1.Progress in basic research of colorectal liver metastases

Liver metastases is the independent risk factor of prognosis of colorectal cancer.In this article we summarized the research about the mechanism of colorectal liver metastases over the latest five years.Studies showed that the specific cell adhesive interaction in metastatic target organs plays an important role in metastases. The metastatic procedure call be roughly assigned to:escape,adhesion,invasion,survival and proliferation.The interactions between tumor cells and nonparenchymal cens in the liver lead to tumor growth.Tumor micro-environ ment is characterized by the lack of oxygen,inflammation,immune suppression,meanwhile,the role played by cell factors in the tumor micro-environment can not be ignored.
2.Expression of CD70 in T lymphocytes from patients with systemic lupus erythematosus

Objective To detect the expression of CD70 in peripheral T lymphocytes of patients with systemic lupus erythematosus (SLE) and the effect of azacitidine, an inhibitor of DNA methylation, on it. Methods Blood samples were obtained from 10 patients with active SLE (SLEDAI score ≥5), 10 patients with nonactive SLE (SLEDAI score < 5) and 10 normal human controls. Peripheral T lymphocytes were isolated and cultured for 72 hours. A part of the T lymphocytes from normal controls, which were cultured in the presence of azacitidine at 1 mol/L, served as the methylation-inhibited group. Semiquantitative reverse transcription PCR and flow cytometry were applied to detect the mRNA expression of CD70 and frequency of CD70+CD4+ cells in the cultured lymphocytes, respectively. Results The frequency of CD70+CD4+ lymphocytes was 14.55%±5.49% in normal control group, 85.25%±14.08% in active SLE group, 77.65% ±18.77% in nonactive SLE group, and 81.54%±8.71% in methylation-inhibited group. Compared with the normal control group, a significant increase was observed in both the frequency of CD70+CD4+ lymphocytes (all P < 0.01) and the expression of CD70 expression (all P < 0.05) in other three groups. There was a positive correlation between the frequency of peripheral CD70+CD4+ lymphocytes and disease activity of SLE in patients (r = 0.72, P < 0.05). Conclusions The elevated expression of CD70 appears to play a significant role in the immunologic disarrangement in SLE, and may act as a indicator of disease activity of this disease.

With the increase of the experience, the incid ence of post- operative complications is subject to further decrease.
5.Preliminary Study of Partial cDNA Cloning for Human Clotting Factor Ⅷ

A cDNA library prepared from human liver was screened for Factor Ⅶ,a clotting factor in the middle phase of blood coagulation,with P114.12 probe.3 recombinant phages were distinguished,and one of the 3recom-binants was further analyzed and extended by 450 bp as compared with the p114.12 probe.The clone 1 (FⅧhc1) was sequenced and homology between 210 bp of FⅧhc1 and 5559-5769 in FⅧ cDNA was found.It was shown that the FⅧhc1 extended forwardly 800~900bp from 17 exon of FⅧ cDNA.
6.ACUTE INTOXICATION OF PYRITHIONE IN NON-ANESTHETIZED DOGS

Five non-anesthetized dogs were intravenously infused with 4% pyrithione at a constant rate of 2 ml per minute for 5 min.During the course of treatment,experimental animals had salivation, nausea, vomiting, urination and defecation. In according with the accumulation of infused drug,the symptoms got worse and worse progressively. Finally, convulsion and respiratory failure appeared and death occured due to heart arrest. One hour after infusion and just before death, the concentration of pyrithione in the blood was 341? 180?g/ml and 657?158?g/ml respectively.The average accumulative lethal dose of pyrithione by intravenous infusion was 0.77?0.07 g/kg for dogs.
7.Clinical nursing significance of electrocardiographic monitor in primary interventional therapy of acute myocardial infarction

