1.Clinical significance of the detection of myocardial enzymes and high sensitive C- reactive protein in chil-dren with herpangina

Objective To evaluate the clinical significance of myocardial enzymes and high sensitive C-reactive protein( hs-CRP) in children with herpangina. Methods A total of 80 children with herpangina( obser-vation group,n=80),at the same time,50 healthy children as control group(n=50)were assigned for the detec-tion of myocardial enzymes(AST,LDH,CK,CK-MB)and hs-CRP. Results The elevated CRP levels were found in 45(56. 25%)patients of observation group and 2(4%)patients of the control group,showing statistical significance(χ2 =31. 55,P<0. 01). Abnormal myocardial enzyme levels were observed in 59(73. 75%)cases, which included AST 51. 6%, CK 40. 1%, CK-MB 21. 4%, LDH 72. 2%. Abnormal myocardial enzyme levels were observed in 6(12%) cases of control group,which included AST l6%,CK 10%,CK-MB 4% and LDH 12%. The significant differences were observed between the two groups ( P<0. 01 ) . Conclusion Herpangina children may have different degrees of myocardial damage. Myocardial enzymes and hs-CRP are useful indicators for myocardial injury which may effectively provide reference for clinical diagnosis and treatment.
2.Application of orthogonal design to optimize real-time quantitative PCR conditions

Objective To investigate the application of orthogonal design to optimize real-time quantitative PCR conditions and perform it for ICAM-1.Methods After model of a skin injury was established in rats,total RNA was isolated from the injured skin tissue and reverse transcriptase reactions were carried out.Using orthogonal design real-time quantitative PCR conditions,the expression level of ICAM-1 gene was studied by use of cDNA as template.The standard of score was set up to amplification and dissociation curves and statistical analysis was done based on the optimized conditions.Results Through 25 tests the best real-time quantitative PCR conditions for ICAM-1 are anneal temperature at 61 ℃,0.4?g of cDNA and concentration of 150?mol/L each primer in the final reaction.There are no interaction among above factors.Conclusion Orthogonal design is a feasible,quick and economical method to optimize real-time quantitative PCR conditions.
3.Study on high-throughput respiratory tract virus detection method based SPR

Objective To develop a biosensor method with strong specificity and high‐throughput by combining with the surface plasmon resonance (SPR) and gene chip technique and aiming at 9 kinds of common respiratory tract viruses including influenza A and influenza B ,(Influ A ,B) ,H1N1 ,respiratory syncytial virus (RSV) ,parainfluenza virus 1 -3 (PIV1 -3) ,adenovirus (ADV) and coronavirus (SARS) leading to severe acute respiratory syndrome .Methods Firstly the software primer 5 was used to design the specific primer and probe of related viruses in the conserved sequence ;the designed nine kinds of corresponding respiratory virus probes were immobilized in the specific region of SPR chip after chemical modification .The SPR technique was applied to conduct the real time monitoring the hybridization process of the probe with the PCR products .Finally the signal amplification was realized by the biotin and streptavidin system .Results The designed gene chip could detect 9 kinds of respiratory tract viruses by high‐throughput with better detection specificity ;the chip surface could be reutilized after certain regeneration condition ,which avoided the influence of intra‐batch difference on the results ;the detection sensitivity reached the nanomole level .Conclusion The prelimi‐nary study results demonstrate that using the SPR biosensor technique to establish a high‐‐throughput detection of respiratory tract viruses has some practicability and feasibility ,and is expected to become a rapid ,large scale and high‐ throughput measure for screening respiratory tract viruses with good application prospect .
4.Preliminary study of endoluminal two-dimensional ultrasonography to evaluate puborectalis muscle of nulliparous women

Objective To investigate the measurement standards and clinical application of observing anatomical morphologies and contractile function of puborectalis muscle on pelvic floor parasagittal plane by endoluminal two-dimensional ultrasonography.Methods The thickness and thickening rate of puborectalis muscle of 78 young nulliparous women were measured on pelvic floor parasagittal plane at three levelsurethral level (anterior),vaginal level (central) and rectal level (posterior),both at rest and during contraction.And the thickening rates of these parts during contraction were compared with each other.Interclass correlation coefficients were calculated to evaluate the consistency of the datas.Results At rest,thickness of anterior part of the left was (9.23 ± 0.20)mm,and that of the right was (8.99 ± 0.20)mm.During contraction,thickness of anterior parts of bilateral puborectalis muscle were significantly thicker than that of central or posterior parts (P <0.05).The interclass coefficients were more than 0.93 and 0.83.Conclusions The endoluminal two-dimensional ultrasonography can be used to observe morphologies and contractile function dynamically of puborectalis on pelvic floor parasagittal plane.It is simple,reproducible and worthy of clinical promotion.
5.The clinical value of four methods of endometrium preparation for frozen-thawed embryo transfer cycle

Objective To estimate the clinical value of 4 methods of endometrium preparation for frozen‐thawed embryo transfer(FET ) cycle .Methods Total 320 FET cycles completed in center of reproductive medicine of Qingyuan people′s hospital from March 2013 to March 2015 were retrospectively analyzed ,and divided into 4 groups :natural cycle group(group A ,n=70) ,o‐vulation‐inducing cycle group(group B ,n=152) ,hormone replacement treatment(HRT) cycle group (group C ,n=74) ,down‐regu‐lated HRT cycle (group D ,n=24) .The pregnancy outcomes were followed and compared among these different 4 groups .Results In FET cycles ,the clinical pregnancy rates and implantation rates of group B ,group C and group D were significantly higher than that of group A (P<0 .05) ,there were no significant differences in the rates of multiple pregnancy ,biochemical pregnancy ,early a‐bortion and ectopic pregnancy among 4 groups(P>0 .05) .However ,the rank of the FET cycle of group D was significantly higher than those of the other three groups(P<0 .05) .Conclusion In FET cycles ,the methods of ovulation‐inducing ,HRT and down‐reg‐ulated HRT all have favorable clinical outcomes ,and the method of down‐regulated HRT is more suitable for patients with previous implantation failure .

