervical cancer

Objective To investigate the expression of RCAS1 in the tissues of normal cervix,cervical intraepithelial neoplasia(CIN)and cervical cancer,and to explore their role in the pathogenesis of cervical carcinoma.Methods The expression of RCAS1 protein was analyzed by immunohistochemical S-P method in 89 cases of cervical carcinoma,82 cases of CIN,and 24 cases of normal cervical specimens.Results Immunohistochemistry showed that the expression rate of RCAS1 in normal cervix,CINⅠ,CINⅡ-Ⅲ and cervical cancer were 0.00,0.314,0.447 and 0.775,respectively.RCAS1 expression tended to increase along with tumor progression.The expression of RCAS1 in cervical cancer was significantly related to the histological grade,the tumor in different sizes(≥4cm or 0.05)betwan patients' age,clinical stage and histological classification.Conclusions The data suggested that RCAS1 might play an important role in the pathoqenesis and development of cervical carcinoma.Over expressed RCAS1 could be regard as an important parameter in malignant degree and poor prognosis of cervical cancer.
3.Expressions of EphB4 and HIF-1? in human gastric carcinoma and their biological implication

Objective To investigate the expressions of EphB4 receptor and hypoxia inducible factor 1? (HIF-1?) protein in human primary gastric carcinoma, and to study the biological implication of their expression. Methods The expressions of EphB4 receptor and HIF-1? protein were detected in 74 specimens of human primary gastric carcinoma (gastric carcinoma group) and in 13 normal gastric mucosa tissues (control group) by immunohistochemistry. Correlations between the expression of these proteins, and clinical and pathological features were respectively analyzed. Result High expressions of EphB4 receptor and HIF-1? protein were found to be 62.2% (46/74) and 52.7% (39/74) respectively in gastric carcinoma group, while the expressions of EphB4 receptor and HIF-1? protein in control group were 15.38% (2/13) and 0% (0/13), respectively. Objective Urinary protein is closely related to the development and progression of major vascular lesionsin diabetic patients.This study was to investigate the correlation between brachial-ankle pulse wave velocity(baPWV) and urinary protein in patients with type 2 diabetes mellitus ( T2DM) . Methods According to the level of 24-hour urinary albumin ( UAL) , 142 T2DM patients were divided into three groups:control (UA<30 mg, n=84), microalbuminuria(UAL<300 mg, n=26), and clinical albuminuria group ( UAL≥300 mg, n=33) .All the patients underwent examination offasting blood glucose, blood lipid, renal function, HbA1c, islet func-tion, C-reactive protein, blood pressure, and BMI.Their baPWV was determined with an Omron atherosclerosis detector and the correla-tion of baPWV with other factors evaluated by Pearson analysis and multivariate linear regression analysis. Results The baPWVwas significantly higher in themicroalbuminuria([1649.19±229.36]cm/s) and clinical albuminuria ([1759.21±291.05]cm/s) than in the control group ([1522.33±248.37]cm/s) (P<0.05).Pearson analysis showed a positive correlation between 24-hour UAL and baPWV ( r=0.347, P<0.01) and multivariate linear regression analysis manifested the same correlation ( P<0.05) . Conclusion The level of urinary protein is closely correlated with baPWV in T2DM patients.
5.Enhancing the Core Competitiveness of University Students through Simulative Employment Training Course

Objective To explore the validity of simulative employment training course on enhancing the core competitiveness of university students.Method We investigate the students with simulative employment trainin, graduates and recruiters through interview, symposium and questionnaire survey. Results The comprehensive quality and occupational capacity of students that completed simulative employment training had been improved. In the simulative employment training course,98%students thought their interview skills had been promoted, 90%recruiters were satisfied with the students,95%graduates who had worked consider that the simulative employment training were helpful. Conclusions Simulative employment training is effective for enhancing the core competitiveness of university students.
6.Expression of KPNA2 in hepatocellular carcinoma

Objective To detect the mRNA expression of KPNA2 in hepatocellular carcinoma(HCC) tissue and to investigate its relationship with the clinicopathological features of HCC so as to provide the basis for predicting HCC infiltration ,metastasis and early treatment .Methods The HCC tissue and paracarcinoma normal tissue from 30 operative cases of HCC in the hepatobiliary surgery department of this hospital from January 2011 to November 2013 were collected and detected KPNA2 expression at the transcriptional level by RT-PCR .The correlation between the mRNA expression of KPNA2 with the clinicopathological factors of HCC was analyzed by consulting the medical record data .Results Among 30 cases of HCC ,the ratio of KPNA2 mRNA in the HCC tissue and the paracarcinoma normal tissue in 25 cases(83 .33% ) was up-regulated (grey value >2 times) .Therefore ,the KP-NA2 gene exhibited the high expression in the HCC tissue and low expression in the paracarcinoma normal tissue .Conclusion KP-NA2 is highly expressed in the HCC tissue and lowly expressed in the paracarcinoma normal tissue ;the high expression of KPNA2 is positively correlated with the stage and pathological grading of HCC ;KPNA2 may be used as an important indicator for early di-agnosing HCC and judging the malignant degree and potential metastais ability of HCC .

