Biocontrol bacteria 308R(pKSH) with hrpN of Erwinia amyloyora. It can secrete the Harpin protein which can induce plant resistance. After successive growth in antibiotic-free LB medium for 50 generations, only 23.1% of the cell still retained the plasmid pKSH, in comparison with 4.75% of the cosmid pCPP430. Sprayed onto the leaves of tomato, the recombinant strain maintained a population density of over 10~ 5 cfu/cm~ 2 when kept under high humidity in thirteen days, but if without humidity, the strain keep 10~ 4 cfu/cm~ 2 over in five days. On the tomato leaves, the stability of the recombinant strain 308R(pKSH) was higher than the strain 308R(pCPP430). The experiment indicated that the recombinant strain 308R(pKSH) had higher stability compared with the strain 308R(pCPP430), but that was not enough. In this paper reasons for unstability and strategies for improving were discussed for the recombinant strain.

Phytoene synthase is a rate-limiting enzyme in the pathway of carotenoid biosynthesis.To aim at establishing a transformation of Ginseng callus cells, elevating the nutritive value through encouraging the composition of corresponding carotenoid,taking Ginseng callus cells as acceptor,psy gene were transformed into cells via Agrobacterium-mediated. The factors affecting genetict ransformation were also investigated seperately from concentrations of A.tumefaciens, age of Ginseng callus cells, infection time and co-culture time. PCR,PCR-Southern and RT-PCR analysis from the transferred gene plants proved that psy gene has been transferred into Ginseng callus cells and can be expressed. The content of ?-carotene was increased by an average of 26 times.A base of improving and raising the contents of carotenoid in the Ginseng callus cell was established.

Objective:To analyze the cause of the blood heat syndrome in psoriasis.Methods: Made the investigation form about information of psoriatic patients through DME method and reference both Chinese and foreign documents,analyzed main component of data by SAS software.Results: The first main component of blood heat syndrome in psoriasis are infection,drinking,dietetic problem,and emotion;the second main component is family history;the third main component is dampness;the forth main component is the allergy to the food;the fifth is outside irritation.Through the corresponding investigation we found that food include hot food,beef,sheep and sea food,emotion problem include over work and stress,dampness include work and living in humid place for long time,outside stimulus meant chemical stimulus.Conclusion: The main cause of the blood heat syndrome in psoriasis are infection,drinking,dietetic problem,and emotion,family history,humidity,allergy to the food and chemical stimulus in order.

Objective To explore whether the presence of juxtapapillary duodenal diverticula (JPDD) risks biliary stone disease and recurrence.Methods 829 patients undergoing ERCP in our hospital from Aug 2008 to Dec 2012 were divided into four groups:biliary stone disease (n =609) non-stone biliary abnormality (n =124) common bile duct malformation with cholelithiasis (n =38) and normal control group (n =58).There were 206 patients with JPDD and 623 patients without JPDD.Biliary stoneformation,post-ERCP pancreatitis,cannulation failure,and stone recurrence were compared between those with JPDD and those without.Results The incidence of JPDD in biliary stone disease group (27.8％) was significantly higher than in non-stone biliary anatomical abnormality group (18.5 ％) (x2 =4.512,P ＜ 0.05).In biliary stone disease group,rates of post-ERCP pancreatitis were significantly higher in JPDD cases (33.7％) compared to those without JPDD (13.8％) (x2 =30.841,P ＜ 0.05).The cannulation failure rate was also higher in patients with JPDD (15.4％) compared to JPDD negative (6.8％) (x2 =0.731,P ＜0.05).Recurrence rates in biliary stone disease were significantly higher in patients with JPDD (19.5％) when compared to JPDD-lacking individuals (9.3％) (x2 =14.51,P ＜ 0.05).Conclusions JPDD is a risk factor for biliary stone formation.JPDD also is associated with post-ERCP pancreatitis,cannulation failure and biliary stone recurrence.

