1.Effect of anti-oncogene Kiss-1 gene on cell proliferation in human melanoma cells

Objective To investigate the effect of Kiss-1 on cell proliferation in human melanoma cancer cell,and screen a target for tumor biotherapy. Methods The eukaryotic expression vectors with Kiss-1 gene were used to transfect A375 cells by liposome-mediated transfection in vitro. The cells were selected by G418. The expression of Kiss-1 gene was confirmed by Western Blotting. FACS was used to detect the ability of cells proliferation and apoptosis. Results There were significantly high expression of Kiss-1 protein in the transfected A375 cells(1.20 ±0.21 vs control 0.60 + 0. 41), whose growth rate was significantly slower than that of control. Flow cytometry showed that apoptosis of A375 cells transfected Kiss-1 were induced(7.23% vs control 4.62%). Conclusion When transfected into A375 cells,exogenous Kiss-1 could cause the cell growth arrest to G1 ,and induce cells apoptosis.
2.Effect of prone positioning on oxygenation and lung mechanics in neonates with mechanical ventilation

Objective To investigate the effects of prone positioning on oxygenation and lung mechanics in neonates with mechanical ventilation.Methods Twenty cases of neonates,selected from Shanghai Children′s Hospital(from May 2008 to Feb 2009),were randomized to supine-prone position group (n=10) and prone-supine position group (n=10) using a crossover design.During 6h,these neonates were placed in the supine position for 3h then in the prone position for 3h or in reversed order.The ventilator parameters,results of arterial blood gas and the lung mechanics were recorded.Results Without obvious changes of ventilator parameters,PaO2 improved significantly when the incubated neonates were placed in the prone position for 3h compared to supine [(74.50±15.37)mm Hg vs (67.60±13.66)mm Hg,1mm Hg=0.133kPa,P<0.05).Similarly,oxygenation index of prone positioning for 3h was (201.86±55.96)mm Hg,which was higher than that of (184.38±63.58)mm Hg,the value of supine positioning for 3 h.There were significant differences (P<0.05).Our results showed that 60% (12/20) of the neonates responded to prone positioning,with oxysenation index increasing as much as 17mm Hg on average.Improvement in minute volume,tidal volume and dynamic compliance of the lung were achieved after prone positioning for 3h (P=0.011,0.020,0.045).The airway resistance did not decrease significantly (P=0.422),but the average decreased in the prone position.Conclusion Oxygenation and the lung mechanics are improved significantly in the neonates with prone position compared with supine position.
3.Clinical observation of Yiling-Qutan Decoction used to treat sub-health

Objective To explore the clinical efficacy of the Yiling-Qutan Decoction on treating Sub-health and the mechanism of traditional Chinese medicine.Methods 219 cases of Sub-health and belonging to dampness-phlegm syndrome and were collected and treated with Yiling-Qutan Decoction for 1 month.The effective rate of syndrome of traditional Chinese medicine and state of Sub-health,syndrome score of traditional Chinese medicine and sub-health status score were observed.Results The effective rate of syndrome of traditional Chinese medicine:7 patients were cured,35 cases were significantly effective,171 cases were effective and 6 cases were unfruitful.The improvement of syndrome score of and Sub-health status were also significantly better (P<0.01).Conclusion It was effective to treat sub-health with Yiling-Qutan Decoction.
4.Systematic evaluation on the treatment of diabetic nephropathy with Tao Hong Si Wu Decoction and its modified prescription

Objective To evaluate the effectiveness and safety of Tao Hong Si Wu decoction in treating Diabetic nephropathy (DN).Methods Randomized controlled trials (RCTs) on Tao Hong Si Wu decoction in treatment of DN were searched in PubMed,EMbase,Cochrane Library,CMCC,VIP,CNKI,CBM.Data were strictly extracted using the fundamental evaluation method recommended in Cochrane Handbook and Meta-analysis was performed using RevMan 5.0.24 software.Results Eleven RCTs of 956 patients of DN were analyzed.The content and methodological quality of these 11 test samples were low,which could not be undertaken high quality research of sensitivity analysis.Meta-analysis results showed that:Tao Hong Si Wu decoction could be reduced to some extent,the early and clinical stage of DN patients with 24 h UPE reduce the Ⅲ,Ⅳ of Scr,and improve renal function to a certain extent.The results also showed that Tao Hong Si Wu decoction could lower TG and CHO,better control FBG and HbAlc of DN patients,besides,serious adverse reactions have not been found during the treatment.Conclusion Tao Hong Si Wu Decoction could be a relatively safe and effective treatment of DN drugs.However,a high probability of selection bias and measurement bias in the included studies was bound to affect the strength of the results.
5.Effect Donglian Capsule on Nonenzymatic Protein Glycation in Diabetes Melitus Rat

Objective To observe the effect of Donglian capsule on nonenzymadc protein glycation.Methods An animal model of DM was established by injecting a little dosage of STZ in high fat-fed rats.When the models were established successfully,corresponding medicines were supplied for 12 weeks and then the blood glucose,glycosylation serum protein(GSP)and aldose reductase(AR)in rats semm were measured.Results Comparing with the model group,the levels of blood glucose,GSP and AR in Metformin Hydrochloride tablets group and Chinese medicine group were decreased markedly (P<0.05).Conclusion Donglian capsule Can restrain nonenzymatic protein glycation and so thus prevent and treat DM.
6.Mechanical mechanism and relative problems of lumbar traction

