1.Research progress of effects of neurotrophic factors in the repair of spinal cord injury

Spinal cord injury(SCI) is a serious nervous system trauma,which may consequently lead to various degrees of paralysis and toilet obstacles,and its disability and the high cost heavily burdened the families and community.Therefore,the research on spinal cord tissue regeneration and the repair after injury has important practical significance.A large number of experimental studies have shown that neurotrophic factor plays an important role in the nerve tissue repair process.This article reviews new research progress about the effects of neurotrophic factor in this field.
2.Determination of Terbutaline Sulfate and Its Combination with β_2 Adrenergic Receptor in Erythrocyte Membrane by High Performance Liquid Chromatography

A high performance liquid chromatographic method with hypersil C_(18) column, 0.020 mol/L potassium dihydrogen phosphate solution(which contains 0.25% triethylamine, pH 5.1)-methanol(20∶ 80, V/V) as mobile phase and fluorescence detection with 280 nm as excitation wavelength and 320 nm as emission wavelength has been developed for the determination of terbutaline sulfate and propranolol hydrochloride in Erythrocyte suspensions. Erythrocyte surface receptor oscillating reacted 1 h with terbutaline in 37 ℃ water bath. Free terbutaline and propranolol were separated from the membrane receptor-binding complex through 1500 r/min centrifugal in 5 min. The linear range was 0.010-3.000 mg/L in Alsever's solutions and the correlation coefficients were 0.9999 and 0.9998. The relative standard deviations(RSDs) of terbutaline and propranolol were 0.8% and 1.2%, respectively. The limits of detection(LODs) of both were 1.00 and 3.00 mg/L. This simple, safety and sensitive method was suitable for the determination of terbutaline and propranolol in erythrocyte suspensions and for the study of receptor-ligand binding.
3.Effects of sodium magnesium fructose diphosphate on free calcium concentration and nitric oxide synthase activity of ischemic synaptosome

Aim To study the effects of sodium magnesium fructose diphosphate (SMFD) on free calcium concentration and nitric oxide synthase activity of ischemic synaptosome, so as to explore the protective mechanisms of SMFD on cerebral ischemia. Methods The synaptosomes from normal rat brain were prepared by phase partition and cultured with oxygen-glucose deprivation to establish ischemic synaptosome model. The intrasynaptosomal free calcium concentration and nitric oxide synthase activity were detected separtately after the synaptosomes were co-incubated with SMFD (1.3 mmol*L-1) or fructose-1,6-diphosphate (FDP, 4.0 mmol*L-1) for 60 min. Results SMFD decreased the free calcium concentration and reduced the activity of nitric oxide synthase (NOS) of ischemic synaptosomes. Its effects were more powerful than those of FDP. Conclusion SMFD may protect neurons from ischemic injury by preventing intracellular Ca2+ overload and inhibiting the activity of nitric oxide synthase.
4.Effects of berberine on Insig-2 and VDR gene expression of HepG2 cell line

AIM: To observe the mRNA expressions of Insulin-induced gene 2(Insig-2) and Vitamin D receptor(VDR) in HepG2 cell line and investigate the possible mechanism of berberine on regulating blood lipid. METHODS: HepG2 cells were incubated with Bet at different concentrations(1,3,10,30,100 ?mol/L),and incubated with 3 ?mol/L berberine for various times(0,12,24,48,72 h).Total RNA was extracted and the mRNA expressions of Insig-2 and VDR from HepG2 cells treated by berberine were quantified by RT-PCR.(RESULTS): After exposure to gradient concentrations of berberine for 24 h,the mRNA expressions of Insig-2 and VDR in HepG2 cells increased obviously peaked at 3 ?mol/L concentration,then the up-regulating effect declined gradually.The relative coefficient between them was(0.753)(P
5.Concentration of quinolinic acid in rat serum by GC-MS

Objective To establish a sensitive and accurate gas chromatography-mass spectrometry method for determination of quinolinic acid in rat serum.Methods The objective compound was purified and derivatized to its dihexafluoroisopropyl ester before quantitative determination by scanning method and quantitative determination by SIM method.Results The endogenous substances in serum had no interference on the measurement of quinolinic acid in the sample.Assay linearity was obtained in the range of 0.05-20 ?mol/L with limit of quantitation(LOQ)of 0.05 ?mol/L.The intra-day relative standard deviation(RSD)was lower than 15%.The inter-day relative standard deviation was lower than 12%.Conclusion It is a sensitive,accurate and convenient method.The dihexafluoroisopropyl ester of quinolinic acid is quite stable,but the serum should not be preserved for a long time before derivation.
6.Role of calcineurin signal transduction pathway in the inhibition of cardiac hypertrophy by L-arginine in vivo and in vitro

