1.Determination of Baicalein in Tangtai Capsule by HPLC

Objective A HPLC method to determine the content of Baicalein in Tangtai capsule was established. Method The separation was performed by Zorbax C18 column with acetonitrile and 0.5% phosphorice acid. The elution program was 1~15 min, 15%~60% acetonitrile. The flow rate was 1 mL/min and column temperature was 25 ℃. Content was detected at a wavelength of 280 nm. Result The linear range was 0.936~18.72 ?g, regression:Y =4849.53X-0.9593 (r=0.9999). The average recovery rate was 98.29% and RSD was 0.97% (n =5). Conclusion The method is simple, accurate and reproductive, and can be used for determining Baicalein in Tangtai capsule.
2.Effects of siRNA Silenced ERK1/2 Gene Collaborated withJiakangning Capsules on FRTL-5 Proliferation

ObjectiveTo observe and explore the effects ofJiakangning Capsules on the expression of ERK1/2 gene and proliferation of FRTL-5 by studying the effects ofJiakangning Capsules collaborated with siRNA on interfering ERK1/2 gene in FRTL-5.Methods FRTL-5 cells in good conditions were divided into control group, negative control group,Jiakangning Capsules collaborated with siRNA group, siRNA interference group, and Jiakangning Capsules group. RT-PCR was performed to detect the expression of ERK1/2 gene in mRNA levels in FRTL-5; Western blotting was performed to detect the expressions of ERK1/2 and p-ERK1/2; CCK8 method was used to detect cell proliferation.Results RT-PCR results showed that the ERK1/2 mRNA expression of the control group and negative control group were of no significant difference (P>0.05); compared with the control group, siRNA interference group andJiakangning Capsules group could inhibit ERK1/2 gene mRNA expression in FRTL-5 (P<0.01). Compared with the control group and negative control group, the ERK1/2 mRNA expression of Jiakangning Capsules collaborated with siRNA group decreased obviously (P<0.01). Compared with the control group and negative control group, the expression of p-ERK1/2, ERK1/2 ofJiakangning Capsules group, Jiakangning Capsules collaborated with siRNA group, and siRNA interference group decreased obviously (P<0.01). At the time of 24 h and 48 h after transfection, according to the results of CCK8, compared with the control group and negative control group, the cell proliferation ofJiakangning Capsules group, Jiakangning Capsules collaborated with siRNA group, and siRNA interference group was inhibited (P<0.01).Conclusion Jiakangning Capsules have blocking effect on the phosphorylation of ERK1/2. After ERK1/2 gene was silenced, the thyroid cell proliferation was inhibited. Jiakangning Capsules can collaborate with ERK1/2-siRNA to inhibit FRTL-5 cell proliferation.
3.Determination of sinomenine and tanshinnone Ⅱ_A of Shuanghuoqufengzhentong Wine by HPLC

AIM: A determination was created of sinomenine and tanshinnone Ⅱ A in Shuanghuoqufengzhentong Wine (Caulis Sinomenii, Radix Salviae Miltiorrhizae, etc.) at the same time by HPLC. METHODS: The method contained a Waters ? PORASIL TM P/N27477 column with methanol water ethylene diamine (90∶10∶0.125) as mobile phase, UV detection wavelength was set up at 262nm. RESULTS: The average recovery of sinomenine was 97.9%, the RSD was 2.83%. The average recovery of Tanshinnone Ⅱ A was 99.0%, The RSD was 1.96%. CONCLUSION: The method is rapid, accurate, sensitive. This study provides a testing method for the quality control of similar products.
4.The early postoperative fluid management experience in infant living-donor liver transplantation

