Objective To investigate the changes in proliferation index (PrI) of gingival fibroblasts in nifedipine-induced gingival hyperplasia ( NIFr-HGF). Methods Gingival fibroblasts were derived from a patient with nifedipine-induced gingival hyperplasia. Cells were induced by 10 ng/mL and 1 000 ng/mL nifedipine ( low- and high-concentration drug intervention groups), respectively. Cells were harvested 18 h and 30 h after intervention, cell cycles were detected by flow cytometry, and Prls were calculated. NIFr-HGF without nifedipine induction were served as blank control. Results After induction for the same time, Prls of NIFr-HGF cell cycle of low- and high-concentration drug intervention groups were significantly higher than those of blank control group (P <0.05) , while there was no significant difference between low and high-concentration drug intervention groups (P > 0.05). In low and high-concentration drug intervention groups, Prls of NIFr-HGF cell cycle after intervention for 30 h were significantly higher than those after intervention for 18 h [(57. 54 ± 0.019)% vs (21.15 ±0.011)%, and (59.36 ±0.031)% vs (19.01 ±0.012) %, respectively] (P < 0.05). Conclusion For patients with nifedipine-induced gingival hyperplasia, PrI of NIFr-HGF cell cycle increases with time of nifedipine intervention, while is not significantly related to drug concentration.

OBJECTIVE:To optimize the formulation and preparation technology of podophyllotoxin derivative(5-FPE) liposomes.METHODS:A thin film-ultrasonic dispersion method was used to prepare the 5-FPE liposomes.Orthogonal experiment was performed to optimize its best formulation and preparation technology with entrapment efficiency as the evaluation index;meanwhile,the entrapment efficiency,drug-loading amount,morphology,particle size distribution,Zeta electric potential and stabilities of the liposomes within 10 days' storing at 25 ℃ or 4 ℃ were investigated.RESULTS:The optimal formulation and prepariton technology of the liposomes has been established in our study.3 batches of liposomes prepared in the optimal conditions had a mean entrapment efficiency of 66.45%,a mean drug-loading amount of 7.97%,mean particle size of 284.7 nm and mean Zeta electric potential of-22.15 mV.The entrapment efficiency of the liposomes decreased more after storing for 10 days at 25 ℃ than at 4 ℃.CONCLUSION:The optimal formulation and preparation technology of 5-FPE liposomes are feasible and which serve as a basis for the further study of 5-FPE preparations.

Objective: To apply chromogenic in situ hybridization (CISH for detection of HER2 gene amplification in breast cancer tissues and to discuss some modifications of the CISH method. Methods: HER2 gene amplification was detected by CISH in 60 breast cancer specimens with an immunohistochemical score over 2+. The correlation between the results of IHC and CISH was analyzed. Our experience in CISH manipulation was summarized and optimization to CISH was discussed. Results: CISH identified gene amplification in 91% (40/44) specimens with an IHC score of 3+ and in 50% (8/16) specimens with an IHC score of 2+. The total concordance rate between IHC and CISH was 80% (48/60, P<0.01). The thickness of sections should be controlled within 4-5 μm; the denaturation should be complete; and the post-hybridization washing temperature and time were also very important and the temperature should be controlled at 70-75 °C. The dyeing time of hematoxylin should also be restrictedly controlled. Positive control should be set up in the experiment for high quality of the experiment. Conclusion: CISH has high concordance rate with IHC in examining HER2 amplification and it may be a new method for detection of HER2 gene. The thickness of the sections, the post hybridization washing temperature and time, and the time of hematoxylin dyeing should be strictly controlled.

Objective: To verify whether hot PBS washing after the enzymatic reaction between tyramine and horseradish peroxidase can improve the sensitivity and specificity of catalyzed signal amplification (CSA). Methods: Using 7 different types of primary antibodies and 2 tumor microarray system, we carried out En Vision, LSAB, Standard CSA, and modified CSA staining and compared their sensitivities and specificities. Results: The modified CSA method was the most sensitive one among the 4 methods, followed by standard CSA, En Vision, and LsAB method. The sensitivity of modified CSA method was 2-3 times higher than that of standard CSA. The 4 methods had similar background staining and had exact localization of cells. Conclusion: The modified CSA is more stable and sensitive than traditional CSA staining method.

