1.Observations on the Efficacy of Electroacupuncture plus Acupuncture Point Injection in Treating Carpal Tunnel Syndrome

Objective To assess the clinical efficacy of electroacupuncture plus acupuncture point injection in treating carpal tunnel syndrome. Method Sixty-three patients with carpal tunnel syndrome were allocated, using a random number table, to a treatment group (32 cases) and a control group (31 cases). The treatment group received electroacupuncture plus acupuncture point injection and the control group, conventional acupuncture. One course of treatment was given. After treatment, the clinical therapeutic effects and the recurrence rates based on the symptoms and signs were compared between the two groups. Result The total efficacy rates in the treatment and control groups were 93.8% and 77.4%, respectively. There was a statistically significant difference in clinical therapeutic effect between the two groups (P<0.05). There was a statistically significant posttreatment difference in the recurrence rate between the two groups of patients (P<0.05). Conclusion Electroacupuncture plus acupuncture point injection is easy to perform and has a good effect in treating carpal tunnel syndrome.
2.Clinical analysis of electroacupuncture combined with massage in the treatment of cervical spondylotic radiculopathy

Objective To observe the clinical effect of electroacupuncture combined with massage in the treatment of cervical spondylotic radiculopathy.Methods 120 patients with cervical spondylotic myelopathy who were treated in Wuhan Traditional Chinese and Western Medicine Hospital from October 2015 to December 2016 were randomly divided into control group and study group.The control group was treated with electroacupuncture.The control group was treated in the control group on the basis of the treatment of massage, compared the two groups of patients with treatment, before and after treatment changes in symptoms, and the treatment of adverse events during the situation.Results The total effective rate was 96.67% in the study group, which was significantly higher than that in the control group(81.67%, P<0.05).The clinical symptoms and work of the two groups before treatment were statistically significant (P<0.05).The scores of the three groups were significantly higher than those of the control group (P<0.05).The adverse events of the study group were significantly higher than those in the control group (P<0.05)The incidence was slightly lower than the control group, the difference was not statistically significant.Conclusion Electroacupuncture combined with massage can effectively improve the total effective rate of treatment, alleviate the clinical symptoms, reduce the incidence of adverse events and improve the safety, and show good application prospects.
3.Transcriptional regulation of GCLC gene in human bronchus epithelial cells

AIM:To investigate the transcriptional regulating function of the catalytic subunit(GCLC)gene of ?-glutamylcysteine synthase(?-GCS),the rate-limiting enzyme of glutathione(GSH)synthetic enzymes.METHODS:The regulating region of the human GCLC gene was cloned by PCR method to construct the wild type plasmid,which expressed luciferase reporting gene.Many mutated plasmids expressing luciferase reporting gene were also constructed by the method of exonuclease Ⅲ with S1 nuclease.The transcription regulation of GCLC gene was investigated by transient transfection of the wild type and the mutated plasmids through the observation of relative activities of luciferase.RESULTS:We found that the-2 515~-2 236 bp,-864~-838 bp,-769~-538 bp,-421~-341 bp regions upside from the transcriptional start site up-regulated the gene transcription are the new positive transcription-regulating regions and the-810~-769 bp(especial the-782~-769 bp)region is the negative transcription-regulating region,which is more precise than ever.CONCLUSION:The results of this experiment provide one good foundation to study the transcription management of the GCLC gene and GSH synthesis.
4.Experimental study on the killing effects of oxymatrine on human colon cancer cell line SW1116

Objective To evaluate the killing effects of oxymatrine(OM) on human colon cancer cell line SW1116 and the mechanism of its anti-neoplastic effect. Methods Methyl thiazolyl tetrazolium analysis, flow cytometry , polymerase chain reaction(PCR)-enzyme linked immunosorbent assay (ELISA) and RT-PCR were used to detect the killing effects of OM and its influence on cell cycle dis- tribution , telomerase activity, expressions of human telomerase reverse transcriptase (hTERT), c-myc ,p53 and mad1 genes in SW1116 cells. Results OM exhibited dose-dependent killing effects on SW1116 cells and induced the increase of G1/G0-phase cells and decrease of S-phase cells. It was found that OM could supress the telomerase activity of SW1116 cells, and the effects were dose- and time-dependent. After OM administration, the expression of hTERT gene in SW1116 cells was decreased, those of p53 and mad1 genes were increased, and the expression of c-myc gene had no marked changes. Conclusions OM has dose-dependent killing effects on SW1116 cells.The anti-neoplastic activity of OM might be due to the inhibition of telomerase activity by means of its influence on hTERT and the up-stream regulation genes.
5.Problem-based learning in clinical teaching ultrasonic therapeutic application

