BACKGROUND: The method of replenishing kidney has becoming one of the therapies for anti-senility in gerontology in Chinese medicine.OBJECTIVE: To probe into the effects of the formula for nourishing kidney yin-zuoguiyin on serum malondialdehyde (MAD) content and superoxide dismutase (SOD) activity in metabolism of free radical.DESIGN: Complete randomized control experiment.SETTINGS: Microbiology and Immunology Department of Guangdong Medical College.PARTICIPANTS: The experiment was performed in Immunology Research Room of Wannan Medical College from September 2002 to July 2004. Forty male senile ICR mice were employed, provided by Institute of Experimental Animal of Medical Scientific Academy in Sichuan. They were randomized into senile control, zuoguiyin group, single shudi (Radix Rehmanniae Praeparata) group (single group) and shudi (Radix Rehmanniae Praeparata) removal group (removal group), 10 mice in each one. Zuoguiyin composes of shudi (Radix Rehmanniae Praeparata), shamyao (Rhizoma Dioscoreae), gouqi (Fructus Lycii), zhi gancao (Radix Glycyrrhizae Praeparata),fuling (Poria) and shangyurou (Fructus Corni) at the ratio of 9:6:6:3:4:5. After decocted routinely, the solution in zuoguiyin group was concentrated to 0.577 5 g/mL of raw herbs contained; that in single group was to 0.157 5 g/mL and that in removal group was to 0.42 g/mL. The mixed medical solutions of 100 g/L were prepared successively in each group.INTERVENTIONS: In zuoguiyin group, single group and removal group,solutions of zuoguiyin and shudi (Radix Rehmanniae Praeparata) and solution with shudi (Radix Rehmanniae Praeparata) removed were administrated successively, 4 times/day, 1.5 mL/time, totally for 75 days. No any drink was provided during the medication. The mice in senile control drank pipe water. After the eyeball extracted, blood of 2 mL collected and serum preserved, the thiobarbituric acid method was used to determine MAD content, the xanthine oxidase method was to determine SOD activity and the colorimetric method was to determine general capacity of anti-oxidation in serum.MAIN OUTCOME MEASURES: ① serum MAD content; ② serum SOD activity; ③ general capacity of anti-oxidation in serum.RESULTS: One mouse was dead in senile control and 3 mice in single group. Terminally, 36 mice entered result analysis. ① MAD content:Serum MAD content in zuoguiyin group and single group were lower than that in senile control [(5.280±1.204) μmol/L and (5.886 2.717) μmol/L,(9.533 3.494) μmol/L, (q=3.641-4.657, P ＜ 0.05)]. Serum MAD content in zuoguiyin group and single group were basically same. ② SOD activity: SOD activity in zuoguiyin group and removal group were higher than senile group [(7.008±0.782) mkat/L and (6.989±0.809) mkat/L, (5.908±0.392) mkat/L,(q=3.641-4.657, P ＜0.05)]. SOD activity in zuoguiyin group and removal group were basically same and that in single group and senile control were basically same. ③ General anti-oxidation capacity: The general capacity of anti-oxidation in serum in zuoguiyin group and single group were stronger than that in senile control, that in removal group was weaker than senile control, but the differences were not significant (P ＞ 0.05).CONCLUSION: Zuoguiyin remarkably reduces MAD content and improves SOD activity in serum of senile mice. It is indicated that zuoguiyin acts remarkably on improving metabolism of free radical in senile body,strengthening SOD activity and reducing organic injuries caused by MAD accumulation.

Objective:To explore the effect of zuoguiyin and its partial prescriptions on cellular immune function in senile mice.Methods:Healthy male ICR senile mice were divided into 4 groups at random: old contrast group, ZGY group, prepared rhizome of rehmannia(PROR) group and none prepared rhizome of rehmannia(NPROR) group. The senile mice had been drunk zuoguiyin and its partial prescriptions for 75 days, then observing the differences through thymic index, spleen index, activity of T lymphocyte and the level of IL-2 after using different drugs.Results:Acted by ZGY, ZGY had enhanced cellular immune function of senile mice, appearing that thymic index, spleen index, activity of T lymphocyte and the level of IL-2 were distinctly higher than those of the old contrast group. Compared to ZGY,prepared rhizome of rehmannia also enhanced the activity of T lymphocyte. However, it had no effect on thymic index, spleen index and the level of IL-2.So its effect on anti-senility was not better than ZGY. None prepared rhizome of rehmannia group had no distinct effect on those indexes.Conclusion:Zuoguiyin had an action of antisenility by improving cellular immune function.

