Objective To study the effect of Iguratimod on the collagen secretion of 3T6 cells. Method The median toxic concen- tration of Iguratimod was determination by MTT Assay. Iguratimod' s median toxic concentration to 3T6 cell was about 1142.6mg/L. Taking into the consideration of the cells performance after administration under spectacles, we chosen the administration concentration as 334.8 mg/L(DMS0:1?l/256?l) ,167.4mg/L,83.7mg/L and 41.85 mg/L. After administration of 12 hours, 24 hours, 36 hours and 48 hours, the supernatant was collected for assayed respectively. The collagens content was detected by the method of chloramine T. 2.Effects of deliberate hypotension combined with acute isovolemic hemodilution on glomerular filtration rate in rabbits

Objective To determine the effects of acute normovolemic hemodilution combined with deliberate hypotension induced by sodium nitroprusside on glomerular filtration rate. Methods Forty rabbits weighing 2.6-4.0kg were anesthetized with intramuscular ketamine(1mg?kg-1). The animals were tracheotomized and intubated.Anesthesia was maintained with 0.5% halothane inhalation and fentanyl infusion (2?g?kg-1?min-1 ) . Carotid artery was cannulated for blood pressure monitoring and blood gas analysis and external jugular vein was cannulated for CVP monitoring and blood sampling. The animals were randomly allocated to one of four groups of ten animals in each group: control group; deliberate hypotension group;hemodilution group and combined hemodilution-deliberate hypotension group. Deliberate hypotension was induced with 0.02% sodium nitroprusside and MAP was reduced to 35-40mm Hg. In hemodilution group and combined group blood was removed from artery until Hct was reduced to 20 % -25 % and normovolemia was maintained by simultaneous infusion of Ringer's lactate solution, 3 times the volume of blood removed. Glomerular filtration rate was assessed by inulin clearance rate. 1% inulin 3 ml was injected I. V. over 30 s and blood samples were taken from CVP line for determination of blood inulin concentration at 1,3,5, 10, 30, 60, and 100 min after injection of inulin. Results Urine output decreased significantly in hypotension group and increased in hemodilution group. There was no significantly difference in urine output between control and combined groups. Inulin clearance decreased significantly in hypotension group as compared with that in control group, but there was no significantly difference in inulin clearance between control and combined groups. Conclusions Deliberate hypotension can decrease renal blood flow and deliberate hypotension combined with hemodilution can maintain kidney microcirculation.

4.Subcellular localization and resistance to Gibberella fujikuroi of AtELHYPRP2 in transgenic tobacco.

The subcellular localization and the resistance to fungal pathogen Gibberella fujikuroi of the protein encoded by Arabidopsis AtELHYPRP2 (EARLI1-LIKE HYBRID PROLINE-RICH PROTEIN 2, AT4G12500) were investigated using transgenic tobacco plants. The coding sequence of AtELHYPRP2 was amplified from genomic DNA of Col-0 ecotype. After restriction digestion, the PCR fragment was ligated into pCAMBIA1302 to produce a fusion expression vector, pCAMBIA1302-AtELHYPRP2-GFP. Then the recombinant plasmid was introduced into Agrobacterium tumefaciens strain LBA4404 and transgenic tobacco plants were regenerated and selected via leaf disc transformation method. RT-PCR and Western blotting analyses showed that AtELHYPRP2 expressed effectively in transgenic tobacco plants. Observation under laser confocal microscopy revealed that the green fluorescence of AtELHYPRP2-GFP fusion protein could overlap with the red fluorescence came from propidium iodide staining, indicating AtELHYPRP2 is localized to cell surface. Antimicrobial experiments exhibited that the constitutive expression of AtELHYPRP2 could enhance the resistance of tobacco to fungal pathogen G. fujikuroi and the infection sites could accumulate H2O2 obviously. The basal expression levels of PR1 and the systemic expression levels of PR1 and PR5 in transgenic tobacco plants were higher than that of the wild-type plants, suggesting AtELHYPRP2 may play a role in systemic acquired resistance.

5.Clinical observation on sedative effect of Tianwang-Buxin Dan for chronic obstructive pulmonary disease patients undergoing mechanical ventilation

Objective To observe the clinical sedativelefficacy of Tianwang-Buxin Dan treating chronic obstructive pulmonary disease patients undergoing mechanical ventilation.Methods 97 patients with chronic obstructive pulmonary disease undergoing mechanical ventilation were randomly recruited into a treatment group (47 cases) and a control group (50 cases).The control group was treated with midazolam and fentanyl,and the treatment group were treated by Tianwang-Buxin Dan on the basis of the control group.The amount of medazolam and fentanyl,duration of MV,and incidence of side-effects such as hypotension,bradycardia,delirium,nausea,etc were observed in both groups.Results The expected sedative and analgesia scores were obtained in patients of both groups.Compared with the control group,the patients in the treatment group were easier to be aroused and be kept in sedation and analgesia state,besides the dose of midazolam (μg/kg·h-1) (9.51 ±5.87 vs 20.01 ±6.24,P<0.01)and fentanyl (μg/kg·h-1) was significantly fewer (0.22 ±0.13 vs 0.32±0.12,P<0.05),duration of MV (days) was shorter(7.13±6.25 vs 12.85±9.13,P<0.01),duration of disable sedatives for extubating (hours)was shorter (4.35 ± 2.57 vs 8.79±4.02,P<0.01),the rates of hypotensionand brdycarcardia were significantly lower,the rates of delirium and nausea were clearly lower(both P<0.05)in the treatment group.Conclusion Sedative effect of Tianwang-Buxin Dan is satisfactory for patients with chronic obstructive pulmonary disease undergoing mechanical ventilation,with the property of easier arousal,lower hypotension rates,lower brdycarcardia rates,lower delirium rates nausea rates,and fewer dosage of midazolam (by 50%) and fentanyl (by 30%) administration.
7.Electrocatalytic Properties of Dopamine and Uric Acid at Vitamin B1 Self-assembled Monolayer Gold Electrode

