1.CdTe Nanoparticles Labeled with Anti-Fluorethene-Antibody and Fluorescent Immunoassay of Fluoranthene in Water Samples

The labeled compounds, CdTe was combined with anti-fluoranthene antibody, had good dispersion and stability with the fluorescence intensity enhancing. A direct competitive fluorescent immunoassay with CdTe-anti-fluoranthene antibody to detect fluoranthene in water sample in the environment was developed. The result showed that fluoranthene can be determined in the concentration range from 0.1 μg/L to 1000 μg/L with a correlation coefficient of 0.9983, a sensitivity of (IC_(50)) of 12.4 μg/L and a detection limit (IC_(20)) of 13.1 ng/L. Trace environmental pollutant in environmental water samples were successfully determined with a good accuracy and suitability. The recovery was between 95.1% and 111.0%, with relative standard deviation less than 9%.
2.Factors Affecting the Effect of Treatment of VCM Based on the Quantity of MRSA for Hospital-Acquired Pneumonia

Objective: To determine the factors affecting the antimicrobial effect of VCM on MRSA.
Study Design: Case series study.
Methods: This study was conducted on pneumonia patients admitted to a university-affiliated hospital between January 2000 and December 2008 and had MRSA in their sputum culture. From seven days prior to the starting VCM administration through the end of the administration, detailed information such as underlying diseases, VCM serum concentration and quantity of MRSA were recorded. Logistic regression analysis was carried out on current diseases, trough concentration, surgery experience, and the detection of Gram-negative bacteria to verify the antimicrobial effect of VCM.
Results: The number of subjects investigated this study was 55 subjects. Multiple logistic regression analysis did not yield any significant factors when carried out using the factors affecting the antimicrobial effect of VCM on MRSA as independent variables.
Conclusions: Multivariable analysis yielded no factors as being significant in affecting the antimicrobial effect of VCM, but did indicate in that patients aged 70 and older, the antimicrobial effect of VCM was poor for MRSA. This suggests that when pharmacists intervene in antimicrobial treatment for improved effects, it is important to consider not only the serum concentration of VCM, but also the background of the patient.
3.Risk factors for paternal vertical transmission of hepatitis B virus

Objective To explore the risk factors of and the influence of different hepatitis B virus (HBV) DNA load on paternal vertical transmission of HBV.Methods Totally,161 HBsAg negative women,whose husband was HBsAg positive,attended the antenatal clinics of the Provincial Maternity and Child Health Hospital of Fujian from September 2007 to December 2008 and their newborns were selected,and the epidemiologic information,the duration of being a HBV carrier,the first class HBV family history of the fathers,HBV markers,HBV DNA load,HBsAb of the gravidas,the outcomes of the newborns were all collected.Cord blood was sampled after delivery for HBV DNA quantification and those with HBV DNA load ≥1.0×103 copy/ml were chosen as the case group and those < 1.0×103 copy/ml as control.Results (1) Among the 161 newborns,36 HBV DNA positive cord blood samples were detected,giving a rate of 22.4% (36/161) for paternal vertical transmission of HBV.The HBV DNA positive rate in cord blood was 32.0% (23/72) in HBeAg-positive fathers and 14.6% (13/89) in HBeAg-negative fathers.(2) Univariate analysis showed that HBeAg-positive,HBV DNA positive,first class family history of HBV and the duration of being a HBV carrier of the fathers were risk factors of paternal HBV vertical transmission[X2= 6.892,29.916,29.499 and 23.821,OR = 2.7,5.2,8.3 and 1.4 (P<0.01)].(3) Multivariate analysis found that paternal serum HBV DNA positive and the first class family history of HBV of the father side were risk factors of paternal vertical transmission of HBV (OR = 11.1,95% CI;4.6-27.1;OR = 17.1,95% CI:3.5-82.6).(4) According to the different serum HBV DNA load of the HBsAg-positive father,7 groups were divided.A dose dependent effect was found that the HBV DNA positive rate of the cord blood increased with the rising of HBV DNA load.No HBV DNA positive cord blood was detected when paternal HBV DNA load was<1.0×104 copy/ml,while 100% of the cord blood were positive when paternal HBV DNA load≥1.0×108 copy/ml.(5) The average birth weight of the newborns in the two groups was the same (3.3±0.4) kg.And the delivery mode,gestational age at delivery,height and Apgar score of the newborns at 1 minute,neonatal pathological jaundice and other complications had no significant difference between the two groups (P > 0.05).No relationship was found between the neonatal outcomes and the paternal HBV vertical transmission (P>0.05).Conclusions HBV DNA load in the serum of HBsAg-positive father,and the paternal first class family history of HBV are risk factors of paternal HBV vertical transmission.When the serum HBV DNA load in HBsAg-positive father is≥1.0×107 copy/ml,the possibility of paternal vertical transmission of HBV would increase.
4.Meta-analysis of Acupuncture Treatment for Type 2 Diabetes

