1.Neurotoxic effects of intrathecal ropivacaine and bupivacaine on the spinal cord in rats

Objective To compare the neurotoxic effects of intratheeal ropivacaine and bupivacaine on the spinal cord in rats.Methods Female SD rats weighing 240-330 g were used in this study.Catheters(PE 10 tubing)were inserted through an incision in the cisternal membrane and advanced caudad until the tip of the catheter reached the lumbar spinal level according to the technique described by Yaksh and Rudy.Fifty-four successfully catheterized rats were randomly assigned tO one of 3 groups(n=18 each):group Ⅰ received normal saline 30μlIT(NS);groupⅡ received 2%bupivacaine 20μl IT(BU)and group Ⅲ 2.7%ropivacaine 20μl IT (RO).Degree of motor block was assessed and scored(0=no block,2=inability to flex the hind limb)before (T1,baseline)and at 10,20,30,60 and 120 min(T2-6)after IT injection.On the 4th day after IT injection specimens were obtained from lumbar spinal cord for microscopic examination.Results The motor block score was significantly higher in group BU and RO than in group NS(P<0.05),and the tissue damage was severe in group BU and RO.The motor block scores were significantly higher at T2 than at T3-6 in beth group BU and RO(P<0.05).There Was no significant difference in motor block score at T2 between groups BU and RO but the motor block scores were significantly lower at T3-5 in group RO than in group BU(P<0.05).The tissue damage was Severer in group RO than in group BU(P<0.05).Conclusion Intrathecal 2.7% ropivacaine is more toxic to the spinal cord than intrathecal 2%bupivacaine in rats.
2.The antagonist of TRPV1 attenuates cytotoxicity of DRG neurons induced by lidocaine

Objective To determine whether or not capsazepine(CPZ),a transient receptor po-tential vanilloid-1(TRPV1)antagonist,attenuates lidocaine-induced cytotoxicity on rat dorsal root ganglion (DRG)neurons in vitro.Methods Adequate DRG neurons from 3-day neonatal Wistar rats were obtained,cultured and purified in vitro.To achieve higher cell viability,we reduced the concen-tration of trypsin to 0.125% compared with others.The purified DRG neurons were incubated with 0,2.5,5,10,20 and 40 mmol/L lidocaine for 10 min,respectively,their viabilities were examined using Cell Counting Kit(CCK-8)assay,and the lethal concentration 50(LC50 )of lidocaine on DRG neurons was calculated.Then,the variation of lidocaine-induced cell viability at LC50 ,when 0,1,10 and 100 μmol/L CPZ were respectively added to the incubations,was examined with CCK-8 assay. Results The purified percentage of DRG neurons was as high as 91% after digesting by 0.125%trypsin and purifying in vitro.Cell viability of DRG neurons in group L1,L2,L3,L4,L5 was signifi-cantly down regulated compared with the control group,to be specific,that of L3,L4,L5 being re-markably lower than that of L1,that of L4,L5 lower than that of L2 and that of L5 lower than that L3 and L4.After lidocaine induced DRG neurons for 10 min,LC50 was 30 mmol/L;10 μmol/L and 100 μmol/L CPZ significantly reduced LC50 DRG neuron toxicity induced by lidocaine (P <0.05). The effect of 10 μmol/L CPZ had reached the maximal effect,decreasing the cell viability decrease from 50% to 35%.Conclusion The novel method in this experiment is effective to obtain good DRG neurons,the LC50 of lidocaine on rat DRG neurons is 30 mmol/L,and CPZ attenuates the cytotoxicity induced by lidocaine on rat DRG neurons.
3.Mutual information-based correlation analysis of herbs against insomnia.