Objective To analyze the curative effect and toxicity of concurrent chemoradiotherapy and sequential chemoradiotherapy in non-small cell lung cancer patients.Methods 64 patients with unresectable stage Ⅲ non-small cell lung cancer in our hospital between May 2010 and Mar 2013 were randomly assigned into concurrent group and sequential group for this test.Concurrent group patients (32 cases) were given Cisplatin and Gemcitabine on the 1,4,7 week during radiotherapy.Sequential group patients accepted 3 cycles chemotherapy before radiotherapy.Results The response rate was 68.8％ in concurrent group,and 43.8％ in sequential group.Difference was significant between the response rates of the two groups (P ＜ 0.05).There was significant difference in haematological toxicity,which was 100％ in concurrent group and 65.6％ in sequential group (P ＜ 0.05).There was no significant difference in rates of radiation pneumonia and radiation esophagitis in the two groups (P ＞ 0.05) Conclusions Concurrent chemoradiotherapy has better curative effect than sequential chemoradiotherapy.Concurrent chemoradiotherapy is a safe and effective treatment for patients with unresectable stage Ⅲ non-small cell lung cancer.

OBJECTIVETo study the overexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells for repairing articular cartilage injury in vivo.

METHODSRabbit bone marrow mesenchymal stem cells (BMSCs) were transduced with lentivirus vector containing Sox9 gene and then cartilage specific molecule was detected by RT-PCR in vitro. Total 48 knee joints of 24 mature New Zealand white rabbits were randomly divided into 3 groups according to different defect treatment. After animals anesthesia,a full-thickness cylindrical cartilage defect of 4 mm diameter and 3 mm deep was created in the patellar groove using a stainlesssteel punch. Meanwhile, the transfected cells were implanted to repair the rabbit model with full-thickness cartilage defects. Cartilage defects tissue was observed with light microscope, electron microscope, HE and immunohistochemistry staining to assess the repair of defects by the complex at 6 weeks or 12 weeks after the implantation.

RESULTSAt 3 days after the transfection, Sox9 gene expression was highest and Sox9 gene expression decreased with the increase of time. At 3 days after the transfection, the expression of collagen type II began and reached the peak at 14 days. It showed that the bone marrow mesenchymal stem cells went into chondrogenic differentiation after transfected by Sox9 gene. Histological observation showed that at 6 weeks after the operation, the defects in the experimental group was filled with hyaline like cartilage tissue, 12 weeks after operation,the defects of cartilage and subchondral bone had satisfactory healing. Both at 6 and 12 weeks postoperatively, the defects were filled with fibrous tissues in control groups. Meanwhile, immunohistochemical staining of sections with type II collagen antibodies showed the proteins in the regenerated tissue stained positive for type II collagen and stronger than the control groups. The histological scoring system indicated that the cartilage repair of experiment groups were better than the two control groups with statistical significances.

CONCLUSIONOverexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells (BMSCs) promote the repair of cartilage defect.

Objective To compare clinical outcome of intracytoplasmic sperm injection(ICSI)cycle by using fresh and cryopreserved-thawed testicular and epididymal spermatozoa in azoospermic patients.Methods Between September 2006 and May 2007,208 azoospermic patients underwent in vitro fertilization(IVF)were treated in Center of Reproductive Medicine,Peking University Third Hospital.Those couples were divided into two groups based on their wishes,including 171 cases in fresh group and 37 cases in cryopreserved-thawed group.The cryopreserved testicular or epididymal spermatozoa were thawed and recovered before ICSI procedure iu thawed group.The outcomes of ICSI in each group were compared.including clinical outcomes(two pronuclear fertilization,high quality embryo,clinical pregnancy and embryo implantation)and pregnancy outcomes(spontaneous miscarriage,gestational weeks and neonatal birth weight).Results (1),The utilization rate were 92%(23/25)in cryopreserved-thawed testicular spermatozoa and 100%(12/12)in epididymal spermatozoa.(2)Between fresh and cryopreserved-thawed groups,no statistical difference was observed in two pronuclear fertilization rate[62.25%(973/1563)vs.64.53%(282/437),P=0.960],high quality embryo rate[78.9%(768/973)vs.79.1%(223/282),P=0.985],clinical pregnancy rate per embryo transfer[44.4%(60/135)vs.46.9%(15/32),P:0.688]and embryo implantation rate[29.3%(84/287)vs.33.3%(23/69),P=0.508].(3)No significant difference between flesh and cryopreserved group was found in spontaneous miscarriage rate (11%vs.7%,P＝1.000),gestational weeks(single birth:39.0 weeks vs.38.7 weeks,P:0.538;twins:36.8 weeks vs.36.3 weeks,P=0.571)and birth weight(single birth:3409 g vs.3350 g,P=0.699;twins:2584 g vs.2635 g,P=0.703).Conclusion It suggested that tissue from the azoospermic patients who underwent diagnostic testieular and epididymal biopsy should be eryopreseved for IVF-ET.