Objective To discuss the effective early postoperative fluid management in infant living-donor liver transplantation. Methods From January 2008 to March 2009, 17 cases of infant living-donor liver transplantation were carried out. According to infant postoperative physiological and hemodynamic characteristics, rehydration was properly controlled on the surgery day, the negative fluid balance was achieved as soon as possible in three days. The infant condition changes, vital signs and urine output were closely monitored. At the same time, the nature and volume of fluid infusion was timely adjusted to maintain the infant in a stable environment in accordance with the laboratory tests. Results The early postopera-donortive hemodynamic stability was effectively maintained in 17 cases of infants, no case appeared with the capacity-related complications. Conclusions It is the key to reduce postoperative complications and mortality with effective circulating blood volume and hemodynamic stability and the negative fluid balance state in the postop-erative 3 days.
5.The Establishment of Gene Gun-Mediated Human GM-CSF Gene Transfection System and It′s Stable Expression in Tumor Cells

Objective: To provide an effective hGM CSF gene transferring vector mediated by gene gun and a basis for study of hGM CSF gene modified tumor cell vaccines. Method: The gastric tumor cell line (SGC) was transfected with eukarytic expression plasmid deoxyribonucleic acid containing the human granulocyte macrophage colony stimulating (hGM CSF) gene using the gene gun. The SGC cell clones (SGC GM CSF 1~5)secreting high hGM CSF level were obtained after G418 resistance selection. The hGM CSF gene had been integrated into chromatosome of SGC by the assay of RT PCR.Results: There was hGM CSF production whose lane was about 30 kD in the culture medium of SGC GM CSF by the assay of SDS PAGE and Western blot. SGC GM CSF had the ability of the high level of GM CSF for a long time(mean 247ng/(10 6 cell?24 h).Conclusion: The hGM CSF gene transferring vector mediated by gene gun was effective and safe. These results provide a basis for study of GM CSF gene therapy for cancer.
6.Effect of Isokinetic Training on Stiff Knees after Fracture Immobilization of Femur

Objective To investigate the effect of isokinetic training on stiff knees after fracture arrest. Methods 38 fracture patients were divided into observation group (n=19) and control group (n=19) by the medical number. Both 2 groups received the Chinese herbs fumigation and joint mobilization. Besides, the observation group received the isokinetic training, while the control group received the instrument training. The range of motion (ROM), maximum peak torque (PT), peak torque/body weight (PT/BW), average power (AP), and hamstrings/quadriceps (H/Q) were tested before and 16 weeks after training. Results The flexion, straightness and flexion radian became significantly better after training. The flexion and the increase of the flexion radian were significantly better in observation group than in control group after training. All indexes of the isokinetic test significantly improved after training (P<0.001) except H/Q (P>0.05). ROM、PT、PT/BW and AP of the knees joint were significantly better in the observation group than in the control group (P<0.001) after training, while there was no significant difference in H/Q between 2 groups (P>0.05) after training. Conclusion Isokinetic training can promote recovery of stiff knees of femur fracture.
7.Effect of Neuromuscular Activation on Low Back Pain Post Operation for Lumbar Vertebra Fracture

Objective To investigate the effect of neuromuscular activation on patients with low back pain after lumbar vertebra fracture operation. Methods 60 low back pain patients who received lumbar vertebra fracture operation were divided into treatment group (n=30)and control group (n=30). Both groups received 20 times of frequency treatments for 4 weeks. While the treatment group received the lumbar stability treatment with neuromuscular activation, and the control group received back muscle strengthen treatment. They were assessed with visual analogue scale (VAS) for pain and Short Form of Health Survey (SF-36) before, 4 weeks and 6 months after treatment. Results The scores of VAS and SF-36 was not significantly different between 2 groups (P>0.05) before treatment, and was significantly different (P<0.05) 4 weeks after treatment and later. Both the scores of VAS and SF-36 improved in both groups after treatment (P<0.05). Conclusion Neuromuscular activation can relieve low back pain after lumbar vertebra fracture operation and improve the quality of life.
8.Effect of Isokinetic, Balance, Weight-support Training on Senile Patients after Total Hip Arthroplasty

