1.Progress in neural stem cells therapy for traumatic brain injury

The neural stem cells (NSCs) can migrate into the injured area and differentiate into neurons or oligodendrocytes.Endogenous neurogenesis may potentially be harnessed as a putative therapy for neural injury.But the complex micro-environment due to TBI will be one of the biggest challenges for endogenous NSCs to perform neural regenerations.Exogenous NSCs have been shown to be able to survive in host tissues and regulate microenvironment via paracrine effects.Thus, transplantation of NSCs to assist neural regeneration has become an attractive option.Recently, rapid advances in the stem cell biology have raised appealing possibilities of replacing damaged or lost neural cells by transplantation of in vitro-expanded stem cells and/or their neuronal progeny.
2.The value of MR diffusion weighted imaging in the diagnosis of invasive ductal carcinoma

Objective To explore the value of diffusion weighted imaging(DWI) in the diagnosis of invasive ductal carcinoma not otherwise specified and find a suitable diagnostic cutoff ADC value.Methods The MRI findings of 69 patients (69 lesions) of invasive ductal carcinoma not otherwise specified and 67 patients benign tumors (69 lesions) confirmed by pathology were evaluated.ADC values of the carcinoma,benign tumors and their contralateral normal breast tissues were obtained.Independent-samples t test and ROC curves were used.Results The ADC values of carcinoma and contralateral normal breast tissues were (0.925 ± 0.268) × 10-3 mm2/s and (1.680 ± 0.446) × 10-3 mm2/s respectively,their difference had statistical significance (t =12.08,P < 0.01) ; The ADC values of benign tumor and contralateral normal breast tissues were (1.350 ± 0.383) × 10-3 mm2/s and (1.690± 0.356) × 10-3 mm2/s respectively,the difference had statistical significance (t =6.64,P < 0.01); The difference between carcinoma and benign tumors had statistical significance as well (t =7.56,P < 0.01).The area under the ROC curve of carcinoma and contralateral normal breast tissues,benign tumors and contralateral normal breast tissues,carcinoma and benign tumors were 0.915,0.794,0.847 respectively.The diagnostic cutoff value for each were 1.185 × 10-3 mm2/s,1.505 × 10-3 mm2/s,1.015 × 10-3 mm2/s respectively;Corresponding sensitivity and specificity were 89.9% and 85.5%,79.7% and 63.5%,75.4% and 87.0%respectively.Conclusions MR-DWI is valuable in the diagnosis of invasive ductal carcinoma not otherwise specified.The best ADC diagnostic cutoff value to differentiate invasive ductal carcinoma not otherwise specified from benign tumors is 1.015 × 10-3mm2/s.
3.Quantitative dynamic contrast enhanced MR in the prediction of response in breast cancer patients undergoing neoadjuvant chemotherapy

Objective To investigate whether quantitative dynamic contrast enhanced MR can predict final pathologic response in primary breast cancer patients undergoing neoadjuvant chemotherapy (NAC).Methods Forty seven patients who were pathologically proved infiltrating ductal carcinoma with core needle puncture biopsy were examined before NAC and after 2 cycles of treatment and the quantitative parameters (Ktrans,Kep and Ve) were analyzed prospectively.Histological response is categorized as non-major histological response (NMHR) and major histological response (MHR).Quantitative parameter changes measured after 2 cycles of NAC were compared between MHR and NMHR using non parametric tests (Mann-Whitney U test) and pretreatment parameters were compared using independent samples t tests.Receiver operating characteristic curve (ROC) was used to determine the best predictor and cutoff value.Results Fifteen patients were grouped into MHR and 32 patients were NMHR.Pretreatment parameters(Ktrans,Kep and Ve) were(1.51 ±0.33) /min,(2.97 ± 1.06) /min and (0.55 ±0.16) in MHR and (1.53 ±0.40) /min,(2.82 ± 0.99) /min and (0.57 ± 0.20) in NMHR.There was no significant difference between the two groups (t values were-0.123,0.450 and-0.380,respectively,P > 0.05).Changes inkinetic parameters(K,Kep and Ve) were-88.2% (-96.0% to 1.5%),-62.5% (-94.3% to -8.7%) and-57.0%(-82.1% to 55.5%) in MHRand-8.1%(-88.5% to 32.4%),-18.2% (-62.1% to 145.9%) and-4.7% (-83.1% to 95.7%) in NMHR.There were significant difference between the two groups (Z values were 4.359,4.359 and 3.332,respectively,P < 0.01).The areas under ROC curve of AKtrans,AKep and Ktrans after 2 cycles of NAC were all 0.898.Sensitivity of the three parameters for predicting NMHR were 87.5%,90.6% and 78.1%,and specificity were 86.7%,80.0% and 93.3%,respectively.Conclusion Quantitative dynamic contrast enhanced MRI can predict final pathologic response in primary breast cancers after 2 cycles of NAC.
4.Proliferation and migration of endogenous neural stem cells in rats after cerebral infarct

