1.Oriented differentiation of adult stem cells into vascular endothelial cells
Chinese Journal of Tissue Engineering Research 2009;13(27):5345-5348
Adult stem cells identified in tissue organs possess obviously plasticity. They can differentiate into the cells in other systems and germ layers, which are known to support the continuous repair and regeneration of tissues. In the vascular tissue engineering, the research was more and more widespread and deep on adult stem cells as seed cells~ Research indicated that there are many kinds of adult stem cells which can differentiate into vascular endothelial cells, such as bone marrow mesenchymal stem cells, hematopoietic stem cells, human adipose derived stem cells, and amnion derived stem calls. Thus, this study reviewed the current situation of adult stem cells as seed cells in vascular tissue engineering.

Objective To observe the alteration of Bcl-2 and Fas-L at various intervals after traumatic brain injury and study the relationship between the alteration and the post-injury interval. Method The rat brain contusion was incurred by falling impact injury, paraffin section was cut after the test group rats were killed after various survival interval and stained with antibody of Bcl-2 and Fas-L, the hemotoxylin and eosin (HE) staining was carried out meanwhile. The staining results were measured quantitatively with computer imaging analysis system. Results Positive staining nerve celis were observed around the contusion area. At 30min after injury, a few Bcl-2 could be found around the wound, the intensity and the quantity of Bcl-2 positive celis increased significantly as post-injury interval extended. At 4h, the intensity came to a peak. Then the staining decreased. Although some Fas-L positive staining celis could be found around the wound at 30min after injury, the staining increased insignificantly from Ih to 4h after the injury. After 4h, the Fas-L positive staining cell increased significantly both in intensity and in quantity as post-injury interval extended. Conclusion There is a rule that the expression of Bcl-2 and Fas-L alters along with post-injury interval extension, which will be of value in time estimation of brain injury.

Objective To study the neuro-protective effects of magnetotherapy after traumatic brain injury of rats. Methods Traumatic brain injury models were created in 20 rats and divided randomly into magnetotherapy and non-magnetotherapy groups. The rats in magnetotherapy group received magnetotherapy on the cranial surface of traumatized region twice daily, each for 20 min, over 10 d. The intensity of magnetic field was O. 15 T at the surface, and 0.1 T during spinning at 3000 rpm. The contraction strength of gastrocnemius of each rat's left hind limb was measured on the 11 th d after trauma. The rats were then sacrificed and brain tissue samples were taken. The infiltra-tions of CD4 + and CD8 + cells and apnptosis in the area around the injury lesion were observed with immunohisto-chemical methods. The relationship among the indexes was analyzed. Results At the 11th d after brain injury, the quantities of CD4 + and CD8 + cells and apoptotic cells in magnetotherapy group were all obviously less than those in non-magnetotherapy group. Average contraction strength of gastrocnemius in magnetotherapy group was significantly stronger than that in non-magnetotherapy group. Conclusions Magnetotherapy can relieve the secondary effects of brain injury and induce neuro-protection effects. This may involve decreasing the local infiltrations of CD4 + and CD8 + and apoptotic cells in the area around the injury lesion.

Objective To evaluate the efficacy of multiple injection of morphine combined with phloroglucinol in the treatment of renal colic. Methods One hundred and twenty patients with severe renal colic were equally divided into three groups by random digits table, with 40 cases each group. The group A was administrated with physiological saline 100 ml and phloroglucinol 80 mg intervenous drop infusion combined with morphine 9 mg intravenous injection fractionated into three times, and the group B was administrated with physiological saline 100 ml and phloroglucinol 80 mg intervenous drop infusion, and the group C was administrated with physiological saline 100 ml intervenous drop infusion and morphine 9 mg intravenous injection fractionated into three times. At 10,20 and 40 min after administration, the antalgic efficacy of three groups were analyzed as well as the side effect. Results At 20 min and 40 min both of the total efficacy rates in group A[82.5%(33/40),95.0%(38/40)] were higher than those in group B [62.5%(25/40),80.0%(32/40)] and group C [60.0%(24/40),77.5%(31/40)] (P< 0.05),and the colic-exclusion rate in group A was also higher than that in group B and group C (P < 0.05). The difference of the efficacy rates in group A in these two time-points had no statistical significance (P >0.05). No increased pain occurred in group A except 1 case of vomiting. Conclusion Multiple injection of morphine combined with phloroglucinol is quick, effective and safe in the early treatment of renal colic.

Objective To investigate the effect of ionizing radiation on the expression of transcription factor STAT3 target genes in the pulmonary adenocarcinoma cell line A549.Methods The expressions of VEGF,Bcl-2 and Survivin in A549 cells with or without STAT3 inhibition were detected by RT-PCR and/or Western blot 24 h after 2 or 4 Gy of γ-ray irradiation. Results After γ-ray irradiation with 2 or 4 Gy,the expression of VEGF and Survivin increased significantly. However, the expression of Bcl-2 was not affected by γ-ray irradiation.Furthermore, the increased expression of VEGF and Survivin induced by radiation was found to be inhibited after radiation-induced activation of STAT3 was blocked by AG490 inhibiter. Conclusions γ-ray irradiation could up-regulate the expression of Survivin and VEGF via activating STAT3.The increase of Survivin might partly inhibit the radiation-induced apoptosis of A549 cells, and the up-regulation of VEGF might contribute to the tumor angiogenesis.