Vitamin B1 self-assembled monolayer modified electrode (VB1-Au/SAMs/CME) was prepared and its electrochemical behavior was studied by cyclic voltammetry. The results showed that a self-assembled monolayer (SAM) was formed on the surface of gold electrode. \3-/4-redox couple was used as the electrochemical probe to study the electrochemical characteristics of SAM modified gold electrode and the results indicated that the existence of Vitamin B1 membrane on gold surface hindered the electron transfer. The electrochemical behaviors of dopamine(DA) and uric acid (UA) were investigated at VB1-Au/SAMs/CME. The results showed that the electrochemical oxidation of DA and UA could be electrocatalyzed. There were linear relationships between the anodic peak current and the concentration of DA(8.0×10-6-4.0×10-4 mol/L) and UA (6.0×10-5-2.2×10-4mol/L) respectively in differential pulse voltammograms. This modified electrode was effective to detect DA and UA in simultaneous determination of these species in a mixture.
8.Organochlorines, Glutathione S-transferase M1 Genetic Polymorphisms and Risk of Breast Cancer

Objective To study the potential effect of gene-environment interaction between glutathione S-transferase M1 (GSTM1) and serum organochlorines residues on the risk of breast cancer in women, in China. Methods Seventy newly pathologically diagnosed female patients with breast cancer from September 2006 to October 2007 were selected as the cases from five large hospitals in Tangshan. The controls were identified at the same hospital as cases. 1∶1 matched case-control study. Between the cases and controls, the difference of age was not over two years and the residence was similar. The organochlorine residues levels in the serum were measured by gas chromatography (GC). Genotypes of GSTM1 polymorphisms were analyzed by multiplex allele-specific polymerase chain reaction (PCR). Interaction indexes (?) were calculated to determine the type of gene-environment interaction. Results After confounding factors adjusted, the result showed that interaction existed in genetic polymorphisms of GSTM1 and DDT, HCH residues, and interaction indexes (?) value were 1.237 and 1.379. Conclusion GSTM1 genetic polymorphisms and DDT, HCH may present an interaction in the development of breast cancer.
9.Summary of the 2008 national autoantibody testing quality control

Objective To consecutively understand the national clinical testing status and to reassure the quality-control of autoantibody detection. Methods Letter or telephone notification were conducted to the participating hospitals or departments. Autoantibodies for quality control survey included anti-nuclear anti-body (ANA), anti-double-stranded DNA (anti-dsDNA), anti-extractable nuclear antigens (A-ENA), anti-mitochondria antibody (AMA)/anti-smooth muscle antibody (ASMA), and anti-citrulline antibody (anti-CCP). Each had 3 control samples, and altogether 15 samples for testing. Sample distribution and data analysis were double-blinded. Results One hundred and two hospitals/departments participated in the national quality-control survey. The accurate rate for this survey was 70%, 88%, 93%, 85%, 79% respectively for ANA, anti-dsDNA, AMA, ASMA and anti-CCP. Anti-ENAs were further divided into anti-RNP, Sm, SSA, SSB and Scl-70 subgroups, and the accurate rate was 88%, 77%, 92%, 93% and 87% respectively. Conclu-sion Compared to the previous 4 national surveys, the accurate rates of ANA, anti-dsDNA, anti-ENAs, AMA in our country's autoantibody testing is improved, and the detection rate of ASMA and anti-CCP antibody is also increased. More hospitals and testing items can test these autoantibodies, which implies that autoantibody testing status has been improving in our country.
10.Effects of Baizhu Huangqi Decoction and Its Effective-part Prescription on Mice Ulcerative Colitis

Objective To observe the effects of Baizhu Huangqi Decoction and its effective-part prescription on mice ulcerative colitis(UC).Methods The mice UC model was induced by clyster with 2,4,6-trinitrobenzenesulfonic acid(TNBS)and the effects were comprehensively evaluated by disease activity index,the macroscopic and histological assessment of colon mucosa damage and the activity of myeloperoxidase(MPO).Results Both Baizhu Huangqi Decoction and its effective-part prescription had effects on disease activity index(DAI),inflammation index and MPO activity.The effect of effective-part prescription was better than that of UC model group(P