BACKGROUND: Conventional opinions believe tissues of central nervous system(CNS) cannot regenerate after injury when they have been developed and maturated. However, recent researches have validated that neural stem cells exist in human and adult animal nervous system, and most of them are in static status in vivo. The therapeutic effects of neural stem cells in cerebral infarct have been a key point in the researches.OBJECTIVE: To observe the reactive process of endogenous neural stem cells after cerebral infarct to explore the effects of endogenous neural stem cell in CNS traumatic rehabilitation, which provides theoretical gist for the self-restoration after cerebral infarct.DESIGN: A randomized controlled trial by using experimental animals as subjects.SETTING: Department of Neurosurgery of Peking Union Hospital.MATERIALS: The study was conducted in the Laboratory of Neurosurgery Department of Peking Union Hospital from March to October in 2003. Totally 82 healthy male Wistar rats with a body mass between 250 g and 300 g were selected.METHODS: Cerebral infarct model was established in rats by thread-ligation method. Rats were divided into post-infarct 1 day, 3 days, 7 days, 14 days and 28 days groups with 14 rats each. Control group was sham-operation group( n= 12). The expressions of bromodeoxyuridine (BrdU) and Nestin in the brain of the rats were dynamically detected by immunohistochemical method.MAIN OUTCOME MEASURES: The changes of BrdU and Nestin positive cell numbers after cerebral infarct in rats.RESULTS: Only very few BrdU and Nestin positive cells had survived in hippocampal dentate gyrus and subventricular zone(SVZ) area in control group. BrdU positive cell significantly increased compared with control group in hippocampus and SVZ area after 1 day of cerebral infarct( P < 0.05),which reached its peak on the 7th day( P < 0.05), started to reduce after 14 days but still in the level significantly higher than normal( P < 0.05), and closed to normal level after 28 days. And moreover, BrdU and Nestin positive cells were significantly more in infarct side than the opposite side ( P< 0.05), which migrated towards the opposite side through callus.CONCLUSION: Cerebral infarct can activate the proliferation in situ and migration of endogenous neural stem cell.
5.Proteomic analysis of hippocampus in the rat

Objective To analyze the protein expression in the rat hippocampus by the proteomic approach.Methods Proteins from hippocampal tissue homogenates of the rat were separated by two-dimensional gel electrophoresis(2-DE),and stained with colloidal Coomassie blue to produce a high-resolution map of the rat hippocampus proteome.Selected proteins from this map were digested with trypsin,and the resulting tryptic peptides were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).The mass spectrometric data were used to identify the proteins through searches of the NCBI protein sequence database.Results 37 prominent proteins with various functional characteristics were identified.The identified brain protein classes covered metabolism enzymes,cytoskeleton proteins,heat shock proteins,antioxidant proteins,signalling proteins,proteasome-related proteins,neuron-specific proteins and glial-associated proteins.Furthermore,3 hypothetical proteins,unknown proteins so far only proposed from their nucleic acid structure,were identified.Conclusion This study provides the first unbiased characterization of proteins of the rat hippocampus and will be used for future studies of differential protein expression in rat models of neurological disorders.
6.The impact of childhood abuse and exposure to family violence on college students' self-esteem

Objective To investigate the impact of childhood abuse and exposure to family violence on college students'self-esteem.Methods Using Childhood Trauma Questionnaire-28 Item Short Form(CTQ-SF),Witness to interparental Violence Questionnaire,Self-esteem Scale(SES),and to investigate 412 college students tional abuse,emotional ignorance,physical ignorance or exposure to interparental physical violence had lower selfesteem(28.04±4.31,28.43±3.81,28.55±3.70,28.66±3.67,28.15±3.87),compared to the students withStudy showed the self-esteem was negatively correlated with childhood physical abuse,emotional abuse,sexual abuse,emotional ignorance,physical ignorance and exposure to interparental physical violence(r=-0.134,-0.216,-0.359,-0.250,-0.170,P<0.01).Study showed most significant correlation between childhood emotional ignorance,childhood emotional abuse and self-esteem.Conclusion The experience of childhood abuse and ignorance,exposure to family violence have side effects on college students'self-esteem.The most important factors are childhood emotional ignorance and emotional abuse.

