1.Relationship between hotspot mutations in basic core promoter of hepatitis B virus and familial clustering of hepatocellular carcinoma in Guangxi

Objective To explore the relationship between mutations in basic core promoter (BCP) of hepatitis B virus (HBV) and familial clustering of hepatocellular carcinoma (HCC) in Guangxi. Methods 153 pairs of members with HBsAg-positive were selected and matched from HCC high-incidence families and carcinoma-free families in Guangxi. The BCP genes were amplified and sequenced. Results The hotspot sites of the previous five mutations in BCP were T1762, A1764, G1775, V1753, G1803. In univariant analysis, HBV DNA≥105 copies/mL, T1762, A1764 and V1753 mutations were associated with the HCC high-incidence (P <0.05). The multivariate logistic analysis showed that HBV DNA≥105 copies/mL and A1764 were independent risk factors for it. Conclusion HBV DNA level, the mutations in BCP showed correlations with familial clustering of HCC in Guangxi.
2.Analysis of hepatitis B virus genotype in regions in Guangxi with high incidence of primary liver cancer

Objective To explore the distribution and characteristics of hepatitis B virus (HBV) genotype in the region in Guangxi with high incidence of primary liver cancer (PLC). Methods 103 pairs of samples from the sex- and age-matched members with HBsAg-positive from PLC-clustering families (the experimental group) and carcinoma-free families (control group) were collected. Nested polymerase chain reaction (PCR) and sequencing methods were applied for the analysis of HBV genotype. Results Four HBV genotypes: B, C, B/C and D, were detected, the percentages of them in the two groups were 31.1%, 63.1%, 1.9%, 1.9% and 30.1%, 55.3%, 6.8%, 2.9%, respectively, showed no significant differences (P > 0.05). HBeAg positive rates were significantly different between genotype C and B (P < 0.05), but no significant differences were showed in such factors as gender, age, ethnic group and HBV DNA level in them (P > 0.05). Conclusions The main genotypes were types B and C besides a small number of combined genotypes B/C and D in the regions of Guangxi with a high incidence of PLC. There may be few relationships between HBV genotypes and the high incidence of PLC for familial clustering in Guangxi.
3.Exploration of the relationship between the expression level of Th1/Th2 cytokines and the familial aggregation of the hepatocellular carcinoma

Objective To investigate the effect of Th1/Th2 cytokines and immune state on the occurrence and familial aggregation of hepatocellular carcinoma(HCC).Methods Ninety-five members whose families have had two or even more HCC patients(high-occurrence families) were selected as the case group,by matching with the same nationality,gender,residential area,age±5 years old,95 members whose families had no any cancer were selected as the control.The level of peripheral blood Th1 type cytokines such as interferon-γ(IFN-γ),interleukin-2(IL-2) and Th2 type cytokines such as interleukin-4(IL-4),interleukin-10 (IL-10) were detected by enzyme linked immunosorbent assay(ELISA).Results There was a Th1/Th2 serum cytokine imbalance profile in members of HCC high-occurrence family.The levels of IFN-γ and IL-2 were significantly lower in members of HCC high-occurrence family than that of the controls.The levels of IL-4 and IL-10 were higher in members of HCC high-occurrence family than that of the controls.Conclusion There was a poor cellular immune state in members of HCC in the high-occurrence families.Th1 type cytokines was inhibited,and Th2 type cytokines was enhanced,so more susceptible to HBV chronic infection.It might be the mechanism of HCC occurrence and familial aggregation.
4.A Novel Recombinant BCG Vaccine Encoding Eimeria tenella Rhomboid and Chicken IL-2 Induces Protective Immunity Against Coccidiosis.

A novel recombinant Bacille Calmette-Guerin (rBCG) vaccine co-expressed Eimeria tenella rhomboid and cytokine chicken IL-2 (chIL-2) was constructed, and its efficacy against E. tenella challenge was observed. The rhomboid gene of E. tenella and chIL-2 gene were subcloned into integrative expression vector pMV361, producing vaccines rBCG pMV361-rho and pMV361-rho-IL2. Animal experiment via intranasal and subcutaneous route in chickens was carried out to evaluate the immune efficacy of the vaccines. The results indicated that these rBCG vaccines could obviously alleviate cacal lesions and oocyst output. Intranasal immunization with pMV361-rho and pMV361-rho-IL2 elicited better protective immunity against E. tenella than subcutaneous immunization. Splenocytes from chickens immunized with either rBCG pMV361-rho and pMV361-rho-IL2 had increased CD4+ and CD8+ cell production. Our data indicate recombinant BCG is able to impart partial protection against E. tenella challenge and co-expression of cytokine with antigen was an effective strategy to improve vaccine immunity.

5.Investigation and analysis of anxiety/depression in patients with asthma-COPD overlap

Objective:We aimed to estimate the incidence of anxiety or depression in asthma-chronic obstructive pulmonary disease (COPD) overlap (ACO) patients and explore its related factors.Methods:Stable patients who were treated or followed up in the outpatient department of respiratory medicine in the First Hospital of China Medical University from March 2018 to March 2019 were enrolled, including 53 ACO patients, 74 asthma patients, and 138 patients with COPD. Hamilton Depression Scale (HAMD) and Hamilton Anxiety Scale (HAMA) were used to evaluating the anxiety and depression status of the three groups of patients, and the risk factors of anxiety and/or depression in ACO patients were analyzed.Results:There were 33 cases (62.3%), 35 cases (47.3%) and 69 cases (50.0%) combined with anxiety in ACO, asthma and COPD group; there were 29 cases (54.7%), 25 cases (33.8%) and 58 cases (42.0%) combined with depression in ACO, asthma and COPD group, respectively. The patients with depression in ACO were significantly higher than those in asthma group ( P<0.05). There were 28 patients (52.8%), 24 patients (32.4%) and 52 patients (37.7%) combined with anxiety and depression in ACO, asthma and COPD group, respectively, with significant difference in ACO group and asthma group ( P<0.05). Multivariate logistic regression analysis showed that chronic obstructive pulmonary disease assessment test (CAT) score≥10 was associated with anxiety ( OR=15.00, P<0.01) or depression ( OR=8.667, P<0.01) in patients with ACO. Conclusions:Anxiety/depression is common in chronic airway disease. Compared with asthma, anxiety and/or depression is more frequent in the patients with ACO. ACO patients with high CAT scores should pay attention to screening for anxiety/depression.