[Objective] To discuss the mechanism and relative problems of lumbar traction. [Method] Relevant articles and retrospect clinical data in the author's hospital were reviewed. Review relevant articles and retrospect clinical data of our hospital. [ Result ] Traction force : 40 kg + 15% ~ 20% of body weight, fineness rate reached 83.5% in 1606 patients being treated. According to course of disease, fineness rate was 90. 1% in the group of less than 3 years, 68.2% in the group of more than 3 years. [ Conclusion] Lumbosacral nerve root leave the peak of the protruding nucleus and establish a new harmonious "root-disc" relationship after traction. The pressure and tension to the nerve root reduces or disappears, meanwhile, the pain of low back and leg is alleviated or eradicated. Appropriate traction weight and correct traction body posture are key factors of good therapeutic effect.
7.Effects of Chinese Herbal Compounds on Levels of Blood Glucose, Froctosamine and Insulin of Diabetic Rats

Objective To observe the effects of Chinese herbal compounds (alkaloid of Coptidis Rhizoma 40%, total saponin of panax notoginseng 35%, and polysaccharide of Ophiopogonis Radix 25%) on levels of blood glucose, fructosamine, and insulin of diabetic rats, and explore mechanism of action.Methods Ten rats were randomly chosen as blank control group, and other rats were used to establish diabetic rat models by STZ intraperitoneal injection. Successfully molding rats were randomly divided into Chinese herbal compounds high, medium, low dose groups, metformin group, and model group, 10 rats in each group. Each rat received gavage for continuous 4 weeks. Then rat blood was collected by cutting head and taking caudal vein. FBG was detected by glucose oxidase method;insulin level was detected by radio-immunity double antibody method;fructosamine content was detected by fructosamine method.Results Compared with the blank control group, model group the levels of FBG, froctosamine content increased, while fasting insulin level were lower after 4 weeks'medication. Compared with the model group, the levels of FBG, froctosamine content in Chinese herbal compounds high and medium dose groups and metformin group were lower, while fasting insulin level increased after 4 weeks'medication (P<0.05).Conclusion TCM herbal compounds can increased the basic insulin level by decreasing the contents of FBG and fructosamine.
8.Review on classification of curative effect evaluation standard to modified Huangqi-Guizhiwuwu Tang on diabetic peripheral neuropathy

There are lack of specific treatment for diabetic peripheral neuropathy (DPN)right now.However, TCM intervention highlights the distinct advantage for the disease.Modified Huangqi-Guizhiwuwu Tang is one of the important recipes for that.But there are no uniform standards curative effect evaluation for it, and some clinical trials do not have some new evaluation standards.We summary the clinical research literatures in recent years and analyze the existing problems, therefore, provide new recommendations as views for medical researchers in the design of clinical trials.
9.Nursing of locally preserved skin of female patients for neurosurgical cranioctomy

Objective To summarize the experience in nursing locally preserved skin of female patients for neurosurgical cranioctomy. Method The nursing key points were given to 66 female patients undergoing local preparation of skin for cranioctomy as follous preoperative preparation, observation over incision and head nursing. Results The time for skin preparation ranged between 3 to 6 minutes. Postoperatively, no drain tube was placed. No intracranial infections occurred. The incisions in all patients were healed at the first class. Conclusion The nursing measures of good preoperative preparation, close observation of incisions and care to the head for female patients undergoing local preparation of skin for cranioctomy can enhance their satisfaction and propel development of standardized nursing.
10.The curative effect study of inhaled corticosteroids in the treatment of infant with wheezy bronchitis

Objective To study the curative effect of inhaled corticosteroids in the treatment of infant with wheezy bronchitis through clinical symptom score. Methods One hundred infant of 4-23 months old with wheezy bronchitis treated were enrolled. According to the clinical symptom score, they were divided into light group (40 cases), moderate group (30 cases) and severe group (30 cases). Patients in each group were divided into atomization group and control group by random digits table. The clinical symptom score was compared between before treatment and after treatment. Results In light, moderate and severe group, the clinical symptom scores 24, 48, 72 h after treatment in atomization group were significantly lower than those in control group (P<0.01 or<0.05). In light, moderate and severe group, the clinical symptom scores in atomization group were significantly decreased 24 h after treatment (P <0.05). In light and moderate group, the clinical symptom scores in control group were significantly decreased 48 h after treatment (P < 0.05), and in severe group, the clinical symptom score in control group was significantly decreased 72 h after treatment (P < 0.05). Factorial design data of variance analysis showed that in the treatment method, there were significantly differences between atomization group and control group (F=66.393, 5.093 and 154.326;P<0.01), in the treatment time aspect, there were significantly differences between atomization group and control group (F = 263.836, 182.568 and 75.587;P<0.01), and in terms of treatment method × time, there were significantly differences between atomization group and control group (F = 13.956, 6.414 and 5.628; P < 0.01). Conclusions Inhaled corticosteroids therapy in light, moderate and severe infant wheezy bronchitis can obviously improve the clinical symptoms and reduce the clinical symptom scores.