8.Expression of FGFR3 in acute lymphoblastic leukemia patients and its contribution to proliferation of circulating endothelial cells

AIM: To evaluate the expression of fibroblast growth factor receptor 3 (FGFR3) in acute lymphocytic leukemia (ALL) patients and its contribution to the proliferation of circulating endothelial cells (CECs).METHODS: The mRNA expression levels of FGFR3 in 44 patients with ALL were assayed by RT-PCR.Overall survival (OS) rates of the patients in FGFR3+ group and FGFR3-group were estimated by Kaplan-Meier analysis.The CECs were sorted from peripheral blood by magnetic-activated cell sorting and then counted by 3-color flow cytometry.The cell counts, antigen expression, growth curve and colony forming rate of the CECs in the 2 groups were determined.The FGFR3 expression of CECs was identified by immunofluorescence staining.RESULTS: The positive rate of FGFR3 mRNA expression was 43.2% in 44 ALL patients with normal karyotype.T-ALL expressed higher level of FGFR3 than B-ALL (P<0.05).FGFR3 was over-expressed in ALL patients with bone marrow blast proportion ≥5% (P<0.05).The probability of OS was significantly lower in FGFR3+ group than that in FGFR3-group (P<0.05).The sorted CECs highly expressed CD31, CD144, VEGFR-2 and CD146, and rarely expressed CD45.The counts of CECs and expression level of CD133 significantly increased in FGFR3+ group compared with FGFR3-group.The same result of the amount of colony formation was observed (P<0.05).There was significant difference at 3 time points of cultured CECs count in vitro between FGFR3+ group and FGFR3-group (P<0.05).The positive rate of FGFR3 expression of CECs from 19 ALL-FGFR3+ patients was (29.00±15.71)%.CONCLUSION: The over-expression of FGFR3 gene in ALL may be helpful to evaluate the prolife-ration of CECs, and become a double target with anti-tumor and anti-angiogenesis effects to offer more choice for molecular therapy in the future.
9.Effects of 3 Types of Antihypertensive Drugs on Cognitive Function in Elderly Hypertensive Patients after Acetabular Surgery

OBJECTIVE:To investigate the effects of 3 types of antihypertensive drugs on cognitive function in elderly hyper-tensive patients after acetabular surgery. METHODS:Ninety hypertensive patients receiving antihypertensive drugs for a long term (not changed antihypertensive drugs within 2 weeks before surgery)undergoing selective acetabular surgery were included sequen-tially and divided into angiotensin receptor blocker(ARB)group,angiotensin converting enzyme inhibitor(ACEI)group and cal-cium channel blocker(CCB)group according to the types of antihypertensive drugs,with 30 cases in each group. All patients re-ceived acetabular surgery under epidural anesthesia. The cognitive function of patients was evaluated by using MMSE 1 d before sur-gery(T0),1 d after surgery(T2)and 3 d after surgery(T3). The concentration of S100β protein serum was determined 1d before surgery (T0),immediately after surgery (T1) and 1 d after surgery (T2). RESULTS:Compared with T0,MMSE score of ARB group at T2,those of ACEI group and CCB group at T2 and T3 were decreased significantly,with statistical significance(P<0.05). Compared with ARB group,MMSE score of ACEI group and CCB group at T2,T3 were decreased significantly,with statistical sig-nificance(P<0.05). Compared with ACEI group,MMSE score of CCB group at T2,T3 were decreased significantly,with statisti-cal significance(P<0.05). Compared with T0,the concentration of S100β protein in serum 3 groups were increased significantly, with statistical significance (P<0.05). Compared with ARB group,the concentration of S100β protein serum in ACEI group and CCB group were increased significantly,with statistical significance(P<0.05). Compared with ACEI group,the concentration of S100β protein serum in CCB group at T1,T2 were increased significantly,with statistical significance(P<0.05). The incidence of cognitive dysfunction was in ascending order of ARB group (30%)
10.Construction and application of the health appropriate technology database in Zhejiang Province

Objective Analyzing the construction process and application of health appropriate technology database in Zhejiang province and proposing related considerations and recommendations for the database construction.Methods To design and construct the technology database system,and using excel software to calculate statistic data.Results Established a health appropriate technology database,which contains 826 technologies that can be selected and promoted by the demonstration bases in the whole province.Conclusions The establishment of the technology database increased the efficiency of health appropriate technology transformation at the provincial level,and also provided a reference for the national-wide generalizing of health appropriate technology.
