1.Clinical significance of homocysteine level detection before and after radiotherapy in patients with nasopharynx cancer

Objective To investigate the clinical significance of detecting the homocysteine (Hcy) level by enzymatic cycling as‐say before and after radiotherapy in nasopharynx cancer .Methods 62 cases of definitely diagnosed nasopharynx cancer in our hos‐pital from September 2013 to September 2014 were included into the observation group and 60 individuals undergoing the physical examination were enrolled as the control group .The venous blood was collected once in the control group and was collected before radiotherapy and at 1 week ,1 month after the radiotherapy beginning in the observation group .The enzymatic cycling assay was used to test the Hcy level .Then the detection results were compared between the two groups .Results The Hcy level had no statis‐tically significantly difference among different stages of nasopharynx cancer before radiotherapy (F= 0 .15 ,P= 0 .86) .The Hcy level before radiotherapy and at 1 week after radiotherapy in the observation group were significantly higher than that in the control group ,the difference was statistically significant (t= - 12 .1 ,P< 0 .05 ;t= - 7 .32 ,P= 0 .00) .The Hcy level at 1 month after radio‐therapy in the observation group had no statistical difference compared with that in the control group (t= - 1 .45 ,P = 0 .15) .The Hcy level at 1 week and 1 month after radiotherapy in the observation group was significantly reduced ,which showed statistical difference compared with before radiotherapy (t= - 5 .22 ,P= 0 .00 ;t= - 13 .56 ,P= 0 .00) .Conclusion The Hcy level in the pa‐tients with nasopharynx cancer is significantly increased ,whereas with the conducting radiotherapy ,which shows the decreasing trend ,indicating that the Hcy level has certain guidance value for the diagnosis and treatment of nasopharynx cancer ,but the Hcy level has no obvious difference among different stages of nasopharynx cancer .
3.Synchronizing presentation of cyclins and merlin in single cell in vestibular Schwannoma tissues with confocal laser microscopy

Objective To explore the expression of merlin and cyclins and the nucleocytoplasmic shuttling of merlin in abiogenous vestibular schwannoma tissues. Methods Four paraffin imbedding vestibular Schwannoma samples were included in the study.The cyclins and merlin were analyzed by immunofluorescence at the same time,and the expression and location were synchronously observed with confocal laser microscopy(CLM) in a single schwannoma cell. Results Cyclins and merlin were perspicuously and synchronously presented in a single cell in vestibular schwannoma tissues.In early G1 phase of the tumor cell,merlin was mainly stained in the nucleus,followed by perinucleus and cytoplasm.In G1/S phase,merlin was concentrated in the perinucleus,less in the nucleus and cytoplasm.Merlin staining showed uniform distribution in the nucleus in G2/M phase,while was absent from the nucleus and concentrated in the perinucleus in late G2/M phase. Conclusion The expression of merlin and various kinds of cyclins can be synchronously presented in abiogenous vestibular schwannoma cells with immunofluorescence and CLM.Merlin is shuttling among the nucleus,cytoplasm and perinucleus along with the progression of cell cycle.

Objective To evluate Feridex-enhanced MRI in the diagnosis of focal hepatic lesions. Methods Conventional plain and Feridex-enhanced MR scanning was performed in 20 cases with suspected hepatic neoplasm.The number of lesions on T 2WI ,and the signal intensity of liver and lesions demonstrated on plain and enhanced scans were observed and analyzed.Results After the injection of Feridex,the signal intensity of normal liver tissue reduced significantly,while the signal intensity of lesions showed no marked changes,Feridex-enhanced MRI clearly displayed more lesions on T 2WI than plain scanning did.Conclusion Feridex-enhanced MRI is very sensitive in detecting focal hepatic lesions.
6.The Experimental Study on MR Imaging of Radiation-induced Hepatic Injuries After Stereotactic Radiotherapy in Hepatocellular Carcinoma

Objective To evaluate the dynamic imaging alteration of radiation-induced hepatic injuries after stereotactic radiotherapy (SRT) in hepatocellular carcinoma.Methods 18 healthy New Zealand white rabbits inoculated VX_2 carcinoma underwent 40 Gy SRT and divided into three groups randomly.The rabbits underwent identical plain and enhanced MR imaging after SRT 3 weeks,9 weeks and 20 weeks respectively. The signal-to-noise ratios of irradiation area and the surrounding normal liver on T_1WI and T_2WI imaging were calculated;The percentage enhancement of the hepatic injuried areas and the surrouding livers after Gd-DTPA enhanced was also calculated.Results After radiotherapy three weeks, the signal-to-noise ratios of irradiation areas on T_1WI and T_2WI were no difference between normal liver tissue and irradiation area; there were significant difference between normal liver tissues in radiotherapy nine weeks and twenty weeks; The difference in the signal-to-noise ratios of irradiation area were significant between after three weeks and nine weeks, twenty weeks. There were no difference in the signal-to-noise ratios of irradiation area between nine weeks and twenty weeks.The percentage enhancement of the hepatic injuries area have significant difference in arterial phase between three weeks and twenty weeks and between nine weeks and twenty weeks;The difference in portal and delay phase were significant between three weeks and nine weeks, twenty weeks, there were no difference between nine weeks and twenty weeks.Conclusion Dynamic enhanced and unenhanced MR imaging can reflect the pathological background of radiation-induced hepatic injuries in different phases.
7.Effects of fluoride on the degeneration and ossification of the ligamentum flavum in rats

[Objective]To study the effects of fluoride on the degeneration and ossification of the ligamentum flavum(OLF).[Method]Thirty-six SD rats were divided into two groups,the experimental group was feeded with water of 100 ppm fluoride in distill water,and the control group with distill water.At the end of three and six months,the rats were analyzed with BMD,and the serum and illiac bone samples were collected for detecting the contents of Calcium,Phosphorus,Magnesium,Zinc,Cuprum,Ferrum and Fluoride.At last,the whole lumbar spine were taken for X-ray and pathological observation.[Result]The results showed dental fluorosis appeared in all objectives as compared to almost normal in control group.BMD of the lumbar spine was also greatly high in six month experimental group.The X-ray analysis showed that there were OLF(OLF appeared)in four rats in six month experimental group.Pathological observations showed in six month group,the changes of the ligamentum flavum were more severe than that in 3 months,and there was OLF oringnated from the attachments,mainly intramembranous ossification.[Conclusion]High content of fluoride could lead to the degeneration and ossification of the ligamnetum flavum in rat's lumbar spine,and might play some important role in the ossification of the ligamentum flavum.

Objective To develop fluorescence quantitative reverse transcription polymerase chain reaction (FQ-RT-PCR) for detection and quantitation of transforming growth factor ?1 (TGF-?1) mRNA in peripheral blood mononuclear cell (PBMC) of patients with chronic hepatic disease.Methods The TGF-?1 recombined plasmid was constructed by conventional molecular biological techniques,and strand-specific cRNA standard was synthesized by T7 RNA polymerase in vitro transcription.The cRNA standard and a TaqMan-MGB probe were used for quantitation of the TGF-?1 mRNA,and the assay was evaluated.Moreover,the plasma TGF-?1 was detected by ELISA.Results Sequence analysis indicated that the recombined plasmid contains the specific 102bp fragment of TGF-?1.
