1.Preliminary research of reducing radiation dose In aortic CT angiography with 80 kV tube kilovoltage

Objective To evaluate the feasibility of 80 kV tube kilovoltage in aortic artery CT angiography with 256-slice CT. Methods A total of 62 patients undergoing aortic artery CTA were enrolled into this study and divided into conventional tube kilovoltage (120 kV, n = 31 ) and low tube kilovoltage (80 kV, n = 31 ). The signal-to-noise ratio (SNR) , contrast-to-noise ratio (CNR), volume CT dose index (CTDIvol) and effective dose (E) were evaluated, respectively. Results The mean image SNR was ( 35.92 ± 5.04) and ( 33.95 ± 8. 30) for conventional tube kilovoltage and low tube kilovoltage,respectively, with no significant difference (t = 1. 131, P =0. 263). The mean image CNR was (30. 32 ±4.78) and (28.71 ± 7.96 ) for conventional tube kilovoltage and low tube kilovoltage, respectively, with no significant difference ( t = 0. 964, P = 0. 339 ). The average effective dose ( E ) was ( 14. 28 ± 0.96 )mSv and (9. 72± 0. 81 )mSv for conventional tube kilovoltage and low tube kilovoltage, respectively, with significant difference ( t = 20. 12, P < 0. 001 ). Conclusions 80 kV tube kilovoltage aortic artery CTA can reduce radiation dose by 31.9% , and contrast dose 50% ,and maintain image quality compared with 120 kV tube kilovoltage.
2.TC-1 Stromal Cells Enhance Retro viral-Mediated Gene Transfer into Murine Marrow Hematopoietic Cells

Objective: To investigate the optimal gene transfer protocols of hematopoietic cells mediated by retrovirus. Methods: Murine bone marrow cells were infected by co-culture with murine bone marrow stromal cell line TC-1 or retro-virus packaging cells or retrovirus supernatant. Human mdr-1 and enhanced green fluorescent protein (EGFP) were used as report genes. Results: Stromal cells could greatly increase the gene transfer efficiency when compared with that of supernatant transfection. Transduction efficiency was highest when infected BM cells were co-cultured with virus producer cells. Conculsion: It may be clinically feasible in gene therapy to perform retroviral transduction by co-culture of target cells with stromal cells or cell lines.
3.Construction of recombinant adenovirus vector with HSV-TK controlled by hTERT promoter and experimental study of anti-hepatoma

Objective To construct the recombinant adenovirus vector with hTERT-HSV-TK and observe the killing effect of Ad-hTERTp-HSV-TK/GCV system on hepatocellular carcinoma cells. Methods A recombinant replication defective adenoviral vector of Ad-hTERTp-HSV-TK was constructed via homologous recombination which both shuttle plasmid pSU-Tp-TK and adenovirus backbone plasmid pBHGE3 transfected into the HEK293 packaging cells. Then the Ad-hTERTp-HSV-TK was amplified and purified through PCR. The activity of the HepG2 cells and the L-02 cells were tested by methyl thiazolyl terazolium (MTT) after they were transfected by the recombinant adenovirus of different multiplicities of infection (MOI) and then were added GCV of different conc.entration. Results The recombinant replication defective adenoviral vector of Ad-hTERTp-HSV-TK were identified by PCR successfully. The viral titer was 1.5×1010 pfu/ml after amplification and purification. The HepG2 cells were targetedly suppressed by Ad-hTERTp-HSV-TK/GCV system. The survival rate of cells decreased gradually along with the increase of the MOI and the GCV' s concentration. Conclusion The recombinant replication defective adenoviral vector of Ad-hTERTp-HSV-TK can inhibit the HepG2 cells significantly, but has not influence on the L-02 cells.
4.Study on motor complications in patients with Parkinson's disease

Objective To survey the prevalence and distribution of dyskinesia and motor fluctuations, and to investigate the factors related to the dyskinesia and motor fluctuations. Methods The detailed information were recorded, patients were rated by Unified Parkinson's disease rating scald (UPDRS) and Hoehn-Yahn stage. The occurrence of dyskinesia and motor fluctuations were recorded according to UPDRS Ⅳ. Results One hundred and twenty-two patients receiving levodopa treatment for at lease 6 months were involved. Fifteen (12.3%) were experiencing dyskinesia and 41(33.6%) motor fluctuations. The age of onset(OR=0.907,P<0.01) and daily levodopa dose (95% CI 1.000-1.004, OR=1.002, P<0.05) were independent factors with dyskinesia; while the age of onset (OR=0.922, P<0.05), levodopa treatment duration (OR=1.234, P<0.05), daily levodopa dose (95% CI 1.002-1.008, OR=1.005, P<0.01) and Hoehn-Yahn stage (OR=1.869, P<0.05) were independent factors of motor fluctuations. Conclusions The rate of motor complications was lower than the results surveyed in European countries. The likelihood of occurrence of dyskinesia and motor fluctuations is increased in those on high daily levedopa dose. The concomitant use of other medication to reduce levedopa dose might delay the motor complications.
5.Effects of deoxynivalenol on apoptosis of human gastric carcinoma cell line SNU in vitro

Objective To explore the effects of deoxynivalenol (DON) on apoptosis of human gastric carcinoma cell line SNU in vitro. Methods SNU cells were treated with DON at different concentrations (50, 100, 1000, 2000 μg/L) for 12 hours, and then cells were harvested for cell apoptosis by flow cytometric (FCM) DNA analysis and the expression of Bax, Bcl-2 and Caspase-3 at protein level with FCM and Western blotting. Results FCM results showed that the apoptosis rates of SNU cells in DON treatment groups were all higher than that in control, especially in DON 1000 μg/L and 2000 μg/L groups (P<0.05). In the concentration range from 50 to 2000 μg/L, a significant concentration-depended response correlation could be found between apoptosis rate and DON concentration (r =0.940, P <0.01). FCM and Western blotting showed Bax and Caspase-3 expression in often SNU cells DON treatment for 12 hours were up-regulated while that of Bcl-2 was down-regulated. Conclusion DON can induce apoptosis of SNU cells in vitro in dose-dependent manner, and possible mechanisms of apoptosis induction effects may be up-regulation of the expression of Bax and down-regulation of that of Bcl-2 and activation of the key enzyme of apoptosis Caspase-3.
6.Clinical experience of laparoscopic resection of adrenal tumors

Objective To investigate the laparoscopie resection of the adrenal tumor effects and clinical experience. Methods Among the 24 cases of Laparoscopic adrenalectomy tumors, there were 8 cases of adrenal adenoma, 7 cases of pheochromoeytoma and 9 eases primary aldosteronism. Results One ease was transit to open operation, the other 23 were successful,each surgerical time was 40~130 min with an average of 98 rain. Hemorrhage was 50~300 mL, 140 ml average, no blood transfusion. It was 1~2 d after sur- gery to remove drainage tubes, and could take some activities and have fresh flow in the second day. The pa- tients didn't appear complications and discharged after 5~7 d. Follow-up of 6~24 months, there was no recurrence and long-term complications. Conclusion Laparoscopic adrenalectomy has the advantages of minor trauma surgery, less bleeding, good demonstrating, convenient operation, fewer complications and quick recovery.
