1.Study on drug addiction ability of cemedex in experiments with white mice

Aims of this study are investigating whether Cedemex could cause addiction and evaluating the safety level of this drug. Researchers used the methods of detoxication promoting or drug hunger stimulating and natural detoxication to evaluate the ability to cause addiction of Cedemex for white mice. Result: White mice were given continuously Cedemex in 9 days, and then they were given Nalotone to stimulate the drug hunger, however, there was not any sign of drug hunger and weight decrease. The natural detoxication group was given Cedemex with gradually increasing dose (2g/kg, 4g/kg, and 8g/kg body weight), drug is pumped into stomach twice a day and continuously in 9 days,, each kind of dose is given in 3 days. After stop giving Cedemex, there was not sign of drug hunger and weight decrease. Therefore, Cedemex does not have the cause drug addiction and has high safety level.

2.Study on the effect of Cedemex on symptoms of stoping syndrome of morphin addicted white house mouse

Subject: to evaluate the effect of cedemex on symptoms of withdrawal syndrome such as jumping time, body weight at lack of morphine. Animals: 50 morphine-addicted white house mice. The results: with appropriate dose, cedemex decreased clearly mouse’s jumping time, prevented effectively the reduction of body weight in mice lacking of morphine. The drug could decrease completely the symptoms of withdrawal syndrome in mice. It is remarkable that, with very high dose, cedemex could affect to digestive function of mice, decreased body weight on the first time, after that recovered but slower than those with medium dose

3.Effects of Ketangte 2 on animal model of hyperglycemia

AIM:To study the effect of Ketangte2 (KTT2)on reducing blood glucose and its mechanism.METHODS:Adrenalin and streptozocin were used to induce hyperglycemia in two mice models,and then treated them with glybenzcyclamide (50 mg/kg),three different dose of KTT2 and normal saline(NS)(0.1 ml/10 g weight)for 15 days.And other healthy mice were set up as control group,During the experiment,the fast blood glucose(FBG),insulin was measured in different time.The pancreas and islets of diabetic mice induced by STZ were studied by pathologic section.RESULTS:In the adrenalin-induced model group,KIT2 could lighten hyperglycemic reaction,which Was significant different from that of model group(P<0.05 or 0.01),while KTT2 high,middle dose group could decrease blood glucose in streptozocin-induced hypoglycemic mice obviously(P<0.01).Histological examination showed that pancreatic island number in pancreas in KTT2 groups increased in comparison with the model group.CONCLUSION:KTT2 can obviously reduce the blood glucose in the two kinds of animal models of hyperglycemia (P<0.05 or 0.01).Its mechanism may be related to the protective effect on islet β cells,thereby increase the insulin secretion.
4.Protective Effects of Ethanol Extracts of Calonyction Acculeatum Beans on Rats with Acute Lung Injury

Objective To investigate the protective effects of ethanol extract of calonyction acculeatum beans( CABE) on rats with acute lung injury ( ALI ) . Methods The ALI model was induced via intravenous injection with 5 mg · kg-1 lipopolysaccharide(LPS). The expression of nuclear factor-кB p65(NF-кB65) was determind by immunohistochemistry(IHC), and the content of interleukin-1β( IL-1β) in the lung tissue was detected by enzyme-linked immunosorbent assay ( ELISA ) . Meanwhile,lung tissue histopathology,the wet-dry weight ratio of lung and the activity of myeloperoxidase(MPO) in the lung tissues were observed. Results Compared with the model group, CABE lessened the lung injury, decreased IL-1β level, reduced the lung dry-wet weight ratio and activity of MPO,and down-regulated the expression of NF-кB/p65(P<0. 05). Conclusion CABE has remarkedly protective effect on ALI induced by LPS in rats. One of the mechanisms may be related to reducing the secretion of IL-1β and interfering with the activity of NF-кB.

Aim To investigate the effects of 1 7-me-thoxyl-7-hydroxyl-benzofuran chalcone(YLSC)on my-ocardial ischemia /reperfusion injury(MI /RI)by mod-ulating PI3K/Akt signaling pathway and the possible mechanisms.Methods Male SD rats were randomly
divided into sham group,model group,YLSC group, wortmannin(WM)group and YLSC +WM group(n =8).The rat model of MI /RI was established by ligation of the left anterior descending artery for 30 min fol-lowed by loosening the ligature for 2 h.After reperfu-
sion,blood samples were obtained to determine serum contents of CK-MB,LDH,NO and TNF-α.The pro-tein levels of total (t)-Akt,phosphorylated (p)-Akt and LC3-Ⅱ were detected by Western blot.Caspase-3,Beclin1 and eNOS mRNA expression was evaluated by FQ-PCR.Results YLSC pretreatment greatly re-duced serum levels of CK-MB,LDH and TNF-α,and elevated NO content.It also inhibited the expression of
caspase-3,Beclin1 and LC3-Ⅱ,while enhanced p-Akt and eNOS expression.Conclusion YLSC protects the heart against MI /RI via inhibition of apoptosis and excessive autophagy,in which protective effect is regu-lated by activation of the PI3K/Akt pathway.
6.Effect of LYS polysaccharides on dementia-related gene expressions in SAMP8 mouse brain

