1.Comparison of liver toxicity of matrine and oxymatrine in m ice

OBJECTIVE To compare the liver toxicity of matrine and oxymatrine ,and to explore their toxic mechanism. METHODS Thirty ICR mice were randomly divided into normal control ,matrine 200 mg · kg-1 and oxymatrine 200 mg · kg-1 groups,10 mice per group. After single ig administration of corresponding drugs or water, animal mortality was calculated at the 15th day. The content of glutamic-pyruvic transami?nase(GPT),glutamic-oxalacetic transamin(GOT),alkaline phosphatase(ALP)and lactate dehydroge?nase (LDH) in serum were detected. Histopathological changes of the liver were examined by HE stain. The content of superoxide dismutase(SOD)and glutathione(GSH)in liver homogenates were detected by ELISA. Hepatocyte apoptosis was detected by Tunel stain. RESULTS The mortality rate of mice in two groups was 80% and 0,respectively. GPT,GOT and ALP contents of dead mice in matrine group were significantly higher than that in normal control group(P<0.05). In oxymatrine group,only the content of ALP was increased(P<0.05). Four of the eight dead mice in matrine group exhibited liver cell necrosis(P<0.05),while only 1/10 mice in oxymatrine group had a mild liver cell necrosis(P>0.05). The content of SOD and GSH of dead mice in matrine group was lower than that in control group(P<0.05,P<0.01). The content of GSH in oxymatrine group was also decreased(P<0.05). The apoptosis rate of liver cells in dead mice in matrine group was increased(P<0.05). CONCLUSION A large dose of matrine and oxymatrine can produce liver toxicity. At an equal dosage,the liver toxicity of matrine is significantly higher than that of oxymatrine. The toxic mechanism is related to oxidative stress and apoptosis.
3.Effect of psychological intervention plus Concentrated Tinidazole Gargles on dental plaque and gingivitis inflammation and influence factor of life quality in elder patients

Objective To investigate the effect of psychological intervention, combined with Concentrated Tinidazole Gargles on dental plaque and gingivitis inflammation and influence factor of life quality in elder patients. Methods The control group was given Concentrated Tinidazole Gargles treatment, the research group was given psychological intervention on the basis of Concentrated Tinidazole Gargles (as control group). Plaque index, gingivitis index and SF-36 scale changes before and after treatment were recorded between two groups of elderly patients with gingivitis. Results There was no significant difference in plaque index, gingival indexand SF-36 score compared with before treatment between two groups; Plaque index, gingivitis index and the SF-36 score were improved in the research group better than the control group after treatment (P<0.05). Conclusion There is positive significance in improving the clinical efficacy of Concentrated Tinidazole Gargles plus psychological intervention and assurance of the quality of life on gingivitis in elder patients.
4.Advantages and application of telemetry compared with the traditional methods in safety pharmacology

Objective To prove the advantages of telemetry by comparing with the traditional methods in safety pharmacology.Methods To monitor continuously the heart rate, respiratory rate, blood pressure and ECG of Beagle dogs by traditional and telemetry methods respectively, analyze and compare the changes between anesthetized and conscious dogs before and after feeding.Results Maintenance of anesthesia changed significantly the heart rate, respiratory rate, blood pressure and QT interval in the ECG of animals.The changes of physiological indicators in 24 h is not obvious in conscious animals, and showed a certain biorhythm.Compared with the conscious animals, the anesthetized dogs’ heart rate was significantly higher, blood pressure increased significantly, QRS and QTcf interval prolonged significantly, respiratory frequency decreased, heart rate increased significantly after feeding, and QTcf interval extended very significantly.Conclusions Traditional methods in safety pharmacology affect animal physiological indicators such as heart rate, blood pressure, respiratory rate and QT interval, which affect the objectivity of drug evaluation.Using conscious animals by telemetry can reduce these errors, however, the interference from outside should be eliminated.
5.Influence of different preservation conditions and times on the measurement of blood biochemical indicators in Beagle dogs

Objective Beagle dogs are commonly used animal for drug safety evaluation .As the necessary parameters , blood biochemical indicators are detected in acute or chronic toxicity tests .This study aims at assessing the influence of different preservation conditions and different preservation time on blood biochemical indicators to ensure the reliability of test results of long-term toxicity assessment .Methods Six Beagle dogs (3 males and 3 females) were used in this study .After collection and preparation of serum samples , biochemical indicators were detected after preservation in refrigerator at 2-8℃for 1, 2, 5, 8, and 12 hours;after preservation in ice transportation boxes at 2-10℃for 2, 5, and 8 hours;and after preservation in refrigerator at -20℃ for 1, 3, and 5 days.The biochemical indicators included alanine aminotransferase ( ALT ) , aspartate aminotransferase ( AST ) and alkaline phosphatase ( ALP ) , total protein ( TP ) , albumin (propagated), urea, creatinine (CREA), glucose (GLU), total cholesterol (TCHO), total bilirubin (TBIL), creatine kinase ( CK ) , gamma pancreatic acyl transferase ( GGT ) , calcium ( CA ) , lactate dehydrogenase ( LDH ) , phosphorus ( P) , high-density lipoprotein cholesterol ( HDL-C) , low density lipoprotein cholesterol ( LDL-C) , triglyceride ( TG) , sodium ( Na+) , potassium ( K+) and chloride ( Cl -) .Results Compared with the results of samples preserved for 1 hour, the LDL-C result of that preserved in refrigerator at -20℃for 5 days was significantly increased (P<0.05), and the coefficient of variation was 39%.The CK and LDH results of specimens preserved in refrigerator at -20℃ for 5 days was increased but not significantly ( P>0.05 ) , and the coefficient of variation of LDH was 41%.Conclusions According to the test results of blood biochemical indicators in the Beagle dogs detected after different preservation conditions and different preservation time in this study , detection test should be done within 1 hour, if not, detection should be done within 12 hours for the samples preserved at 2~8℃, or within 3 days for the sample preserved at -20℃.For transportation of serum samples , the serum samples should be placed in the ice box at 2~10℃, and detection test should be done within 8 hours .
6.Isolation of human genomic DNA from archival dried blood spots for neonatal disease screening and its application to methylation detection