Objective To explore the cause and prevention of skin flap necrosis after radical operation for breast cancer and reduce the incidence of skin flap necrosis.Methods The data of 158 patients with breast cancer who had surgical treatment were analysed.The data included the thickness of the skin flap,tension of the skin flap,and the mode of the operation and their relation with necrosis of the skin flap.Results Among the 158 operated cases,32 cases(20.25%) had skin flap necrosis.The incidence of the flap necrosis in the thick skin flap group was lower than in the thin skin flap group(10.87%vs28.95%,P
8. Acupuncture Alleviated Depressive-like Behavior by Down-regulating NF-κB, iNOS and NO Levels in Prefrontal Cortex of Depression Rats

OBJECTIVE: To observe the effect of acupuncture intervention on depressive-like behavior and nuclear factor-κB (NF-κB), inducible nitric oxide synthase (iNOS) and nitric oxide (NO) levels in the prefrontal cortex in chronic stress induced depression rats, so as to explore its mechanism underlying antidepressant effect. METHODS: SD rats were randomly divided into control, model, acupuncture and medication (Fluoxetine) groups (8 rats/group). The depression model was established by chro-nic unpredictable stress stimulation (fasting, water deprivation, cold water swimming, tail clamping, constraining, etc., and solitary raising) for 28 days. Acupuncture treatment was applied to "Baihui" (GV 20) and unilateral "Neiguan"(PC 6) once every other day for 14 days. Fluoxetine (10 mg/kg) was given to rats of the medication group by gavage (p.o.) before every stress stimulation, once every day for 28 days. The animals' behavior was tested by using sucrose intake and open field tests (number of crossings and rearings in 5 min). The expression of NF-κB in the prefrontal cortex was detected by Western blot, and the contents of prefrontal iNOS and NO were detected by ELISA and nitric reductases, respectively. RESULTS: After modeling, the sucrose intake and the numbers of crossing and rearing times were significantly decreased (P<0.01), and the expression of NF-κB protein and the contents of iNOS and NO in the prefrontal cortex were significantly increased in the model group compared with the control group (P<0.01). Following the treatment, the sucrose intake and the numbers of crossing and rearing times were significantly increased in both acupuncture and medication groups compared with the model group (P<0.05, P<0.01), and the expression level of NF-κB, and iNOS and NO contents in the prefrontal cortex were significantly lower in both acupuncture and medication groups than in the model group (P<0.01). There were no significant differences between the acupuncture and medication groups in the crossing number and rearing times, NF-κB expression, and iNOS and NO levels (P>0.05). CONCLUSION: Acupuncture treatment can improve the depressive-like behavior of depression rats, which is closely related to its effects in down-regulating the levels of NF-κB protein, iNOS and NO in the prefrontal cortex to reduce brain inflammatory damage.
9.Antagonism of Bacillus spp.B_1 and B_2 Strains Against Fusarium oxysporum f.sp. pisi

The antagonism of two strains B1 and B2 of Bacillus spp. against pea root rot pathogen Fusarium oxysporum f. sp. pisi was studied. The result of pairing culture showed that B1 and B2 strains of Bacillus spp. had strong antifungal activity to the pathogen. The colonial color of the pathogen changed from gray to white, aerial hyphas increased and entangled into group after treatment with the cell-free fermentational filtrate of B1 or B2. Observation under optical microscope showed that hyphas and spores of the pathogen swelled and distorted with concentrated cytoplasm after treatment, the spores could not germinate or germinated abnormally . A lot of vesicles appeared at the top of the hyphas, and the hyphas stopped growing and broke finally, their cytoplasm spilled from the cell. The cell-free fermentational filtrate of B1 or B2 strains contained 1795.53?g/mL and 1345.93?g/mL protein respectively, from which two antifungal proteins of 103.5 kD (B1 ) and 127.6 kD (B2) were purified.
10.Construction and expression of a prokaryotic vector of recombinant human adiponectin global domain.

OBJECTIVETo construct and express the recombinant human adiponectin (gAd) global domain.
METHODSgAd complementary DNA (cDNA) was obtained from human fat tissue by RT-PCR. The PCR product was cloned into the vector pMD18-T and the prokaryotic expression vector pET32a(+). The recombinant vector was identified by digestion with double restriction endonucleases SalI and EcoRI, PCR and sequence analysis. The recombinant plasmid containing gAd gene was transformed into E. coli BL21 (DE3), and the expression of the fusion protein His-gAd was induced by IPTG.
RESULTSThe gAd cDNA of 412 bp was obtained from the total RNA of the fat tissue and verified by sequence analysis.
CONCLUSIONThe recombinant plasmid could stably express the 34-kD fusion protein His-gAd in the engineered bacteria in the form of inclusion bodies.
