2.Evaluation of the regeneration of chemically extracted acellular facial nerve allograft for the whole facial nerve defect in rat with multiple retrograde tracing

[Objective]To evaluate the axonal transport function of the regenerate nerve of buccal branch,marginal mandibular branch and cervical branch after chemically extracted acellular facial nerve allograft to the whole facial nerve defect in rat using fluoro-gold(FG),quantum dot(QD) and fast blue(FB).[Method]The extracranial section of facial nerve and branches were dissected under light microscope,and 10-mm-long segments of facial nerve gap were made on the left side.The defect was repaired by acellular facial nerve allografts with epineural suture method.Two months later,fluoro-gold(FG),quantum dot(QD) and fast blue(FB) were injected into distal bridged side of buccal branch,marginal mandibular branch and cervical branch.After three days,the slices of brain stem were collected and the facial motoneuron was observed under fluorescences microscope.[Result]There were three kinds of different fluorescences in the frozen sections of the facial nerve originated from the brainstem nuclei under the fluorescence microscope.Fluoro-gold(FG),quantum dot(QD) and fast blue(FB) labeled neurons were shown yellow,red and blue colors,respectively.[Conclusion]According to estimation of axonal branching by triple retrograde labeling,it was found that the regenerate facial nerve repaired the gap,and nerve continuity obtained restoration.The evaluation method of this study is simple,reliable and easy.It is an ideal evaluation method.
3.Expressions of bFGF, PDGF and TSP-1 mRNA in small intestinal adenoma and adenocarcinoma and their clinico-pathological significance

Objective To study on the expressions of bFGF, PDGF and TSP-1 mRNA in small intestinal carcinoma and adenoma and their clinico-pathological significance. Methods The specimens were continuously sliced into pieces. The expression of bFGF, PDGF and TSP-1 mRNA was determined by in situ hybridization. Results bFGF, PDGF and TSP-1 was positive in 57%,48% and 36% respectively in 44 cases of small intestinal adenocarcinoma, and was 20%, 30% and 90% in 10 of adenoma respectively. The positivity rates of bFGF mRNA and PDGF mRNA were significantly lower in adenocarcinoma of Ⅰ~Ⅱ grade, without invading muscular layer or invading superficial layer, maximal diameter of tumor mass
4.The effect of early enteral nutrition on hospital acquired pneumonia in patients with hypertensive cerebral hemorrhage

Objective To investigate the clinical value of early enteral nutrition (EN) on hospital acquired pneumonia (HAP) in postoperative patients with severe hypertensive cerebral hemorrhage.Methods One hundred and forty postoperative patients with severe hypertensive cerebral hemorrhage were divided into treatment group (70 cases) and controll group (70 cases) by random digits table. The treatment group was given EN from the second day after operation, while the control group was given total parenteral nutrition (TPN). The incidence and duration of HAP,the incidence of superinfection,the percent and duration of mechanical ventilation and the mortality rate of HAP was observed. Results The incidence of HAP, superinfection and using mechanical ventilation in treatment group [30.0% (21/70), 12.9% (9/70),35.7%(25/70)] were significantly lower than those in control group [47.1%(33/70) ,27.1%(19/70) ,47.1%( 33/70 )] (P < 0.05 ). The duration of HAP and using mechanical ventilation in treatment group[(6.4 ± 2.3 ),(6.4 ± 0.5 ) d] were significantly lower than those in control group [( 15.6 ± 2.1 ), ( 11.4 ± 0.3 ) d] (P < 0.01or < 0.05 ). The mortality rate of HAP in treatment group was significantly lower than that in control group [8.6% (6/70) vs. 18.6% (13/70),P <0.05]. Conclusion Early EN not only effectively decreases the incidence of HAP and superinfection,but also decreases the incidence of mechanical ventilation, shortens the duration of mechanical ventilation and decreases the mortality rate of HAP.

8.Effective components of Semen Ziziphi Spinosae for sedative-hypnotic based on receptor ligand binding assay

Aim To determine the effective compo-nents of Semen Ziziphi Spinosae for sedative-hypnotic and its mechanism. Methods The extraction of Se-men Ziziphi Spinosae and the rat brain homogenates were prepared. High concentrations of Diazepam com-petitively replaced the ligand compounds of Semen Ziz-iphi Spinosae combining BDZ receptor in brain tissue, and all the compounds with sedative and hypnotic effects were collected and identified by HPLC and LC-MS technique, as the compounds extracted from the brain tissue were administered with Semen Ziziphi Spi-nosae. The brain tissue was administered with Diaze-pam, and with Semen Ziziphi Spinosae and Diazepam. Results The HPLC chromatograms show that the peak time of BDZ receptor ligand compounds was 2. 71 min and 46. 87min, when compared with Diazepam. And the LC-MS chromatograms display the relative molecu-lar weight of the ligand compounds was 274. 28 m/z, 453. 34 m/z,496. 34 m/z and 608. 38 m/z respective-ly. According to the fingerprint of Semen Ziziphi Spi-nosae, these compounds may be fatty acid substances and lupine pill triterpene compounds. Conclusions On the basis of the principle of receptor ligand bind-ing, we established a way to quickly analyze and iden-tify the role of natural products in the same drug target compounds. The method not only can clearly define the effective components of natural products, but also clar-ify the mechanism of action of the compounds. The ac-tive ingredient of calm hypnosis in Semen Ziziphi Spi-nosae may be fatty acid substances Palmitic acid ( C16 H32 O2 ) and lupine pill triterpene compounds Alphitolic acid( C30 H48 O4 ) and Spinosin( C28 H32 O15 ) . They exert their sedative and hypnotic effects by combining with BDZ receptor, and the research has laid a theoretical foundation for the further study about mechanism of Se-men Ziziphi Spinosae.
9.Application of electrocochleography and ABR in the diagnosis of auditory neuropathy.

OBJECTIVE:
To analyze and compare the clinical application of alternating click evoked electrocochleography (ECochG) and auditory brainstem response (ABR) in the diagnosis of auditory neuropathy (AN) subjects.
METHOD:
ECochGs and ABRs were recorded in 16 patients (32 ears) with AN/AD as AN group and 20 patients (26 ears) with sensorineural hearing loss(SNHL) as control group. Test stimuli were alternating polarity clicks. (1) To compare the occurrence rate of ECochGs and ABRs in AN patients. (2) To compare the peak latency of CAPs, absolute amplitude of CAPs, and amplitude ratios of -SP and CAP between AN group and control group.
RESULT:
ECochGs (--SPs or CAPs) presented in 78.1% (25 among 32 ears) of AN patients. CAPs latency of AN group was shorter than control group (P < 0.05), CAP absolute amplitude of AN group was lower than normal group (P < 0.05), while --SP/CAP was higher than control group (P < 0.01). Deformed ABRs presented in only 5 ears among 32 tested AN ears, while relative good ABR waveforms were evoked in control group. The occurrence rate of ABRs (15.6%) was significantly lower than that of ECochGs in AN patients (P < 0.01).
CONCLUSION
Both ECochGs and ABRs play important role in the diagnosis of AN. The capacity of temporal processing is significantly impaired while the intensity perception related capability is intact in AN patients.
