1.Establishment and expression of multidrug resistance-related genes of human colon carcinoma LoVo/Adr cell line

Objective To establish human colon carcinoma LoVo/Adr cell line with multidrug resistance (MDR) and to study its MDR mechanism. Methods MDR cell line (LoVo/Adr) was induced by stepwise selection on exposure to increasing doses of adriamycin (ADR). The MDR of LoVo/Adr was detected by MTT assay and the distribution of its cell cycle was detected by flow cytometry. The expression of MDR related genes, including mdr1, MRP, GST ? and TopoⅡ was measured by RT PCR and the level of P gp was detected by immunohistochemistry. Results Compared with parental cells, the resistance line had a slower growth rate and longer doubling time. It was larger and mixed with giant cells in different sizes and the number of cells in S phase decreased while that in G1, G2 phase increased. The LoVo/Adr cell line showed 61 fold, 14 fold, 3 fold, 9 fold and 1 fold higher resistance to ADR, VCR, MMC, CTX and 5 FU respectively than its parental cell line. It was also cross resistant to VCR, MMC and CTX, but not to 5 FU. The parental LoVo cells showed no mdr1 expression and the level of mdr1 mRNA expression increased gradually according to the concentration of ADR in resistant cell lines, and the level of GST ? mRNA was only increased significantly in the induced initial stage, although the parental LoVo cells expressed a low level of GST ?. MRP mRNA expression was not detected in both parental cell line and resistant cell lines. The level of Topo ⅡmRNA remained stable. Conclusions LoVo/Adr cell line offers a model with a typical MDR phenotype for the study of MDR in human colon cancer. Its drug resistance was mediated by mdr1 and GST ?, not MRP and TopoⅡ.
2.ANTI-ARRYTHMIC EFFECTS OF TETRAHYDROBERBERINE

The action of Tetrahydroberberine ( THB ) on arrhythmias of the animal models were studied. The arrhythmias induced by CaCl2 and BaCl2 were significantly counteracted by THB ( 30mg/kg ip ) without markedly changing the hemodynamic parameters of rats, but those evoked by aconitine or ouabain were not inhibited in rats or guinea pigs. THB ( 40mg/kg ip) was used to be prevented the arrhythmias induced by ischaemia-reperfusion of left anterior des-cending coronary artery in rats. It could decrease the incidence of vetricular fibrillation, short restored time of the sinus rhythm and diminish the mortality. THB ( 10mg/kg iv ) effectively reduced the incidence of the ventricular arrhythmia induced by hypothalamic electric stimulation in rabbits. The results indicate that THB possess significant antiarrhythmic effect. It may be concerned the action of THB to blok calcium entry
3.Investigation in psychological stress of young nurses facing patients' death

Objective To explore the psychological stress of young nurses facing patients' death. Methods Young nurses in emergency department, ICU and hospice care ward (96 cases, research group) and young nurses in other departments(94 cases,control group)from 8 general hospitals were investigated by self-rating anxiety scale (SAS)and serf-rating depression scale (SDS).The results underwent comparison and analysis. Results SAS and SDS score,incidence of anxiety and depression in the research group were higher than those of the control group (P < 0.05). Conclusions Young nurses who face patients' death frequently have high incidence rate of psychological stress.They show low psychological health level which deserves the close attention of nursing managers.
4.Influence of different concentration of ropivacaine on heart rate variability following intersalene brachial plexus block

