3.The expression level of carcinoembryonic antigen under hypoxic condition in breast cancer ;microenvironment and its prognostic value of breast cancer

Objective To investigate the correlationbetween thecarcinoembryonic antigen ( CEA) level and hypoxic status of breast cancer, and itspotential of being a prognostic factor in breast cancer. Methods Breast cancer cell line MDA-MB-231 was cultured under hypoxic or normoxic condition to determine the protein level of CEA using Western blotting.30 breast cancer tissue sections were collected in Tianjin Medical University Cancer Institute and Hospital from April 2013 to June 2015.and were detected the expression of CEA, hypoxia-inducible factor-1α( HIF-1α) and carbonic anhydrase-9 ( CA-9 ) by immunohistochemistry.Serum CEA were detected by immunofluorescence assay of 166 breast cancer patients collected from April 2009 to June 2011.The correlation between the expression levels of CEA and breast cancer patients'clinical data was analyzed by Logistic univariate analysis.Kaplan-Meier method was employed to calculate the progression-free survival of patients with breast cancer. Log-rank test was conducted to compare the difference between groups.Results CEA levels were significantly higher in breast cancer cells under hypoxic condition in a time-dependent manner, compared to normoxiccontrol. Immunohistochemical staining indicated that CEA was coexpressed with HIF-1αand CA-9 in breast cancer and linear regression analysis revealed that CEA expression correlated with HIF-1α( P =0.0096 );Preoperative serum levels of CEA were closely related with tumor size ( P =0.015 ) andLymph node metastasis(P=0.032); CEA positive patients achieved a progression-free survival of 29.85months(versus 39.08 months in CEA negative patients, P=0.003).Conclusions Hypoxia upregulates CEA expression in breast cancer and the levels of preoperative serum CEA is a potential prognostic factor ofbreast cancer.
4.Optimization of the Preparation Technology of Buqi Tongmai Capsule by Orthogonal Experiment

OBJECTIVE: To optimize the preparation technology of Buqi tongmai capsule.METHODS: Orthogonal experiment of exeracting Radix Astragali was conducted with crude drugs soaking time,extracting times,amount of water added and decocting duration as factors and the content of astragaloside as index.Orthogonal experiment of exeracting Salvia miltiorrhiza,Ligusticum chuanxiong,Fructus Aurantii Immaturus,Carthamus tinctorius was conducted with crude drugs soaking time,amount of water added and decocting duration as factors and the content of tanshinol as index.RESULTS:Water was adopted for extraction and the concentration of alcohol for precipitate was 70 percent.The optimal technology of exeracting Radix Astragali was to soak the crude drugs for 30 min,decoct for 90 min in 10 times of water for 2 times.The optimal technology of exeracting S.miltiorrhiza,L.chuanxiong,Fructus Aurantii Immaturus,C.tinctorius was to decoct the crude drugs for 90 min,8 times of water for 2 times.CONCLUSION:The preparation processes is feasible,stable and meet the demands in the clinic.
5.Analysis on Utilization of Antibiotics and Non-steroidal Anti-inflammatory Drugs in 3 Hospitals of Guang’an from Jan. 2005 to Jan. 2008

OBJECTIVE: To evaluate application of antibiotics and non-steroidal anti-inflammatory drugs in 3 hospitals of Guang’an and find out the reason of unreasonable use of drug. METHODS: 120,000 prescriptions were randomly collected from inpatient and outpatient departments of 3 hospitals from Jan. 2005 to Jan. 2008, which included 20 000 pediatrics prescriptions. The DDDs and DUI of antibiotics and non-steroidal anti-inflammatory drugs were analyzed statistically. RESULTS: The DUI of 10 kinds of antibiotics and 3 kinds of non-steroidal anti-inflammatory drugs were above 1, which indicated that drug abuse occurred. CONCLUSION: Physicians’ prescribing behavior lack medical rationality and the situation of drug feeding hospital is still critical in primary hospitals.
6.CT features and misdiagnosis analysis of retroperitoneal fibrosis

Objective To assess the value of CT in the diagnosis of PRF,and analyze the reason of misdiagnosis. Methods Retrospectively analyze the CT data of ten patients with retroperitoneal fibrosis,which were confirmed by the clinical pathology.All the patients underwent CT scan and enhanced scan.Two experienced physicians using the blind method to evaluate the location, boundary, density, invasion on the surrounding tissue and enhancement of retroperitoneal fibrosis lesions respectively.Results All patients with CT scan findings were retroperitoneal irregular-shaped soft tissue lesions,which was similar to muscle density.6 cases with surrounding the retroperitoneal vessel, 2 cases with expansion and hydrops of renal pelvis and ureter.By enhanced scan,9 cases with different degrees of enhancement,1 case without obvious enhancement.6 cases were misdiagnosed as retroperitoneal schwannoma or lymphoma. Conclusion CT can show the characteristics of retroperitoneal fibrosis. Comprehensively analyze various imaging findings is helpful for the diagnosis of retroperitoneal fibrosis. Misdiagnosis reason is mainly due to retroperitoneal fibrosis is a rare disease,and understanding of this disease in imaging findings is insufficient in the daily work.
7.Cloning and Sequencing of Cathepsin L1(FheCL1) Gene cDNA of Fasciola hepatica

Objective To search for a candidate DNA vaccine of Fasciola hepatica. Methods Using RT-PCR and digestion with Hind III and BamHI, Fasciola hepatica secreted cathepsin L1(FheCL1) cDNA was cloned into the expression vector pcDNA3.1. Results The cloning was successful, the cDNA sequence and its deduced amino acid sequence were analyzed. There was much difference between the cloned FheCL1 and the published one. But the first 20 residues of their amino acid sequences were the same. Conclusion The recombinant plasmid pcDNA3.1-FheCL1 may be a new type of candidate DNA vaccine candidate for Fasciola hepatica. It is possible that Fasciola hepatica presents different sub-species but their amino acid residues (1 to 20) encoded by FheCL1 might build up membrane spanning helix.
8.The effects of Nimotop on the permeability of blood-brain barrier and cerebral infarction size after cerebral ischemic reperfusion in rats

Objective To study the effect of Nimotop on the permeability of blood-brain barrier (BBB) and cerebral infarction size after cerebral ischemic reperfusion (CIR) in rats.Methods CIR models were established by occluding unilateral middle cerebral arteries(MCA) of rats for 2 hours and then reperfused. The models were divided into 2 groups:Nimotop group and the saline control group,each group had CIR 6、12、24、48 and 72 h sub-groups. As soon as reperfusion,Nimotop or saline was abdominal abministrated(2 mg/kg weight) respectively per 12 h. Fluorophotometry and transmission electron microscopy were used to detect the permeability of BBB,the rate of infarction size was calculated by 2,3,5-triphenyl tetrazolium chloride(TTC) dyeing.Results Following reperfusion,the permeability of BBB and the rate of cerebral infarction size were increased. The permeability of BBB had two peaks. The first peak took place at 12h after CIR. The second peak was at 48 h after CIR. These changes were more severe in Nimotop group than in saline group( P
9.Strict Control of Expenditure over Budget by Using Database Trigger

Objective To solve the problem of army hospital expenditure over budget software controlling.Methods In the army hospital accounting reckoning system,database trigger mechanism was used to intelligently control expenditure over budget.Results Expenditure was controlled strictly and intelligently.Financial problems were checked effectively.Conclusion The methods that automatically control budget by trigger is simple and effective.It enhances budget management of hospital effectively.
10.Bioethical Issue in Epidemiological Study of AIDS