ObjectiveTo evaluate the effect of the comprehensive rehabilitation training, including multi-joint isokinetic muscle strength, balance, and weight-support training, on the senile patients after total hip arthroplasty. Methods50 patients after total hip arthroplasty were divided into group A (n=25) and group B (n=25). The group A received the comprehensive rehabilitation training combined with the 3 systems, while the group B received the routine rehabilitation training. They were assessed with the Harris scale and the life satisfaction before and 4, 8 and 12 weeks after rehabilitation. ResultsCompared with that before rehabilitation, the Harris score of all the patients improved 8 weeks after rehabilitation (P<0.05), while it improved more in the group A than in the group B (P<0.05). The incidence of hip joint recovery was more in the group A than in the group B as well (P<0.05). The life satisfaction improved in both groups 4 weeks after rehabilitation (P<0.01), and it was more in the group A than in the group B 12 weeks after training (P<0.05). ConclusionThe rehabilitation combined with isokinetic, balance, weight-support training can promote the functional recovery after total hip arthroplasty
9.Effect of age on body composition in healthy Beijing women

Objective To observe the effect of ageand menstrual statuson body composition in healthy Beijing women.Methods We measured body composition with dual-energy X-ray(GE Lunar Prodigy)in 316 healthy Beijing females aged 20 to 74 years(5-7 cases per age).Parameters provided by the software were as following:total body bone mineral content,lean mass,fat mass and fat percentage(% fat).Local regions measured included arm,leg,trunk,android region and gynoid region.Body mass index (BMI),fat mass index(FMI),free fat mass index(FFMI)and A/G were calculated.Volunteers were assigned to 6 groups according age by every ten years a group.Results BMC peaked during the 4th decade,LM peaked during the 5th decade,with a decline of 18.1%and 5.2%respectively at age 74 years.Total body fat mass and % fat showed a general increase with aging throughout the studied age range.Total body fat mass increased from 16±5 kg at age 20-29 years to 24±6 kg at age 70-74 years,while % fat increased from 31.3%to 39.5%.All local region % fat increased with aging at different extents.Android region % fat showed the largest raise extent(32.2%).BMI increased gradually from 21.1 kg/m2 at age 20-29 years to 26.1 kg/m2 at age 70-74 years.FMI changed more obviously than FFMI.A/G increased from 0.85 at age 20-29years to 1.02 at age 70-74 years.Different menstrual status in women of 40-59 vears had obvious eflfect on A/G and BMC(P<0.05),while it had no significant effect on BMI.body weight and waist circumference(P>0.05). Conclusions Aging and menstrual status have evident effect on body composition distribution in healthy Beijing women.
10.Inhibition of notoginsenoside R1 on SOCE in pulmonary arterial smooth muscle cells of pulmonary hypertension rats

Aim To evaluate the effects of notoginsen-oside R1 on store-operated calcium entry ( SOCE ) in pulmonary arterial smooth muscle cells ( PASMCs ) of chronic hypoxia ( CH)-and monocrotaline ( MCT)-in-duced pulmonary hypertension ( PH) rats. Methods Mn2+ quenching of Fura-2 and measurement of intra-cellular free calcium concentration ( [ Ca2+] i ) using fluo-3 were examined in PASMCs of CH-exposed and MCT-treated rats. Results ①CH-exposed and MCT-treated rats exhibited profound PH when examined 3 weeks after hypoxia exposure or MCT injection, respec-tively. ②In the presence of 3 μmol·L-1 nifedipine, 10 μmol · L-1 notoginsenoside R1 significantly re-duced cyclopiazonic acid ( CPA )-induced the percent reduction in Fura-2 fluorescence measured 500 sec af-ter application of Mn2+, the maximal rate of Mn2+quenching, the amplitude of the Ca2+ influx transient and the resting [ Ca2+] i in PASMCs of CH-exposed and MCT-treated rats. Conclusion Notoginsenoside R1 inhibits SOCE and reduces resting [ Ca2+] i in PASMCs of CH-and MCT-induced PH rats.
