1.Study on HPLC fingerprint of Caulis Spatholobi

ObjectTo establish HPLC fingerprint for the identification and evaluation of Caulis Spatholobi. Methods HPLC system was used to obtain the chromatograms of Caulis Spatholobi and the clustering analysis was applied for data analysis. Results There are 11 peaks in total, among them five are common, the relative retention time is 0.046, 0.664, 1.000, 1.198, 1.383, the relative peak area is 0.819,0.221,1.000,0.331,0.458. There was obvious difference among chromatographic fingerprints of Caulis Spatholobi and its easily-confused species. The 11 measured peaks could be used as the fingerprint features. Conclusion Chromatographic fingerprint of HPLC can be used for identifying the Caulis Spatholobi and its easily-confused species.
2.Nonlinear Finite Element Analysis of Lumbar Spine Stress at Various Loading

Objective In order to probe the possible biomechanical mechanism of lumbar vertebral injuries, we studied the nonlinear stress distribution on lumbar vertebrae at 1500N axial compression force and 15Nm bending moments during hambar flexion, extension, lateral bending and axial torsion using an available nonlinear finite element (FE) model of functional spine unit. Methods Three-dimensional nonlinear FE model was created based on CT data of L3-L4 motion segment. The segment mechanical responses to different loadings were evaluated by the range of disc bulging, the stress level of fibrousrings, facet loadings and equivalent stress distribution of posterior structure. Results Compression and flexion induced disc bulging in anterior region, whereas extension and lateral bending induced disc bulging posteriorly and posterolaterally. Axial torsion caused not only higher unilateral facet stress but also the concentrated stress on posteroleteral fibrousrings stress. The loadings in extension and axial compression were lower than in torsion but higher than in lateral bending and anterior flexion. Higher equivalent stress was found when compression and extension loadings were applied to the pedicle of vertebral arch and interarticularis. Conclusion Intervertebral disc beared higher compressive than tensile. The posterolateral regions of lumbar vertebrae were prone to be deformity and at higher risk of fibrousring failure. Stress failure could be occured when facets were overloaded with axial torsion,extension and extension force due to the importance of facets during lumbar movement.
3.Positive selection caused change of hepatitis C virus quasispecies distribution in the natural history of he patitis C infection

Objective To investigate the evolution of hepatitis C virus (HCV) quasispecies in persistent and self limited infection patients. Methods Peripheral blood HCV preserved for 10 years from 8 naive individuals once infected by HCV were analyzed. HCV hypervariable region 1 (HVR1) genes were cloned and sequenced. Entropy, genetic distances, nonsynonymous mutations per nonsynonymous site (K A ), synonymous mutations per synonymous site(K S ) were evaluated for evolutionary analysis. Results The entropy values and genetic distances of HCV quasispecies were lower in self limited infection indivi duals than that of persistent infection individuals. In 3 of the 4 persistent infection individuals there exist no difference between within group and between group genetic distances in HCV HVR1. The ratios of K A /K s were higher than 1 in 7 of these 8 individuals. Conclusions The complexity and diversity degree of HCV quasispecies may affect the outcome of HCV infection. Positive Darwinian selection may be the major cause for the change of HCV quasispecies distribution in hepatitis C infection natural history.

Object To study the protective effect of Rodobryum roseum (Hedw) Limpr. on acute myocardial ischemia. Methods Rat acute myocardial ischemia models were prepared by ligating their left coronary arteries. Changes of their S-T segment was observed on lead Ⅱ dynamic ECG. Degree of ischemia was assessed by the extent of S-T segment elevation. OBJECTIVETo study on pharmacologic actions on quail hyperlipidemia and atherosclerosis model.
METHODTo duplicate quail hyperlipidemia model by ectogenesis cholesterol and high fat forage, induce to atherosclerosis model, observe influence of sugarcane alkane alcohol to model animals' blood fat level, formation of atherosclerosis plaque, pathological changes of coronary vessels and vascular intimal.
RESULTTC, TG, LDL-C level in blood serum of quail hyperlipidemia markedly decreased after administered sugarcane alkane alcohol by dose of 30, 15, 7.5 mg x kg(-1), proliferation of aorta and brachiocephalic artery tunica intima foam cells was suppressed.
CONCLUSIONSugarcane alkane alcohol has satisfactory pharmacologic actions on hyperlipidemia and atherosclerosis animal model by regulating blood fat.

6.Transfusion of activated immune cells induced fluctuations of HCV titer without mutations in hepatitis C virus core and E2 region CTL epitopes.

OBJECTIVETo investigate the variations of HCV core and E2 region epitopes during transfusion of activated immune cells.
METHODSFour patients receiving transfusion of activated immune cells were under continuously observation. HCV titers were measured by quantitive PCR. HCV core and E2 regions were cloned and sequences were analyzed by computer software.
RESULTSDuring the follow-up the serum ALT levels and the HCV virus titers fluctuated greatly in each of these persons. After transfusion, no significant variations were observed in HCV core and E2 coding regions.
CONCLUSIONSThe alteration of host immune attacks could induce the fluctuations of HCV load without any mutations in the currently observed coding genes of the epitopes.

