1.Extraction Optimization of Volatile Oil in Myrrh by Uniform Design

Objective:To optimize the extraction technology of volatile oil in myrrh. Methods: Using the extraction quantity of volatile oil as the index, the volatile oil was extracted by an essential oil extractor, and U6 (64 ) uniform design was used to investigate the influencing factors such as the immersion time ( A) , the mesh number ( B) , the extraction time ( C) and the additive water amount ( D) . Results:The optimized extraction technology was as follows: myrrh was crushed to obtain 10 mesh powder, soaked for 10 mi-nutes with 8-fold amount of water, and then extracted 3 hours. Conclusion:The quantity of volatile oil extracted from myrrh is accu-rate, the technology is reasonable, and the research provides scientific basis for the extraction of volatile oil in myrrh.
2.Optimization of Water Extraction Technology for Shugan Xiaocuo Granules by Multi-index Comprehensive Evaluation

Objective:To study the optimal water extraction method for Shugan Xiaocuo granules. Methods: The study was car-ried out according to the orthogonal tablet. The water amount, extraction time,extraction times and soak time as the influencing factors, and the extractive yield and baicalin content as the indices in a comprehensive evaluation, the optimal extraction technology was screened. Results:The optimal extraction technology was as follows:soaked 30 minutes before cooking, 8-fold water,extracted for 3 times with 1h for every time. Conclusion:The verification test showed that the optimum extraction process was stable,reasonable and feasible.
3.Study on Optimization of Extraction Technology of Jiuwei Yiwei Mixture by Orthogonal Design

OBJECTIVE:To optimize water reflux extraction technology of Jiuwei yiwei mixture. METHODS:With the over-all score of the content of salvianoli acid B,a water-soluble component of the principal drug Salvia miltiorrhiza,the content of pae-oniflorin in Paeonia Radix Rubra and dry ointment yield rate as the observed index,and taking soaking time,extraction time, amount of water added,and extraction times as the factors,L9(34)orthogonal test was designed to optimize the extraction technolo-gy of Jiuwei yiwei mixture. And the verification test was conducted. RESULTS:The optimal technology was to add water with an amount 8 times as much as that of the drugs to soak them for 1 h and to conduct extraction twice,1 h for each. The overall scores of 3 batches of samples in the verification test were respectively 98.793,99.078 and 99.220(RSD<2%,n=3). CONCLUSIONS:The optimal technology for Jiuwei yiwei mixture is stable and feasible.
4.Optimization of Ethanol Reflux Extraction Process for Aitong Cataplasm by Weighted Score Orthogonal Test

Objective:To optimize the ethanol reflux extraction process for Aitong cataplasm .Methods: Orthogonal design was used to investigate the effect of solid-liquid ratio,extraction time and times on the extraction technology of Aitong cataplasm with the content of asarinin and dry extract yield as the indices .Results:The best extraction conditions were as follows:extracted 3 times with 8-fold amount of 70%ethanol, and 1 hour for each time.Conclusion: The optimized extraction process is stable and feasible , which can be used to extract Aitong cataplasm .
5.Determination of Release Rate and in vitro Transdermal Rate of Asarinin in Cancer Pain Cataplasm

OBJECTIVE:To determinate the release rate and in vitro transdermal rate of asarinin in Cancer pain cataplasm. METHODS:Using homemade devices and modified France diffusion,isolated skin of rats as barrier,normal saline as solvent,the content of asarinin was determined by HPLC. Release rate of Cancer pain cataplasm within 20,50,80 and 120 min and transder-mal amount within 2,4,8,12,24 h were investigated,and accumulative release rate and accumulative transdermal rate were cal-culated. RESULTS:Accumulative release rate by 120 min of asarinin in Cancer pain cataplasm was 73.01%;24 h in vitro transder-mal rate was 26.01%,and transdermal kinetics equation of asarinin was Q=5.717 7t1/2-0.385 4(r=0.979). CONCLUSIONS:Cancer pain cataplasm has good release and transdermal performance. Its transdermal kinetics is in line with Higuchi equation.
6.Determination of Drug Release and in vitro Transdermal Rate of Tetrahydropalmatine in Xiaoji Aitong Cat-aplasmas in Rats

Objective:To determine the release and in vitro transdermal rate of tetrahydropalmatine in Xiaoji Aitong cataplasmas in rats. Methods:Using the self-made drug release determination devices and the modified Franz diffusion cells, and the skin of rats as the barrier, the release and in vitro transdermal rate of tetrahydropalmatine in Xiaoji Aitong cataplasmas were detected by HPLC. Re-sults:Tetraydropalmatine within the range of 0. 51-10. 22 μg showed a good linearity (r=0. 999 7), and the average recovery was 98. 49%(RSD=0. 84%,n =9). The release of tetrahydropalmatine in 80 min was 63. 60%, and the skin permeation rate was 23. 15% in 24h. Conclusion:Xiaoji Aitong cataplasmas have good drug release and transdermal performance.
7.Study on Optimization Supercritical CO2Extraction Technology of Paniculate Swallowwort Root by Design-response Surface Method

