Chinese Journal of Medical Education Research 2003;0(02):-
The transformaton of medical mode brings forward the new demand for orthopedics education. We should think highly of the cultivation of orthopedics post-graduates' professional ethics so as to promote their communication between medical staff and patients and give emotional concern to patients.Strengthening the cultivation of Orthopedics post-graduates' clinical skill,evidence-based medical thinking and scientific research competence is the key to cultivate the orthopedics talents with high quality and ability.

4.Effects of Various Dosage and Multiple Course of Dexamethasone on Expression of WNT Signal Transduction Pathway in Lung of Premature Rats
Journal of Applied Clinical Pediatrics 2006;0(14):-
Objective To explore the effects of various dosage and multiple course of Dexamethasone(DEX) on the expressions of WNTs,?-catenin and glycogen synthase kmase-3?(GSK-3?) genes in the lung of premature rats on the 19th day of embryo. Methods Twelve pregnant SD rats were divided into 3 groups randomly:small dose DEX group, large dose DEX group and control group with 4 rats in each group. The rats in control group were injected with saline 9 g/L;rats in small dose DEX group were injected with DEX 0.4 mg/(kg?d),and the rats in large dose DEX group were injected with DEX 0.8 mg/(kg?d) after DEX was diluted to 0.5 mL with saline. On the 19th day of gestation, fetuses were surgically taken out.The reverse transcription polymerase chain reaction PCR method was used to detect expressions of WNT7b,WNT5a,WNT2,GSK-3? and ?-catenin genes mRNA.Results The expressions of WNT7b(0.55?0.19,0.64?0.54)and ?-catenin(2.03?0.58,2.40?0.89)genes mRNA in small dose DEX group and large dose DEX group were significantly higher than those of control group(WNT7b:0.18?0.10,?-catenin:1.77?0.54)(Pa

6. Comparison of tear film stability and corneal sensitivity between laser-assisted subepithelial keratectomy with laser in situ keratomileusis
Academic Journal of Second Military Medical University 2010;28(4):404-407
Objective:To compare the changes in tear film stability and corneal sensitivity after laser-assisted subepithelial keratectomy (LASEK) and laser in situ keratomileusis (LASIK). Methods: Forty eyes of 20 patients underwent LASIK and 40 eyes of 20 patients underwent LASEK to correct mypia. The patients were observed for corneal sensitivity, tear break-up time (BUT) and Schirmer test under local anesthesia before and 1, 3 and 6 months after operation. The results were analyzed statistically. Results: The tear film stability and corneal sensitivity were decreased to some extent in both groups after operation. Compared with before operation, there were significant differences in corneal sensitivity 1, 3 and 6 months after operation and in BUT 1, 3 months after operation in LASIK group (P<0.05); there were also significant differences in corneal sensitivity 1, 3 months after operation and in BUT 1 month after operation in LASEK group (P<0.05). The corneal sensitivity and BUT values were obviously different between the 2 groups 1, 3 months after operation (P<0.05), with the decrease of corneal sensitivity and BUT values more significant in LASIK group. There was no significant changes in Schirmer test results before and after operation in both groups (P>0.05). Conclusion: Both LASIK and LASEK influence the corneal sensitivity and tear film stability after operation; the influence diminishes with the recovery of the cornea. The influence in LASIK group is more serious and more persistent than that in LASEK group.
7. Construction of luciferase reporter gene cell model and its application in screening rhein derivatives targeting RAC1
Chinese Pharmacological Bulletin 2019;35(7):1025-1029
Aim To establish luciferase reporter gene expression cell models of CNEi-RACI-Luc2 based on the target of RAC1 promoter, and explore the application of screening anti-tumor active of rhein derivatives targeted regulating RAC1 at transcriptional level. Methods The lentiviral carrying luciferase reporter vector was designed and synthesized using RAC1 promoter se-quence, and CNE1 cells were infected with recombinant plasmid lentiviral to obtain cell lines that stably expressed firefly luciferase. Luciferase reporter assay was used to detect the luciferase luminescence value after stimulating with RAC1 activator PMA and inhibitor NSC23766 that targeted regulating the RAC1 promoter activities in cells, and RAC 1 expression was verified by Western blot. The effect of series of rhein derivatives on the lu-cifcrase activity of RAC1 promoter was observed, and RAC1 expression was determined by Western blot. Results The identification result of double enzyme digestion showed that a lentiviral expression vector carrying luciferase reporter vector rc-combined with RAC1 promoter was successfully constructed. Lcntivirus-infcctcd CNE1 cells were screened by puromycin, the CNE1-RAC1-Luc2 cells stably expressing firefly luciferase were obtained, and the Iransfection efficiency was over 90%. The RAC1 luciferase reporter assay system was sensitive to FMA and NSC23766 and consistent with the result of RAC1 protein expression by Western blot. The regulation of series of rhein derivatives to RAC1 luciferase activity of CNE1-RAC1-Luc2 cells was consistent with the results of Western blot. Conclusions The cell model of luciferase reporting system containing RAC1 promoter can be successfully constructed, which provides a practical platform for high throughput screening of RAC1-targeted drugs.

Hedgehog (Hh) signaling is critical for embryonic development and in differentiation, proliferation, and maintenance of multiple adult tissues.Deregulation of the Hh pathway is associated with Multiple pathological steps of PDAC(Pancreatic ductal adenocarcinoma).A prominent desmoplastic stromal reaction is the pathological hallmark of PDAC,it contributes to the formation of the hypovascular and hypoxic microenvironment of PDAC.And Hedgehog signaling is one of the most crucial pathways that maintains and activates the stroma.Clinical trials of Hedgehog signaling inhibitors in treatment of PDAC has been conducted, but results have been disappointing.Recently,to undercover the reason of the failure of clinical trial, more work has been done to uncovering the underlying role of stroma in PDAC.

Objective To investigate different methods in parathyroid adenoma tissue storage and to provide theoretical support for standardized construction of parathyroid adenoma tissue data. Methods 22 samples of sporadic parathyroid adenoma in Beijing Union Hospital from Aug.2010 to Nov.2011 were collected. All cases obtained pathological diagnosis, among which 11 were kept in frozen nitrogen (group A)and the rest in RNAlater(group B).The ratio of OD 260/280 was measured by ultraviolet spectrophotometer. The integrity of RNA was examined by gel electrophoresis. The expression of β-actin was measured by Realtime-PCR technique. Results RNAlater group had no advantage than frozen nitrogen group in terms of the purity, concentration, and integrity of the extracted RNA(P >0.05),but had a higher expression level of the β-actin(P <0.05).Conclusion The method of RNAlater has no significant advantages compared with frozen nitrogen in terms of RNA prevention and RNA degradation, but is more effective in longer storage of samples.
