1.THE IMMUNOHISTOCHEMICAL STUDY ON ?-EP,5-HT,5-HTR CELLS IN PANCREATIC ISLETS OF RATS DURING EXPERIMENTAL GASTRIC ULCER

Objective Studies on the changes of ? EP, 5-HT, 5-HTR immunoreactive cells in pancreatic islets during experimental gastric ulcer. Methods The immunohistochemical technique was used. Results The numerical densities on area of ? EP, 5-HT, 5-HTR cells in experimental gastric ulcer group were higher than control group on the 4day,10day after operation.The peak of the numerical densities on area of ? EP and 5-HT cells appeared on the 4th day, while the peak of 5-HTR cells as on the 10th day. Part of ? EP, 5-HT,5-HTR immunoreactive material were located in A or B cells neighbourly section. Conclusion The present work proved that the ? EP, 5-HT,5-HTR cells of pancreatic islets were directly or indirectly participated in the regulation balance of endocrine during the self-healing process of the experimental gastric ulcer and here we provided morphological evidence.;

Objective Studies on the changes of gastrin and SS immunoreactive cells in pancreatic islets during experimental gastric ulcer. Methods The immunohistochemical ABC technique was used. Results The gastrin immunoreactive cells were located in most of the pancreatic islet. The mumber of G cells in experimental gastric ulcer group were higher than that of control group on the 4th and 10th day after operation.The D cells raised on the 10th day.Conclusion The present work provides the evidence that the G and D cells of pancreatic islets might be involed in the self-healing process of the experimental gastric ulcer by endocrine or paracrine regulation.
3.Clinicopathologic and prognostic study of 404 colon cancer cases

Objective To identify clinicopathologic characteristics and prognostic factors through a retrospective analysis of 404 colon cancer patients.Methods A total of 404 patients(209 of them were followed up with inquisition) with colon cancer were surgically treated from 1993 to 2003 and studied by univariate and multivariate analysis.Results Using univariate analysis age of patients,it was found that differentiation degree of the tumors,lymphatic invasion,abdominal and distant metastases,TNM stage and adjuvant radiotherapy were associated with outcome.(2)Multivariate analysis showed that only abdominal and distant metastases and TNM stage were associated with prognosis.Conclusion Multivariate analysis proved following list are independent prognostic factors: abdominal,distant metastases and TNM stage.
4.Mesenteric lymph duct ligation relives hepatic injury subjected to two-hit in rats

Objective To explore the mechanism of mesenteric lymph duct ligation relieving hepatic injury in rats by two-hit of hemorrhage and LPS.Methods Forty-five Wistar rats were divided into three groups: ligation group,non-ligation group and sham group,and the two-hit model was established by hemorrhage and LPS,mesenteric lymph was blocked by ligating mesenteric lymph duct in ligation group.After 24 hours of operation,took out the liver for pathological section,and the hepatocellular apoptosis rate was determined by method of TUNEL,the expression of BCL-2 and BAX protein was determined by immunohistochemical test.At the same time,taking out liver for homogenate of 10 percent,the activity of MPO and ATPase and the contents of TNF-? and IL-6 were determined in hepatic homogenate.Results After two-hit,the hepatocellular apoptosis rate and expression of BAX protein in non-ligation group were significantly increased as compared with sham group and ligation group,and expression of BCL-2 protein was significantly lower.The contents of MPO,TNF-? and IL-6 in hepatic homogenate of non-ligation group were significantly increased than that of sham group,and the activity of ATPase in hepatic homogenate was significantly lower.But the ATPase in hepatic homogenate of ligation group were significantlyincreased and MPO,TNF-? and IL-6 in hepatic homogenate of ligation group were significantly lower as compared with non-ligation group.Conclusion The mechanism of mesenteric lymph duct ligation relieving hepatic injury of rats was related to the mesenteric lymph blockage reduces the TNF-? and IL-6 and improves the expression of BCL-2 protein and the activity of ATPase in liver.

Here we reported a research project based on Black-board to integrate medical curriculum .The key points of this research is application of clinical cases as teaching data and facilitate learning of knowledge following the principle of learning by doing and , input the concept of precision medicine and informatics in learning process with an individually designed framework of learning .The learning outcome is evaluated with big data tech-nology and thus creates a student-centered pathway of medical education .

7.Effect of WTAPgene knockdown on malignant biological behaviors of lung adenocarcinomaA549 cells

Objective: To investigate the effects of Wilms’tumor 1-associating protein (WTAP) on proliferation, migration and invasion of human lung adenocarcinoma A549 cells. Methods: Human lung adenocarcinoma cell line A549 and HEK293T cells were chosen for this study. Two sets of shWTAP interference sequences were designed to construct lentiviral vector plasmid. Human lung adenocarcinomaA549 cells were infected after packaging lentivirus in HEK293T cells, and the control group was transfected with 277 empty vector plasmid. The mRNAand protein expression levels of WTAPinA549 cells were detected by qPCR and WB. Changes in proliferation, migration and invasion of A549 cells were detected by BrdU assay, cell scratch healing assay and Transwell assay, respectively. Results: Two plasmids, shWTAP-1 and shWTAP-2, were successfully constructed. Compared with the control group, the mRNA and protein expression levels of WTAP were significantly down-regulated inA549 cells with WTAP knockdown (both P<0.05), and the proliferation, migration and invasion ability of cells were significantly decreased (all P<0.05). Conclusion: Knockdown of WTAP significantly inhibited the proliferation, migration and invasion of human lung adenocarcinoma A549 cells. The expression of WTAP gene is associated with the occurrence and development of lung adenocarcinoma. WTAP may be a potential target for the diagnosis and treatment of lung adenocarcinoma.