ObjectiveTo explore the PBL mode in training students' ability,improve the quality of teaching effect,in the teaching of Clinical therapeutic ultrasound and the teaching method based on problems with discussion method.MethodsTo 31 subjects of study who were randomly divided into five groups,each 6 people,a PBL teaching mode was used for 12 periods by proposing and study,group discussion,the whole class exchange and commenting on summary.ResultsThe survey shows that 93.6% students think the teachers' design has gained their ends,71% students are impressed by the knowlege and 96.8% of the students show the good learning attitude,64.5% of the students raised their interest in learning,which shows teaching results are effective.ConclusionPBL mode is superior to LBL teaching mode in training students' ability,improving the quality of teaching effect.
6.Inhibition of physiological concentration of glucocorticoids on LPS-induced inflammation in rat alveolar epithelial cells

Aim To investigate the role of physiological concentration of glucocorticoids on the inflammation mediator IL-6 expression in response to LPS in rat alveolar epithelial cells(CCL149).Methods The CCL149 were treated with LPS,H2O2 and glucocorticoid respectively.Flammtory mediator IL-6 protein expression was measured with ELISA,and the activity of histone deacetylase(HDAC) was measured using colorimetric HDAC activity assay kit.Results IL-6 protein levels were increased in cells exposed to 10 mg?L-1 LPS.Hydrocortisone decreased IL-6 protein expression induced by LPS.Such effect of hydrocortisone was blunt by HDAC inhibitor trichostatinA treatment(10 ?g?L-1).LPS decreased HDAC activity.Hydrocortisone increased HDAC activity.The expression of IL-6 protein induced by LPS was further enhanced by H2O2 treatment.Pretreatment with H2O2 resulted in the inhibition of antiflammtion effect of glucocorticoids.Conclusion Physiological concentration of glucocorticoids could suppress inflammatory response,and this effects requires recruitment of HDAC.Oxidants such as H2O2 may cause the failure of glucocorticoids to function effectively,and the reason may be related to the reduction of HDAC activity.This mechanism may contribute to the pathogenesis of pulmonary disorder.
7.Study on Sedative and Hypnotic Effects of Different Eluents of Shuangxiatang

This article was aimed to study the sedative and hypnotic effects of different eluents of Shuangxiatang (SXT). The effects of SXT water decoction, water eluent, 20%, 70% and 95% alcohol eluent on spontaneous ac-tivity and the sleeping induced by subthreshold dose of pentobarbital sodium were measured. The results showed that the SXT decoction, 20% and 95% alcohol eluent can significantly decrease the number of rearing in mice with the percentage of 78.5%, 78.3% and 62.5%, respectively. SXT water eluent and 70% alcohol eluent can significantly decrease the spontaneous activity of mice (P < 0.01), the number of rearing (P < 0.01) and grooming time (P < 0.05). SXT water decoction can significantly shorten sleep latency (P < 0.05), prolong sleep time (P <0.05), and increase rates of sleeping in mice. SXT water eluent can significantly shorten sleep latency in mice (P< 0.05), increase rates of sleeping in mice. SXT water decoction and water eluent have the sedative and hypnotic effects. And the effects are more than alcohol eluents.
8.Effects of isoflurane delayed preconditioning on nuclear factor-κB during myocardial ischemia reperfusion injury in rabbits