Objective To establish an effective DNA isolation method for neonatal disease screening,so as to explore its application to the methylation detection.Methods The 20 dried blood spots samples were randomly divided into 2 groups according to the gender:the traditional method group (n =10) and the improved kit method group(n =10).The DNA quality was evaluated based on its concentration,integrity and whether it could be used in polymerase chain reaction (PCR).These DNA samples with or without bisulfite treatment were used as template in the methylation-specific polymerase chain reaction (MSP).The methylation levels of Leptin and tumor necrosis factor-α (TNF-α) gene promoter region were detected.Results DNA concentration of the improved kit method [(5.70 ± 0.81) mg/L] was significantly higher than that of the traditional method [(3.50 ± 0.45) mg/L] (t =2.79,P < 0.05),and biochemical analyzer analysis showed a better DNA integrity.Agarose gel electrophoresis revealed that 18S gene fragment could be successfully amplified by PCR method,suggesting its potential application to PCR study.MSP results showed different DNA methylation levels of Leptin and TNF-α genes promoter regions from various samples.Conclusions The improved kit method can effectively extract DNA from dried blood spots samples,and these DNA can be used in methylation research.The study can provide a new research direction and technical method to reveal the pathogenesis of disease from the perspective of DNA methylation.

8.Study on Kinetics of Absorbing Timosaponin by AB-8 Resin

Objective To investigate the kinetics of absorbing timosaponin by AB-8 resin.Methods Static absorption experiment,dynamic absorption experiment and series dynamic absorption experiment were carried out,and then the content of timosaponin was detected by spectrophotometry.Finally,the static absorption curve and the dynamic absorption curve were figured out.Results The result of static absorption experiment showed that the parameters of Lagergren equation are Kad=0.0186,r=0.9986,and the parameters of Dumwals-Wagner are K=0.0081,r=0.9958.In dynamic absorption experiment,the leaking of timosaponin is found and the penetrating point is at 600mL(0.1g/mL).Conclusion The mechanism of AB-8 resin absorbing timosaponin is characterized as liquid membrane diffusion at the early phase and intragranular diffusion at the later phase.Leaking phenomena in dynamic absorption experiment can be solved by three tubes in series.Therefore,this method can be used for the research of Chinese herbal medicine.
9.MRI manifestations of pyometra and comparison with pathologic findings

Objective To discuss the MRI features of pyometra and its correlation with pathologic.Methods MRI and DWI data of 12 cases pyometra proved by surgery and pathology were analyzed retrospectively, and a comparision with pathologic findings was made.Mean ADC values were calculated.Results According to the homogeneity of MRI signal: Uniform signal type in 2 cases,T1WI showed slightly low signal,T2WI showed slightly high signal,DWI showed high signal,and the distribution of pus compositions is relatively uniform;Signal mixed type in 5 cases, T2 showed at the bottom of the pus or peripheral mixed slightly short signal, DWI showed mixed high signal, there was some sediment in the bottom of the pus;5 cases of liquid layer type, the upper displayed signal of water,and the lower signal was lower than the upper signal on T2WI, of which 3 cases showed uniform signal of the lower, DWI showed a uniform high signal, the distribution of pus compositions is uniform, the other 2 cases showed mixed signal of the lower, which was pyometra with bleeding,DWI showed high signal and low signal mixed together.The average ADC value of the 12 patients were 0.532×10-3 mm2/s.12 cases of uterine volume were increased, including 10 cases of uterine wall thinning, the other 2 cases with inflammatory invasion, the uterine wall thickening.8 cases with cervical cancer.5 cases with pelvic effusion.Conclusion MRI findings of pyometra are characteristic, its MRI manifestations and pathological components are highly correlated,and the ADC value is of great value in the diagnosis of pyometra.
10.Effect of 3,4-oxo-isopropylidene-shikimic acidon free radical metabolism in the brain issue of rats subjected to middle cerebral artery thrombosis

AIM To study effect of 3,4 oxo isopropylidene shikimic acid(ISA) on free radical metabolism in the brain issue of rats subjected to middle cerebral artery thrombosis.METHODS Adopt a middle cerebral artery thrombosis model to determine the activity of T SOD, Cu Zn SOD, MDA and GSH px of brain issue of rats by the way of colorimetry.RESULTS ISA 100 and 200 mg?kg -1 ig for 2 times in 24 hours raised the activity of T SOD;ISA 50, 100 and 200 mg?kg -1 ig,increased the activity of Cu Zn SOD and GSH px in rats brain issue ;ISA 50, 100 and 200 mg?kg -1 ig for 2 times in 24 hours,decreased the content of MDA in rats brain issue. CONCLUSIONS ISA protect brain issue from the ischemic damage by modifying the free radical metabolism.