This paper aims to analyze Professor Guo Rongjuan's medication experience on insomnia therapy based on the Traditional Chinese Medicine (TCM) Inheritance Support Plat. First, TCM formulae prescribed by Professor Guo for insomnia therapy were collected from the TCM Inheritance Support Plat. Next, unsupervised data mining algorithms, including apriori, modified mutual-information, and entropy clustering of complex system were applied to obtain the frequencies for different herbs and identify the association rules among the herbs. Accordingly, we can gain new insights into Professor Guo's medication experience on insomnia therapy. Based on analysis of 3 084 formulae, we determined the frequencies for herbs in the formulae and identified the association rules among these herbs. At last, 41 core combinations and 7 new formulae were obtained. The identified medication experience conform with Professor Guo's views on the etiology and pathogenesis of insomnia: "pathogenic fire derived from stagnation of liver-QI (Gan Yu Hua Huo)" is the core pathogenesis of insomnia; "liver stagnation and spleen deficiency" and "chronic illness transferred to kidney" are the main features for insomnia. The TCM Inheritance Support Plat is of great practical value for mining clinical experience of famous TCM doctors.

5.Morphological and biomechanic characters of the sacrum fractures

Objective To discuss morphology and biomechanics of the sacrum fractures so as to provide scientific basis for corresponding clinical treatment. Methods A total of 10 fresh pelvis specimens were collected for dynamic impact test and static destruction test, in the former one of which, the dynamic parameters were measured to make sure the dynamic characters of the fractures. Meanwhile, the fractures of the sacrum wing, the sacral foramina and the sacrum edge were made decalcification, slice of paraffin wax and staining (Masson, Mallory, HE) in order to make a cytological observation of the tissue. Results (1) The form of sacrum fracture or acetabulum fracture, crista iliaceis fracture was relevant to the impact energy. Low impact energy usually caused fractures of the ilium, acetabulum or sacroiliac crest. High impact energy resulted in following three kinds of fractures, just as the classification of Denis: sacral ala fractures belonged to typeⅠfractures, sacral hiatus fractures to type Ⅱ fractures and central vertebral canal fractures to type Ⅲ fractures. All three types of fractures might involve lateral or bilateral nerve roots. (2) There was a significant mechanic difference in regard of the mechanism of both dynamic destruction and static destruction of pelvis, ie, not only the limit pressure differed but also the former increased rapidly with the higher rate of the strain. The clash energy beyond 25 J would beget the cleft fractures of the sacrum via sacrum hiatus and even involve the nerve roots. The clash energy under 20 J usually resulted in fractures of the ilium and the sacrum. The clash energy between 20 J and 25 J more easily caused type Ⅰ fractures. While fracture of the ilium and the acetabulum would happen most in static destruction. (3) The cross section of the sacrum was cracked and the bone board of Haversian system is brittle, as led to separation of bone board and malposition of a few cross bone boards. Conclusions Under dynamic state, the sacrum fractures mostly belong to type Ⅰ and type Ⅱ (Denis classification of sacral fractures), usually involving the nerve roots. The sacrum fracture is relevant to the microstructure, the distribution of the bone trabecula, the osseous lacuna and the Haversian system of the sacrum. The fractures of the ilium and the acetabulum more frquently appear in static state, with slight wound of peripheral tissues.
6.Determination of Trinitrotoluene in Air by Capillary Gas Chromatography

Objective To establish a capillary gas chromatography method for the determination of trinitrotoluene (TNT) in workplace air.Methods Using dichloromethane as the eluent,the air was drawn through a glass fiber filter to collect TNT,TNT was extracted from the filter with dichloromethane by an ultrasonic shaker,and the sample was analyzed by OV-17 elastic quartz electron capture detector capillary gas chromatography.Results Under the optimal condition,the lowest detection limit was 0.006 ?g/ml,the lowest detection concentration was 0.001 3 mg/m~3 (based on 45 L air sample).When the concentration of standard solution was 0.040-2.0 ?g/ml,the linear equation was good,r=0.999 8,RSDs were between 0.66%-3.62%,the recovery rates were 90.4%-95.5%.When sample was collected by fiberglass filter paper,the efficiency of desorption was more than 90%,and was stable for at least 7 days at 2℃-8℃.Conclusion This method is applicable to the determination of trinitrotoluene in workplace air.
8.Determination of perfluorocarboxylic acids in fish by ultra performance liquid chromatography tandem mass spectrometry

