1.Proliferation and differentiation of rat bone marrow mesenchymal stem cells under different mechanical strains

BACKGROUND:In vitro and in vivo studies of cel response to a variety of mechanical loadings have demonstrated the stimulation of bone formation by loads. However, the effects of different mechanical strains on the same cel s have never been adequately studied by far.
OBJECTIVE:To investigate the effects of different mechanical strains on rat bone marrow mesenchymal stem cel s.
METHODS:Rat bone marrow mesenchymal stem cel s were isolated and cultured in vitro. Bone marrow mesenchymal stem cel s were subjected to different stimulations including dynamic stretch, static stretch and hybrid stretch through the use of custom-made mechanical stretch device. Cel ular proliferation, alkaline phosphatase activity and mRNA expression of Runx2 of bone marrow mesenchymal stem cel s were detected and the secretion of osteocalcin was evaluated under three different stretch modes respectively.
RESULTS AND CONCLUSION:Compared to the control group, cel proliferation increased by 18.67%, however, alkaline phosphatase activity, Runx2 expression and osteocalcin secretion were not changed obviously in the static stretchgroup. Compared to the control group, alkaline phosphatase activity, Runx2 expression and osteocalcin secretion increased by 60.33%, 49.67%and 48%respectively;however, cel proliferation was inhibited, in the dynamic stretch group. Compared to the control group, cel proliferation was slightly, but not significantly, increased in the hybrid stretch group, and the alkaline phosphatase activity, Runx2 expression and osteocalcin secretion increased although the increases were not as apparent as those in the dynamic stretch group. These findings suggest that static mechanical strain can significantly promote cel proliferation, the dynamic mechanical strain more greatly promotes osteogenic differentiation of bone marrow mesenchymal stem cel s, and the hybrid mechanical strain promotes the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cel s.
2.Pharmaceutical Care for One Case of Rheumatoid Arthritis Complicated with Severe Pneumonia

Objective:To explore the role of clinical pharmacists in the drug treatment of one patient with rheumatoid arthritis and severe pneumonia. Methods: The drug treatment of one patient with rheumatoid arthritis and severe pneumonia involved in clinical pharmacists was retrospectively analyzed. Clinical pharmacists provided rational suggestions for the adjustment of anti-infection treat-ment,selection of medication, evaluation of therapeutic effect and monitoring of adverse drug reactions. Results: Clinical pharmacists participated in the whole medication treatment process, and their suggestions on anti-infection drugs choices, dose adjustment and ad-verse reactions treatment were accepted by doctors. Conclusion:Taking use of self professional competence, clinical pharmacists play a positive role in the optimization and adjustment of drug treatment regimen, which provides security for the medication safety and clini-cal rational drug use.
3.One Case Analysis of Insect Bites Induced Rhabdomyolysis Complicated with Acute Renal Failure

Objective: To reveal the harm seriousness of bug bites induced rhabdomyolysis complicated with acute renal failure to human body and provide case evidence for clinical workers to improve the understanding and treatment level of insect bites related diseases.Methods: The clinical diagnosis and treatment of one case of insect bites induced rhabdomyolysis complicated with acute renal failure was tracked, and the individualized diagnosis and treatment were performed and summarized according to the clinical symptoms and the conditions of the patient.Results: All the indicators of the patient returned to normal with satisfactory curative effect and promising prognosis.Conclusion: The treatment of rhabdomyolysis complicated with acute renal failure needs quick identification and reversible factors correction, which are beneficial to the improvement of organ function and prognosis.
4.Research on and design of visual prosthesis based on visual information processing.

Induced by a variety of retinopathy, visual loss has become the most serious form of disability, which influences the quality of human life. With the rapid development and crossing among the information science, microelectronics, material science and biomedical disciplines, the visual prosthesis makes reparation possible for the visual blindness caused by retinitis pigmentosa, age-related macular degeneration, and other eye, retina, optic nerve and visual cortex lesions. With technology innovation, the prosthesis design, manufacturing and surgical technique are no longer the biggest obstacles to the future development of the visual prosthesis, but how to construct effective transmission of information between the brain and the prosthesis. However, due to the complex structure of the human visual system, the visual prosthesis manufacturing and visual information signal mapping are facing some difficulties. Thus, we can only study the representation strategy of image information and micro-electrode array stimulation basing on limited pixels of simulated prosthesis visual information. By studying the visual information processing of the visual prosthesis, we propose a visual prosthesis design which is based on biological, mechanical, and electronic integration.

6.Animal models of white matter lesions

White matter lesion is a major subtype of cerebral small vessel disease. Its pathophysiology and mechanism remain unclear. Because the risk factors often coexist in clinical research, it is difficult to judge the relationship between certain risk factors and white matter injury. Moreover, due to the differences in animal and human brain tissue structure, there is currently a lack of reproducible animal models of white matter lesions. Therefore, establishing a practical animal model and further exploring the pathogenesis and risk factors of white matter lesions from the basic research level is crucial for the preclinical study of the treatment of white matter damage. This article reviews the characteristics, optimization measures, and application prospects of the white matter lesion models.
7.Construction and validation of a risk prediction model for postoperative delirium in primary liver cancer patients aged 60 years or older