Objective To investigate the influence of ropivacaine with isodosage on heart rate variability (HRV ) following interscalene brachial plexus block under ultrasound guidance. Methods Forty-five ASA grade Ⅰ patients (28 males,1 7 females,aged 18-50 years)scheduled for upper limb fracture internal fixation removal were randomized into group R1 (0.375% ropivacaine 40ml),group R2(0.5% ropivacaine 30ml)and group R3 (0.75% ropivacaine 20 ml),1 5 in each group.Changes in HRV,such as LF,HF,LF/HF,SDNN,RMSSD,PNN50% were evaluated with dynamic electrocardiogram.The degree and duration of sensory and motor block were recorded 30 min after drug injection.Results The anesthetic effect and maintaining time had no significant difference among the groups.The incidence rate of Horner syndrome had no significant difference among groups,group R1 was 26.7%,group R2 20% and group R3 20%.LF(30 min after drug injection)in both group R2 and group R3 were significantly decreased compared with that before drug injection, and the decreased degree of LF had significant difference among the three groups.SDNN(30 min after drug injection)in group R2 and PNN50% in group R3 were significantly decreased compared with that before drug injection.The time required for LF,the most obvious changeable HRV parameter, returning to the pre injection level was (876.2 +43.7)min for group R1,(798.8 + 61.5 )min for group R2, group R3 (712.2 + 45.7 ) min, the difference between the three groups was not statistically significant.Conclusion ISBPB can affect the stability of the cardiovascular autonomic ac-tivities by reducing the cardiac sympathetic nerve function and increasing parasympathetic nerve func-tion,possibly through blockade of stellate ganglion or branches of vagus nerve.Along with the in-crease of the dose of anesthetic,the influence on HRV is greater.
5.Genetic polymorphisms of interleukin-10 promoter region in patients with chronic HCV infection

Objective To investigate the correlation between gene polymorphisms in IL-10 promoter region and the susceptibility and clinical phenotypes of HCV infection. Methods Two groups of subjects were studied. Group 1 included 85 treatment naive patients with chronic HCV infection, and group 2 included 75 healthy blood donors. Genotyping of IL-10 promoter-592 was performed by PCR-RFLP. Results No significant difference was found in IL-10 promoter-592 genotypes between the healthy individuals and HCV-infected patients. However, IL-10 promoter-592 genetype A/A appeared more frequently in patients with ALT≥80U/L than in those with ALT
6.Purification and application of major allergens of Metapenaeus ensis

Objective Isolating and purifying the major allergens from Metapenaeus ensis and identifying its immunoreactivity for diagnostic application. Methods The proteins extracted from Metapenaeus ensis were purified by DEAE-52 ion-exchange chromatography and Sephadex G-100 gel filtration. The eluted peaks were detected by dot-ELISA,and the immunoreactivity of the purified allergens was detected by ELISA with the sera from patients allergic to shrimp. The molecular weights of the purified allergic proteins were detected by SDS-PAGE. Results Two major allergic proteins which have highly immunoreactivity in ELISA detection were purified from Metapenaeus ensis. The molecular weights of which were 36 and 68kDa,respectively. Conclusion The major allergens of Metapenaeus ensis purified by ion-exchange chromatography and gel filtration have effective immunoreactivity and can be used as specific antigen in ELISA reagent.
7.THE EFFECT OF IRON SUPPLEMENTATION ON PREGNANT WOMEN AND NEWBORNS WITH IRON DEFICIENCY

Objective: To study iron deficiency (ID) of newborns caused by ID in pregnant women and the effect of iron supplementation. [WT5FZ]Methods: [WT5BZ]Forty four pregnant women were divided into control and iron groups randomly and began to take placebo or iron during median phase of pregnancy. Several indices including serum iron (SI), total iron binding capacity (TIBC), transferrin saturation (TS) and serum ferritin (SF) were measured. Hemoglobin concentrations were obtained from clinical records. [WT5FZ]Results: [WT5BZ]The Hb,SI,TS and SF of newborns in control group were lower than those of newborns in iron group. The incidence of anemia and insufficiency of iron store of newborns in control group were 80% and 54.5% respectively, which were higher than those of newborns in iron group. Except SI of newborns and SF of pregnant women, the SI, TS and SF of newborns were positively related to those of pregnant mothers. Fewer women in iron group suffering from iron deficiency anemia by the end of pregnancy compared to the control were noted. From median phase to the end of pregnancy the Hb, SI, TS and SF of pregnant women in control group remained constant or lower, while the Hb, SI and TS of the pregnant women in iron group got higher and their SF didnt decrease progressively. [WT5FZ]Conclusion: [WT5BZ]The relationship between the nutritional status of iron of pregnant women and newborns is closely related. ID of pregnant woman can lead to the ID of her newborn. Iron supplementation can reduce ID of pregnant women and newborns.
8.Research of impacts on medical visit behaviors of patients due to the two reforms of the clinic-pharmacy-separation at a hospital in Beijing