Objective To systematically assess the efficacy and safety of acupuncture in treating type 2 diabetes.Methods Related literature was retrieved with a computer. After the data were sifted out from the literature according to the established criteria, they were collected and crossly checked by two researchers independently. The data were input to RevMan5.2 for statistical analysis. Results A total of ten studies were included with a total of 743 patients. The results of Meta analysis showed 1) Post-treatment total efficacy rate was higher in the acupuncture or combined acupuncture and medicine group than in the control group [OR=3.60, 95%CI (2.11,6.13)] and there was a statistically significant difference (Z=4.71,P<0.00001); 2) Post-treatment range of decrease in fasting blood glucose (FBG) was significantly higher in the experiment group than in the control group [WMD=-1.20,95%CI (-1.38,-1.02)] and there was a statistically significant difference (Z=12.88,P<0.00001); 3) Post-treatment range of decrease in 2-h postprandial blood glucose (2hPBG) was significantly higher in the experiment group than in the control group [WMD=-1.27, 95%CI (-1.47,-1.06)] and there was a statistically significant difference (Z=12.02,P<0.00001).Conclusion Clinically acupuncture or combined acupuncture and medicine is more effective than Chinese herbal medicine or Western medication alone in treating T2DM and has no toxic and side effects.
5.Relationship of C677T polymorphisms of MTHFR gene with diabetic nephropathy in southeast Guangxi

Objective To study the relationship between the C677T polymorphisms of N5,N10-methylenetetrahydrofolate re-ductase(MTHFR)gene and type 2 diabetic nephropathy(T2DN).Methods By using PCR-RFLP method,the C677T polymor-phisms of MTHFR was analyzed in 163 cases of type 2 diabetes mellitus(T2DM),which included diabetic nephropathy(DN group, n=82)and diabetes without complications(DM group,n=81),and 77 cases of healthy people as the control group(CON).Plasma total Hcy levels were also measured for all the subjects.Results The frequencies of MTHFR TT and CT homogenetic type and al-lele T(4.9%,37.8%,23.8%)in the DN group were significantly higher than those in the DM(2.5%,28.4%,16.7%)group or the CON group(0.0%,29.8%,14.9%).However,there was no significant difference in MTHFR genotype and allele frequency be-tween the DM group and the CON group.Moreover,plasma Hcy levels were markedly higher in the patients with allele TT and CT genotype than those in the patients with CC genotype,the difference showing statistical significance (P <0.01).The univariate lo-gistic regression analysis showed that the C677T polymorphism of MTHFR gene was closely associated with the development of DN.The odds ratio was 1 .660 and the 95% confidence interval was 1.038 and 2.655 respectively.Conclusion The C677T polymor-phisms of MTHFR gene is associated with T2DN in Southeast Guangxi and the allele T might be the susceptibility gene for DN.
6.Effectiveness and safety of ultrasound-enhanced thrombolysis for acute ischemic stroke

Objective To evaluate the effectiveness and safety of ultrasound-enhanced thrombolysis for acute ischemic stroke.Methods Fifty stroke patients with acute middle cerebral artery occlusion were randomly divided into either a ultrasound-enhanced thrombolysis group (recombinant tissue-plasminogen activator [rtPA] +2 MHz ultrasound monitoring for 2 h) or a standard thrombolysis group (rtPA alone).The demographic characteristics,vascular risk factors,blood pressure before treatment,thrombolysis in brain ischemia (TIBI) grade before thrombosis,and vascular occlusion site of the patients were collected.The primary outcome endpoint was the good outcome rate (defined as the modified Rankin Scale score 0-1) at 3 months.The secondary outcome endpoints were complete recanalization at 2 h after thrombolysis,sustained complete recanalization,symptomatic intracerebral hemorrhage,and mortalitY.Results The good outcome rate of the ultrasound-enhanced thrombolysis group at 3 months after treatment was significantly higher than that of the standard thrombolysis group (64% vs.36%;P=0.011).The sustained complete recanalization rate (40% vs.8%;P =0.018) and complete recanalization rate (48% vs.12%;P =0.012) of the ultrasound-enhanced thrombolysis group were significantly higher than those of the standard thrombolysis group,but there were no significant differences in the reocclusion rate (8% vs.12%;P =0.637),incidence of symptomatic intracerebral hemorrhage (4% vs.4%;P=1.000),and mortality (4% vs.4%;P=1.000) compared with the standard thrombolysis group.Conclusions Ultrasoundenhanced thrombolysis can improve the sustained complete recanalization rate,complete recanalization rate,and good outcome rate after using rtPA within 2 h,and it does not increase the risks of symptomatic cerebral hemorrhage and death.It is a safe and effective adjunctive thrombolytic therapy.
7.Cause and management of delayed hemorrhage after minimally invasive percutaneous nephrolithotomy

