Journal of Chinese Physician 2001;0(10):-
Objective To explore the effects and molecular mechanisms of arsenic trioxide(As 2O 3) in inducing apoptosis of colonic cancer cell line SW480. Methods The morphologic changes and apoptotic rate of SW480 cells induced by As 2O 3 were observed with fluorescent microscopy and flow cytometry. The bcl-2 and Fas expressions induced by As 2O 3 in SW480 cells were examined by immunohistochemical method. Results 24 hours after exposed to As 2O 3, SW480 cells exhibited typical morphologic changes of apoptosis, the apoptotic rate of which was 2.1%~10.6%. A typical subdiploid peak before G 1 phase was observed by flow cytometry, and As 2O 3 mainly acted in G 2/M phase. As 2O 3 decreased bcl-2 expression and increased Fas expression in SW480 cells. Conclusion As 2O 3 could induce apoptosis of SW480 cells. The molecular mechanism of As 2O 3-induced apoptosis of SW480 cells might be down-regulating the bcl-2 expression and up-regulating the Fas expression.

Objective To study the effects of ?-asarone on mitochondrial membrane potential(MMP) of cardiac myocytes with ischemia reperfusion injury. Methods The cardiac myocytes model of myocardial ischemia reperfusion injury (MIRI) was induced with sodium dithionite (Na2S2O4). The primary cultured cardiac myocytes of SD neonatal rats were randomized into six groups:normal group, model control group,high-dose ?-asarone (25 ?g/mL),middle-dose ?-asarone group(12.5 ?g/mL),low-dose ?-asarone group (6.25 ?g/mL) and medium group. Mitochondrial membrane potential(MMP) of myocardial cells were measured by flow cytometry. Results Compared with the normal group,MMP in the model control group was obviously decreased(P

Objective To study the protective effects of ?-asarone against the myocardial ischemia/ reperfusion injury (MI/RI) in cultured cardiac myocytes. Methods MI/RI model was set up with Na2S2O4, which make use of rat cardiac myocytes. The viability of cardiac myocytes (using MTT method), dehydrogenase (LDH, CK) activitices in the medium were measured. Results In MI/RI model, ?-asarone can obviously advance the viability of cardiac myocytes, and decrease the dehydrogenase (LDH, CK) activitices in the medium. Conclusion ?-asarone showed protective effects against the myocardial ischemia/reperfusion injury in cultured cardiac myocytes.

[Objective] To screen an optimal method for the extraction of volatile oil from Naokangling Capsule (NC) and processing techniques of ?-cyclodextrin (?-CD) inclusion compound of NC. [Methods] The study was carried out with orthogonal design . Oil-extraction ratio, oil-bearing rate and inclusion rate of ?-CD inclusion compound of NC were used as indexes. [Results] The optimal process for oil extraction was adding 10-fold water in medicinal materials and then soaking for one hour and distilling for 5 hours. The optimal processing conditions for ?-CD inclusion compound of NC were: the proportion of ?-CD and volatile oil being 6:1, the inclusion compound being stirred for 2 hours at 40℃ . [Conclusion] This method is simple and practical and can increase the stability of preprations. It can be used for the production of ?-CD inclusion compound in a large scale.

Objective Thymidine phosphorylase (TP) cDNA was transfected into colorectal cancer cell lines LOVO with the lentiviral vector, the anticancer effeciency of 5'-deoxy-5-fluorouridine (5'-DFUR)and 5-fluorouracil (5-FU) on LOVO cells were evaluated. Methods TP cDNA were transfected into LOVO cell line with the lentiviral vector pLenti6.3_MCS_IRES2-EGFP,and the transfection efficiency was analyzed by flow cytometer and immunohistochemistry.Cells were divided into six groups:LOVO,LOVO-TP,LOVO-control,LOVO + INF-α2a,LOVO-TP + INF-α2a,LOVO-control + INF-α2a.TP protein expression and the relative quantitative analysis for TP mRNA in transfections cells were detected by Western blot and RT-PCR respectively.Volumes of converted 5-FU from either in the medium containing different concentration of 5'-DFUR,in which all cells were cultured,or in cells lysate, were detected by high performance liquid chromatography (HPLC).Results The transfection efficiency of TP cDNA in LOVO cells was 95%.The Mean gray value of TP protein expression in LOVO-TP and LOVO-TP + INF-α2a were 198.15 folds and 243.22 folds higher than LOVO cell, respectively (P < 0.01).The RQ values of TP mRNA expression in LOVO-TP and LOVO-TP + INF-α2a were also 18.56 folds and 59.61 folds higher than wild LOVO cell, respectively (P < 0.01).The IC50 values of 5'-DFUR on LOVO-TP and LOVO-TP + INF-α2a were 1 660 μ mol/L and 813 μ mol/L,respectively,significantly lower than 4 462.59 μ mol/L in wild LOVO (P < O.01).The 5-FU volumes detected from media contained series concentration of 5'-DFUR for culturing LOVO-TP and LOVO-TP + INF-α2a were 2.0-5.3 folds, and 2.9-10.4 folds more than wild LOVO,respectively (P < 0.01).Conclusions Transfected TP cDNA into colorectal cancer cell line LOVO with lentiviral vector increases the expression of TP mRNA and TP protein and the amount of 5-FU converted from 5'-DFUR,enhancing its anticancer effect.

Objective To evaluate the quality of life status in patients aged 70 years and over following off-pump coronary artery bypass (OPCAB) grafting. Methods Seventy-eight patients with coronary heart disease [mean age (74.6 ± 5.3) years, 66 males, 12 females] were investigated retrospectively. Three questionnaires about the quality of life, including Seattle Angina Questionnaire (SAQ), Nottingham Healthy Profile (Part Ⅰ NHP) and Duke Activity Status Index (DASD, were used to investigate patients before and after OPCAB. Results Prior to OPCAB, there was lower quality of life index in males than in females [SAQ: (65.3±5.1) vs. (69.5±8.1); NHP: (89.4±17.3) vs. (125.2±19.9), P<0.01; DASI: (4. 1±1.1) vs. (4.3± 1.3)]. At the 12th months after OPCABG, there were significant improvements in all patients. The effects were less pronounced in females than in males [SAQ: (83.1 ±5.8) vs. (88.5±4.5), P<0.05; NHP: (84.7± 11.7) vs.(91.4±13.7), P<0.05; DASI: (4.7±1.4) vs. (5.4±1.1)]. Conclusions Our study shows that OPCAB improves quality of life in elderly patients with coronary heart disease. The benefits of OPCAB are even more pronounced in male patients.