Aim To study the role of calcineurin signal transduction pathway in the anti-hypertrophic effect of L-arginine in vivo and in vitro.Methods The hypertrophic effects was assayed by calculating the right ventricular hypertrophy index(RVHI=right ventricle weight/left ventricle and septum weight),and atrial natriuretic peptide(ANP)mRNA expression in rat right ventricle hypertrophy model induced by monocrotaline(MCT) or by measuring the cell diameter,protein content,and ANP mRNA expression in hypertrophic cardiomyocyte induced by prostaglandin F2?(PGF2?).For mechanism studies,the intracellular free calcium concentration([Ca2+]i) in cultured cardiomyocytes was measured by using Fura 2/AM as a fluorescent indicator.ANP and CaN mRNA expressions,and expressions of CaN and its downstream effectors,NFAT3 and GATA4 proteins were assayed by RT-PCR and Western blot,respectively,in vivo and in vitro.Results In MCT-hypertrophic model,prevention-and treatment-administration of L-arginine,a nitric oxide(NO) precursor,200 mg?kg-1?d-1,could obviously inhibit the elevated RVHI and ANP mRNA expression;similar to that found in vivo.Addition of L-arginine 1 mmol?L-1 could markedly inhibit the increased cell diameter,protein content and the expression of ANP mRNA in the hypertrophic cardiomyocyte induced by PGF2? 100 nmol?L-1,and it could also decrease the elevated [Ca2+]i in vitro;notably,the above dose or concentration of L-arginine could blunt the elevated expressions of calcineurin mRNA and the calcineurin-,NFAT3-,GATA4-proteins induced by MCT or by PGF2?.These effects of L-arginine were blocked by NG-nitro-L-arginine-methyl ester,a NO synthase inhibitor,in vivo and in vitro.Conclusion These results suggest that calcineurin signal transduction pathway may play an important role in the NO-induced inhibition of cardiac hypertrophy.The anti-hypertrophic effects of L-arginine may involve the decrease of [Ca2+]i,and then inhibit the activated calcineurin pathway,through the release of NO.
7.Study on Hypoglycemic Effect of Domestic Glimepiride in Animals

AIM:To study the effects of domestic glimepiride(Gli) on blood sugar,glucose tolerance and release of insulin in rabbits and rats METHODS:Taken the changes of blood sugar level and glucose tolerance before and after giving drug as parameters,the hypoglycemic effect of Gli was observed in rabbits and rats RESULTS:Gli dose-dependly decreased the levels of blood glucose in rats with a decrease of 23% in high-dose group and seemed to be similar to glibenclamide in potency of lowering blood sugar Gli increased release of insulin by 40%~70% in rats and rabbits and was twice as potent as glibenclamide;Glucose tolerance test showed that Gli could restrain the rising of blood glucose at 30 min and 60 min after glucose was administrated ig in rabbits Time-response relation displayed that blood glucose began to drop 30 min after Gli 1 0mg/kg administered and the levels of blood glucose dropped by 40% 3 h later CONCLUSION:The domestic Gli has similar effects to glibenclamide on lowering blood sugar However,its effect on lowering blood sugar seems to be quicker,shorter and more moderate than that of glibenclamide
8.Effects of tetrandrine and fructose-1,6-diphosphate on the elevated intrasynaptosomal [Ca~2+]_i induced by excitatory amino acids

AIM: To study the effects of tetrandrine(Tet) and fructose-1,6-diphosphate(FDP) on the elevated intrasynaptosomal [Ca 2+ ] i induced by excitatory amino acids (EAA). METHODS: A rapid method for preparing synaptosomes was used, and intrasynaptosomal free calcium ([Ca 2+ ] i) was measured by using the fluorescent indicator quin-2. RESULTS: L-glutamate (Glu, 100 ?mol/L), aspartate (Asp, 100 ?mol?L -1 ), N-methy1-D-aspartate (100 ?mol/L) and Glu (50 ?mol/L) plus Asp (50 ?mol/L) all elevated intrasynaptosomal [Ca 2+ ] i in a dose-dependent manner. Pretreatment with Tet (10,30,60 ?mol/L) , FDP (15, 30, 75, 150 ?mol/L), MK-801 (10, 20 ?mol/L) and Tet (15, 30 ?mol/L) plus FDP(15, 30 ?mol/L) all attenuated the increase in intrasynaptosomal [Ca 2+ ] i induced by EAAs mentioned as above in a dose-dependent manner, and the effect of Tet plus FDP was most significant. CONCLUSION: Both Tet and FDP inhibited a rise in intrasynaptosomal [Ca 2+ ] i induced by EAAs, which may be one of mechanisms that Tet and FDP pretect cerebral tissues against ischemia injury.
9.Preventive and Therapeutic Effects of Extract of Curcuma on Experimental Hyperlipidemia Models

OBJECTIVE:To evaluate the preventive and therapeutic effects of extract of Curcuma on experimental hyper?lipidemia models in rats and rabbits.METHODS:Hyperlipidemia models of healthy rats and rabbits,fed on a forage containing cholesterol,fat and etc,were established.TG,TC,LDL,HDL and LDL/HDL in serum of animals were measured.RESULTS:The preventive administration of extract of Curcuma could significantly reduce the serum TC,LDL,LDL/HDL and increase serum HDL in hyperlipidemic rats in a dose-dependent manner.Its hypolipidemic action was stronger than that of positive contrast agent lovastatin.The serum levels of TC,TG,LDL and HDL not of LDL/HDL obviously decreased in hyperlipidemic rabbits after either preventive or therapeutic administration and its hypolipidemic action was weaker than that of positive con?trast agent lovastatin.CONCLUSION:Extract of Curcuma could obviouly reduce hyperlipidemic in rats and rabbits.Its action is stronger than that of lovastatin in rats,but weaker in rabbits.
10.Effect of icariin on expression of cytochrome C oxidase subunit Ⅱ mRNA in mice of cerebral ischemia/reperfusion

Objective To evaluate the effects of cerebral ischemia/reperfusion or/and icariin on expression of cytochrome C oxidase subunitⅡ(COⅡ) mRNA in mice. Methods The mouse model of transient cerebral ischemia/reperfusion was made by bilateral occlusion of common carotid arteries and ischemic hypotension/reperfusion.