Objective To investigate the effect of natriuretic peptide on vascular endothelial function and inflammatory factors of patients with heart failure complicating myocardial infarction.Methods Two hundred and twenty-six patients with heart failure complicating myocardial infarction admitted into Hanzhong Central Hospital from March 2012 to March 2016 were randomly divided into experimental group (n =138) and control group (n =128).Patients in control group were treated with conventional drug,and patients in experimental group were treated with natriuretic peptide based on conventional drug treatment.The vascular endothelial function,contents of NO and eddthelin 1,and inflammatory factors including CRP,IL-6,and TNF-α were detected.Results After 2 weeks of treatment,the flow-mediated vasodilation (FMD) and NO in experimental group were remarkably higher than those in control group,and eddthelin 1 was significantly lower than those of control group (P ＜ 0.05).Additionally,CRP,IL-6,and TNF-α in experimental group were significantly lower than those in control group after treatment for 5 d (P ＜ 0.05).Conclusion Natriuretic peptide could improve the vascular endothelial function of patients with heart failure complicating myocardial infarction via increasing the content of NO and decreasing content of eddthelin 1,and was benefit to cardiac function by inhibiting release of inflammatory factors.

Objective To investigate the eyelid and orbital development of congenital microphthalmia. Design Prospective case se- ties. Participants 23 eyes of 23 patients with congenital microphthalmia aged from 6 months to 78 months. Methods The parameters of palpebral fissure length (PFL), palpebral fissure height(PFH) were measured in all patients. Also, combining the computer tomography (CT) imaging with the computer-aided design system, the anteroposterior axis of eyeball and orbital volumn were calculated automatically. To e- valuate the influence factor of eyelid and orbital growth, correlations between age, anteroposterior axis of eyeball and PFL, PFH, orbital volumn were analyzed. Main Outcome Measures PFL, PFH, anteroposterior axis of eyeball and orbital volumn. Results The PFL, PFH, anteroposterior axis of eyeball and orbital volumn in micro-side were (16.44?3.24) mm, (2.47?1.48) mm, (11.99?3.33) mm, (14.19?2.37) ml respectively. These parameters above were all significantly smaller than the unaffected side (all P=0.000). Moreover, orbital volume showed a strong linear correlation with the anteroposterior axis of eyeball and age(r=0.62, 0.63;p=0.037, 0.035). Both PFL and PFH were corelated with the anteroposterior axis of eyeball (r=0.54, 0.53; P=0.030,0.034), while showing no significant difference with age. According to the an- teroposterior axis of eyeball, two groups, including group less than or equal to 12 mm (10 cases) and group more than 12 mm (13cases), whose PFL,PFH and orbital volumn were (13.50?1.97) mm, (1.08?0.66) mm; (12.73?0.95) ml, (18.20?2.49) mm; (3.30?1.16) mm, (15.05?2.57) ml, respectively. They had statistically significant difference in PFL, PFH and orbital volumn (P=0.001, 0.003,0.024). Conclusion The development of eyelid and orbit with congenital microphthalmia is obviously retarded. The smaller the eyeball is, the much worse in- fluence in eyelid and orbital growth is. The intervention therapy in early stage as far as possible is fundamental approach in simulating the eyelid and orbital development, or preventing and reducing the incidence of orbital deformity.

Object To develop a simple and reliable method to detect the presence of uronic acids in polysaccharides Methods Uronic acid containing polysaccharides and standard glucons were subjected to acid hydrosis for 2 h and 4 h respectively. Samples of the products, before and after hydrolysis were analyzed by TLC using standard uronic acid as the control and aminobenzen alizaric acid as the chromogenic agent, to compare the sensitivities in the detection of uronic acid used in the present conventional analysis. Results The fluorescent spots used as criterion in traditional TLC method were not shown by uronic acids, but by CF 3COOCH 3 and other by products in reaction 0 5 ?g of uronic acid could be detected with our method, without false positive reaction Conclusion The traditional TLC method seemed to lack theorectical background and may mislead structural study The present method in more reliable to detect uronic acids in polysaccharides by analyzing the complete hydrolytic products of polysaccharides using TLC with standard uronic acids as control and aminobenzene alizaric acid as chromogenic agent

Objective To explore the pattern of cognitive impairments mainly caused by the white matter lesions(WML) in the frontal lobe in patients with subcortical ischemic vascular disease (SIVD).Methods Fifty SIVD patients were divided into severe WML group (visual score >3, n=27) and mild WML group(visual score ≤3, n=23) according to their severities of the WML in frontal lobe .Seven patients without SIVD were collected as controls .All patients underwent a set of neuropsychological battery ,and the results were analyzed .Results There was no statistical significance among three groups on basic data .Compared with mild WML group and control group , non frontal white matter scores and numbers of lacunes in frontal lobe of severe WML group were significantly higher ( all P=0.000).Compared with mild WML group,the Montreal cognitive assessment scale in severe WML group were significantly lower ( P=0.047 ) , and scores related to the executive function were significantly lower ( P=0.006 ) , even after adjusting the numbers of lacunes in frontal lobe ,there was statistically significant difference (P=0.038). Multiple regression confirmed that the Z scores of executive functions were mainly affected by white matter lesions located in the frontal lobe ( P=0.000 ) .Conclusion WML located in the frontal lobe mainly affect the executive function in patients with SIVD .