Objective:
To develop the ultra performance liquid chromatography tandem mass spectrometry ( UPLC-MS/MS ) for the determination of perfluorocarboxylic acids ( PFCAs ) in fish.
Methods:
The fish samples were extracted with tert-butyl methyl ether and purified by WAX columns. The WAX cartridges were rinsed with methanol and 25 mmol/L ammonium acetate, and the target compound residues were eluted with 0.5% ammonia methanol and then redissolved with 50% methanol aqueous solution after nitrogen blowing to nearly dry. Nine kinds of PFCAs were simultaneously quantified by UPLC-MS/MS with 1 mmol/L ammonium acetate-methanol solution as the mobile phase.
Results:
The extraction was separated well in UPLC BEH C18 column. There were good linear correlations of nine kinds of PFCAs in the range of 1.0-200.0 ng/mL, with the coefficients all more than 0.99. The limits of detection and quantification were 0.06-0.19 μg/kg and 0.19-0.62 μg/kg, respectively. The recovery rates were 70.08%-117.24% at different spiked levels ( 5.0, 25.0, 50.0 μg/kg ), and the relative standard deviations were 2.31%-19.68%.
Conclusion
Through optimizing the pretreatment conditions, the mobile phase of liquid chromatography and the detection conditions of mass spectrometry, the UPLC-MS/MS could meet the monitoring requirements of PFCAs in fish.

10.Impacts on analgesia and detumescence in ankle sprain treated with acupuncture at Xiaojie point combined with tendon-regulation manipulation.

OBJECTIVETo compare the efficacy difference of analgesia and detumescence on ankle sprain among acupuncture at Xiaojie point combined with tendon regulation manipulation, acupuncture at Xiaojie point and tendon regulation manipulation.
METHODSSixty cases of ankle sprain were randomized into a combined therapy group, a Xiaojie point group and a tendon-regulation manipulation group, 20 cases in each one. The combined therapy of acupuncture at Xiaojie point and tendon regulation manipulation, the acupuncture at Xiaojie point and the simple application of tendon-regulation manipulation were applied respectively in the three groups, once a day, 3 treatments were required. The symptom score such as pain, ecchymosis, swelling and motor dysfunction and the total score were observed before and after treatment in the three groups. The different values of pain and swelling scores were compared before and after treatment in the three groups. The efficacy was compared among the groups.
RESULTSThe total effective rate was 100.0% (20/20) in each group. But the curative rate was 85.0% (17/20) in the combined group, 65.0% (13/20) in the Xiaojie point tion manipulation group. After treatment, the symptom scores of pain, ecchymosis, swelling and motor dysfunction and the total score were all improved as compared with those before treatment in the three groups (P < 0.01, P < 0.05). The pain score in either the combined therapy group or Xiaojie point group was lower than that in the tendon-regulation manipulation group after treatment (0.20 -/+ 0.41, 0.15 +/- 0.37 vs 0.60 +/- 0.50, both P < 0.05). Swelling score in the Xiaojie point group was different significantly from that in the tendon-regulation manipulation 0.49 vs 06.4 vs. 20+0.41, P < 0.05). The different value of pain score in either the combined therapy group or Xiaojie point group was higher than that in the tendon-regulation manipulation group before group after treatment (0.65 bined therapy group or Xiaojie point group was higher than that in the tendon-regulation manipulation group before and after treatment (2.35 +/- 0.59, 2.45 +/- 0.51 vs 2.00 +/- 0.46, both P < 0.05). The different value of swelling score in the tendon-regulation manipulation group was higher than that in the Xiaojie point group before and after treatment (2.30 +/- 0.57 vs 1.60 +/- 0.60, P < 0.05).
CONCLUSIONAcupuncture at Xiaojie point combined with tendon-regulation manipulation achieve an apparent effect of analgesia and detumescence on ankle sprain.