The treatment effect of primary and metastatic hepatic carcinoma after local intervention?al therapy is closely related to the prognosis of patients. Traditional imaging modalities such as CT, MRI and ultrasound can only provide anatomical information in monitoring treatment response. In recent years, PET/CT has been widely used in monitoring treatment response for tumors. Many studies have compared the effi?cacy of PET/CT with that of traditional imaging modalities in monitoring the response of primary and meta?static hepatic carcinoma after interventional therapy. This review summarizes recent progress in this field.

Objective To establish a method for determination of total hardness in drinking water by using automatic potentiometric titrator.Methods Dynamic equivalence point titration(DET)and monotonic equivalence point titration(MET)mode was used to determine the high total hardness and low total hardness of drinking water samples respectively.Results Used DET mode to determine the high total hardness,the relative standard deviation(RSD)was 0.69%～1.72% and the recovery rate was 101.5%～102.2%.Used MET mode to determine the low total hardness,RSD was 3.49%～4.00% and the recovery rate was 95.8%- 103.6%.Conclusion This method is rapid,simple,accurate,precise and applicable to the determination of total hardness in drinking water in low and high levels.

Objective To analyze the mediating effect of mental resilience on the relationship between work stress and sleep quality in psychiatrists. Methods A total of 221 front-line psychiatrists from four mental health centers in Shannxi Province were selected as the study subjects using convenience sampling method. The questionnaires of Scale for Occupational Stressors on Clinician，Chinese Version of Connor-Davidson Resilience Scale and Pittsburgh Sleep Quality Index Scale were used to investigate the work stress，mental resilience and sleep quality of the psychiatrists. We conducted Bootstrap mediation test to analyze the mediating effect of mental resilience using SPSS PROCESS V3.5 macro program. Results The total score of work stress of psychiatrists was 97.0±17.5，and the medium（P25，P75）of mental resilience and sleep quality scores were 84.0（75.5， 94.0）and 6.0（5.0，9.0）respectively. The detection rate of sleep disorders among psychiatrists was 33.9%（75/221）. The total score of work stress of psychiatrists was negatively correlated with the total score of mental resilience ［rank correlation coefficient（rS ）=−0.34，P<0.01］，and was positively correlated with the total score of sleep quality（rS =0.48，P<0.01）. The total score of mental resilience was negatively correlated with that of sleep quality （rS = − 0.39，P<0.01）. The work stress of psychiatrists had a positive predictive effect on sleep quality［standardized regression coefficient（β）=0.41.P<0.01］，and a negative predictive effect on mental resilience（β）=−0.38，P<0.01）. Mental resilience had a negative predictive effect on sleep quality（β）=−0.24，P<0.01. Mental resilience played a partial mediating role between work stress and sleep quality，and the mediating effect accounted for 22.0% of the total effect. Conclusion Both work stress and mental resilience of psychiatrists can directly affect their sleep quality，and the mental resilience has a partial mediating role in the effect of work stress on sleep quality.

Full-speed development of the primary health care centering on community health service is key to the ongoing health reform in China. Building a new management mechanism for these grass-root health centers is an innovative approach of such a reform as carried out in Zhejiang province.The authors described the present management mechanism in the province, and focused on the specific measures and main outcomes of the nine Group-scale model as used by a district health bureau in Hanzhou since end of 2007. The paper aims at building an ideal model for group-scale management of community health centers, and improving the new city primary health care system centering on community health service.

Objective To find out the living condition of the elderly adults and their needs for community bealthcare services so as to provide evidence-based guidance for our govemment. Methods A total of 925 participants aged ＞ 60 years were personally interviewed for questionnaire and ADL scale evaluation. Chi-square test and Kmskal-Wallis H test were used for data analysis. Results The subjects showed relatively lower education level and the illiteracy rate was high. Their marriage was stable and a decreased divorce rate was found. The economic situation was satisfactory. Their life satisfaction was farbeyond the health satisfaction. Most of the elderly did not live with their offspring,although 21.8% could not take care of themselves. The prevalence rate of chronic diseases was 81.5% ,which resulted in more demands for social activities, health consultation, rehabilitation nursing, and home healtheare services in the community. Conclusion They need to pay more attention to physical and mental health of the elderly. A community healthcare system that could provide services in an-all-round way,including medical care, should be established and improved.
