1.Inhibitory Effect of Platycodin D on Proliferation of SGC7901 Cells and Moleclar Mechanism

[Objective] To investigate the effects of platycodin D(PD) on the proliferation and apoptosis of human stomach cancer SGC7901 and the related mechanism.[Methods] SGC7901 was cultured in virto and was treated with 5~20μm·L-1 concentrations of PD.Cell proliferation was examined by MTT assay.Cell apoptosis was detected by Annexin V FITC/PI double staining.The change of mitochondrial trans-membrane potential was measured by JC-1 staining.The potein expression of cleaved caspase-3,cleaved caspase-9,cleaved PARP,bcl-2,bax,p-ERK,ERK,p-JNK,JNK,p-p38 and p38 detected by Western blot.[Results] MTT results showed that PD inhibited the growth of SGC7901 cells in a dose-dependent manner at 24h and 48h.SGC7901 cells treated with PD for 24h showed significantly enhanced apoptosis and weakened mitochondrial membrane potential compared with the control cells.Western blot results showed that PD could up-regulate expression of cleaved PARP,cleaved caspase-3,cleaved caspase-9,bax,p-JNK,p-p38 protein,decreased bcl-2,p-ERK protein,the expression of ERK,JNK,p38 protein did not change significantly.[Conclusion] PD may inhibit the proliferation and induce the apoptosis of SGC7901 cells.These findings indicated that PD inhibited cell proliferation by inhibiting the ERK signaling.PD effect on bax and bcl-2 by activation of JNK and p38 signaling pathway resulted in the decrease of mitochondrial membrane potential and activation of caspase,which induced the apoptosis of cancer cells.

3.Macular light sensibility in idiopathic epiretinal membrane measured by scanning laser ophthalmoscope

Objective To observe the effect of scanning laser ophthalmoscope (SLO) measuring macular light sensibility on evaluating the visual function in idiopathic epiretinal membrane (IERM), and analyze the relationship among the macular light sensibility, central visual acuity, and the thickness of fovea. Methods Procedure of microperimetry of SLO was performed on 44 patients (55 eyes) with IERM diagnosed by indirect and direct ophthalmoscope and optical coherence tomography (OCT). The light sensibility at 10? macular central area was measured. The results were compared with 31 healthy control eyes which underwent the same examinations simutaneously. The correlation among the macular light sensibility, the thickness of fovea measured by OCT, and the results of logarithm visual acuity was anaylzed. Horizontal impacted lower third molars are often adjacent to the inferior alveolar nerve,direct extraction may cause trauma and complications,the most serious complication is inferior alveolar nerve damage.This article reports one case of horizontal impacted lower third molar adjacent to the mandibular nerve canal,the molar was extracted by minimal anchorage pulling for 1 week and then removed smoothly.
5.Cytotoxicity and antibacterial effect of silicone-based soft liner varnish containing Ag/TiO2: an in vitro study.

OBJECTIVEWe aim to evaluate the cytotoxicity and antibacterial effects of Silagum-Comfort (SLC) silicone-based soft liner varnish containing Ag/TiO2 in vitro.
METHODSSilicone-based liner circular specimens, with a diameter of 10 mm and a thickness of 2 mm, were created and divided into six groups randomly. Ag/TiO2 was incorporated into the SLC varnish based on the agent/material weight rate of 0, 0.5%, 1.0%, 1.5%, 2.0%, and 2.5%. The mixed varnish was used to polish the SLC specimens. Antibacterial specimens (ATSLC) were created. Methyl thiazolyl diphenyl-tetrazolium bromide assay method was used to evaluate the 3T3 cell cytotoxicity of ATSLC in vitro. Surface roughness (Ra) measurements of ATSLC were obtained. The antibacterial effect of ATSLC on Candida albicans was examined using the pelliclesticking method.
RESULTSThe cytotoxicity of ATSLC was graded from 0 to 2. The Ra of ATSLC increased when Ag/TiO2 agent concentration increased. However, the differences were not statistically significant compared with the 0 group. With increasing Ag/TiO2 concentration, the antibacterial agent concentration and the antibacterial rate of ATSLC also increased. When the Ag/TiO2 antibacterial agent concentration reached 2.0% and 2.5%, the antibacterial rate increased to 97.5% and 96.5, respectively.
CONCLUSIONVarnish containing Ag/TiO2 has excellent antibacterial effect and can significantly improve the antibacterial effect of silicone-based soft liner.

6.Design and application of digital mobile quality management system for central sterile supply department

Objective To design and apply the quality tracking system for central sterile supply department,complete the quality tracking of sterile medical products,rental surgical devices and high level disinfected products,conform the second-class storage management and human resource management.Methods By using the modern technology of WiFi wireless network,mobile PDA technology and two dimensional barcode,through establishing the team,collecting the data,installing the hardware,debugging the system,training and education,the development and application of the system was completed.Results The system has been operated for two years nearly in central sterile supply depamnent and other departments in hospital,it showed great practicability and convenience,covered all the daily work in central sterile supply department,achieved the information management in mobile digital way.Conclusions The application of digital mobile quality management system accelerates the central sterile supply department in standard way and realizes scientific in strategic decision and management.
7.Bioequivalence of progesterone sustained release suppository in rabbits.