7.Analysis of digital mammography and clinical manifestations of rare diseases in male breast

Objective To probe the X-ray imaging features and clinical manifestations of some rare diseases in male breast.Methods The data of 12 patients with some rare diseases in male breast were collected.The X-ray manifestations and clinical features of the diseases were analyzed.Results The postoperative pathology showed the lipogranuloma in 1 patient,plasma cell mastitis in 2,secondary infection of epidermal cyst in 1,intraductal papilloma in 1,breast cancer in 6 (invasive ductal carcinoma in 4,poor-differentiated adenocarcinoma in 1,and mucinous carcinoma in 1),and breast lymphoma in 1.The digital mammography showed the intramammary solitary masses in 1 1 and multiple ones in 1.10 masses were located at the rear of the areola,and other 2 were opposite.Unclear edge of the lesion was found in 9,spiculation in 1,lobulation in 1,and micro-calcification in 1.Nipple retraction and areola skin thickening in 10 and enlarged axillary lymph node in 1 were also found.The total accurate rate of X-ray diagnosis was 41% (5/12),while that of breast cancer was 83.3%.The total misdiagnosis rate was 59% (7/12),and that of benign lesion was 80% (4/5).Conclusion The diseases in male breast are rare and various,and the X-ray and clinical manifestations are not specific.
8.Evaluation on lnsulin Combined HCG Improved Method in Establishment of Obese PCOS Rat Models

This study was aimed to establish and evaluate obese polycystic ovary syndrome (PCOS) rat model. Rats were randomly divided into the blank control group, high-fat model group, insulin (INS) combined human chorionic gonadotropin (HCG) model group, and INS combined HCG plus high fat emulsion model group. The obese PCOS rat model was induced by subcutaneous injection of INS and HCG on the nape, respectively. The intragastric administration of high fat emulsion was also used in the PCOS rat model establishment. The estrous cycle of rat was monitored. The detection was also made the weight increasing rate of rats, luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol (E2), testosterone (T), high-density lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TG), cholesterol (TC), fasting plasma glucose (FPG), fasting insulin (FINS), 2 h blood glucose and 2 h INS. Calculation was made on the HOMA index, related viscera index and bilateral ovaries HE staining. The results showed that when INS combined HCG improved method (INS combined HCG plus high fat emulsion model group) rats were sacrificed, the body weight, weight increasing rate, ovary viscera index, T, LH, and HOMA index were significantly higher than that of the blank control group, high-fat model group and INS combined HCG model group (P < 0.05,P < 0.01); HDL was significantly decreased (P < 0.05,P < 0.01). FPG, FINS, and 2 h INS of the INS combined HCG improved method group were significantly higher than that of the high-fat model group (P <0.01). It was concluded that INS combined HCG improved method was one of the ideal animal model establishment methods in the pathogenesis study of PCOS.
9.Relation between Injury Time and the Expression of COX6C mRNA in Skeletal Muscle of Rats after Contusion

Objective T o investigate the relation betw een injury tim e and the expression of cytochrom e c oxidase subunitⅥc (COX6C) m R N A in skeletal m uscle of rat after contusion. Methods A total of fifty-four SD rats w ere divided into the control group and the contusion groups (0.5,1,6,12,18, 24, 30, and 36 h after contusion), random ly. T he contusion m odel w as established by free fall drop of gravity ham-m er. A t corresponding tim e point after contusion, the regular histology w as exam ined and expression level of COX6C m R N A w as tested by real-tim e PC R after extraction of total R N A from the tissues. Results T he m ain pathological features of 6 h after injury included edem a and hem orrhage in m yocytes w ith no inflam m atory cells found. A fter 6 hours, the findings included m yocyte degeneration and necro-sis, inflam m atory cells infiltration, and fibrous connective tissue proliferation in the contused zone. T he expression level of COX6C m R N A w as higher than that of the control group w ithin 6 h after contusion. T he expression level w as low er than that of the control group from 6-36 h after contusion. Conclusion T he level of COX6C m R N A expresses in a regular w ay after contusion. It m ay be useful for estim ating w ound age in com bination w ith the results of pathological features.
10.Relative Expression of Indicators for Wound Age Estimation in Forensic Pathology

Objective In order to understand which kind of function genes play an important role for es-timating wound age, the variation of difference genes’ mRNA expression were compared after injury. Methods T he mRNA expression levels of seven candidate genes (ICAM-1, NF-κB, MX2, MT1, MT2, sTnI, and Cox6c) were analyzed in contused rat skeletal muscle at different time points using real-time fluorescent quantitative PC R (R T-qPC R ). T he rawC t values were normalized relative to that of RPL32 mRNA , and converted to standard C t values. A t each time point after injury, the standard deviations (SD ) of the standard C t values were calculated by SPSS. Results T he expression trends of the seven genes were all found to be related to wound age, but there were lower variation coefficients and greater reliability of sTnI and Cox6c when compared with other genes. Conclusion T he genes encoding struc-tural proteins or proteins that performbasic functions can be suitable for wound age estimation.