Objective To study the clinical nursing significance of ECG monitor in primary PCI for AMI patients and the relation of changes in ECG and the reperfusion of infarction related artery.Methods Regressional analysis was made in ECG changes including different types of arrthymia,degree of ST segment regression,and their relation between the reperfusion of infarction related artery before and after interventional therapy in AMI patients.Results In 468 AMI patients,142 patients had ventricular arrhythmia before therapy.After interrentional therapy,285 patients were co-morbid with accelerated ventricular tachycardia,203 patients with transient ventricular tachycardia or R-on-T phenomenon,62 patients with ventricular fibrillation and received asynchronous electriacal cardioversion.Twenty four patients had new onsent of Ⅱ-Ⅲ degree atrio-ventricular block and 45 patients had bradycardia or sinus arrest.The atrio-ventricular blook in 10 patients before PCI disappeared after the operation.There are 301 patients with apparent ST segment regression(ST segment regressed ≥50%,57 patients with partial ST segment regression,and 29 patients without ST segment changes.Conclusion Arrhythmias always accompanied with IRA reperfusion.Careful monitoring of ECG is essential for patients' safety after AMI.Obvious ST segment is observed in sufficient IRA reperfusion.However,poor regression of ST segment as well as haemodynamics disorder and cardiogenic shock is observed in patients with poor reperfusion or subacute occlusion of IRA.
8.Optimization of Extraction Process of Volatile Oil from Wenyang Purgation Granules by Orthogonal Design

Objective To optimize the extraction conditions of volatile oil from Angelica Sinensis and Cassia Bark in Wenyang purgation granules.Methods The volatile oil was extracted from Angelica Sinensis and Cassia Bark by the method of steam distillation light oil device with diethyl ether extraction. The yield of the volatile oil was chosen as the evaluation index. The time of dip in water, the ratio of water to herbal medicine material and the time of distillation were used as the main factors. The optimum extraction conditions were investigated by the L9(34) orthogonal design. Results The optimal conditions for extraction process of volatile oil from Angelica Sinensis and Cassia Bark in Wenyang purgation granules were as follows:the time of dip in water was 3 h;the ratio of water to herbal medicine material was 10∶1;the time of distillation was 6 h. Conclusion The optimized conditions of extraction process are stable and feasible.
9.Relationship among HLA-A alleles,supertype and the susceptibility of cervical cancer

Objective To study the relationship among HLA-A alleles,supertype,HPV infection and cervical cancer in Tu Nationality of Hubei province.Methods As a case-control surevy. The comparisons included the comparison between HPV positive cases and HPV positive women in control group,and the comparison between HPV positive cases and HPV negative women in control group. Number of cases was 100(HPV positive in 86),and control was 187 (HPV positive in 95 and HPV negative in 92). The most polymorphism of 2 and 3 exons of the HLA-A alleles were analyzed by the high-resolution typing method-sequence-based typing(SBT).Results Comparison between HPV positive cases and HPV positive control women. Supertype HLA-A3(Pcorrected=0.005,OR=2.36,95% CI=1.45~3.85) was risk factors. Comparison between HPV positive cases and HPV negative control women,HLA-A*0206 alleles (Pcorrected=0.025,OR=0.20,95% CI=0.07~0.58)supertype HLA-A2 (Pcorrected=0.005,OR=0.57,95% CI=0.37~0.88)was protective factor. Supertype HLA-A3 (Pcorrected=0.005,OR=2.36,95% CI=1.45~3.85) was also related to the susceptibility of cervical carcinoma.Conclusion Supertype HLA-A3 is a risk factor of cervical cancer.
10.Isolation of specific humanized anti-D-dimer scFv fragments from scFv phage libraries

Objective To isolate specific humanized anti-D-dimer scFv(single chain Fv) antibody from scFv phage libraries. Methods Isolate anti-D-dimer positive clones from Tomlinson I + J phage libraries by three rounds of panuing, then sequence monoclonal genes by bideoxy-mediated chain termination and express soluble scFv antibody; Pick out anti-D-dimer antibodies with high specificity and affinity by ELISA.Results After three rounds of selection from human scFv phage libraries Tomlinson I and J, 38 monclonal specific anti-D-dimer scFv fragments were selected. By polyclonal and monoclonal phage ELISA and gene sequencing, 20 different full-length monoclonal scFv phages were identified, the result of soluble scFv ELISA showed that 20 full-length monoclonal scFv were expressed smoothly. According to the result of soluble scFv ELISA, in 5 scFv antibodies with high value of A450 selected, 3 scFv antibody fragments showed high specific and affinity. Conclusion Antibody phage display was an effective, rapid method to isolate anti-D-dimer antibodies with high specificity and affinity.