Aim To observe the effects of LYS polysaccharides on expressions of APP, PS1, PS2 and ApoE in SAMP8 mouse brain. Methods 50 6-month-old SAMP8 mice were used and divided randomly into 5 groups: SAMP8 untreated control group, huperzine A control group, low-, mid-and high-dose groups of polysaccharides, with 10 mice in each group. 10 6-month-old SAMR1 mice were used as normal control. After each group was treated by corresponding drug for 40 days continuously, the mRNA contents of APP, PS1, PS2 and ApoE in brain tissue were assayed by reverse transcription polymerase chain reaction.Results The low-, mid-and high-dose groups of polysaccharides could reduce the mRNA contents of APP, PS1 and PS2, which showed a dose-effect relationship in some degrees. But it could not affect the content of ApoE. Conclusion LYS polysaccharides could inhibit the expressions of APP, PS1, and PS2 in SAMP8 mouse brain.
7.Study on Extraction Technique of Sidiming Capsule

OBJECTIVE:To optimize the extraction technique for Sidiming capsules.METHODS:The technical conditions for the water decoction and the alcohol precipitation were optimized respectively by the orthogonal experiment design L9(34)with hydrosoluble extract used as the index for the water decoction and the catalpol extract for alcohol precipitation.The content of Catalpol was determined by HPLC.RESULTS:The optimum conditions were as follows:decocting the crude drugs twice with 8-fold water,1 h each time.The physic liquor extracted by water was filtered,mixed and concentrated to 1.0 g?mL-1(crude drug),and then precipitated by 75% concentration of alcohol for 24 h.Then the physic liquor was filtered,concentrated and dried by microwave vacuum concentration dryer to obtain the dry ointment.CONCLUSION:The optimum extraction procedure is stable and reliable,and it can be used as the optimal extraction procedure of Sidiming capsules.
8.Effects of Millettia pulchra Extracts on TNF-?、PGE_2、NO in Pleuritis Model Rats

OBJECTIVE: To investigate the effect of Millettia pulchra extracts on tumor necrosis factor ? (TNF-?), prostaglandin E2(PGE2) and nitric oxide (NO) in pleuritis model rats. METHODS: 56 Wistar male rats were randomly divided into control group, model group, dexamethasone (DEX) group, water extract of M.pulchra (TYLS) group (high dose and low dose) and total flavonoids of M.pulchra(FYLS) group (high dose and low dose). After preoperational intragastric administration for 7 days, the pleuritis model was induced by injecting carrageenan into pleural cavity in 30 minutes after the last medication. The amounts of pleurorrhea, leucocyte, TNF-?,PGE2 and NO in the pleurorrhea were measured at 8 hours after modeling. RESULTS: As compared with the model group, in TYLS and FYLS group the pleurorrhea volume, leukocyte amount, contents of TNF-? and PGE2 reduced markedly, but the synthesis of NO had little change.CONCLUSION:M.pulchra Extracts show a marked inhibitive effect on pleuritis. Their anti-inflammation effects may be related to inhibiting the increase of TNF-? and the infiltration and transmigration of leucocytes, but not associate with the synthesis of NO.
9.Protective Effect of Yulangsan Polysaccharide on Acute Alcohol Hepatic Injury in Mice

OBJECTIVE:To study the protective effect of Yulangsan polysaccharide(YLSPS) on acute alcoholic hepatic injury in mice.METHODS:50% alcohol at a dose of 10 mL?kg-1 was administered (ig) to mice to establish acute alcoholic hepatic injury model.The activities of serum transaminase and superoxide dismutase(SOD) and the triglyeride(TGL) level were determined and a pathohistological study was performed. RESULTS:The activity of serum transaminase and the content of triglyceride in mice were significantly down-regulated by different doses of YLSPS,but the activity of superoxide dismutase in YLSPS-treated groups was significantly up-regulated and the number of fat droplets was reduced. Fatty degeneration in acute alcoholic hepatic injury model mice was significantly abated by high-dose and middle-dose YLSPS.CONCLUSION:The protective effect of YLSPS on alcohol-induced acute hepatic injury in mice was attributed to its action in lessening the effect of alcohol on lipometabolism.
10.Study on Extraction Technique of Compound Liuyuexue Granules

OBJECTIVE:To optimize the conditions of the extraction technique for compound Liuyuexue gran?ules.METHODS:The herbs in formulation were first extracted by ethanol percolating,then the dregs were decocted with water to be extracted once more.Taking oleanolic acid and total flavones as evaluation markers,the extraction technique in two-step process was evaluated by orthogonal design.RESULTS:The optimum condition of ethanol percolating process was that the crude herb powder was soaked by8-fold80%ethanol for24hours and the optimum condition of water decocting process was that the dregs were decocted for3times with12-fold water,1.5hour each time.CONCLUSION:The repeated experimental results are stable,and the contents of components for markers are high.The optimum conditions can be used for enlarged pro?duction.