BACKGROUND:Previous researches have focused on the effect of phosphorus compounds on stem cels from animals or from human. But there is no study on the effect of phosphorus ions on human bone marrow mesenchymal stem cels under three-dimensional culture. OBJECTIVE:To explore the effect of phosphorus ions on human bone marrow mesenchymal stem cels under three-dimensional culture. METHODS:There were six groups in the experiment. Human bone marrow mesenchymal stem cels were inoculated in three-dimensional polystyrene scaffolds and then subjected to serum-free growth medium (group 3-GM) or serum-free growth medium containing 4 mmol/L (group 3-4P), 8 mmol/L (group 3-8P) phosphorus ions for 21 days, respectively. Cels cultured on the two-dimensional polystyrene scaffolds were used as control groups (groups 2-GM, 2-4P, 2-8P). Celular proliferation was examined by cel counting kit-8; the mRNA expressions of osteogenic marker genes were assessed by RT-PCR; the formation of mineralized nodules for the osteogenic differentiation of human bone marrow mesenchymal stem cels was examined by Alizarin red staining. RESULTS AND CONCLUSION:Compared with the two-dimensional culture, the growth of human bone marrow mesenchymal stem cels induced by phosphorus ions were more obvious in the three-dimensional polystyrenes scaffolds at days 4, 7, 14 and 21 (P < 0.05). Compared with the group 3-GM, the mRNA expression of colagen type I in groups 3-4P and 3-8P increased more significantly at days 7 and 14 (P < 0.05); the mRNA expression of alkaline phosphatase in groups 3-4P and 3-8P increased more significantly at day 14 (P < 0.05); the mRNA expression of osteocalcin in groups 3-4P and 3-8P increased more significantly at days 14 and 21 (P < 0.05). Mineralized nodules were formed in groups 3-4P and 3-8P but not in group 3-GM at day 21. So we concluded that phosphorus ions can promote proliferation and osteogenic differentiation of human bone marrow mesenchymal stem cels in three-dimensional polystyrenes scaffolds. Compared with the two-dimensional cel culture, the promoting growth effect of phosphorus ions on human bone marrow mesenchymal stem cels in three dimensional polystyrenes scaffolds are more obvious.

Objective To determine the three dimensional motion data of each segment of cervical vertebrae and analyze the characteristics of the intervertebral coupled motion during cervical axial rotation under physiological weight bearing. Methods A total of 16 healthy volunteers (ranging from 22 to 29, median age, 23 years) were recruited to our study. Any cervical spine disorder history, pain or other discomfort and malformations were excluded so as to avoid abnormal neck motion. These subjects underwent CT scans of their cervical segments in a supine position, and 3D models of C1-C7 were constructed. Next, each subject was asked to sit up straight and was positioned in the following sequence:maximal left and right twisting, while double oblique images by DFIS were taken simultaneously at each of the positions. Then, the CT models were matched to the osseous outlines of the images from the two oblique views to quantify the position of cervical vertebraes in 3D at each position. Through local coordinate systems at the center of vertebral bodies, changes of position and angle of each cephalad vertebrae relative to the cauddal one were calculated before and after the axial rotation. Results (1) In the axial rotation of the cervical spine, the contribution of C1/2 accounted for the most of the total cervical rotation range. For the lower levels, axial rotation was found to be maximal at C3/4 and C5/6, minimal at C2/3. (2) In cervical axial motion, C1/2 demonstrated a coupled lateral bending opposite to the axial rotation direction, while each segment of C2-7 demonstrated coupled lateral bending towards the same side of the axial rotation. Among these segments the lateral bending angle of C2/3 was smaller than angles of C3/4, C4/5 and C5/6. Conclusion This study investigated the cervical coupling behavior using the noninvasive 2D-3D matching technique and obtained the motion data at each cervical spinal segment. These findings will help to improve the understanding on physiological cervical spine movement and potential biomechanical mechanism and treatment of cervical spondylosis. Also our data may provide useful reference for the prosthesis design.