Objective To explore the mechanism of the catdioprotection of isoflurane delayed preconditioning on myocardial ischemia reperfusion injury in rabbits.Method Thirty male New Zealand white rabbits,weighing 2.0 to 2.5 kg,were randomly divided into three groups(ten for each group):Control group(group C),I/R group(I/R group) ,2.0% isoflurane group(group S) .Group S was exposed to 2.0% isoflurane-100% oxygen for2 h.Group C and I/R group were exposed 2 h to 100% oxygen served as untreated controls.Twenty-four hours later I/R group and group S underwent 40 rain of coronary occlusion followed by 2 h of reperfusion.Blood samples were taken from arterial line at 20 min before occlusion(T1) ,20 rain after occlusion(T2) ,40 rain after occlusion(T3) ,1 h after reperfusion(T4) and 2 h after reperfusion(TS) for determination of plasma IL-10 levels and TNF-alevels by ELISA.At the end of the reperfusion,infarct size and area at risk were defined by Evans and TTC staining.The heart was harvested and levels of the nuclear factor kappa β(NF-κB)activity were determined by Western Blot,and ultrastructures were observed by electron microscopy.The data was expressed as,and were analyzed by using oneway ANOVA test with SPSS 13.0.P value less than 0.05 indicated statistical significance.Results The NF-κB activity of group S was significantly lower than that of group I/R(P<0.05).Group S significantly(P<0.05)reduced infarct size(19.7%±2.8% in group S) of the left ventricular area at risk as compared with control (37.8 %±1.7 % in I/R group).The injury of I/R group was worse than that of group S from the changes of the cellular structure under light microscope.Group S had a lower levels of TNF-α and also had a higher level of IL-10.Conclusions Isoflurane can inhibit NF-κB activity during myocardial ischemia reperfusion and modulate the cytokine expression,which may be one of molecular mechanisms of Isoflurane delayed preconditioning on cardioprotection.
9.Effects of isoflurane delayed preconditioning on myocardial proteome in rabbits with myocardial ischemia-reperfusion injury

Objective To investigate the effects of isoflurane delayed preconditioning(IDP) on myocardial proteomin rabbits with myocardial ischemia-reperfuaion(I/R).mjury.Methods Eight New Zealand white rabbits of both sexes weighing 2.0-2.5 kg were randomly divided into 2 groups(n=4 each):I/R group and IDP group.Myocardial I/R Wgg induced by occlusion of left anterior descending artery for 40 min followed by 120 min repednsion.In group IDP.the animals inhaled 2%isoflurane for 2 h,undergoing I/R 24 h later.At the end of 120 min reperfusion,the myocardium of left ventricle anterior wall was removed for two-dimensional gel electrophorvsis.The different protein spots were analyzed by means of mass chromatography.Results There were 13 different protein spots between group I/R and IDP.Of the 13 proteins,the expression of 10 spots Was up-regulated and 3 spots down-regulated in quantity.Eleven protein spots of all spots were identified by means of MALDI-TOF-MS.Conclusion IDP can aNenuate myocardial I/R injury in rabbits and it may be related to the alteration in proteome of myocardium.
10.Dynamical distribution of bone marrow mesenchymal stem cells in rat model of chronic aristolochic acid nephropathy.

OBJECTIVETo investigate the distributive path and proliferative rule of marrow mesenchymal stem cells (MSCs) in the rat transplanted via caudal vein from male rat to female rats model of chronic aristolochic acid nephropathy (CAAN).
METHODSCells taken from femoral bone marrow of male Wistar rats were made into single cell suspension, cultured, purified and identified as MSCs. MSCs were transplanted via caudal vein into 50 female Wistar CAAN model rats allocated in the test group, they were killed, 10 rats in a batch, at various time points (6 h, 48 h, 10 d, 30 d and 60 d after transplantation). Besides, 10 rats allocated in the control group were killed on the 30th day after received sham-transplantation. Kidney tissue of all rats was taken for detecting cells originated from the donors by fluorescence in situ hybridization test with FAM-labeled sex determining region of Y chromosome (SRY FISH) probe, and their number in SRY was counted using SRY PCR.
RESULTSMSCs were mainly distributed in the glomerular capillaries at the time points of 6 h and 48 h, but the number of MSCs in glomerular capillaries decreased and those in renal mesenchyma increased at the time points from 10 d to 60 d gradually, then tended to a steady state, meanwhile it showed a stable increasing trend in renal tubule. Cell colony of MSCs could be found in mesenchyma with a slowed down increasing between 30 d to 60 d, but the increasing in tubule was still steady.
CONCLUSIONMSCs originated from the donor can enter the kidney of acceptor and distribute from blood capillary to renal mesenchyma and tubule, and they can long time inhabit there and make propagation.
