1.The establishment of 3 cell strains secreting anti-acute non-Iymphocytic Ieukemia M_2-type McAb and their characterization
Chinese Journal of Immunology 1985;0(02):-
By the application of hybridoma technique we have established 3 cell strains designated as 1 D 1, 1 C 2 and 1 D6, which all the strains secrete anti-acute nonlymphocytic leukemia M2-type monoclonal antibodies individually. These strains secrete the McAb steadily after passed on propagation over 11 months. The McAb secreted by these cell strains all give positive reactions with the leukemia cells from actue non-lymphocytic leukemia M2-type pationt, but not with cells from M1, M3, M4, and M5 type patients. These McAb also can not react with cells from chronic myelocytic leukemia and its acute transformation phase and acute lymphocytic leukemia patient. The McAb can not react with white cells from normal adults too. From the above metioned we may conclude that the McAb secreted by these cells possess a remarkable specificity. Therefore it is applicable for the immuno-typing diagnosis of acute non-lymphoc yt ic leukemia and researches on target directed therapeutics.

Journal of Environment and Health 1992;0(04):-
Objective To study the effects of static magnetic field (SMF) on development of rat embryonic spinal cord neurons. Methods Primary cultured embryonic spinal cord neurons of Wistar rat were exposed to 1.0, 10.0, 50.0, 100.0 and 200.0 mT static magnetic field. The morphological structure, cell's differentiation and proliferation of the embryonic spinal cord neurons were observed and the contents of MDA, superoxide dismutase (SOD) and protein contents in the neurons were determined. Results Static magnetic field at density of 50-200.0 mT could inhibit the differentiation and proliferation of the cells and the phenomena such as cell aggregation, detouchment of some cells, decrease of clone-formation rate and the size of the cells were observed. The contents of MDA in the cells were increased, while the activities of SOD and the level of protein were decreased. Conclusion Static magnetic field might damage the development of embryonic spinal cord neurons by enhancing the lipid peroxidation.

Orthopedic Journal of China 2006;0(06):-
[Objective]To establish a method for obtaining well-differentiated goat chondrocytes quickly in large scale by RCSS using microcarriers technique. [Method]Articular chondrocytes were harvested from goat by sequential digestion with trypsin and collagenase , and then grown in RCSS bioreactor culture system which contained the Cytodex-3 microcarriers in the culture medium (DMEM) . Growth of chondrocytes on Cytodex-3 microcarriers was observed dynamically under phase contrast microscope . Immunocytochemical analysis was performed for type Ⅰcollagen and type Ⅱcollagen. [Result]The articular chondrocytes attached rapidly to the surface of Cytodex-3 microcarriers. Quick growth of these cells was observed after they fully spread onto the microcarriers. The density of chondrocytes increased by 15~17 times in later stage of culture as compared with the initial density. The harvested chondrocytes had no detectable staining for collagen type Ⅰ, but stained intensively for collagen type Ⅱ. [Conclusion]Microcarrier culture of chondrocytes can yield a large quantity of goat cells within a short time ,which will be of benefit for banking cartilage cells for reconstruction of impaired cartilage by way of tissue engineering.

Journal of Environment and Health 1989;0(06):-
Objective To approach the effect of acrylonitrile (ACN) on the proliferation and differentiation of the lung fibroblasts of mice. Methods The purification and primary culture of the lung fibroblasts(LFb)of new born mice were conducted. The cells were treated with ACN added in the medium at the varying doses of 0.01, 0.5, 1.0, 10.0, 50.0, 100.0 and 200.0 ?g/ml. The cytomorphological methods were used, the protein, malonaldehyde (MDA) and the activity of superoxide dismutase (SOD) were determined. Results No inhibitted effect on the lung fibroblasts was observed at the doses of 0.01-10.0 ?g/ml. At the doses of 50.0-200.0 ?g/ml, acrylonitrile could inhibit the differentiation and proliferation of the cells, the volume of cells became small, the rate of cell clusters decreased, at the doses of 10.0-200.0 ?g/ml, the content of protein and the activity of SOD decreased, MDA content increased. Conclusion Acrylonitrile can inhibit the proliferation and differentiation of the lung fibroblasts in mice, protein synthesis inhibition and lipid peroxidation are considered as the related factors.

China Oncology 2009;19(8):597-601
Background and purpose: It has been reported that activation of Notch1 could strongly inhibit proliferation of HPV (human papilloma virus)-positive HeLa cells by down-regulation of the E6 and E7 genes. The aim of this paper was to investigate the role of the Notch signaling pathway in growth arrest of EC109 cells in vitro and the molecular mechanism. Methods: EC109 cell lines, a well differentiated human ESCC (esophageal squamous cell carcinoma) cell line with HPV18-positive, was used in the study. Exogenous intracellular domain of Notch1(ICN) was transfected into cultured EC109 cells by lipofectamine transfection, the proliferation of the transfected cells was measured by an MTT assay. Cell cycle distribution was analyzed by flow cytometry. Human papilloma virus type 18 (HPV18) E6/E7 mRNA expression was detected by RT-PCR, and p53 protein expression was detected by Western blot. Results: Activation of Notchl signaling resulted in inhibition of EC109 cell proliferation with the induction of G_2/ M arrest. There was a significant difference in terms of the percentage of G_2/M phase cells among the ICN-transfected group (42.57±1.57)% and the non-transfected group (1.88±0.66)% or the empty plasmid transfected group (1.99±1.02)% (P<0.01). Down modulation of HPV18 E6/E7 gene expression and upregulation of p53 expression was (2.15±0.23) in ICN-transfected group higher than non- transfected group (0.45±0.07) and empty plasmid transfected group (0.46±0.02) (P<0.01). Conclusion: Repression of HPV18 E6/E7 expression by Notch1 signaling results in growth suppression of HPV18-positive EC109 cells with concomitant activation of p53-mediated pathways.