Objective To discuss the cause and management of delayed hemorrhage after minimally invasive percutaneous nephrolithotomy (PCNL). Methods From 2004 to 2009,there were 13 cases (10 men and 3 women, mean age 47 years) developed severe bleeding following PCNL. The cause of hemorrhage and treatments were retrospectively analyzed and summarized. Results The time of hemorrhage was 5-40 d. In 7 of 13 cases, the bleeding were was controlled by complete bed rest,hemostatic,balloon compression and clamped nephrostomy tube. Six of 13 underwent selective renal arteriography after ineffective with conservative treatment. The DSA showed 4 were false aneurysm and 2 were arteriovenous fistula. All the 6 cases were treated by the super-selective arteriolar embolization. The hematuria was disappeared 1- 3 d later. The intravenous pyelogram revealed the renal function kept well during follow-up visit. Conclusions Delayed hemorrhage is one of the severe complications, which may be caused when the renal puncture passage established as forming false aneurysm and arteriovenous fistula. The renal arteriography and super-selective arteriolar embolization could be a safe and effective treatment for the severe hemorrhage after PCNL.
8.Study on the relationship between semen HBV-DNA load and offspring-paternal-vertical-transmission of HBV

Objective To explore the relationship between HBV-DNA load and the offspring vertical transmission of HBV.Methods 138 families who had taken the examination between August 2009 and November 2011 but the HBsAg of the housewife was negative,were chosen as research objects.Blood from the couples and sperms from the husbands during pregnancy were followed and collected for detection on related indicators.Cord blood was sampled after delivery for HBVM and HBV-DNA quantification.Those with HBV-DNA load ≥5 × 102 copies/ml were chosen as cases while those <5 × 102 copies/ml were formed as controls,respectively.Results 1) The positive rates of HBV-DNA was 34.8% (48/138) in the neonatal cord blood while the positive rates of cord blood HBsAg and HBeAg were 28.3% (39/138) and 15.2% (21/138) respectively.2) The positive rate of semen HBV-DNA was 21.0% (29/138) while the positive rates of paternal serum HBV-DNA and HBeAg were 76.8% (106/138)and 42.8% (59/138).3)Among the positive ones on paternal serum HBV-DNA,paternal serum HBeAg,semen HBV-DNA,items as measures taken for HBV vertical transmission and prevention on the fathers and the first class family histories on HBV appeared to be the risk factors for HBV paternal transmission (P<0.05).4)Data from Multivariate analysis showed that positivities on patemal serum HBV-DNA,paternal serum HBeAg and semen HBV-DNA were risk factors for HBV paternal transmission (OR=5.7,95%CI:1.1-29.1 ; OR=4.2,95%CI:1.7-10.0; OR=6.7,95% CI:2.4-18.9).5)Dose-response relationships were seen between levels of paternal serum HBV-DNA load and cord blood HBV-DNA load,between levels of paternal serum HBV-DNA load and semen HBV-DNA load,between levels of semen HBV-DNA load and cord blood HBV-DNA load.6)Results from the analysis on ROC curve showed that paternal serum HBV-DNA load level (105 copies/ml) and semen HBV-DNA load level (103 copies/ml)were better demarcation points to forecast the occurrence of paternal transmission of HBV,because of the better sensitivity and specificity they had.Conclusion Items as positives on paternal serum HBV-DNA,paternal serum HBeAg and semen HBV-DNA were risk factors for HBV paternal transmission.When paternal serum HBV-DNA load > 105 copies/ml and semen HBV-DNA load > 103 copies/ml appeared,the positive rate of HBV paternal transmission would increase.
9.Role of Erbin in sepsis-associated encephalopathy in mice and the relationship with NLRP3 inflammasomes