7.Role of mesenchymal stem cells and their exosomes in Th17/Treg-related pathogenesis of psoriasis

T helper 17 (Th17) cell/regulatory T (Treg)cell imbalance has been widely considered as one of immunopathogeneses of psoriasis.Mesenchymal stem cells (MSC) can inhibit the proliferation and differentiation of Th 17 cells,but induce the proliferation of Treg cells.Exosomes secreted by MSC are a kind of important carrier for intercellular communication and material transportation.Studies have found that exosomes from MSC and MSC have similar immunoregulatory function,which can inhibit the proliferation of Th17 cells and induce the differentiation of Treg cells.In recent years,many studies have shown that abnormalities of MSC in bone marrow and skin lesions of psoriasis patients are related to the occurrence of psoriasis,but the mechanism is still unclear.This review elaborates the role of normal MSC and their exosomes in the maintenance of Th 17/Treg balance,which provides ideas for studying how abnormalities of MSC in bone marrow and skin lesions of psoriasis patients participate in the occurrence of psoriasis.
8.Precise percutaneous sacroplasty assisted by a Renaissance robot for sacral insufficiency fractures

Objective To evaluate the precise percutaneous sacroplasty (PSP) assisted by a Renaissance robot for sacral insufficiency fractures (SIF).Methods The clinical data of 12 SIF patients were retrospectively analyzed who had been treated from March 2016 to March 2018 at Department of Spinal Surgery,Honghui Hospital.They were 5 males and 7 females,aged from 55 to 76 years (average,67.5 years).They all received PSP assisted by a Renaissance robot.Their operation time,hospital stay and intraoperative radiation were recorded.The clinical efficacy was evaluated by comparing their visual analogue scale (VAS) and Oswestry disability index (ODI) before surgery,1 day,3 and 12 months after surgery.Results All the 12 patients underwent surgery successfully with no complications like cement leakage.Their operation time ranged from 32 to 47 minutes (mean,36.8 minutes),their hospital stay from 12 to 25 hours (mean 17.5 hours) and their intraoperative exposure to radiation from 0.87 to 1.53 mSv (mean,1.27 mSv).All the patients were followed up for 12 to 18 months (mean,15.8 months).Their VAS (1.7 ± 0.7) and ODI (22.8 ± 4.1) one day after surgery were significantly decreased than the preoperative values (7.6 ±0.9 and 43.7 ±4.6) (P < 0.05).At 3 and 12 months after surgery,their VAS scores were 2.0 ± 0.8 and 2.4 ±0.8 and their ODI scores 21.5 ±4.3 and 23.0 ±4.6,respectively,showing no significant differences from the values at 1 day after surgery (P > 0.05).Conclusion The PSP assisted by a Renaissance robot is safe and leads to satisfactory clinical efficacy for SIF as bone cement can be accurately injected into the target area of the fracture.
9.Effect of robot navigation system combined with 3D printing in assisting percutaneous vertebroplasty for Kümmell disease

Objective:To compare the effect of robot navigation system (Tian Ji robot system) plus 3D printing and traditional C-arm X-ray fluoroscopy in assisting percutaneous kyphoplasty (PKP) or percutaneous vertebroplasty (PVP) for treatment of Kümmell disease.Methods:A retrospective case-control study was conducted to analyze the clinical data of 40 patients with Kümmell disease treated at Honghui Hospital Affiliated to Xi'an Jiaotong University School of Medicine from December 2017 to February 2019, including 12 males and 28 females, with an average age of 56.4 years (range, 42-71 years). In observation group, 20 patients underwent PKP or PVP assisted by the robot navigation system and 3D printing. In control group, 20 patients underwent PKP or PVP assisted by the traditional C-arm X-ray fluoroscopy. The operation time and incidence of complications were observed. The visual analogue scale (VAS), Oswestry disability index (ODI), Cobb angle and anterior vertebral height were compared before operation, 1 day and 3 months after operation.Results:All patients were followed up for 3.5-8.6 months (mean, 6.7 months). The operation time in control group was (32.2±5.8)minutes, compared with (26.7±3.6)minutes in observation group ( P<0.05). The incidence of cement leakage was 0% (0/20) in the observation group and 5% (1/20) in control group ( P>0.05). One day after operation, in observation group and control group, the VAS was (2.1±0.3)points and (3.7±0.8)points, the ODI was 14.3±1.8 and 25.5±5.7, the Cobb angle was (20.6±1.2)° and (22.4±0.6)°, and the anterior height of vertebral body was (21.2±0.8)mm and (17.6±0.7)mm, respectively, showing significant improvement compared with those before operation ( P<0.01). Three months after operation, in observation group and control group, the VAS was (1.8±0.4)points and (2.8±0.8)points, the ODI was 12.3±1.5 and 21.6±2.3, the Cobb angle was (18.1±0.8) ° and (20.5±1.6)°, and the anterior height of vertebral body was (20.1±1.8)mm and (16.8±1.3)mm, showing no significant difference compared with those at day 1 after operation ( P>0.05). There were significant differences in the VAS, ODI, Cobb angle and anterior vertebral height between the two groups 1 day and 3 months after operation ( P<0.01). Conclusion:For Kümmell disease, with assistance with the robot navigation system combined with 3D printing, PKP or PVP can more effectively reduce the pain of patients, improve the quality of life, restore the anterior height of vertebral body, and realize the individualized treatment in comparison with the traditional C-arm X-ray fluoroscopy.
10.Development of a novel chemokine signaling-based multigene signature to predict prognosis and therapeutic response in colorectal cancer.

Colorectal cancer (CRC) is the most lethal gastrointestinal cancer in both males and females worldwide (Sung et al., 2021). Because of the high heterogeneity of tumors, robust prognostic biomarkers are urgently needed in CRC management (Koncina et al., 2020). Chemokine signaling is a well-known pivotal player in immunity, inflammation, and cancer metastasis (Lacalle et al., 2017; Poeta et al., 2019; Do et al., 2020), and multiple genes involved in chemokine signaling have been demonstrated as potential prognostic biomarkers for CRC (Cabrero-De Las Heras and Martínez-Balibrea, 2018; Ottaiano et al., 2020; Yu et al., 2020). Therefore, the aim of our study was to develop a chemokine signaling-based multigene signature (CSbMgSig) that could effectively predict overall survival (OS) and therapeutic response for patients with CRC.