Objective To study the impacts and the difference made by the pilot reform and comprehensive reform of clinic-pharmacy-separation on the medical visit behaviors of outpatients in Beijing.Methods Invoice information of outpatients′ fee payment during the December 2012 reform and April 2017 reform at the hospital was collected.Information of these patients at the same time as controls was collected to analyze patients′ fee categories, so as to study the effect on the behavior of these patients.Results The outpatient visits decreased by 5.29% as a result of the pilot reform.The ratio of medical insurance patients rose by 6.86%, and that of such patients seeking only prescriptions rose by 4.30%;number of outpatient visits decreased by 2.03% after the comprehensive reform, while the ratio of medical insurance outpatients dropped by 2.61%, and those seeking prescription only dropped by 2.54%.Conclusions The comprehensive reform of clinic-pharmacy-separation has influenced the outpatient′s medical visit behavior, evidencing a positive outcome of the healthcare reform.
9.Up-regulation of CD40 and ICAM-1 expression treated with lipopolysaccharide through the activation of NF-κB in rat peritoneal mesothelial cells

Objective To investigate the expression of CD40 and intercellular adhesion molecule-1 (ICAM-1) treated with lipopolysaccharide (LPS) in rat peritoneal mesothelial cells(RPMC) and the role of NF-κB signal transduction pathway. Methods RPMCs were harvested from Sprague-Dawley rat peritoneal cavity and maintained under defined in vitro conditions. The cells were exposed respectively to different concentrations of LPS for 12 h or treated with LPS (5 μg/ml) for different time points. To observe the effect of LPS on the expression of CD40 and ICAM-1, the RPMCs were treated with LPS (5 μg/ml) for different time points. To observe the effect of LPS on the expression of NF-κB and p-NF-κB protein, the RPMCs were treated by LPS or pretreated with BAY11-7085 (5 μmol/L or 1 μmol/L ) for 3 h, then treated with LPS for another 3 h, respectively. Expression of CD40 and ICAM-1 mRNA was examined by RT-PCR. Expression of NF-κB and p-NF-κB protein was detected by Western blot. Results Compared with medium control group, stimulation of RPMCs with 1 μg/ml and 5 μg/ml of LPS resulted in a significant increase in the expression of CD40 and ICAM-1 mRNA(P<0.05). 10 μg/ml of LPS had strongest effect on CD40 and ICAM-1 expression compared with that of 1 μg/ml and 5 μg/ml of LPS. Treatment with 5 μg/ml of LPS resulted in time-dependent increase in the gene level of CD40 and ICAM-1, with the peak at 3 h. However, after that time point, the gene level of them was gradually attenuated. Following treatment with LPS (5 μg/ml), the level of p-NF-κB began to increase at 15 min, gradually reached the peak at 1 h, and then decreased. But the level of p-NF-κB at 2 h was still significantly higher than that of medium control. 5 μmol/L of BAY11-7085 decreased significantly the up-regulation of CD40 and ICAM-1 induced by LPS. Conclusion LPS enhanced the expression of CD40 and ICAM-1 on RPMCs in a concentration-dependent and a time-dependent manner. LPS induced expression of CD40 and ICAM-1 depend on the NF-κB signal transduction pathway.

Objective To investigate the influence of different volumes of ropivacaine with isod-osage on diaphragmatic excursion following ultrasound-guided interscalene brachial plexus block. Methods Sixty ASA Ⅰ-Ⅱ patients scheduled for right ulnoradial fracture internal fixation removal were randomized into group A (0.5% ropivacaine 20 ml)and group B (0.75% ropivacaine 13.3 ml) (n=30,each).Diaphragmatic excursion were evaluated by M type ultrasound before,1 5 min and 30 min after drug injection. The degree and duration of sensory and motor block were recorded. Results The anesthetic effect and maintain time had no significant difference between groups;Com-pared with group A,group B had lower incidence of diaphragmatic paralysis (P <0.05).There were 28 (93%)and 22 (73%)cases of patients in group A and group B respectively showed reduction of diaphragmatic excursion more than 50% or even complete paralysis 30 min after drug injection (P <0.05).Conclusion Ultrasound-guided interscalene brachial plexus with 0.5% ropivacaine 20 ml or 0.75% ropivacaine 13.3 ml both can provide excellent block.Compared with 0.75% ropivacaine 13.3 ml,0.5% ropivacaine 20 ml is more likely to lead to diaphragmatic paralysis.