To study the bioequivalence of a kind of progesterone sustained release suppository, a randomized cross-over study was conducted in 12 rabbits. A single rectal dose of 2.75 mg/kg progesterone sustained released suppository (tested formulation, T) and progesterone suppository (reference formulation, R) was administered; a multiple dose of 2.75 mg/kg was given up to seven times with an interval of 8 h. Concentrations in serum were determined by a competitive enzyme immunoassay. The main parameters of T were: for single and multiple doses, Cmax was 48.8 +/- 11.8 ng/mL and 43.5 +/- 9.4 ng/mL, Tmax was 0.5 +/- 0.3 h and 0.4 +/- 0.3 h, AUC(0-24 h) was 362.4 +/- 143 ng x h x mL(-1) and 310.6 +/- 70.3 ng x h x mL(-1), respectively. The relative bioavailability of T to R were (104.2 +/- 13.4)% and (111.4 +/- 19.1)%, respectively. Statistical analysis showed that the two formulations were bioequivalent and T had sustained released feature.

8.Pharmacokinetics and bioavialability of nimesulide dispersible tablet

Aim To study the pharmacokinetics and bioequivalence of nimesulide dispersible tablet and its normal tablet. Methods 20 healthy volunteers were treated with a single oral dose of domestic nimesulide dispersible tablet or normal tablet (control) in a randomized crossover study and the plasma drug concentration was determined by HPLC. Results The plasma concentration time curve was fitted to the one compartment model. The pharmacokinetic parameters obtained were: c max ( 3.91 ? 0.74) ?g ?ml -1 , t (1/2)? ( 3.40 ? 0.78) h , t max ( 3.15 ? 0.67) h , AUC 0~24 ( 31.92 ? 6.36) ?g ?ml?h -1 , there was no significant difference between the active and control groups. The relative bioavailability obtained was ( 96.43 ? 8.41 ) %. Conclusion The pharmacokinetic profile for the 2 tablets was similar so it may be concluded that they are bioequivalent.
9.Therapeutic effect of glucosides of chaenomeles speciosa on adjuvant arthritis in rats

AIM To investigate the therapeutic effect and mechanism of glucosides of Chaenomeles speciosa (GCS) on adjuvant arthritis (AA) in rats. METHODS Complete Freund's adjuvant was used to induce AA in rats. Secondary paw swelling of AA rats was measured with volume meter. Pain response and polyarthritis index were scored. Meanwhile, ConA induced thymocyte proliferation and LPS induced splenocyte proliferation were assayed by 3 (4,5 dimethylthiazol 2 thiazolyl) 2,5 diphenyl 2H tetrazolium bromide (MTT) assay. IL 1 production was measured by thymocyte proliferation assay; TNF ? and PGE 2 production were determined by radio immunoassay. RESULTS GCS (30, 60, 120 mg?kg -1 ) and Actarit (60 mg?kg -1 ) were given by intragastric administration for 8 days from the 17th day after immunization. Treatment with GCS (60, 120 mg?kg -1 ) and Actarit (60 mg?kg -1 ) for 5 days significantly diminished the secondary hind paw swelling, as well as relieved the pain response and the polyarthritic symptoms of the whole body as compared with AA model group. GCS (120 mg?kg -1 ) and Actarit (60 mg?kg -1 ) restored the diminished thymocytes proliferation ConA induced in AA rats. GCS reduced the enhanced productions of IL 1,TNF ? and PGE 2 from peritoneal macrophages in AA rats. CONCLUSION GCS has therapeutic effect on AA in rats, which may be related to regulation function of thymocyte T cells and inhibiting the productions of proimflammatory cytokines secreted by peritoneal macrophages.
10.Experimental study on the differentiation of SHI-1 cells induced by puerariae radix flavones in vitro

Objective To explore the possible effects on differentiation of SHI-1 cells induced by puerariae radix flavones(PRF)in vitro.Methods SHI-1 cells were treated with PRF in various concertration,then the inhibitory effects of cell proliferation were detected by MTT assay,the cell cycles were analyzed by flow cytometry(FCM),the cells reduction rates were detected by NBT reduction test,and the expression of CD11b and CD14 were tested by FCM.Results 10-50 μg/ml PRF could inhibit the proliferation of SHI-1 cells in a time-and dose-dependent manner,and the cell cycles were arrested in S phase.When SHI-1 cells were treated with 10,30 and 50 μg/ml PRF in 48 houres respectively,the NBT reduction rates of cells were increased in a dose-dependent with PRF(P<0.05),and the expression of cells surface differentiation antigen CD14 was also increased along with the concentration of PRF.Conclusion The SHI-1 cells could be induced to differentiation partially after treated with 10,30 and 50 μg/ml PRF in vitro.