Objective To optimize the supercritical CO2extraction technology condition of radix cynanchi paniculati. Methods The paeonol extract was used as indexes.The content of paeonol was determined by high performance liquid chromatography method.The extraction condition was optimize tby design-response surface method. Results The optimum process conditions for the extraction pressure was 10.81 MPa,extraction temperature was 55 ℃,flow rate of CO2was 59.91 L·h-1,extraction time was 2.42 h. Conclusion The content of paeonol with optimum extraction process of radix cynanchi paniculati is high,process is stable and feasible.
8.Reconstruction of palmar finger soft tissue defects by using first dorsal metatarsal artery Flow-through flap with digital artery and nerve anastomosis

Objective To investigate the treatment of outcomes of repairing soft tissue defects of the palmar finger accompanied by proper digital artery and nerve defects.Methods From January,2014 to June,2016,7 patients(4 males and 3 females.Patients'age ranged from 18 to 45 years,with an average of 28.5 years) with soft tissue defects on the palmar side of the proximal and middle phalanx of the fingers accompanied by proper digital artery and nerve defects were treated by first dorsal metatarsal artery Flow-through flap,application of color Doppler ultrasound was used in the detection of vascular type before operation.The flap area was from 2.0 cm×2.5 cm to 3.5 cm×5.5 cm.All the donor site of the flap were sutured directly.The patients were followed-up in 1 month,3 months,6 months,12 months,24 months after the surgery,and the results were evaluated according to the Upper Extremity Functional Evaluation Standard set up by Hand Surgery Branch of Chinese Medical Association.Results All flaps survived.These cases were followed-up for 6 to 24 months,average 12 months.All the flaps got satisfactory appearance and good sense function,and 2-PD of the flap averaged 7 mm,ranging from 6 mm to 12 mm.All injured fingers got satisfactory appearance and good sense function,2-PD of the injured fingers averaged 8 mm,ranging from 6 to 15 mm.The donor site incision healed well no obvious scar hyperplasia,good function.Conclusion Application of the first dorsal metatarsal artery Flow-through flap to reconstruct soft tissue defects of the palmar finger accompanied by proper digital artery and nerve defects,can achieve good clinical effects.This method can restore the appearance,blood supply and sensation of the injured finger.
9.Effect of Bixie deacidification fang on hyperuricemia mouse model and its effect on the expression of renal urate transporter

Objective To investigate the anti-hyperuricemia effects of Bixie deacidification fang on hyperuricemia mice and its mechanism of renal protein transport. Methods The effects of Bixie deacidification fang were investigated on hyperuricemia mice induced by potassium oxonate. Bixie deacidification fang was administered to hyperuricemia mice daily at doses of 220, 440 and 880 mg/kg for 10 days, and allopurinol (5mg/kg) was given as positive control. Serum and urine levels of uric acid and creatinine were determined by colorimetric method. Simultaneously, protein levels of urate transporter 1 (URAT1) and organic anion transporter 1 (OAT1) in the kidney were analyzed by Western blot. Results Compared with the model group, high-dose of Bixie deacidification fang inhibited xanthine oxidase (XOD) activities in serum (18.12±1.33 u/L) and that in liver (70.15±5.20 u/g protein) (P<0.05), decrease levels of serum uric acid (2.04 ± 0.64mg/L) (P<0.05) and serum creatinine (0.35±0.18µmol/L) and blood urea nitrogen (BUN)(8.83±0.71mmol/L) (P<0.05), ncreased levels of urine uric acid (38.34±8.23mg/L), urine creatinine (34.38±1.98mmol/L), down-regulated of URAT1 and up-regulated of OAT1 protein expressions (P<0.05) in the renal tissue of hyperuricemia mice. Conclusion Bixie deacidification fang recipe may promote the excretion of uric acid in the kidney by up-regulating the expression of OAT1 protein to promote the excretion of uric acid, and down-regulating the expression of URAT1 protein to inhibit the reabsorption of uric acid.
10.Characterization of collagen/polyvinyl alcohol complex membrane crosslinked by UV-riboflavin.

The objective of this investigation was to study the characteristics and biocompatibility of collagen/polyvinyl alcohol (PVA) membrane crosslinked by UV-riboflavin. Membranes that were made into complex ones with different mass ratios of collagen to PVA (1:1 and 2:1) were synthesized, and crosslinked with UV-riboflavin. The surface characteristics were analyzed using the omnipotent materials instrument, IR, SEM, water absorption test, gas permeability test, and degradation test, respectively. The biocompatibility of membrane complex and rat bone marrow mesenchymal stem cells (BMSCs) was evaluated after 7 d and 14 d, respectively. The collagen/PVA complex membranes showed good homogeneity, mechanical property, degradation ratio, water absorption, gas permeability, etc. The biocompatibility of the collagen/PVA (2:1) complex membrane crosslinked with UV-Riboflavin was higher than that of without crosslinking and collagen/PVA (1:1) membrane. It could be well concluded that collagen/PVA complex membranes crosslinked with UV-riboflavin would have a potential application in biomedicine.