ObjectiveTo construct a risk prediction model for postoperative delirium in elderly patients with primary liver cancer, and to validate its application value. MethodsA retrospective analysis was performed for 175 elderly patients with primary liver cancer who were admitted to Tianyou Hospital Affiliated to Wuhan University of Science and Technology from March 2020 to January 2023. The incidence rate of postoperative delirium was recorded, and the univariate and multivariate regression analyses was performed for factors that may affect the onset of delirium. A prediction model was constructed, and the clinical application value of the prediction model was analyzed and validated. The independent-samples t test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups. The univariate and multivariate logistic regression analyses were performed for factors that may affect the onset of delirium in elderly patients with primary liver cancer, and the receiver operating characteristic (ROC) curve was used to investigate the value of the model in predicting the onset of delirium. ResultsAmong the 175 elderly patients with primary liver cancer, 41 experienced postoperative delirium, with an incidence rate of 23.43%. The univariate analysis showed that age, presence of more than two underlying diseases, Child-Pugh class of liver function, preoperative blood lactate, time of operation, preoperative hemoglobin, and preoperative serum albumin were associated with the onset of postoperative delirium (t=3.534, χ2=12.000, χ2=4.938, t=7.561, t=5.768, t=5.141, t=6.148, P<0.05). The multivariate logistic regression analysis of the factors with statistical significance in the univariate analysis showed that time of operation, preoperative hemoglobin, preoperative serum albumin, and age were included in the regression model (P<0.05), and they were independent risk factors for the onset of postoperative delirium in elderly patients with primary liver cancer. According to the results of the multivariate logistic regression analysis, a prediction model for postoperative delirium in elderly patients with primary liver cancer was constructed as follows: -2.222+3.678×time of operation-2.441×preoperative hemoglobin-3.904×preoperative serum albumin+1.807×age. The prediction performance of this model was analyzed, with an area under the ROC curve of 0.931 (95% confidence interval: 0.890 — 0.971, P<0.001) and an optimal cut-off value of -1.604 (with a sensitivity of 87.80% and a specificity of 87.30%). A total of 56 elderly patients with primary liver cancer who underwent radical surgery in Tianyou Hospital Affiliated to Wuhan University of Science and Technology from February 2023 to June 2023 were enrolled in a prospective study for model validation. According to the above risk prediction model, there were 14 patients in the high-risk group and 42 patients in the low-risk group, and the high-risk group had a significantly higher incidence rate of postoperative delirium than the low-risk group (71.43% vs 11.90%, χ²=16.056, P<0.05). ConclusionAge, time of operation, preoperative serum albumin, and preoperative hemoglobin are important influencing factors for the onset of postoperative delirium in elderly patients with primary liver cancer. The risk prediction model based on these factors has a good prediction performance, which holds promise for further in-depth research.
8.Study on inhibitory effects of Wubao capsule on airway inflammation in asthmatic model mice by regulating the upstream and downstream cytokines of type Ⅱ innate lymphoid cells

OBJECTIVE To study the effects of Wubao capsule on airway inflammation in asthmatic model mice by regulating upstream and downstream cytokines of type Ⅱ innate lymphoid cells (ILC2s). METHODS Totally 40 female BABL/c mice were randomly divided into normal group, model group, positive control group (dexamethasone 1 mg/kg), Wubao capsule low-dose and high-dose groups (0.5, 1 g/kg), with 8 mice in each group. Asthma models were induced by ovalbumin (OVA) sensitization and nebulization. Each group was given normal saline or drug intragastrically for 7 consecutive days. The contents of IgE and OVA-IgE in serum, the contents of interleukin 5 (IL-5), IL-9, IL-13, IL-25, IL-33, thymic stromal lymphopoietin (TSLP) and mucin 5AC (MUC5AC) in bronchoalveolar lavage fluid (BALF) were determined by ELISA. HE staining was used to observe the pathological changes of lung tissues in mice. PAS staining was used to observe the changes of goblet cell proliferation in each group. The number of ILC2s in lung tissue was determined by flow cytometry (except for Wubao capsule low-dose group). RESULTS Compared with model group, the contents of IgE and OVA-IgE in serum and the contents of IL-5, IL-9, IL-13, IL-25, IL-33, TSLP and MUC5AC in BALF were significantly reduced in Wubao capsule high-dose and low-dose groups (P<0.01). The infiltration of inflammatory cells and the thickening of basement membrane in lung tissue was alleviated to varying degrees, and the proliferation of goblet cells was inhibited; the number of ILC2s in lung tissues of mice in Wubao capsule high-dose group was significantly reduced (P<0.01). CONCLUSIONS Wubao capsule could effectively reduce the number of ILC2s in lung tissue, the contents of upstream and downstream cytokines of ILC2s in BALF of asthmatic model mice, so as to inhibit the airway inflammation and improve asthma.

The many-banded krait, Bungarus multicinctus, has been recorded as the animal resource of JinQianBaiHuaShe in the Chinese Pharmacopoeia. Characterization of its venoms classified chief phyla of modern animal neurotoxins. However, the evolutionary origin and diversification of its neurotoxins as well as biosynthesis of its active compounds remain largely unknown due to the lack of its high-quality genome. Here, we present the 1.58 Gbp genome of B. multicinctus assembled into 18 chromosomes with contig/scaffold N50 of 7.53 Mbp/149.8 Mbp. Major bungarotoxin-coding genes were clustered within genome by family and found to be associated with ancient local duplications. The truncation of glycosylphosphatidylinositol anchor in the 3'-terminal of a LY6E paralog released modern three-finger toxins (3FTxs) from membrane tethering before the Colubroidea divergence. Subsequent expansion and mutations diversified and recruited these 3FTxs. After the cobra/krait divergence, the modern unit-B of β-bungarotoxin emerged with an extra cysteine residue. A subsequent point substitution in unit-A enabled the β-bungarotoxin covalent linkage. The B. multicinctus gene expression, chromatin topological organization, and histone modification characteristics were featured by transcriptome, proteome, chromatin conformation capture sequencing, and ChIP-seq. The results highlighted that venom production was under a sophisticated regulation. Our findings provide new insights into snake neurotoxin research, meanwhile will facilitate antivenom development, toxin-driven drug discovery and the quality control of JinQianBaiHuaShe.