Objective:To evaluate the role of ErbB2 interacting protein (Erbin) in sepsis-associated encephalopathy (SAE) in mice and the relationship with nod-like receptor thermoprotein domain associated protein 3 (NLRP3) inflammasomes.Methods:Sixty SPF-grade healthy male wild-type C57BL/6 mice and 60 Erbin (-/-)C57BL/6 mice, aged 8-10 weeks, weighing 20-25 g, were divided into 4 groups ( n=30 each) by a random number table method: wild-type sham operation group (WT+ Sham group), wild-type SAE group (WT+ SAE group), Erbin (-/-) sham operation group (EKO+ Sham group) and Erbin (-/-) plus SAE group (EKO+ SAE group). The model of SAE was established by cecal ligation and perforation in anesthetized mice.Open field test (total distance moved) was performed at 7 days after establishing the model, new object recognition test (recognition index) was performed at 8 days after establishing the model, and Morris water maze test (time of staying at target quadrant) was performed at 10 days after establishing the model.The mice were sacrificed, and hippocampal tissues were removed for microscopic examination of pathologic changes (by HE staining) and for determination of neuron count, expression of NLRP3, caspase-1 and apoptosis-associated speck-like protein containing a CARD (ASC) (by Western blot), the number of NLRP3 positive cells (by immunohistochemistry), and contents of interleukin-1beta (IL-1β), tumor necrosis factor-alpha (TNF-α) and IL-18 (by enzyme-linked immunosorbent assay). The cell survival rate was calculated. Results:Compared with group WT+ Sham, the time of staying at target quadrant was significantly shortened, the recognition index and cell survival rate were decreased, the contents of IL-1β, IL-18 and TNF-α and the number of NLRP3 positive cells were increased, and the expression of NLRP3, caspase-1 and ASC was up-regulated in group WT+ SAE ( P<0.05). Compared with group EKO+ Sham, the time of staying at target quadrant was significantly shortened, the recognition index and cell survival rate were decreased, the contents of IL-1β, IL-18 and TNF-α and the number of NLRP3 positive cells were increased, and the expression of NLRP3, caspase-1 and ASC was up-regulated in group EKO+ SAE ( P<0.05). Compared with group WT+ SAE, the time of staying at target quadrant was significantly shortened, the recognition index and cell survival rate were decreased, the contents of IL-1β, IL-18 and TNF-α and the number of NLRP3 positive cells were increased, and the expression of NLRP3, caspase-1 and ASC was up-regulated in group EKO+ SAE ( P<0.05). There was no significant difference in total distance moved between the four groups ( P>0.05). Conclusion:Erbin can exert endogenous protection by inhibiting the activation of NLRP3 inflammasomes in mice with SAE.
10.Effect of"Five-in-One"of traditional Chinese medicine combined with low concentration of atropine in prevention and control of myopia in adolescents

Objective To explore the safety and effectiveness of the comprehensive therapy of"Five-in-One"of traditional Chinese medicine combined with low concentration of atropine in the pre-vention and control of myopia in adolescents.Methods Eighty adolescents diagnosed with mild myo-pia were included in the study.According to the wishes of the subjects and their guardians,they were divided into control group with 40 cases and traditional Chinese medicine intervention group with 40 cases.The control group received 0.01%atropine sulfate eye drops in both eyes once before bedtime every night,the traditional Chinese medicine intervention group underwent twice-weekly comprehen-sive therapy of"Five-in-One"of traditional Chinese medicine on the basis of local application of med-ication in the control group.Both groups received a 12-month intervention course.Follow-up visits were conducted before intervention and at 3,6,and 12 months after treatment.The international standard visual acuity chart was used to examine the changes in unaided distance visual acuity before and after treatment in both groups.The changes of distant visual acuity of naked eye before and after treatment were measured in two groups.The changes in equivalent spherical lens power before and after treatment in both groups were measured by mydriatic refraction combined with a computer op-tometer.The changes in axial length before and after treatment were measured using an optical bi-ometer.The adverse reactions after treatment in both groups were compared.Results In the con-trol group,the uncorrected distance visual acuity decreased compared with that before intervention as the treatment time prolonged,while was gradually increased in the traditional Chinese medicine intervention group compared with that before intervention(P<0.05).At 12 months after treat-ment,the uncorrected distance visual acuity in the traditional Chinese medicine intervention group was higher than that in the control group(P<0.05).With the prolongation of treatment time,the equivalent spherical lens power gradually increased in both groups(P<0.05).At 12 months after treatment,the equivalent spherical lens power in the traditional Chinese medicine intervention group was lower than that in the control group(P<0.05).The axial length gradually increased with the prolongation of treatment time in both groups(P<0.05).At 6 and 12 months after treatment,the axial length in the traditional Chinese medicine intervention group was shorter than that in the control group(P<0.05).After using 0.01%atropine,8 children in the control group developed outdoor photophobia symptoms;after treatment with the comprehensive therapy of"Five-in-One"of tradi-tional Chinese medicine combined with 0.01%atropine sulfate eye drops in the traditional Chinese medicine intervention group,4 children developed outdoor photophobia symptoms,but the symptoms disappeared after 2 weeks of medication.Conclusion The comprehensive therapy of"Five-in-One"in traditional Chinese medicine combined with low-concentration atropine is significantly more effective than low-concentration atropine alone in effectively controlling the progression of refractive myopia and reducing its adverse reactions.
