2.THE EXPRESSION OF AQP4 IN RAT EYES
Acta Anatomica Sinica 2002;0(06):-
Objective To define the expression of AQP4 in normal rat eyes at transcriptional and translational level, so as to provide morphological bases for the mechanisms of water transport in eyes. Methods Immunohistochemical SABC method and in situ hybridization were used to detect the expression and localization of AQP4 in normal rat eyes. Results The corneal epithelium, the iridal pigment epithelium cells, the nonpigment epithelium cells of ciliary body, the lens epithelium cells, the lens fiber cells, the outer nuclear layer cells, and the inner nuclear layer cells, the ganglion cells of the retina showed positive immunoreactivity of AQP4. The reactive substance was distributed in membrane but not in nuclei of all positive cells. The resuIts of in situ hybridization were identical with that of immuohistochemistry method. AQP4 mRNA could be detected in the cytoplasm of positive immunoreactive cells, however, nuclei were negative. Conclusion The results implied that AQP4 could coordinate with other AQP4 to play an important role in the secretion and regulation of aqueous humor, the maintenance of the diaphaneity of cornea and lens, the motivation and transmission of optesthesia.;

Chinese Journal of Anesthesiology 2001;21(1):10-13
Objective This study was designed to determine the clinical pharmacokinetics ofcisatracurium in Chinese patients. The purpose of this study was twofold: the determination of ED50 and ED95 of cisatracurium and the intubating conditions and changes in BP and HR produced by 2×ED95 and 3 ×ED95 . Methods 100 patients scheduled for elective surgery, aged 30-65 years, served as the experimental subjects. Anesthesia was induced with propofol 2mg/kg and fentanyl 4μg/kg. 5 min after BP and HR were stabilized the study was started. The study consisted of two parts according to the purpose of the study. In part Ⅰ 70 patients were included and randomized to 7 groups based on the doses of cisatracurium the patients received. The doses were 25, 30, 35, 40, 45, 50 and 55 μg/kg from group Ⅰ to Ⅶ. Neuromuscular transmission was assessed by measuring the responses of adductor pollicis muscle to stimulation of ulnar nerve with TOF pulses.The maximal depression of Tt was measured after administration of cisrtracurium, and ED50 and ED95 were calculated. In part Ⅱ 30 patients were included and divided into two groups to receive 2×ED95 (group Ⅷ ) or 3×ED95 (group Ⅸ ) of cisatracurium. The intubating conditions were evaluated. The onset time and time of recovery from paralysis as well as the changes in BP and HR after administration of cisatracurium were recorded. ResultsED50 and ED95 for cisatracurium were 30.7μg/kg and 53.4μg/kg respectively. The onset times for 2×ED95 and 3×ED95 were (254±84)s and (190±59)s. The durations of action was (38.8±6.1)min and (53.28.8)min respectively. Recovery indexes were (16.4±4.9) min and (15.6±5.2) min. There were little changes in BP and HR after administration of cisatracurium in both group Ⅷ and Ⅸ. The intubating conditions were not significantly different. ConclusionsED50 and ED95 for cisatracurium are 30.7μg/kg and 53.4μg/kg in Chinese patients. The onset time is shorter and duration of action longer with 3×ED95 which does not significantly affect BP and HR.

Journal of Guangzhou University of Traditional Chinese Medicine 2015;(3):485-488
Objective The dynamic accumulation trend of secondary metabolite in cultivated Spatholobus suberectus Dunn at different growth time was studied to provide scientific basis for confirming its reasonable harvesting time. Methods Ethanol-soluble extractives were measured by the method recorded in Chinese Pharmacopoeia ( A) Appendix XA ( 2010 edition). The content of total flavonoids was tested by ultraviolet spectrophotometer. The contents of protocatechuic acid, catechin, and epicatechin in Spatholobus suberectus Dunn were determined by high performance liquid chromatography ( HPLC). Results The contents of ethanol-soluble extractives, total flavonoids and the major components such as protocatechui cacid, catechin and epicatechin in Spatholobus suberectus Dunn showed increasing trend with the prolongation of growth time. The changes of contents of alcohol-soluble extract, total flavonoids and epicatechin in samples collecting at different months in one year showed a parabolic trend, and the contents reached the peak values from July to September. Catechin content kept increasing continually. The content of ethanol -soluble extractives of Spatholobus suberectus Dunn aged 4-6 years met the requirements of the current edition of Chinese Pharmacopoeia when harvested from July to September. Conclusion It is preliminarily recommended that Spatholobus suberectus Dunn cultivated for more than 7 years is suitable for harvesting, but the optimal harvest period and the harvest month still need to be further studied.

Medical Journal of Chinese People's Liberation Army 2001;0(12):-
Objective To localize the gene of a Chinese autosomal-dominant non-syndrome deafness (DFNA) family by linkage analysis. Methods Pedigree was drawn after genetic investigation. All family members were checked up, and detail audiology examination proceeded (including pure tone audiometer, acoustic immittance measurement, auditory brainstem response). Vestibular function was evaluated. At first, the similarity of the family's locus linkage with the known DFNA loci should be excluded, and then whole genome scan was carried out. Results Deafness gene of this family was localized at 2q13-q14.2. A maximum LOD score of 3.22 (?=0.00) was obtained with the microsatellite marker D2S363. Haplotype analysis placed the novel locus within a 8.4cM region interval defined by markers D2S1888 and 2S2224. DNA sequencing of coding regions and exon/intron boundaries of a candidate gene PAX8 in this interval did not reveal disease-causing mutation in this family. Conclusion A novel DFNA locus was defined in a Chinese Neimeng family.
